CD4+CD25+ regulatory T cells (Tregs) plays a key role in maintaining immune tolerance. IgAN has a close relationship with the immune response. However the significance of CD4+CD25+ T cells to improve ...renal function of IgAN patients is not clear. The purpose of this study is to evaluate renal function of experimental IgAN rats treated by CD4+CD25+ Tregs cells.
CD4+CD25+ Tregs were separated from the blood of SD rats by immunomagnetic beads, and amplified in vitro. The amplified in vitro and cultured 2*106 CD4+CD25+ Tregs were infused intravenously into IgAN rat model, 3 times every other day. The serum creatinine, urea nitrogen, urinary protein and red blood cells were detected in the fourth and eighth week. The glomerular damage was evaluated by pathological analysis.
Tregs cells can be amplified largely in vitro. After CD4+CD25+ T cells were infused into IgAN rat model, urine protein and red blood cells were improved. The glomerular injury can be improved by pathological analysis.
CD4+CD25+ regulatory T cells can significantly improve the symptoms of immunoglobulin A nephropathy (IgAN) rat model, and have clinical application prospect.
This study aimed to investigate the effects of Wuzhi capsule on blood concentration of tacrolimus after renal transplantation. Sixty patients after allogenic renal transplantation were enrolled in ...this study and randomly divided into an experimental group and a control group. One oral Wuzhi capsule was taken in the morning and evening for patients in the experimental group, while none was given to the control group, maintaining the trough blood concentration of tacrolimus in the normal range. After 3 weeks, the changes of tacrolimus dosage and hepatorenal function in the two groups were compared. Comparisons of drug dosage and blood concentration C0 value of tacrolimus before initiating the experiment showed that there was no statistically ignificant difference (P>0.05) between the two groups. The differences of blood concentration of tacrolimus and hepatorenal function for patients in both two groups after 3 weeks treatment also showed no statistical significance (P>0.05), whereas a statistically significant decrease was demonstrated in the tacrolimus dosage of the Wuzhi capsule group compared with that of the control group (P=0.0083). After renal transplantation, Wuzhi capsules were added so as to enable tacrolimus to reach a suitable blood concentration, which can prevent the occurrence of renal transplantation rejection, thus alleviating the economic burden of patients and producing larger social benefits.

To study the characteristics of gene mutations in Chinese myelodysplastic syndromes (MDS) patients.
A total of 511 Chinese patients with MDS performed 112-gene targeted sequencing were ...retrospectively analyzed.
Eighty-three distinct mutant genes were found in 511 patients with MDS. Amongst these, the most frequent mutations was associated with epigenetics (50%) , followed by spliceosome (37%) , signal transduction (34%) , transcription factors (24%) and cell cycle/apoptosis (17%) . 439 subjects (86%) had at least one gene mutation. The mean number of mutations in refractory anemia with unilineage dysplasia (RCUD) was 1.25, refractory anemia with multilineage dysplasia (RCMD) was 1.73, refractory anemia with ring sideroblasts (RARS) was 2.79, refractory anemia with excess blasts-1 (RAEB-1) was 2.22, RAEB-2 was 2.34, MDS with isolated 5q- was 2.67, MDS, unclassified (MDS-U) was 2.00. A total of 906 consecutive newly diagnosed patients with PV seen at the Blood Diseases Hospital, ...Chinese Academy of Medical Sciences, from June 2007 to February 2020 were included, and their data were collected. PV was diagnosed according to 2016 World Health Organization (WHO) diagnostic definitions. OS and prognostic factors were retrospectively analyzed.

To investigate the clinical manifestation, cytogenetics, gene mutations and prognostic factors of chronic neutrophilic leukemia (CNL) .
16 CNL cases, according to WHO (2016) -definition, were ...reviewed retrospectively. Identifications of the CSF3R, ASXL1, SETBP1, CALR and MPL mutations were performed by direct sequencing. JAK2 V617F mutation was detected by AS-PCR.
Of the 16 CNL patients, the median age was 64 (43-80) years with a male predominance of 75% (12/16) . The median hemoglobin was 114 (81-154) g/L, with median WBC of 41.20 (26.05-167.70) (10(9)/L and median PLT of 238 (91-394) ×10(9)/L.The median level of marrow fibrosis (MF) was 1 (0-3) degree. There was no other cytogenetic abnormalities except t (1;7) (p32;q11) , +21 and 14ps+ for each. All the 16 CNL patients harbored CSF3R T618I mutation. ASXL1 mutations were identified in 81% (13/16) , while SETBP1 mutations were confirmed in 63% (10/16) . The CALR K385fs*47 mutation was found. There was no mutation in JAK2 V617F or MPL in the above 16 patients. CSF3R T618I mutation was specific for CNL. The median OS of CNL patients was 24 months, and WBC≥50×10(9)/L at diagnosis was an unfavorable prognostic factor.
Leaves of Nagafu No. 6 were transformed by the Agrobacterium tumefaciens-mediated method, and 158 hyg-resistant buds were obtained. Using fresh leaf tissue, seven positive lines were obtained by ...polymerase chain reaction (PCR) with total DNA, and four by reverse transcriptase–polymerase chain reaction (RT–PCR) with total RNA. The results showed that the MhTGA2 gene was successfully transformed into Nagafu No. 6 plants and expressed. At 4 months after transplanting, photosynthetic parameters of young leaves were determined using transgenic and control plants prior to the determination of leaf surface characteristics (epidermal structure and stomatal distribution) by scanning electronic microscopy. The results showed that the net photosynthesis rate, the instantaneous carboxylation rate and stomatal conductance of transgenic plants were higher than the control, whereas the internal carbon dioxide (CO2) concentrations of the transgenic plants were lower. Under non-saline conditions, the stomata of the transgenic plants were better distributed than in the control. Under saline conditions, the epidermal structure of the control leaves was severely hypohydrated and the quantity of stomata was significantly decreased. Conversely, the epidermal structure of transgenic leaves was not significantly altered, indicating salt-stress tolerance. The overall results suggested that MhTGA2 may play a crucial role in the response to salt stress in Nagafu No. 6.
