The ever-increasing presence of contaminants in environmental waters is an alarming issue, not only because of their harmful effects in the environment but also because of their risk to human health. ...Pharmaceuticals and pesticides, among other compounds of daily use, such as personal care products or plasticisers, are being released into water bodies. This release mainly occurs through wastewater since the treatments applied in many wastewater treatment plants are not able to completely remove these substances. Therefore, the analysis of these contaminants is essential but this is difficult due to the great variety of contaminating substances. Facing this analytical challenge, electrochemical sensing based on molecularly imprinted polymers (MIPs) has become an interesting field for environmental monitoring. Benefiting from their superior chemical and physical stability, low-cost production, high selectivity and rapid response, MIPs combined with miniaturized electrochemical transducers offer the possibility to detect target analytes in-situ. In most reports, the construction of these sensors include nanomaterials to improve their analytical characteristics, especially their sensitivity. Moreover, these sensors have been successfully applied in real water samples without the need of laborious pre-treatment steps. This review provides a general overview of electrochemical MIP-based sensors that have been reported for the detection of pharmaceuticals, pesticides, heavy metals and other contaminants in water samples in the past decade. Special attention is given to the construction of the sensors, including different functional monomers, sensing platforms and materials employed to achieve the best sensitivity. Additionally, several parameters, such as the limit of detection, the linear concentration range and the type of water samples that were analysed are compiled.

In this study, powder flowability is assessed during granular compaction under free surface conditions by using Hausner ratio (HR) as flowability index. Different kinds of pharmaceutical and food ...powders were examined. The assessment of the flow properties of these powders according to HR is obtained using three compaction devices: DensiTap®, GranuPack® and a homemade Vibratory device (particle dumper).
The comparison of the HR values obtained with each compaction test provided a deeper understanding of HR. Here we show that HR is strongly dependent on the energy supplied to the system. This energy is the result of the vibration wave transmitted to the powder bed, and is strongly dependent on the vibration amplitude, i.e. on the device used.
HR analysis as a function of the energy supplied to the powder bed allowed us to determine a maximal HR value, denoted ultimate HR (HRU), obtained when reaching a maximal compaction state. Thus, a new approach is proposed to determine flow behavior, through HR, as a function of the energy supplied to the powder bed.

