The achievement of a strong and stable bond between composite resin and dentin remains a challenge in restorative dentistry. Over the past two decades, dental materials have been substantially ...improved, with better handling and bonding characteristics. However, little attention has been paid to the contribution of collagen structure/stability to bond strength. We hypothesized that the induction of cross-linking in dentin collagen improves dentin collagen stability and bond strength. This study investigated the effects of glutaraldehyde-and grape seed extract-induced cross-linking on the dentin bond strengths of sound and caries-affected dentin, and on the stability of dentin collagen. Our results demonstrated that the application of chemical cross-linking agents to etched dentin prior to bonding procedures significantly enhanced the dentin bond strengths of caries-affected and sound dentin. Glutaraldehyde and grape seed extract significantly increased dentin collagen stability in sound and caries-affected dentin, likely via distinct mechanisms.
The Muon Portal Project has built a prototype of a real size detector (6m×3m×7 m) for the inspection of containers by muon tomography. This technique may provide 2D and 3D images of the interior of a ...container, to identify the presence of high-Z materials. In the present Project, 4800 extruded scintillator strips were arranged such as to cover four X–Y detection planes (6m×3 m), two placed above and two below the container to be inspected. Silicon photomultipliers were used as photosensors, to collect the light transported by Wave Length Shifter (WLS) fibres embedded in the scintillator strips. First tomographic images are here presented.
Growing evidence indicates that microRNAs (miRNAs or miRs) are involved in basic cell functions and oncogenesis. Here we report that miR-133 has a critical role in determining cardiomyocyte ...hypertrophy. We observed decreased expression of both miR-133 and miR-1, which belong to the same transcriptional unit, in mouse and human models of cardiac hypertrophy. In vitro overexpression of miR-133 or miR-1 inhibited cardiac hypertrophy. In contrast, suppression of miR-133 by 'decoy' sequences induced hypertrophy, which was more pronounced than that after stimulation with conventional inducers of hypertrophy. In vivo inhibition of miR-133 by a single infusion of an antagomir caused marked and sustained cardiac hypertrophy. We identified specific targets of miR-133: RhoA, a GDP-GTP exchange protein regulating cardiac hypertrophy; Cdc42, a signal transduction kinase implicated in hypertrophy; and Nelf-A/WHSC2, a nuclear factor involved in cardiogenesis. Our data show that miR-133, and possibly miR-1, are key regulators of cardiac hypertrophy, suggesting their therapeutic application in heart disease.
The serine-threonine kinase Akt seems to be central in mediating stimuli from different classes of receptors. In fact, both IGF-1 and IL6-like cytokines induce hypertrophic and antiapoptotic signals ...in cardiomyocytes through PI3K-dependent Akt activation. More recently, it was shown that Akt is involved also in the hypertrophic and antiapoptotic effects of β-adrenergic stimulation. Thus, to determine the effects of Akt on cardiac function in vivo, we generated a model of cardiac-specific Akt overexpression in mice. Transgenic mice were generated by using the E40K, constitutively active mutant of Akt linked to the rat α-myosin heavy chain promoter. The effects of cardiac-selective Akt overexpression were studied by echocardiography, cardiac catheterization, histological and biochemical techniques. We found that Akt overexpression produced cardiac hypertrophy at the molecular and histological levels, with a significant increase in cardiomyocyte cell size and concentric LV hypertrophy. Akt-transgenic mice also showed a remarkable increase in cardiac contractility compared with wild-type controls as demonstrated by the analysis of left ventricular (dP/dtmax) in an invasive hemodynamic study, although with graded dobutamine infusion, the maximum response was not different from that in controls. Diastolic function, evaluated by left ventricular dP/dtmin, was not affected at rest but was impaired during graded dobutamine infusion. Isoproterenol-induced cAMP levels, β-adrenergic receptor (β-AR) density, and β-AR affinity were not altered compared with control mice. Moreover, studies on signaling pathway activation from myocardial extracts demonstrated that glycogen synthase kinase3-β is phosphorylated, whereas p42/44 mitogen-activated protein kinases is not, indicating that Akt induces hypertrophy in vivo by activating the glycogen synthase kinase3-β/GATA 4 pathway. In summary, our results not only demonstrate that Akt regulates cardiomyocyte cell size in vivo, but, importantly, show that Akt modulates cardiac contractility in vivo without directly affecting β-AR signaling capacity.
Cosmic ray tomography is a technique which exploits the multiple Coulomb scattering of highly penetrating cosmic ray-produced muons to perform non-destructive inspection of high-Z materials without ...the use of artificial radiation. A muon tomography detection system can be used as a portal monitor at border crossing points for detecting illegal targeted objects. The Muon Portal Project is a joint initiative between Italian research and industrial partners, aimed at the construction of a real size detector prototype (6×3×7m3) for the inspection of cargo containers by the muon scattering technique. The detector consists of four XY tracking planes, two placed above and two below the container to be inspected. After a research and development phase, which led to the choice and test of the individual components, the construction and installation of the detection modules is almost completed. In this paper the present status of the Project is reported, focusing on the design and construction phase, as well as on the preliminary results obtained with the first detection planes.

Pb block was positioned inside the volume to be inspected, in order to quantitatively investigate the appearance of the Poca signal. Signal-to-noise ratio and significance of the Poca signal were investigated by means of mono-dimensional spectra of the Poca components, for different values of the scattering angle between the incoming and outgoing muon tracks and with different angle-dependent weights. A systematic scan of two-dimensional maps was also carried out, as a strategy to search for possible enhancements to the Poca signal. A comparison was also done between the results obtained from the two half-volumes, one containing the Pb block and one left empty, to take into account the response of the detector and some aspects of the Poca strategy.
The Muon Portal Project has as its goal the design and construction of a real-size working detector prototype in scale 1:1, to inspect the content of travelling containers by means of the secondary ...cosmic-ray muon radiation and to recognize high-Z hidden materials (i.e. U, Pu). The tomographic image is obtained by reconstructing the input and output trajectories of each muon when it crosses the container and, consequently, the scattering angle, making use of two trackers placed above and below the container. The scan is performed without adding any external radiation, in a reasonable time (few minutes) and with a good spatial and angular resolution. The detector consists of 8 planes each segmented in 6 identical modules. Each module is made of scintillating strips with two WaveLength Shifting fibers (WLS) inside, coupled to Silicon photomultipliers. The customized read-out electronics employs programmable boards. Thanks to a smart read-out system, the number of output channels is reduced by a factor 10. The signals from the front-end modules are sent to the read-out boards, in order to convert analog signals to digital ones, by comparison with a threshold. The data are pre-analyzed and stored into a data acquisition PC. After an intense measurement and simulation campaign to carefully characterize the detector components, the first detection modules ( 1 ×3 m 2 ) have been already built. In this paper the detector architecture, particularly focusing on the used electronics and the main preliminary results will be presented.
The design of a detector for tracking charged particles is presented together with the characterization techniques developed to extract the main design specifications. The goals for the final ...detector are to achieve real-time imaging performances, a large detection area, and a high spatial resolution, particularly suitable for medical imaging applications. This paper describes the prototype of the tracker plane, which has a 20 × 20 cm2 sensitive area consisting of two crossed ribbons of 500 μm square scintillating fibers. The information about the hit position extracted real-time tracker in an innovative way, using a reduced number of the read-out channels to obtain a very large detection area but with moderate costs and complexity. The performances of the tracker have been investigated using β sources, cosmic rays, and a 62 MeV proton beam.
