CLiC-it 2015 is held in Trento on December 3-4 2015, hosted and locally organized by Fondazione Bruno Kessler (FBK), one the most important Italian research centers for what concerns CL. The ...organization of the conference is the result of a fruitful conjoint effort of different research groups (Università di Torino, Università di Roma Tor Vergata and FBK) showing the nationwide spreading of CL in Italy. As in the first edition, the main aim of the event is at establishing a reference forum on CL, covering all the aspects needed to describe the multi-faceted and cross-disciplinary reality of the involved research topics and of the Italian community working in this area. Indeed the spirit of CLiC-it is inclusive, in order to build a scenario as much as possible comprehensive of the complexity of language phenomena and approaches to address them, bringing together researchers and scholars with different competences and skills and working on different aspects according to different perspectives. The large number of researchers that have decided to present their work at CLiC-it and the number of directions here investigated are proof of the maturity of our community and a promising indication of its vitality. We received a total of 64 paper submissions, out of which 52 have been accepted to appear in the Conference Proceedings, which are available online and on the OpenEdition platform. Overall, we collected 129 authors from 15 countries.
•Octogenarians and nonagenarians with AML can be treated successfully with VEN-HMA, and a subset of patients (∼25%) have prolonged survival.•Reduced dosing duration may be appropriate in this ...population, adjusted on a patient-by-patient basis to minimize myelosuppression.

Venetoclax (VEN) plus a hypomethylating agent (HMA) regimen is the standard of care for older adults with acute myeloid leukemia (AML); however, it is associated with significant myelosuppression and complications, potentially limiting its use in those who are very old. We performed a multicenter retrospective analysis of VEN-HMA treatment in octogenarians and nonagenarians to further understand the tolerability, feasibility, dosing considerations, and clinical efficacy in this unique group. Patients with AML aged ≥80 years who received VEN-HMA between March 2015 and April 2022 were reviewed. VEN-HMA dosing was determined by treating physician, accounting for CYP3A4 drug interaction dose adjustments. In total, 154 patients were included, with a median age of 82 years (range, 80-92), who received treatment with VEN-HMA (83% with azacitidine and 17% with decitabine). Most patients (53%) had European LeukemiaNet 2017 adverse risk AML, 33% had intermediate, 8% had favorable, and 6% were unknown. With a median follow-up of 7.7 months, 36 patients (23%) remained in remission, with 31 (20%) still on VEN-HMA. The 30-day and 60-day mortality rates were 8.5% and 17%, respectively. The composite complete remission (CRc) rate for patients with newly diagnosed AML without prior myelodysplastic syndrome was 73% (48 of 66). Median overall survival (OS) was 8.1 months, and in patients who achieved a response (CRc), median OS was 13.2 months. Landmark analysis from the time CRc was first achieved showed that patients receiving VEN for ≤14 days had improved OS; median, 24.0 months. Patients who are very old can be treated safely with combination VEN-HMA with expectations of dose reductions and cycle extensions to ensure tolerability over the long term.

American cutaneous leishmaniasis, an endemic disease in the northwestern region of Paraná, Brazil, is transmitted by phlebotomines to man and animals like dogs, armadillos, opossums and wild rodents. This disease has been occurring in places where forests have been felled and on the banks of rivers and lakes with arboreal vegetation, where man comes into contact with infected insects. This disease is a public health problem because of the occurrence of epidemic outbreaks throughout Paraná. Because of the importance of finding out about endemic areas, this study used epidemiological file data on patients attended between 1999 and 2004. These data were correlated with areas of forest felling that were identified by means of satellite remote sensing techniques and products. The results showed that the occurrences of cases in the municipalities of this region coincided with the presumed likely areas for patient infection.