Traumatic stress in early-life increases the risk for cognitive and neuropsychiatric disorders later in life. Such early stress can also impact the progeny even if not directly exposed, likely ...through epigenetic mechanisms. Here, we report in mice that the offspring of males subjected to postnatal traumatic stress have decreased gene expression in molecular pathways necessary for neuronal signaling, and altered synaptic plasticity when adult. Long-term potentiation is abolished and long-term depression is enhanced in the hippocampus, and these defects are associated with impaired long-term memory in both the exposed fathers and their offspring. The brain-specific gamma isoform of protein kinase C (Prkcc) is one of the affected signaling components in the hippocampus. Its expression is reduced in the offspring, and DNA methylation at its promoter is altered both in the hippocampus of the offspring and the sperm of fathers. These results suggest that postnatal traumatic stress in males can affect brain plasticity and cognitive functions in the adult progeny, possibly through epigenetic alterations in the male germline.
The appropriate choice of reference genes is essential for accurate normalization of gene expression data obtained by the method of reverse transcription quantitative real-time PCR (RT-qPCR). In ...2009, a guideline called the Minimum Information for Publication of Quantitative Real-Time PCR Experiments (MIQE) highlighted the importance of the selection and validation of more than one suitable reference gene for obtaining reliable RT-qPCR results. Herein, we searched the recent literature in order to identify the bacterial reference genes that have been most commonly validated in gene expression studies by RT-qPCR (in the first 5 years following publication of the MIQE guidelines). Through a combination of different search parameters with the text mining tool MedlineRanker, we identified 145 unique bacterial genes that were recently tested as candidate reference genes. Of these, 45 genes were experimentally validated and, in most of the cases, their expression stabilities were verified using the software tools geNorm and NormFinder. It is noteworthy that only 10 of these reference genes had been validated in two or more of the studies evaluated. An enrichment analysis using Gene Ontology classifications demonstrated that genes belonging to the functional categories of DNA Replication (GO: 0006260) and Transcription (GO: 0006351) rendered a proportionally higher number of validated reference genes. Three genes in the former functional class were also among the top five most stable genes identified through an analysis of gene expression data obtained from the Pathosystems Resource Integration Center. These results may provide a guideline for the initial selection of candidate reference genes for RT-qPCR studies in several different bacterial species.
In this work, a disposable electrochemical (voltammetric) molecularly imprinted polymer (MIP) sensor for the selective determination of diclofenac (DCF) was constructed. The proposed MIP-sensor ...permits fast (30 min) analysis, is cheap, easy to prepare and has the potential to be integrated with portable devices. Due to its simplicity and efficiency, surface imprinting by electropolymerization was used to prepare a MIP on a screen-printed carbon electrode (SPCE). MIP preparation was achieved by cyclic voltammetry (CV), using dopamine (DA) as a monomer in the presence of DCF. The differential pulse voltammetry (DPV) detection of DCF at MIP/SPCE and non-imprinted control sensors (NIP) showed an imprinting factor of 2.5. Several experimental preparation parameters were studied and optimized. CV and electrochemical impedance spectroscopy (EIS) experiments were performed to evaluate the electrode surface modifications. The MIP sensor showed adequate selectivity (in comparison with other drug molecules), intra-day repeatability of 7.5%, inter-day repeatability of 11.5%, a linear range between 0.1 and 10 μM (
= 0.9963) and a limit of detection (LOD) and quantification (LOQ) of 70 and 200 nM, respectively. Its applicability was successfully demonstrated by the determination of DCF in spiked water samples (river and tap water).
In recent years, analytical chemistry has been facing new challenges, particularly in developing low-cost, green, and easy-to-reproduce methods. In this work, a simple, reproducible, and low-cost ...electrochemical (voltammetric) molecularly imprinted polymer (MIP) sensor was designed specifically for the detection of trazodone (TZD). Trazodone (TZD) is an antidepressant drug consumed worldwide since the 1970s. By combining electropolymerization (surface imprinting) with screen-printed electrodes (SPCEs), the sensor is easy to prepare, is environmentally friendly (uses small amounts of reagents), and can be used for in situ analysis through integration with small, portable devices. The MIP was obtained using cyclic voltammetry (CV), using 4-aminobenzoic acid (4-ABA) as the functional monomer in the presence of TZF molecules in 0.1 M HCl. Non-imprinted control was also constructed in the absence of TZD. Both polymers were characterized using CV, and TZD detection was performed with DPV using the oxidation of TZD. The polymerization conditions were studied and optimized. Comparing the TZD signal for MIP/SPCE and NIP/SPCE, an imprinting factor of 71 was estimated, indicating successful imprinting of the TZD molecules within the polymeric matrix. The analytical response was linear in the range of 5-80 µM, and an LOD of 1.6 µM was estimated. Selectivity was evaluated by testing the sensor for molecules with a similar structure to TZD, and the ability of MIP/SPCE to selectively bind to TZD was proven. The sensor was applied to spiked tap water samples and human serum with good recoveries and allowed for a fast analysis (around 30 min).

Naloxone (NLX) is a pharmaceutical used as opioid antagonist. A molecular imprinted polymer electrochemical sensor for simple and rapid detection of NLX was prepared through the modification of commercial available screen printed carbon electrode (SPCE). The SPCE was modified with multi-walled carbon nanotubes (MWCNT) by drop coating to increase the signal response and improve the sensitivity. The MIP preparation was carried out via in situ electropolymerization using 4-aminobenzoic acid (4-ABA) as functional monomer. The morphology of the obtained sensor was characterized by scanning electron microscopy (SEM). Several parameters controlling the preparation and performance of the MIP sensor were studied and optimized. The electrochemical behavior of NLX at MIP and control non-imprinted (NIP) sensor was evaluated by differential pulse voltammetry (DPV), demonstrating a better MIP response and the success of the imprinting. The proposed MIP/MWCNT/SPCE sensor showed a linear relationship between peak current intensity and NLX concentration in the range between 0.25 and 10.0μM, with limits of detection (LOD) and quantification (LOQ) of 0.20μM and 0.67μM respectively. The repeatability and reproducibility were also tested with relative standard deviations (RSD) of 4.6 and 9.6% respectively. Moreover, the applicability of the method was successfully confirmed with detection of NLX in biological samples (urine and human serum). The sensor is promising to be used for screening NLX in point-of-care people with opioid overdose.

