Acyl-CoA binding domain-containing 7 (Acbd7) is a paralog gene of the diazepam-binding inhibitor/Acyl-CoA binding protein in which single nucleotide polymorphism has recently been associated with ...obesity in humans. In this report, we provide converging evidence indicating that a splice variant isoform of the Acbd7 mRNA is expressed and translated by some POMC and GABAergic-neurons in the hypothalamic arcuate nucleus (ARC). We have demonstrated that the ARC ACBD7 isoform was produced and processed into a bioactive peptide referred to as nonadecaneuropeptide (NDN) in response to catabolic signals. We have characterized NDN as a potent anorexigenic signal acting through an uncharacterized endozepine G protein-coupled receptor and subsequently via the melanocortin system. Our results suggest that ACBD7-producing neurons participate in the hypothalamic leptin signalling pathway. Taken together, these data suggest that ACBD7-producing neurons are involved in the hypothalamic control exerted on food intake and energy expenditure by the leptin-melanocortin pathway.
Background
Patients with spontaneous or traumatic active mesenteric bleeding cannot be treated endoscopically. Transarterial embolization can serve as a potential alternative to emergency surgery. ...Literature on transarterial embolization for mesenteric bleeding remains very scarce. The objective of this study was to evaluate the safety and efficacy of transarterial embolization for mesenteric bleeding. We reviewed all consecutive patients admitted for mesenteric bleeding to the interventional radiology department, in a tertiary center, between January 2010 and March 2021. Mesenteric bleeding was defined as mesenteric hematoma and contrast extravasation and/or pseudoaneurysm visible on pre-operative CT scan. We evaluated technical success, clinical success, and complications.
Results
Among the 17 patients admitted to the interventional department for mesenteric bleeding, 15 presented with active mesenteric bleeding requiring transarterial embolization with five patients with hemodynamic instability. Mean age was 67 ± 14 years, including 12 (70.6%) males. Technical success was achieved in 14/15 (93.3%) patients. One patient with technical failure was treated by percutaneous embolization with NBCA-Lipiodol mixture. Three patients (20%) had early rebleeding: two were treated by successful repeat embolization and one by surgery. One patient (6.7%) had early death within 30 days and two patients (13.3%) had late death after 30 days. Mean length of hospitalization was 12.8 ± 7 days. There were no transarterial embolization-related ischemic complications.
Conclusion
Transarterial embolization is a safe and effective technique for treating mesenteric bleeding even in patients with hemodynamic instability. Transarterial embolization doesn’t close the door to surgery and could be proposed as first intention in case of mesenteric bleeding.
Focal epilepsy is characterized by repeated spontaneous seizures that originate from cortical epileptogenic zone networks (EZN). Analysis of intracerebral recordings showed that subcortical ...structures, and in particular the thalamus, play an important role in seizure dynamics as well, supporting their structural alterations reported in the neuroimaging literature. Nonetheless, between‐patient differences in EZN localization (e.g., temporal vs. non‐temporal lobe epilepsy) as well as extension (i.e., number of epileptogenic regions) might impact the magnitude as well as spatial distribution of subcortical structural changes. Here we used 7 Tesla MRI T1 data to provide an unprecedented description of subcortical morphological (volume, tissue deformation, and shape) and longitudinal relaxation (T1) changes in focal epilepsy patients and evaluate the impact of the EZN and other patient‐specific clinical features. Our results showed variable levels of atrophy across thalamic nuclei that appeared most prominent in the temporal lobe epilepsy group and the side ipsilateral to the EZN, while shortening of T1 was especially observed for the lateral thalamus. Multivariate analyses across thalamic nuclei and basal ganglia showed that volume acted as the dominant discriminator between patients and controls, while (posterolateral) thalamic T1 measures looked promising to further differentiate patients based on EZN localization. In particular, the observed differences in T1 changes between thalamic nuclei indicated differential involvement based on EZN localization. Finally, EZN extension was found to best explain the observed variability between patients. To conclude, this work revealed multi‐scale subcortical alterations in focal epilepsy as well as their dependence on several clinical characteristics.

Motivation
Density Peaks is a widely spread clustering algorithm that has been previously applied to Molecular Dynamics (MD) simulations. Its conception of cluster centers as elements ...displaying both a high density of neighbors and a large distance to other elements of high density, particularly fits the nature of a geometrical converged MD simulation. Despite its theoretical convenience, implementations of Density Peaks carry a quadratic memory complexity that only permits the analysis of relatively short trajectories.
Results
Here, we describe DP+, an exact novel implementation of Density Peaks that drastically reduces the RAM consumption in comparison to the scarcely available alternatives designed for MD. Based on DP+, we developed RCDPeaks, a refined variant of the original Density Peaks algorithm. Through the use of DP+, RCDPeaks was able to cluster a one-million frames trajectory using less than 4.5 GB of RAM, a task that would have taken more than 2 TB and about 3× more time with the fastest and less memory-hunger alternative currently available. Other key features of RCDPeaks include the automatic selection of parameters, the screening of center candidates and the geometrical refining of returned clusters.

