We compared the effects of salt-stresses (SS, 1: 1 molar ratio of NaCl to Na₂SO₄) and alkali-stresses (AS, 1: 1 molar ratio of NaHCO₃ to Na₂CO₃) on the growth, photosynthesis, solute accumulation, ...and ion balance of barley seedlings, to elucidate the mechanism of AS (high-pH) damage to plants and the physiological adaptive mechanism of plants to AS. The effects of SS on the water content, root system activity, membrane permeability, and the content of photosynthetic pigments were much less than those of AS. However, AS damaged root function, photosynthetic pigments, and the membrane system, led to the severe reductions in water content, root system activity, content of photosynthetic pigments, and net photosynthetic rate, and a sharp increase in electrolyte leakage rate. Moreover, with salinity higher than 60 mM, Na⁺ content increased slowly under SS and sharply under AS. This indicates that high-pH caused by AS might interfere with control of Na⁺ uptake in roots and increase intracellular Na⁺ to a toxic level, which may be the main cause of some damage emerging under higher AS. Under SS, barley accumulated organic acids, Cl⁻, SO₄ ²⁻, and NO₃ ⁻ to balance the massive influx of cations, the contribution of inorganic ions to ion balance was greater than that of organic acids. However, AS might inhibit absorptions of NO₃ ⁻ and Cl⁻, enhance organic acid synthesis, and SO₄ ²⁻ absorption to maintain intracellular ion balance and stable pH.
Bacterial metabolosomes are a family of protein organelles in bacteria. Elucidating how thousands of proteins self-assemble to form functional metabolosomes is essential for understanding their ...significance in cellular metabolism and pathogenesis. Here we investigate the de novo biogenesis of propanediol-utilization (Pdu) metabolosomes and characterize the roles of the key constituents in generation and intracellular positioning of functional metabolosomes. Our results demonstrate that the Pdu metabolosome undertakes both "Shell first" and "Cargo first" assembly pathways, unlike the β-carboxysome structural analog which only involves the "Cargo first" strategy. Shell and cargo assemblies occur independently at the cell poles. The internal cargo core is formed through the ordered assembly of multiple enzyme complexes, and exhibits liquid-like properties within the metabolosome architecture. Our findings provide mechanistic insight into the molecular principles driving bacterial metabolosome assembly and expand our understanding of liquid-like organelle biogenesis.
ABSTRACT Mini-Neptunes and volatile-poor super-Earths coexist on adjacent orbits in proximity to host stars such as Kepler-36 and Kepler-11. Several post-formation processes have been proposed for ...explaining the origin of the compositional diversity between neighboring planets: mass loss via stellar XUV irradiation, degassing of accreted material, and in situ accumulation of the disk gas. Close-in planets are also likely to experience giant impacts during the advanced stage of planet formation. This study examines the possibility of transforming volatile-rich super-Earths/mini-Neptunes into volatile-depleted super-Earths through giant impacts. We present the results of three-dimensional hydrodynamic simulations of giant impacts in the accretionary and disruptive regimes. Target planets are modeled with a three-layered structure composed of an iron core, silicate mantle, and hydrogen/helium envelope. In the disruptive case, the giant impact can remove most of the H/He atmosphere immediately and homogenize the refractory material in the planetary interior. In the accretionary case, the planet is able to retain more than half of the original gaseous envelope, while a compositional gradient suppresses efficient heat transfer as the planetary interior undergoes double-diffusive convection. After the giant impact, a hot and inflated planet cools and contracts slowly. The extended atmosphere enhances the mass loss via both a Parker wind induced by thermal pressure and hydrodynamic escape driven by the stellar XUV irradiation. As a result, the entire gaseous envelope is expected to be lost due to the combination of those processes in both cases. Based on our results, we propose that Kepler-36b may have been significantly devolatilized by giant impacts, while a substantial fraction of Kepler-36c's atmosphere may remain intact. Furthermore, the stochastic nature of giant impacts may account for the observed large dispersion in the mass-radius relationship of close-in super-Earths and mini-Neptunes (at least to some extent).

Exosomes are membranous nanovesicles of endocytic origin that carry and transfer regulatory bioactive molecules and mediate intercellular communication between cells and tissues. Although ...seminal exosomes have been identified in human seminal plasma, their exact composition and possible physiologic function remain unknown. The objective of this study was to perform a comprehensive proteomics analysis of exosomes derived from human seminal plasma. Seminal exosomes were isolated and purified from 12 healthy donors using a 30% sucrose cushion‐based exosome‐isolation protocol, followed by characterization by western blot, transmission electron microscopy, and nanoparticle tracking analysis before performing extensive liquid chromatography tandem mass spectrometry proteomics analysis. The identified proteins were analyzed by bioinformatics analysis, and seminal exosomes‐associated proteins were selectively validated by western blot. A total of 1474 proteins were identified in all seminal exosomes samples, with Gene Ontology analysis demonstrating that these identified seminal exosomes‐associated proteins were mostly linked to ‘exosomes,’ ‘cytoplasm,’ and ‘cytosol.’ Bioinformatics analysis indicated that these proteins were mainly involved in biologic processes, including metabolism, energy pathways, protein metabolism, cell growth and maintenance, and transport. Of these identified proteins, PHGDH, LGALS3BP, SEMG1, ACTB, GAPDH, and the exosomal‐marker protein ALIX were validated by western blot. This study provided a more comprehensive description of the seminal exosomes proteome and could also be a resource for further screening of biomarkers and comparative proteomics studies, including those associated with male infertility and prostate cancer.
