The potato rot nematode (Ditylenchus destructor) is a very economically important nematode in agronomic and horticultural plants worldwide. In this study, 43 populations of D. destructor were ...collected from different hosts across China, including 37 populations from Chinese herbal medicine plants. Obtained sequences of ITS-rDNA and D2–D3 of 28S-rDNA genes of D. destructor were compared and analyzed. Nine types of significant length variations in ITS sequences were observed among all populations. The differences in ITS1 length were mainly caused by the presence of repetitive elements with substantial base substitutions. Reconstructions of ITS1 secondary structures showed that the minisatellites formed a stem structure. Ten haplotypes were observed in all populations based on mutations and variations of helix H9. Among them, 3 known haplotypes (A–C) were found in 7 populations isolated from potato, sweet potato, and Codonopsis pilosula, and 7 unique haplotypes were found in other 36 populations collected from C. pilosula and Angelica sinensis compared with 7 haplotypes (A–G) according to Subbotin’ system. These unique haplotypes were different from haplotypes A–G, and we named them as haplotypes H–N. The present results showed that a total of 14 haplotypes (A–N) of ITS-rDNA have been found in D. destructor. Phylogenetic analyses of ITS-rDNA and D2–D3 showed that all populations of D. destructor were clustered into two major clades: one clade only containing haplotype A from sweet potato and the other containing haplotypes B–N from other plants. For further verification, PCR-ITS-RFLP profiles were conducted on 7 new haplotypes. Collectively, our study suggests that D. destructor populations on Chinese medicinal materials are very different from those on other hosts and this work provides a paradigm for relevant researches.
We present precise measurements of the growth rate of cosmic structure for the redshift range 0.1 < z < 0.9, using redshift-space distortions in the galaxy power spectrum of the WiggleZ Dark Energy ...Survey. Our results, which have a precision of around 10 per cent in four independent redshift bins, are well fitted by a flat Λ cold dark matter (ΛCDM) cosmological model with matter density parameter Ωm= 0.27. Our analysis hence indicates that this model provides a self-consistent description of the growth of cosmic structure through large-scale perturbations and the homogeneous cosmic expansion mapped by supernovae and baryon acoustic oscillations. We achieve robust results by systematically comparing our data with several different models of the quasi-linear growth of structure including empirical models, fitting formulae calibrated to N-body simulations, and perturbation theory techniques. , where g is the galaxy overdensity field), and demonstrate that the WiggleZ galaxy-mass cross-correlation is consistent with a deterministic (rather than stochastic) scale-independent bias model for WiggleZ galaxies for scales k < 0.3 h Mpc−1. Measurements of the cosmic growth rate from the WiggleZ Survey and other current and future observations offer a powerful test of the physical nature of dark energy that is complementary to distance-redshift measures such as supernovae and baryon acoustic oscillations.

A(z) = (1205 ± 114, 1380 ± 95, 1534 ± 107) Mpc and H(z) = (82.6 ± 7.8, 87.9 ± 6.1, 97.3 ± 7.0) km s−1 Mpc−1 at these three redshifts. Further combining our results with other baryon acoustic oscillation and distant supernovae data sets, we use a Monte Carlo Markov Chain technique to determine the evolution of the Hubble parameter H(z) as a stepwise function in nine redshift bins of width Δz = 0.1, also marginalizing over the spatial curvature. Our measurements of H(z), which have precision better than 7 per cent in most redshift bins, are consistent with the expansion history predicted by a cosmological constant dark energy model, in which the expansion rate accelerates at redshift z < 0.7.
We present measurements of the baryon acoustic peak at redshifts z= 0.44, 0.6 and 0.73 in the galaxy correlation function of the final data set of the WiggleZ Dark Energy Survey. We combine our ...correlation function with lower redshift measurements from the 6-degree Field Galaxy Survey and Sloan Digital Sky Survey, producing a stacked survey correlation function in which the statistical significance of the detection of the baryon acoustic peak is 4.9σ relative to a zero-baryon model with no peak. We fit cosmological models to this combined baryon acoustic oscillation (BAO) data set comprising six distance-redshift data points, and compare the results with similar cosmological fits to the latest compilation of supernovae (SNe) and cosmic microwave background (CMB) data. The BAO and SNe data sets produce consistent measurements of the equation-of-state w of dark energy, when separately combined with the CMB, providing a powerful check for systematic errors in either of these distance probes. Combining all data sets we determine w=−1.03 ± 0.08 for a flat universe, consistent with a cosmological constant model. Assuming dark energy is a cosmological constant and varying the spatial curvature, we find Ωk=−0.004 ± 0.006.