The increase in worldwide travel is making imported malaria a growing health concern in non-endemic countries. Most data on the pathophysiology of malaria come from endemic areas. Little is known ...about cytokine profiles during imported malaria. This study aimed at deciphering the relationship between cytokine host response and malaria severity among imported cases in France. This study reports cytokine profiles in adults with Plasmodium falciparum malaria included in the PALUREA prospective study conducted between 2006 and 2010. The patients were classified as having uncomplicated malaria (UM) or severe malaria (SM), with this last further categorized as very severe malaria (VSM) or less severe malaria (LSM). At hospital admission, eight blood cytokines were assayed in duplicate using Luminex
technology: interleukin (IL)-1α, IL-1β, IL-2, IL-4, IL-10, tumor necrosis factor (TNF)α, interferon (IFN)γ, and macrophage migration inhibitory factor (MIF). These assays were repeated on days 1 and 2 in the SM group. Of the 278 patients, 134 had UM and 144 SM. At hospital admission, over half the patients had undetectable levels of IL-1α, IL-1β, IL-2, IL-4, IFNγ, and TNFα, while IL-10 and MIF were significantly higher in the SM vs. the UM group. Higher IL-10 was significantly associated with higher parasitemia (R = 0.32 0.16-0.46; P = 0.0001). In the SM group, IL-10 elevation persisting from admission to day 2 was significantly associated with subsequent nosocomial infection. Of eight tested cytokines, only MIF and IL-10 were associated with disease severity in adults with imported P. falciparum malaria. At admission, many patients had undetectable cytokine levels, suggesting that circulating cytokine assays may not be helpful as part of the routine evaluation of adults with imported malaria. Persisting high IL-10 concentration was associated with subsequent nosocomial infection, suggesting its possible interest in immune monitoring of most severe patients.
Land-based transport corridors and related infrastructure are increasingly extending into and across the Arctic in support of resource development and population growth, causing large-scale ...cumulative changes to northern socio-ecological systems. These changes include the increased mobility of people, goods and resources, and environmental impacts on landscapes and ecosystems as the human footprint reaches remote, unindustrialized regions. Arctic climate change is also generating new challenges for the construction and maintenance of these transport systems, requiring adaptive engineering solutions as well as community resilience. In this review article, we consider the complex entanglements between humans, the environment, and land transportation infrastructure in the North and illustrate these interrelations by way of seven case studies: the Baikal–Amur Mainline, Bovanenkovo Railway, Alaska–Canada Highway, Inuvik–Tuktoyatuk Highway, Alaska Railroad, Hudson Bay Railway, and proposed railways on Baffin Island, Canada. As new infrastructure is built and anticipated across the circumpolar North, there is an urgent need for an integrated socio-ecological approach to impact assessment. This would include full consideration of Indigenous knowledge and concerns, collaboration with local communities and user groups in assessment, planning and monitoring, and evaluation of alternative engineering designs to contend with the impacts of climate change in the decades ahead.
This paper presents long term creep data from high performance concretes (HPC) sourced from different research programs or civil engineering projects carried out at IFSTTAR’s Paris laboratory, ...France. The authors are now making all this data publicly available through a data archive. The experimental procedures are described in detail, in particular that used to verify the constancy over time of the specimens’ compressive stress. It is shown that the majority of the concretes exhibit linear long-term behavior when plotted against log (time). Indeed, the paper describes modelling sourced from the literature, based on a logarithmic function of time which, despite its simplicity, matches the experimental data quite well. Using this modelling, we studied the behavior of HPC prepared from the same set of constituents. We have shown that the logarithmic change in creep is a quasi linear function of the water to cement ratio and aggregate concentration. Finally, the accuracy of the fib MC2010 model code has been tested on IFSTARR’s data bank.
Anaerobic digestion of food waste still faces important challenges despite its world-wide application. An important fraction of food waste is composed of organic material having a low hydrolysis rate ...and which is often not degraded in digesters. The addition of this less hydrolysable fraction into anaerobic digesters requires a longer hydraulic residence time, and therefore leads to oversizing of the digesters. To overcome this problem, the conversion of the highly biodegradable liquid fraction from fruit and vegetable waste in a up-flow anaerobic sludge blanket (UASB) digester is proposed and demonstrated. The more easily biodegradable fraction of the waste is concentrated in the liquid phase using a 2-stage screw press separation. Then, this liquid fraction is digested in a 3.5 L UASB digester at a high organic loading rate. A good and stable performance was observed up to an organic loading rate (OLR) of 12 g COD/(Lrx.d), with a specific methane production of 2.6 L CH
/(Lrx.d) and a degradation of 85% of the initial total COD. Compared to the conversion of the same initial waste with a continuously stirred tank reactor (CSTR), this new treatment strategy leads to 10% lower COD degradation, but can produce the same amount of methane with a digester that is twice as small. The scale-up of this process could contribute to reduced costs related to the anaerobic digestion of food waste, while reducing management efforts associated with digestate handling and increasing process stability at high organic loading rates.
