Diabetic foot ulceration (DFU) is a devastating complication of diabetes whose pathogenesis remains incompletely understood. Here, we profile 174,962 single cells from the foot, forearm, and ...peripheral blood mononuclear cells using single-cell RNA sequencing. Our analysis shows enrichment of a unique population of fibroblasts overexpressing MMP1, MMP3, MMP11, HIF1A, CHI3L1, and TNFAIP6 and increased M1 macrophage polarization in the DFU patients with healing wounds. Further, analysis of spatially separated samples from the same patient and spatial transcriptomics reveal preferential localization of these healing associated fibroblasts toward the wound bed as compared to the wound edge or unwounded skin. Spatial transcriptomics also validates our findings of higher abundance of M1 macrophages in healers and M2 macrophages in non-healers. Our analysis provides deep insights into the wound healing microenvironment, identifying cell types that could be critical in promoting DFU healing, and may inform novel therapeutic approaches for DFU treatment.
Moral judgments are often viewed as the outcome of affective and deliberative processes that could be impacted by social factors and individual characteristics. The purpose of this study was to ...examine the interaction between gender and social context on moral judgment. Participants included 315 undergraduate students (67.3% female). The participants completed the Moral Decision-Making Task while seated at row tables facing the front of the room or round tables facing other participants. The results indicated that males responded in a more utilitarian manner (harm one to save five) than females for moral impersonal (MI) and moral personal (MP) dilemmas regardless of seating arrangements. When seated at round tables, all participants were more likely to respond deontologically (cause no harm) to the moral impersonal dilemmas. In addition, we calculated a moral reasoning difference score for each participant as the difference between the MI and MP scores to represent additional reactivity due to the idea of taking direct action. The moral reasoning difference score was consistent for females but indicated a more deontological response from males at round tables and a more utilitarian response from males at row tables. These results suggest that males are more utilitarian than females and are more likely to be influenced by social context when responding to moral dilemmas. More broadly, the current results indicate that moral judgments are affected by social context particularly in males in ways that have not been incorporated in many models of moral decision making.
The interaction between sleep and work-related behaviors influence many aspects of employee performance, safety, and health as well as organizational-level success. Although it is well established ...that quantity and quality of sleep can affect different types of task performance and personal health, the interactions between sleep habits and organizational behaviors have received much less attention. It is important to examine how sleep habits and workplace behaviors relate and the role of the underlying circadian rhythm on the potential impact of sleep and sleepiness in the workplace. Developing a deeper understanding of how sleep habits and sleepiness impact workers and the organization can help provide the necessary background for human resource management to develop more progressive support networks for employees that benefit both the worker and the organization. Human resources and employees should emphasize the impact of good sleep and sleep habits on organizational and individual productivity and safety.
Pregnancy in HoFH females is associated with further elevation of already markedly elevated low density lipoprotein cholesterol (LDL-C) levels, particularly if lipid-lowering therapy is discontinued, ...placing the mother and fetus at increased cardiovascular risk. Lipoprotein apheresis is the current recommended treatment for pregnant HoFH patients. However, this is costly, time consuming, and is not available in many countries. Alternative treatment strategies to control hypercholesterolaemia during pregnancy in HoFH patients are necessary.
This study was a retrospective review of 39 pregnancies from a cohort of 20 genotypically confirmed female HoFH patients.
No maternal cardiac complications or deaths occurred during the pregnancies or during the first year postpartum. Twenty five pregnancies were exposed to lipid-lowering therapy, of which 18 were exposed to statin therapy, just prior to or during the pregnancy. Thirty three (84%) pregnancies carried to term, 3 (8%) premature deliveries and 3 (8%) miscarriages were observed. Complications associated with pregnancy in these HoFH patients, did not differ from those reported during pregnancies of otherwise healthy woman.
HoFH is a severe disease impacting significantly on life expectancy. However, for many females with HoFH, despite the high cardiovascular risk, pregnancy is not uncommon. In resource poor settings and when LA is not available, lipid lowering therapy, particularly statin therapy during pregnancy, appears to be safe for both mother and fetus and is an acceptable alternative for LDL-C reduction in these high risk patients.
•Statin exposure during pregnancy might not be as detrimental to the fetus as previously thought.•Pregnancy complications in homozygous familial hypercholesterolaemia patients did not differ from healthy woman.•Birth weights between statin exposed and non-exposed pregnancies was similar.•No severe skeletal abnormalities or developmental delay occurred in statin exposed fetusses.
A meta-analysis to mathematically summarize the effect of hot and cold temperature exposure on performance was completed. The results from 515 effect sizes calculated from 22 original studies suggest ...that hot and cold temperatures negatively impact performance on a wide range of cognitive-related tasks. More specifically, hot temperatures of 90°F (32.22°C) Web Bulb Globe Temperature Index or above and cold temperatures of 50°F (10°C) or less resulted in the greatest decrement in performance in comparison to neutral temperature conditions (14.88% decrement and 13.91% decrement, respectively). Furthermore, the duration of exposure to the experimental temperature, the duration of exposure to the experimental temperature prior to the task onset, the type of task and the duration of the task had di

ects on performance. The current results indicate that hot and cold temperature exposure have a negative impact on performance and that other variables (e.g., length of exposure to the temperature or task duration) may modify this relationship.
