•LINC00511 was up-regulated in glioma.•SiLINC00511suppressed the biological behavior of glioma cells.•SiLINC00511 inhibited glioma cell progression via miR-15a-5p/AEBP1 axis.
A strong relationship ...between long intergenic non-protein coding RNA 511 (LINC00511) and glioma has been previously reported but the mechanism of LINC00511 in glioma is yet to be determined. This study examined the mechanism of LINC00511 in glioma.
The expression of LINC00511 in glioma was determined by bioinformatics analysis and real-time quantitative PCR (RT-qPCR) analysis. The target relationship between genes was predicted by starBase, TargetScan, and was verified by dual-luciferase. Subsequently, siRNA targeting LINC00511 (siLINC00511) and miR-15a-5p mimic were transfected into glioma cells to examine the effect on biological characteristics using cell counting kit-8, clone formation, flow cytometry, wound-healing, and transwell. MiR-15a-5p inhibitor and AEBP1 were used for in vitro rescue experiments, and tumorigenesis assay and immunohistochemical assays were performed for in vivo experiments. Epithelial-mesenchymal transition (EMT) and p65 phosphorylation were examined by Western blot.
LINC00511 was predicted and verified to be up-regulated in glioma. SiLINC00511 suppressed cell viability, proliferation, migration and invasion, accelerated apoptosis of glioma cells. Mechanically, siLINC00511 promoted E-cadherin expression but suppressed N-cadherin and Snail expressions. MiR-15a-5p bound to LINC00511, and miR-15a-5p inhibitor partially reversed the effect and regulation of siLINC00511 on glioma cells. AEBP1, a target gene of miR-15a-5p, could activate p65 phosphorylation to promote EMT protein expression and partially reverse the inhibitory effect of miR-15a-5p mimic on the malignant phenotype of glioma cells. SiLINC00511 inhibited tumor growth, down-regulated miR-15a-5p expression and up-regulated AEBP1 and Ki67 expressions in vivo.
LINC00511 knockdown inhibits glioma cell progression via miR-15a-5p/AEBP1 axis.
Ribociclib (RIB, LE011, Kisqali®), an orally administered inhibitor of cyclin-dependent kinase-4/6 (CDK-4/6) complex, is clinically effective for the treatment of several malignancies, including ...advanced breast cancer. However, information regarding the effects of RIB on membrane ion currents is limited. In this study, the addition of RIB to pituitary tumor (GH3) cells decreased the peak amplitude of erg-mediated K+ current (IK(erg)), which was accompanied by a slowed deactivation rate of the current. The IC50 value for RIB-perturbed inhibition of deactivating IK(erg) in these cells was 2.7 μM. In continued presence of μM RIB, neither the subsequent addition of 17β-estradiol (30 μM), phorbol 12-myristate 13-acetate (10 μM), or transforming growth factor-β (1 μM) counteracted the inhibition of deactivating IK(erg). Its presence affected the decrease in the degree of voltage-dependent hysteresis for IK(erg) elicitation by long-duration triangular ramp voltage commands. The presence of RIB differentially inhibited the peak or sustained component of delayed rectifier K+ current (IK(DR)) with an effective IC50 of 28.7 or 11.4 μM, respectively, while it concentration-dependently decreased the amplitude of M-type K+ current with IC50 of 13.3 μM. Upon 10-s long membrane depolarization, RIB elicited a decrease in the IK(DR) amplitude, which was concomitant with an accelerated inactivation time course. However, the inability of RIB (10 μM) to modify the magnitude of the hyperpolarization-activated cation current was disclosed. The mean current–voltage relationship of IK(erg) present in HL-1 atrial cardiomyocytes was inhibited in the presence of RIB (10 μM). Collectively, the hyperpolarization-activated cation current was observed. RIB-mediated perturbations in ionic currents presented herein are upstream of its suppressive action on cytosolic CDK-4/6 activities and partly participates in its modulatory effects on the functional activities of pituitary tumor cells (e.g., GH3 cells) or cardiac myocytes (e.g., HL-1 cells).
This paper presents novel stability criteria for neutral systems with time-varying delay. By developing a delayed decomposition approach, information of the delayed plant states can be taken into ...full consideration, and new delay-dependent sufficient stability criteria are obtained in terms of linear matrix inequalities (LMIs). Then, based on the Lyapunov method, delay-dependent stability criteria are devised by taking the relationship between terms in the Leibniz-Newton formula into account. Criteria are derived in terms of LMIs, which can be easily solved by using various convex optimization algorithms. Three illustrative numerical examples are given to show less conservatism of our obtained results and the effectiveness of the proposed method.
The complexity in stock index futures markets, influenced by the intricate interplay of human behavior, is characterized as nonlinearity and dynamism, contributing to significant uncertainty in ...long-term price forecasting. While machine learning models have demonstrated their efficacy in stock price forecasting, they rely solely on historical price data, which, given the inherent volatility and dynamic nature of financial markets, are insufficient to address the complexity and uncertainty in long-term forecasting due to the limited connection between historical and forecasting prices. This paper introduces a pioneering approach that integrates financial theory with advanced deep learning methods to enhance predictive accuracy and risk management in China’s stock index futures market. The SF-Transformer model, combining spot-forward parity and the Transformer model, is proposed to improve forecasting accuracy across short and long-term horizons. Formulated upon the arbitrage-free futures pricing model, the spot-forward parity model offers variables such as stock index price, risk-free rate, and stock index dividend yield for forecasting. Our insight is that the mutual information generated by these variables has the potential to significantly reduce uncertainty in long-term forecasting. A case study on predicting major stock index futures prices in China demonstrates the superiority of the SF-Transformer model over models based on LSTM, MLP, and the stock index futures arbitrage-free pricing model, covering both short and long-term forecasting up to 28 days. Unlike existing machine learning models, the Transformer processes entire time series concurrently, leveraging its attention mechanism to discern intricate dependencies and capture long-range relationships, thereby offering a holistic understanding of time series data. An enhancement of mutual information is observed after introducing spot-forward parity in the forecasting. The variation of mutual information and ablation study results highlights the significant contributions of spot-forward parity, particularly to the long-term forecasting. Overall, these findings highlight the SF-Transformer model’s efficacy in leveraging spot-forward parity for reducing uncertainty and advancing robust and comprehensive approaches in long-term stock index futures price forecasting.