The aryl hydrocarbon receptor (AHR) is a ligand-dependent transcription factor activated by environmental agonists and dietary tryptophan metabolites for the immune response and cell cycle ...regulation. Emerging evidence suggests that AHR activation after acute stroke may play a role in brain ischemic injury. However, whether AHR activation alters poststroke astrogliosis and neurogenesis remains unknown.
We adopted conditional knockout of AHR from nestin-expressing neural stem/progenitor cells (AHRcKO) and wild-type (WT) mice in the permanent middle cerebral artery occlusion (MCAO) model. WT mice were treated with either vehicle or the AHR antagonist 6,2',4'-trimethoxyflavone (TMF, 5 mg/kg/day) intraperitoneally. The animals were examined at 2 and 7 days after MCAO.
The AHR signaling pathway was significantly upregulated after stroke. Both TMF-treated WT and AHRcKO mice showed significantly decreased infarct volume, improved sensorimotor, and nonspatial working memory functions compared with their respective controls. AHR immunoreactivities were increased predominantly in activated microglia and astrocytes after MCAO compared with the normal WT controls. The TMF-treated WT and AHRcKO mice demonstrated significant amelioration of astrogliosis and microgliosis. Interestingly, these mice also showed augmentation of neural progenitor cell proliferation at the ipsilesional neurogenic subventricular zone (SVZ) and the hippocampal subgranular zone. At the peri-infarct cortex, the ipsilesional SVZ/striatum, and the hippocampus, both the TMF-treated and AHRcKO mice demonstrated downregulated IL-1β, IL-6, IFN-γ, CXCL1, and S100β, and concomitantly upregulated Neurogenin 2 and Neurogenin 1.
Neural cell-specific AHR activation following acute ischemic stroke increased astrogliosis and suppressed neurogenesis in adult mice. AHR inhibition in acute stroke may potentially benefit functional outcomes likely through reducing proinflammatory gliosis and preserving neurogenesis.
Growth differentiation factor 11 (GDF11) is a member of the transforming growth factor beta 1 (TGF-β1) superfamily that reverses age-related cardiac hypertrophy, improves muscle regeneration and ...angiogenesis, and maintains progenitor cells in injured tissue. Recently, targeted myocardial delivery of the
GDF11
gene in aged mice was found to reduce heart failure and enhance the proliferation of cardiac progenitor cells after myocardial ischemia–reperfusion (I–R). No investigations have as yet explored the cardioprotective effect of exogenous recombinant GDF11 in acute I–R injury, despite the convenience of its clinical application. We sought to determine whether exogenous recombinant GDF11 protects against acute myocardial I–R injury and investigate the underlying mechanism in Sprague–Dawley rats. We found that GDF11 reduced arrhythmia severity and successfully attenuated myocardial infarction; GDF11 also increased cardiac function after I–R, enhanced HO-1 expression and decreased oxidative damage. GDF11 activated the canonical TGF-β signaling pathway and inactivated the non-canonical pathways, ERK and JNK signaling pathways. Moreover, administration of GDF11 prior to reperfusion protected the heart from reperfusion damage. Notably, pretreatment with the activin-binding protein, follistatin (FST), inhibited the cardioprotective effects of GDF11 by blocking its activation of Smad2/3 signaling and its inactivation of detrimental TGF-β signaling. Our data suggest that exogenous GDF11 has cardioprotective effects and may have morphologic and functional recovery in the early stage of myocardial I–R injury. GDF11 may be an innovative therapeutic approach for reducing myocardial I–R injury.
Astronomical observations suggest that today's Universe is dominated by a dark energy of unknown physical origin. One of the most notable results obtained from many models is that dark energy should ...cause the expansion of the Universe to accelerate: but the expansion rate as a function of time has proved very difficult to measure directly. We present a new determination of the cosmic expansion history by combining distant supernovae observations with a geometrical analysis of large-scale galaxy clustering within the WiggleZ Dark Energy Survey, using the Alcock-Paczynski test to measure the distortion of standard spheres. Our result constitutes a robust and non-parametric measurement of the Hubble expansion rate as a function of time, which we measure with 10-15 per cent precision in four bins within the redshift range 0.1 < z < 0.9. We demonstrate, in a manner insensitive to the assumed cosmological model, that the cosmic expansion is accelerating. Furthermore, we find that this expansion history is consistent with a cosmological-constant dark energy.