Although researchers have investigated the impact of social isolation on well-being, the recent quarantines due to COVID-19 resulted in a social isolation environment that was unique to any examined ...in the past. Because sleep is one of the endogenous drives that impacts short and long-term health and well-being, it is important to consider how social isolation during the COVID-19 government-mandated quarantines affected sleep and sleep habits. A number of researchers have addressed this question during the last 2 years by examining several concepts related to possible changes in sleep during the quarantines. To best understand these recent results, the current mini review provides a background on the pre-pandemic literature on the effects of social isolation and loneliness with a focus on sleep and then summarizes the recent literature on sleep and sleep habits. In general, sleep was negatively impacted for many people during the pandemics but not all. One group that seemed to benefit from the pandemic in terms of sleep patterns, were younger people who could more easily adapt their sleep times to match their internal chronobiology. Given the potential broad impact of sleep on health and well-being, better understanding how social isolation impacts sleep is an important consideration for individuals, work organizations, and governments.
Self-control is an intricate component of decision making and effectively managing day-to-day life. Failing to maintain adequate self-control can have negative effects on many desired goals and ...social experiences. As such, understanding how different facets of the human experience may affect self-control is an important undertaking. One area that is yet unclear is the possible relationships between social support and self-control. Research suggests that social support can be an effective resource in reducing stress and promoting health and well-being. Research has also indicated that stress can be a limiting factor on self-control. In contrast, few studies have focused on social support as a potential resource for self-control. The goal of this mini-review article is to explore the intersections between self-control and social support and encourage integration of these two relatively independent areas of research. This review will help provide a broader understanding of self-control resources and how we can better understand the relationships between social well-being and our ability to monitor and utilize our capacity to maintain self-control.
Volcanic eruptions contribute to climate variability, but quantifying these contributions has been limited by inconsistencies in the timing of atmospheric volcanic aerosol loading determined from ice ...cores and subsequent cooling from climate proxies such as tree rings. Here we resolve these inconsistencies and show that large eruptions in the tropics and high latitudes were primary drivers of interannual-to-decadal temperature variability in the Northern Hemisphere during the past 2,500 years. Our results are based on new records of atmospheric aerosol loading developed from high-resolution, multi-parameter measurements from an array of Greenland and Antarctic ice cores as well as distinctive age markers to constrain chronologies. Overall, cooling was proportional to the magnitude of volcanic forcing and persisted for up to ten years after some of the largest eruptive episodes. Our revised timescale more firmly implicates volcanic eruptions as catalysts in the major sixth-century pandemics, famines, and socioeconomic disruptions in Eurasia and Mesoamerica while allowing multi-millennium quantification of climate response to volcanic forcing.
Good sleep habits and effective self-control are important components of successful functioning. Unfortunately chronic sleep loss and impaired self-control are common occurrences for many individuals ...which can lead to difficulty with daily self-control issues such as resisting impulses and maintaining attentive behavior. Understanding how self-control is depleted and how good sleep habits may help replenish and maintain the capacity for self-control is an important issue. A sleep-deprived individual who has expended the necessary resources for self-control is at an increased risk for succumbing to impulsive desires, poor attentional capacity, and compromised decision making. To date, few studies have investigated how sleep and self-control are inter-related. The goal of this mini-review is to explore the intersection between sleep habits and self-control and encourage researchers to focus on a new area of research that integrates what are at present largely separate areas in psychology and human neurosciences.
Fisheries bycatch is a widespread and serious issue that leads to declines of many important and threatened marine species. However, documenting the distribution, abundance, population trends and ...threats to sparse populations of marine species is often beyond the capacity of developing countries because such work is complex, time consuming and often extremely expensive. We have developed a flexible tool to document spatial distribution and population trends for dugongs and other marine species in the form of an interview questionnaire supported by a structured data upload sheet and a comprehensive project manual. Recognising the effort invested in getting interviewers to remote locations, the questionnaire is comprehensive, but low cost. The questionnaire has already been deployed in 18 countries across the Indo-Pacific region. Project teams spent an average of USD 5,000 per country and obtained large data sets on dugong distribution, trends, catch and bycatch, and threat overlaps. Findings indicated that >50% of respondents had never seen dugongs and that 20% had seen a single dugong in their lifetimes despite living and fishing in areas of known or suspected dugong habitat, suggesting that dugongs occurred in low numbers. Only 3% of respondents had seen mother and calf pairs, indicative of low reproductive output. Dugong hunting was still common in several countries. Gillnets and hook and line were the most common fishing gears, with the greatest mortality caused by gillnets. The questionnaire has also been used to study manatees in the Caribbean, coastal cetaceans along the eastern Gulf of Thailand and western Peninsular Malaysia, and river dolphins in Peru. This questionnaire is a powerful tool for studying distribution and relative abundance for marine species and fishery pressures, and determining potential conservation hotspot areas. We provide the questionnaire and supporting documents for open-access use by the scientific and conservation communities.