Herein, we describe a novel methodology for the regio- and stereoselective convergent synthesis of 2-amino-2-deoxy-dithioglycosides via one-pot relay glycosylation of 3-O-acetyl-2-nitroglucal donors. ...This unique organo-catalysis relay glycosylation features excellent site- and stereoselectivity, good to excellent yields, mild reaction conditions, and broad substrate scope. 2-Amino-2-deoxy-glucosides/mannosides bearing 1,3-dithio-linkages were efficiently obtained from 3-O-acetyl-2-nitroglucal donors in both stepwise and one-pot glycosylation protocols. The dithiolated O-antigen of E. coli serogroup 64 was successfully synthesized using this newly developed method.
Retinoblastoma (Rb) protein is a tumor suppressor that is dysregulated in a majority of human cancers. Rb functions to inhibit cell cycle progression in part by directly disabling the E2F family of ...cell cycle-promoting transcription factors. Because the de novo synthesis of multiple glutamine-derived anabolic precursors is required for cell cycle progression, we hypothesized that Rb also may directly regulate proteins involved in glutamine metabolism. We examined glutamine metabolism in mouse embryonic fibroblasts (MEFs) isolated from mice that have triple knock-outs (TKO) of all three Rb family members (Rb-1, Rbl1 and Rbl2) and found that loss of global Rb function caused a marked increase in (13)C-glutamine uptake and incorporation into glutamate and tricarboxylic acid cycle (TCA) intermediates in part via upregulated expression of the glutamine transporter ASCT2 and the activity of glutaminase 1 (GLS1). The Rb-controlled transcription factor E2F-3 altered glutamine uptake by direct regulation of ASCT2 mRNA and protein expression, and E2F-3 was observed to associate with the ASCT2 promoter. We next examined the functional consequences of the observed increase in glutamine uptake and utilization and found that glutamine exposure potently increased oxygen consumption, whereas glutamine deprivation selectively decreased ATP concentration in the Rb TKO MEFs but not the wild-type (WT) MEFs. In addition, TKO MEFs exhibited elevated production of glutathione from exogenous glutamine and had increased expression of gamma-glutamylcysteine ligase relative to WT MEFs. Importantly, this metabolic shift towards glutamine utilization was required for the proliferation of Rb TKO MEFs but not for the proliferation of the WT MEFs. Last, addition of the TCA cycle intermediate α-ketoglutarate to the Rb TKO MEFs reversed the inhibitory effects of glutamine deprivation on ATP, GSH levels and viability. Taken together, these studies demonstrate that the Rb/E2F cascade directly regulates a major energetic and anabolic pathway that is required for neoplastic growth.
The tumor suppressor p53 is a master regulator of apoptosis and also plays a key role in cell cycle checking. In our previous studies, we demonstrated that p53 directly regulates Bak in mouse JB6 ...cells (Qin et al. 2008. Cancer Research. 68(11):4150) and that p53-Bak signaling axis plays an important role in mediating EGCG-induced apoptosis. Here, we demonstrate that the same p53-Bak apoptotic signaling axis executes an essential role in regulating lens cell differentiation. First, during mouse lens development, p53 is expressed and differentially phosphorylated at different residues. Associated with p53 expression, Bak is also significantly expressed during mouse lens development. Second, human p53 directly regulates Bak promoter and Bak expression in p53 knockout mice (p53-/-) was significantly downregulated. Third, during in vitro bFGF-induced lens cell differentiation, knockdown of p53 or Bak leads to significant inhibition of lens cell differentiation. Fourth, besides the major distribution of Bak in cytoplasm, it is also localized in the nucleus in normal lens or bFGF-induced differentiating lens cells. Finally, p53 and Bak are co-localized in both cytoplasm and nucleus, and their interaction regulates the stability of p53. Together, these results demonstrate for the first time that the p53-Bak apoptotic signaling axis plays an essential role in regulating lens differentiation.

One-dimension MnCo2O4 nanowire arrays are synthesized on nickel foam by a facile hydrothermal method. The MnCo2O4 nanowires are highly crystalline with an average diameter of 70nm and exhibit excellent properties for electrochemical energy storage. Impressively, the MnCo2O4 nanowire array exhibits noticeable pseudocapacitive performance with a high capacitance of 349.8 F g−1 at 1 A g−1 and 328.9 F g−1 at 20 A g−1 as well as excellent cycling stability. As an anode material for Li-ion batteries, the MnCo2O4 nanowire array delivers an initial specific discharge capacity of 1288.6 mAh g−1 at 100mAg−1, with reversible capacity retention of 92.7% after 50 cycles. The outstanding electrochemical performances are mainly attributed to its nanowire array architecture which provides large reaction surface area, fast ion and electron transfer and good structure stability.
