Las inverosimilitudes y desfases históricos de la Historia Roderici como los indicios filológicos que apuntan a situar su composición a finales del siglo XII, desmienten la teoría de que su autor ...fuera partícipe de los hechos o de que su información procediera de testigos presenciales. El concepto de “archivo cidiano”, por su parte, además de carecer de base empírica y diplomática, tiene poquísimo rendimiento heurístico si se desestiman los numerosos documentos supuestamente perdidos del cartulario cidiano ideado por Menéndez Pidal. A partir de ahí, conviene devolver la obra a un marco de estudio más habitual: el de cualquier crónica cristiana de la Edad Media alta y central, con sus fuentes documentales, historiográficas y memorísticas así como con sus valoraciones, intenciones y parte de invención. El concepto de “memoria linajística cidiana” permite salvar las dificultades planteadas tanto por la teoría de un autor cercano a los hechos como por la del uso que pudo hacer este de un problemático archivo cidiano, manteniendo la posibilidad de que le llegase un eco vivo –auténtico, deformado o amañado– de la vida del Campeador. No obstante, su manejo es difícil por las arduas indagaciones históricas que supone penetrar la carne viva del recuerdo en personajes cuya huella se perdió en gran parte, y no puede más que servir de recurso auxiliar para completar la información procedente de la historiografía y de la documentación. La relación de la Historia Roderici con la historiografía árabe es la que me lleva al enfrentamiento más contundente y arriesgado con la doxa pidaliana. Sin embargo, de aceptarse una datación tardía de la Historia, dicha relación no puede ser sino la de una derivación textual. La misma conformidad de los relatos, estrecha, detallista, fraseológica a veces, desmiente que estos puedan constituir reflejos independientes de una misma realidad.
El segmento 1 a 5 de la Historia Roderici, en el que se exponen la genealogía de Ruy Díaz y sus primeros hechos bajo el reinado de Sancho II de Castilla, comparte sus datos con gran parte de la rica ...tradición historiográfica del siglo XII hispano, en particular con tres obras también dedicadas, en totalidad o en parte, a relatar los hechos del Campeador: el Carmen Campidoctoris, la Chronica naiarensis y el Libro de las generaciones y linajes de los reyes. Esta red de concordancias textuales depara al estudioso una situación muy propicia a sacar enseñanzas crono-genéticas del cotejo de la Historia con obras de datación menos incierta. Estas enseñanzas son las siguientes: La Historia Roderici es posterior a la Chronica naiarensis, que se considera compuesta en la década de los 1180, posiblemente en los años 1188-1189. En cambio, es anterior al Linage de Rodric Diaz, eso es a la versión más antigua del Libro de las generaciones y linajes de los reyes, culminada posiblemente antes del 27 de junio de 1194, o, como más tarde, antes del 15 de agosto de 1209. Nos encontramos pues ante una obra escrita entre 1188 y 1194 (o incluso 1209), eso es ante una crónica tardía y de redacción probablemente muy cercana en el tiempo –y quizá en el espacio– a la del Libro.

Muy al contrario de lo que se afirmó antaño y alguno sigue opinando, la producción historiográfica del reinado de Alfonso III no tuvo los “pobres” y “bárbaros” albores de algún esbozo cronístico ...“seco”, “frío”, “lacónico” y “oscuro” –ni aun “sencillo” ni “sobrio”–. Tampoco se formó progresivamente, completando con retazos informativos alguna crónica de anteriores reinados. Surgió de golpe, por los años 877-881, con una obra amplia, densa, estructurada, razonada, culta e informadísima a la vez que inventiva, y de ambicioso alcance político: la versión “rotense” de la Crónica de Alfonso III. Sobre esta espléndida base, en el fervoroso scriptorium de la iglesia del Salvador, donde clérigos de procedencia astur, goda o mozárabe colaboraban a su manera en la conquista de un territorio otrora cristiano y en la construcción de la infraestructura gubernativa de un reino en pleno desarrollo, nació primero, en 881, un compendio historiográfico que vinculaba a Alfonso III y a los anteriores monarcas asturianos con los emperadores romanos y los reyes godos. En 883, este entró a formar parte de un memorándum o manual de saberes naturales e históricos que integró la monarquía asturiana en las grandes coordenadas físicas, históricas y culturales del ámbito universal e hispánico. Entre tanto, en el mismo año 883, se había forjado una profecía destinada a afianzar y alentar el impulso reconquistador de Alfonso III. Seguidamente, entre 883 y 890, un historiador regio que tenía a mano el conjunto de los textos conservados o producidos en Oviedo procedió a realzar la dignidad formal de la versión primitiva de la Crónica de Alfonso III, a expurgarla de sus contenidos molestos o escandalosos, y a centrarla más decididamente en la exaltación de la dinastía reinante.
RNA-specific nucleotidyl transferases (rNTrs) are a diverse family of template-independent polymerases that add ribonucleotides to the 3'-ends of RNA molecules. All rNTrs share a related active-site ...architecture first described for DNA polymerase beta and a catalytic mechanism conserved among DNA and RNA polymerases. The best known examples are the nuclear poly(A) polymerases involved in the 3'-end processing of eukaryotic messenger RNA precursors and the ubiquitous CCA-adding enzymes that complete the 3'-ends of tRNA molecules. In recent years, a growing number of new enzymes have been added to the list that now includes the "noncanonical" poly(A) polymerases involved in RNA quality control or in the readenylation of dormant messenger RNAs in the cytoplasm. Other members of the group are terminal uridylyl transferases adding single or multiple UMP residues in RNA-editing reactions or upon the maturation of small RNAs and poly(U) polymerases, the substrates of which are still not known. 2'-5'Oligo(A) synthetases differ from the other rNTrs by synthesizing oligonucleotides with 2'-5'-phosphodiester bonds de novo.
Through alternative polyadenylation, human mRNAs acquire longer or shorter 3′ untranslated regions, the latter typically associated with higher transcript stability and increased protein production. ...To understand the dynamics of polyadenylation site usage, we performed transcriptome-wide mapping of both binding sites of 3′ end processing factors CPSF-160, CPSF-100, CPSF-73, CPSF-30, Fip1, CstF-64, CstF-64τ, CF Im25, CF Im59, and CF Im68 and 3′ end processing sites in HEK293 cells. We found that although binding sites of these factors generally cluster around the poly(A) sites most frequently used in cleavage, CstF-64/CstF-64τ and CFIm proteins have much higher positional specificity compared to CPSF components. Knockdown of CF Im68 induced a systematic use of proximal polyadenylation sites, indicating that changes in relative abundance of a single 3′ end processing factor can modulate the length of 3′ untranslated regions across the transcriptome and suggesting a mechanism behind the previously observed increase in tumor cell invasiveness upon CF Im68 knockdown.