Metal-organic frameworks (MOFs) possess large surface area, high porosity, tunable structures, and flexible tailored properties, enabling their use as adsorbents and sensors for food contamination. ...Specifically, MOFs have been regarded as promising materials in instrumental analysis of food contamination via separation and extraction of target analytes based on their high surface area with high adsorption capacity. Premature ovarian failure (POF) is characterized by hormonal disorders, amenorrhea, and premature loss of fertility potential in women of reproductive age. Several studies have been conducted on the ...effectiveness of traditional Chinese medicine (TCM) in treating POF. TCM relied primarily on apoptosis, immunity, and aging to treat POF based on the studies of domestic and foreign literature. Zuogui pills inhibited mitochondrial-dependent apoptosis in the treatment of POF. Huyang Yangkun formula regulated the downstream of the Bcl-2 family to resist apoptosis through the aquaporin-1 protein. Modified Bazhen decoction regulated apoptosis in POF by regulating X-linked inhibitors of apoptosis protein. Bushen Tianjing recipe was effective in treating POF by promoting angiogenesis and preventing apoptosis. As for immunity, Bushen Jianpi prescription and Er-Xian decoction cured autoimmunity POF models and increased follicular development-related protein expression. Bushen Huoxue Tang improved ovarian function and reduced ovarian inflammation by regulating the Nrf2/Keap1 signaling pathway and T lymphocytes. Taohong Siwu decoction promoted the proliferation and differentiation of granulosa cells of POF mice by regulating the TGF-β1/Smads signaling pathway. In addition, ginsenoside Rg1 and Jiajian Guisheng formula treated POF by regulating cell aging-related mechanisms. Si Wu Tang treated POF by activating the angiogenesis-related proteins. The goal of this review is to serve as a reference for in-depth research into the treatment of POF with TCM and provide inspiration for new diagnostic methods and treatment options.
Background A higher risk of developing dementia is observed in patients with atrial fibrillation (AF). Results are inconsistent regarding the risk of dementia when patients with AF use different ...anticoagulants. We aimed to investigate the risk of dementia in patients with AF receiving non-vitamin K antagonist oral anticoagulants (NOACs) compared with those receiving warfarin. Methods and Results We conducted a nationwide population-based cohort study of incident cases using the Taiwan National Health Insurance Research Database. We initially enlisted all incident cases of AF and then selected those treated with either NOACs or warfarin for at least 90 days between 2012 and 2016. First-ever diagnosis of dementia was the primary outcome. We performed propensity score matching to minimize the difference between each cohort. We used the Fine and Gray competing risk regression model to calculate the hazard ratio (HR) for dementia. We recruited 12 068 patients with AF (6034 patients in each cohort). The mean follow-up time was 3.27 and 3.08 years in the groups using NOACs and warfarin, respectively. Compared with the HR for the group using warfarin, the HR for dementia was 0.82 (95% CI, 0.73-0.92;
=0.0004) in the group using NOACs. Subgroup analysis demonstrated that users of NOAC aged 65 to 74 years, with a high risk of stroke or bleeding were associated with a lower risk of dementia than users of warfarin with similar characteristics. Conclusions Patients with AF using NOACs were associated with a lower risk of dementia than those using warfarin. Further randomized clinical trials are greatly needed to prove these findings.
In this paper, we consider the problem of delay-interval-dependent robust stability and stabilization of a class of linear uncertain neutral-type systems with time-varying delay. By constructing a ...candidate Lyapunov–Krasovskii functional (LKF), that takes into account the delay-range information appropriately, less conservative robust stability criteria are proposed in terms of linear matrix inequalities (LMIs) to compute the maximum allowable upper bounds (MAUB) for the delay-interval within which the uncertain neutral-type system under consideration remains asymptotically stable. The verifiable stabilizability conditions and memoryless state feedback control design are stated. Finally, numerical examples are also designated to demonstrate the effectiveness and reduced conservatism of the developed results.
•This paper has presented improved delay-interval-dependent robust stabilization criterion for uncertain neutral-type time-varying delays systems.•The delay interval is divided into two subintervals with an unequal width, which is different from the existing methods.•Much less conservative delay-interval-dependent robust stability criteria are derived compared with existing methods.
This paper considers the problem of delay-range-dependent stability analysis for a class of delayed recurrent neural networks (DRNNs) with a time-varying delay in a range. Based on the ...Lyapunov-Krasovskii functional and derive the time derivative of this with integral inequality approach (IIA), new delay-dependent stability criteria for the system are established in terms of linear matrix inequalities (LMIs), which can be solved easily by various efficient convex optimization algorithms. Information about the lower bound of the delay is fully used in the Lyapunov functional. Two examples are given to illustrate the effectiveness and the reduced conservatism of the proposed results.