Ewing's sarcoma shows a strong tendency to metastasize to the lungs or the skeleton, or both. A peculiar feature of the secondary involvement of bone with this tumor is that it may also appear in the ...absence of clinically evident lung metastases, both at clinical presentation and during the course of the disease. Although osseous metastases are critically relevant for prognosis, the pathogenesis of this peculiar feature of Ewing's sarcoma is poorly understood, partly due to the lack of appropriate experimental in vivo models. We show that the intravenous injection of TC-71 Ewing's sarcoma cells into athymic 4-5-week-old Crl/nu/nu (CD1) BR mice reproducibly colonizes specific sites of the skeleton in addition to the lungs and lymph nodes. The distribution and the morphologic appearance of these experimental bone metastases mimic the pattern of skeletal involvement observed in humans. This experimental model of bone metastasis of Ewing's sarcoma may be the basis for future studies aimed at understanding the pathophysiology and treatment of Ewing's sarcoma.

The surface expression of given HLA class I alleles protects target cells from lysis mediated by natural killer (NK) clones specific for these (or related) alleles. We could define two groups of NK ...clones specifically recognizing either Cw4 and related C alleles ("group 1") or Cw3 and related C alleles ("group 2"), respectively. Monoclonal antibodies (mAb) to class I molecules should interfere with the interaction between NK receptors and class I molecules, thus resulting in lysis of protected target cells. However, none of the numerous available mAb to class I molecules had this effect. Therefore, we attempted to select new mAb on the basis of their ability to induce lysis of Cw4- or Cw3-protected lymphoblastoid cell lines by "group 1" or "group 2" NK clones, respectively. From mice immunized with phytohemagglutinin (PHA)-activated lymphocytes expressing either Cw3 or Cw4 alleles, two mAb were selected, the 6A4 (IgG1) and the A6-136 (IgM), on the basis of their ability to induce lysis of protected target cell. Both mAb immunoprecipitated molecules which, in sodium dodecyl sulfate-polyacrylamide gel electrophoresis, gave two bands of 45 and 12 kDa, typical of the class I heavy chain and beta 2 microglobulin, respectively. It has been proposed (but not proven), that self major histocompatibility complex class I molecules protect normal cells from autologous NK cell lysis. Thus, we used the 6A4 and A6-136 mAb to assess this possibility directly. Cw4-specific ("group 1") and Cw3-specific ("group 2") NK clones were isolated from donors expressing the corresponding (or related) protective C alleles. None of these clones lysed autologous PHA-induced blasts, used as target cells. However, addition of the F(ab')2 of 6A4 mAb or the A6-136 mAb resulted in lysis of autologous target cells by "group 1" or "group 2" NK clones, respectively. These data provide direct evidence that the expression of class I molecules protects normal cells from lysis by autologous NK cells.
First Ba(Zr,Ti)O3 (BZT) ceramics with various compositions prepared by solid state, with high density, homogeneous microstructures and grain sizes in the range (0.7-4) ?m were studied. Besides the ...dielectric and ferroelectric investigations, First Order Reversal Curves method was employed to describe the changes of the switching properties induced by composition and grain size, related to the ferroelectric-relaxor crossover. The dielectric and ferroelectric data for ceramics with similar grain sizes demonstrated the expected ferroelectricrelaxor crossover induced by increasing x. For a given composition (x = 0.10), the relaxor character increases whit reduction of the grain size. The FORC distribution shows almost zero reversible contribution and well separated sharp irreversible component for larger grains, while more diffuse distribution with a continuous extension from Ec=0 (reversible) to Ec?0 (irreversible, switching) is typical for finer grains. .

The effect of different pre-slaughter procedures on behavioural and blood parameters were evaluated on 120 pigs reared in one farm and delivered in groups of 40 subjects to three slaughterhouses. Due ...to the different attitude of the personnel involved, differences in handling were evident at loading and at unloading where the difficulties to drive the pigs increased the behavioural events. Blood analysis parameters showed that different resting time did not reduce the physical stress experienced by the pigs, which seems related "per se" to loading, transport and unloading and not to the different handling applied in each slaughter plant.