This study demonstrates the ability of an electrochemical sensor based on molecularly imprinted polymers (MIPs) to selectively quantify 3,4-methylenedioxymethamphetamine (MDMA), also known as ecstasy, in biological samples. The device was constructed using ortho-phenylenediamine (o-PD) as the MIP’s building monomer at the surface of a screen-printed carbon electrode (SPCE). The step-by-step construction of the SPCE-MIP sensor was characterized by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). Density functional theory (DFT) calculations and modelling were performed not only to understand template-monomer interaction but also to comprehend which possible polymer structure - linear or ramified poly(o-PD) – indeed interacts with the analyte. The prepared sensor worked by directly measuring the MDMA oxidation signal through square-wave voltammetry (SWV) after an incubation period of 10 min. Several parameters were optimized, such as the monomer/template ratio, the number of electropolymerization scanning cycles, and the incubation period, to obtain the best sensing efficiency. Optimized sensors exhibited suitable selectivity, repeatability (2.6%), reproducibility (7.7%) and up to one month of stable response. A linear range up to 0.2 mmol L−1 was found with an r2 of 0.9990 and a limit of detection (LOD) and quantification (LOQ) of 0.79 and 2.6 μmol L−1 (0.15 and 0.51 μg mL−1), respectively. The proposed sensor was successfully applied to human blood serum and urine samples, showing its potential for application in medicine and in forensic sciences.

The incidence of breast cancer has been increasing over the years. To control and monitor this disease several tumor biomarkers have been proposed for early diagnosis, patient follow-up and/or treatment guidance. The only serum breast cancer biomarker in current use is the cancer antigen 15-3 (CA 15-3). In this work a molecularly imprinted polymer (MIP)-based electrochemical (voltammetric) sensor to monitor breast cancer was developed, based on direct surface imprinting of CA 15-3 on a screen-printed gold electrode (Au-SPE). The imprinting was performed in two steps: (1) adsorption of CA 15-3 on the surface of the Au-SPE and (2) electropolymerization of 2-aminophenol around the adsorbed protein. After extraction of the imprinted protein voltammetric analysis was conducted using hexacyanoferrate(II/III) as redox probe, measuring the signals before and after protein binding. The sensor was characterized by voltammetric techniques and electrochemical impedance spectroscopy, and the analytical responses of imprinted and non-imprinted polymer sensors were studied. A linear relationship between the peak current intensity of the redox probe and the logarithm of CA 15-3 concentration was established between 5 and 50UmL−1, achieving a limit of detection of 1.5UmL−1. The prepared MIP-sensor provides fast (15min) analysis and is cheap, easy to prepare, disposable and could easily be integrated in small portable point-of care devices.

Determination of antibiotics in environmental waters is an important global issue. Although furazolidone (FZD) was banned from use in food-producing animals, owing to its mutagenic and carcinogenic effects, this antibiotic has been illegally used across the world and its presence in environment have being noted.
In this work, the first selective molecularly imprinted polymer (MIP) was developed for electrochemical detection of FZD. It was constructed based on the modification of the traditional carbon paste electrode (CPE) with MIP microparticles, followed by introduction of multi-walled carbon nanotubes (MWCNTs). Quantum mechanical (QM) calculations and molecular dynamics (MD) simulations were performed to allow rational selection of an appropriate functional monomer and to simulate the best pre-polymerisation conditions, respectively. The MIP were synthetized by polymerization using 2-acrylamido-2-methyl-1-propanesulfonic acid (AMPS) as monomer and FZD as template molecule. The MIP microparticles were then incorporated on CPE-MWCNTs and the electrochemical analysis of FZD were evaluated by differential pulse voltammetry (DPV). After optimisation of experimental conditions, the MIP-CPE-MWCNTs sensor exhibited a good linear response over the concentration range of 0.01 μM to 1 μM with a correlation coefficient of 0.9995. The limit of detection (LOD) was found to be 0.03 μM (S/N = 3). Due to high imprinting efficiency the sensor displayed selectivity to recognise FZD molecules and it was successfully applied in water samples where excellent recovery values (over 90 %) were obtained. The proposed sensor provides an efficient and promising sustainable strategy for monitorisation of FZD in environmental waters.