Correlations between the intrinsic shapes of galaxy pairs, and between the intrinsic shapes of galaxies and the large-scale density field, may be induced by tidal fields. These correlations, which ...have been detected at low redshifts (z < 0.35) for bright red galaxies in the Sloan Digital Sky Survey (SDSS), and for which upper limits exist for blue galaxies at z∼ 0.1, provide a window into galaxy formation and evolution, and are also an important contaminant for current and future weak lensing surveys. Measurements of these alignments at intermediate redshifts (z∼ 0.6) that are more relevant for cosmic shear observations are very important for understanding the origin and redshift evolution of these alignments, and for minimizing their impact on weak lensing measurements. We present the first such intermediate-redshift measurement for blue galaxies, using galaxy shape measurements from SDSS and spectroscopic redshifts from the WiggleZ Dark Energy Survey. Our null detection allows us to place upper limits on the contamination of weak lensing measurements by blue galaxy intrinsic alignments that, for the first time, do not require significant model-dependent extrapolation from the z∼ 0.1 SDSS observations. Also, combining the SDSS and WiggleZ constraints gives us a long redshift baseline with which to constrain intrinsic alignment models and contamination of the cosmic shear power spectrum. Assuming that the alignments can be explained by linear alignment with the smoothed local density field, we find that a measurement of σ8 in a blue-galaxy dominated, CFHTLS-like survey would be contaminated by at most +0.02
−0.03 (95 per cent confidence level, SDSS and WiggleZ) or ± 0.03 (WiggleZ alone) due to intrinsic alignments. We also allow additional power-law redshift evolution of the intrinsic alignments, due to (for example) effects like interactions and mergers that are not included in the linear alignment model, and find that our constraints on cosmic shear contamination are not significantly weakened if the power-law index is less than ∼2. The WiggleZ sample (unlike SDSS) has a long enough redshift baseline that the data can rule out the possibility of very strong additional evolution.
The influence of a galaxy's environment on its evolution has been studied and compared extensively in the literature, although differing techniques are often used to define environment. Most methods ...fall into two broad groups: those that use nearest neighbours to probe the underlying density field and those that use fixed apertures. The differences between the two inhibit a clean comparison between analyses and leave open the possibility that, even with the same data, different properties are actually being measured. In this work, we apply 20 published environment definitions to a common mock galaxy catalogue constrained to look like the local Universe. We find that nearest-neighbour-based measures best probe the internal densities of high-mass haloes, while at low masses the interhalo separation dominates and acts to smooth out local density variations. The resulting correlation also shows that nearest-neighbour galaxy environment is largely independent of dark matter halo mass. Conversely, aperture-based methods that probe superhalo scales accurately identify high-density regions corresponding to high-mass haloes. Both methods show how galaxies in dense environments tend to be redder, with the exception of the largest apertures, but these are the strongest at recovering the background dark matter environment. We also warn against using photometric redshifts to define environment in all but the densest regions. When considering environment, there are two regimes: the 'local environment' internal to a halo best measured with nearest neighbour and 'large-scale environment' external to a halo best measured with apertures. This leads to the conclusion that there is no universal environment measure and the most suitable method depends on the scale being probed.
Founder parents have contributed significantly to the improvement of wheat breeding and production. In order to investigate the genetic characteristics of founder parents and widely planted ...cultivars, Mazhamai (M), Biyumai (B) and six sibling lines (BM1–6) derived from the cross M×B were phenotyped for eight yield-related traits over multiple years and locations and genotyped using the the wheat 90K single nucleotide polymorphism (SNP) assay. BM4 has been used as a founder parent, and BM1 has been widely planted, whereas BM2, 3, 5, and 6 have not been used extensively for breeding or planting in China. Phenotypic comparisons revealed that BM4 and BM1 displayed a better overall performance than the other sibling lines. BM1 showed higher thousand-grain weight than BM4, whereas BM4 exhibited lower coefficient of variation for most of the yield-related traits across different years and locations, indicating that BM4 was widely adaptable and more stable in different environments. SNP analysis revealed that BM4 and BM1 inherited similar proportions of the M genome but are dissimilar to BM2, 3, 5, and 6. Both BM1 and BM4 have specific alleles that differ from the other BM lines, and most of these alleles are concentrated in specific chromosomal regions that are found to associate with favorable QTLs, these SNPs and their surrounding regions may carry the genetic determinants important for the superior performance of the two lines. But BM4 has more genetic diversity than BM1 with more specific alleles and pleiotropic regions, indicating that the genome of BM4 may be more complex than the other sibling lines and has more favorable gene resources. Our results provide valuable information that can be used to select elite parents for wheat and self-pollinating crop breeding.