Alternative cleavage and polyadenylation generate mRNAs with 3′ untranslated regions of different lengths. Keller, Zavolan, and colleagues mapped the binding sites of ten 3′ end processing proteins by PAR-CLIP and identified the 3′ end processing sites by A-seq. CstF-64 and CF Im68 proteins showed the highest positional specificity. Knockdown of CF Im68 induced a systematic shift toward proximal polyadenylation sites, indicating that changes in the relative abundance of a single 3′ end processing factor can modulate the length of 3′ untranslated regions transcriptome-wide.

Alternative polyadenylation (APA) is a general mechanism of transcript diversification in mammals, which has been recently linked to proliferative states and cancer. Different 3' untranslated region ...(3' UTR) isoforms interact with different RNA-binding proteins (RBPs), which modify the stability, translation, and subcellular localization of the corresponding transcripts. Although the heterogeneity of pre-mRNA 3' end processing has been established with high-throughput approaches, the mechanisms that underlie systematic changes in 3' UTR lengths remain to be characterized. Through a uniform analysis of a large number of 3' end sequencing data sets, we have uncovered 18 signals, six of which are novel, whose positioning with respect to pre-mRNA cleavage sites indicates a role in pre-mRNA 3' end processing in both mouse and human. With 3' end sequencing we have demonstrated that the heterogeneous ribonucleoprotein C (HNRNPC), which binds the poly(U) motif whose frequency also peaks in the vicinity of polyadenylation (poly(A)) sites, has a genome-wide effect on poly(A) site usage. HNRNPC-regulated 3' UTRs are enriched in ELAV-like RBP 1 (ELAVL1) binding sites and include those of the CD47 gene, which participate in the recently discovered mechanism of 3' UTR-dependent protein localization (UDPL). Our study thus establishes an up-to-date, high-confidence catalog of 3' end processing sites and poly(A) signals, and it uncovers an important role of HNRNPC in regulating 3' end processing. It further suggests that U-rich elements mediate interactions with multiple RBPs that regulate different stages in a transcript's life cycle.
Understanding the regulation of gene expression, including transcription start site usage, alternative splicing, and polyadenylation, requires accurate quantification of expression levels down to the ...level of individual transcript isoforms. To comparatively evaluate the accuracy of the many methods that have been proposed for estimating transcript isoform abundance from RNA sequencing data, we have used both synthetic data as well as an independent experimental method for quantifying the abundance of transcript ends at the genome-wide level.
We found that many tools have good accuracy and yield better estimates of gene-level expression compared to commonly used count-based approaches, but they vary widely in memory and runtime requirements. Nucleotide composition and intron/exon structure have comparatively little influence on the accuracy of expression estimates, which correlates most strongly with transcript/gene expression levels. To facilitate the reproduction and further extension of our study, we provide datasets, source code, and an online analysis tool on a companion website, where developers can upload expression estimates obtained with their own tool to compare them to those inferred by the methods assessed here.
As many methods for quantifying isoform abundance with comparable accuracy are available, a user's choice will likely be determined by factors such as the memory and runtime requirements, as well as the availability of methods for downstream analyses. Sequencing-based methods to quantify the abundance of specific transcript regions could complement validation schemes based on synthetic data and quantitative PCR in future or ongoing assessments of RNA-seq analysis methods.
Cette étude se situe à la confluence d’une histoire politique de l’historiographie et d’une histoire sociale des femmes de pouvoir. Elle est portée par le souci de discerner l’importance et, au ...préalable, les lieux d’une intervention politique des femmes dans les royaumes hispaniques médiévaux. Pour l’espace et la période considérés – le royaume de León aux XIIe et XIIIe siècles –, l’entreprise n’a rien de facile et oblige à prendre en compte, au-delà de quelques données positives, des éléments indiciels permettant de formuler des hypothèses défendables. Dans le cadre de ce questionnement et de ces contraintes, on suivra, sous les formes fondamentales du patronage et de l’incitation, les traces d’une contribution féminine à l’émergence d’une grande historiographie royale et, conséquemment, à l’élaboration d’une pensée politique, puisque, au cours de cette période, celle-ci trouva dans l’histoire sa meilleure expression. Accessoirement, ces quelques réflexions pourront aussi être versées au dossier de l’histoire de l’infantat et du monastère-collégiale de Saint-Isidore de León.
