Nanoscale materials have recently gained wide attention due to their potential to revolutionize many technologies and industrial sectors, including information technology, homeland security, ...transportation, energy, food safety, environmental science, catalysis, photonics and medicine. Among various nanoparticles, platinum nanoparticles (PtNPs) are widely used for biomedical applications, including imaging, implants, photothermal therapy and drug delivery. Indeed, PtNPs possesses intrinsic antimicrobial, antioxidant, and anticancer properties. Also, due to their remarkable catalytic activity, they are able to reduce the intracellular reactive oxygen species (ROS) levels and impair the downstream pathways leading to inflammation. Various approaches, including both physical and chemical methods, are currently employed for synthesis of PtNPs. However, the use of hazardous reaction conditions and toxic chemicals in these processes poses a potential threat to the environment and severely compromise the biocompatibility of the nanoparticles. Hereby, increasing need for exploitation of novel routes for synthesis of PtNPs has led to development of biological fabrication using microbes, specifically bacteria. , etc. An overview of the underlying mechanisms of both enzymatic and non-enzymatic methods of synthesis is included. Moreover, this review highlights the scope of developing optimized process to control the physicochemical properties, such as the nanoparticle surface chemistry, charge, size and shape, which, in turn, may affect their nanotoxicity and response at the biointerface for nanomedicine applications.

Angiogenesis plays a key role in the wound healing process, involving the migration, growth, and differentiation of endothelial cells. Angiogenesis is controlled by a strict balance of different ...factors, and among these, the angiogenin protein plays a relevant role. Angiogenin is a secreted protein member of the ribonuclease superfamily that is taken up by cells and translocated to the nucleus when the process of blood vessel formation has to be promoted. However, the chemical signaling that activates the protein, normally present in the plasma, and the transport pathways through which the protein enters the cell are still largely unclear. Copper is also an angiogenic factor that regulates angiogenin expression and participates in the activation of common signaling pathways. The interaction between angiogenin and copper could be a relevant mechanism in regulating the formation of new blood vessel pathways and paving the way to the development of new drugs for chronic non-healing wounds.

In this review, we focus on the peculiar properties of the layer formed when biomolecules, especially proteins and peptides, face two-dimensional (2D) nanomaterials, to provide an overview of the state-of-the-art knowledge and the current challenges concerning the biomolecule coronas and, in general, the 2D nano-biointerface established when peptides and proteins interact with the nanosheet surface. Specifically, this review includes both experimental and simulation studies, including some recent machine learning results of a wide range of nanomaterial and peptide/protein systems.
Noble metal nanomaterials, such as gold or silver nanoparticles, exhibit unique photonic, electronic, catalytic and therapeutic properties. The high versatility in their synthesis, especially size ...and shape features, as well as in the surface functionalization by, e.g., physisorption, direct chemisorption of thiol derivatives and covalent binding through bifunctional linkers or specific affinity interactions, prompted their widespread and rising use as multifunctional platforms for theranostic purposes. In this paper, the recent developments of gold and silver nanoparticles for application in biosensing, medical imaging, diagnosis and therapy is reviewed from the following five aspects: (1) the gold and silver nanomaterials intrinsic properties of biomedical interest; (2) the synthesis of noble metal nanoparticles by chemical, physical and biological/green processes; (3) the applications of gold and silver nanoparticles in imaging, diagnostic and therapeutic mode; (4) the surface functionalization processes for targeting, controlled drug loading and release, triggered pathways of cellular uptake and tissue distribution; and (5) nanotoxicity. The historical developments and the most recent applications have been focused on, together with suggested strategies for future more efficacious, targeted delivery.

has been reported to have beneficial effects such as antioxidant, antimicrobial activity and wound repair capacity. To enhance antioxidant activity of snail mucus, it was extracted in a hydroalcoholic solution and consequently freeze-dried. The obtained snail mucus extract (SME) was indeed endowed with higher antioxidant activity observed in cell-free models, however it was not possible to test its effects in cellular models as it creates a thick film on the cell surface. Therefore, in order to enhance beneficial effects of snail mucus and extend its potential use, SME was used to develop snail mucus extract-coated gold nanoparticles (AuNPs-SME) which exhibited anti-inflammatory properties on non-tumorigenic cells. LPS-induced inflammation in human NCTC keratinocytes was used as model to investigate the
in vitro
cytoprotective effects of nanoparticles. Co-treatment with LPS and AuNPs-SME significantly reduced pro-inflammatory cytokine transcription. Moreover, we demonstrated that AuNPs-SME not only can be used for anti-inflammatory treatments, but also as a sunscreen and antioxidant for potential cosmetic applications. Furthermore, AuNPs-SME's ability to selectively inhibit the growth of two human melanoma cell lines without affecting immortalized human keratinocyte viability in the same conditions was assessed. Thus, we demonstrated that snail mucus is suitable for creating innovative formulations and it can be considered a valid candidate for cosmeceutical applications to enrich the snail mucus based anti-age and sunscreen products already present on the market. Moreover, innovative formulations containing snail mucus can be potentially used for the treatment of specific skin neoplasms.

Our knowledge of the molecular events underlying type 2 diabetes mellitus—a protein conformational disease characterized by the aggregation of islet amyloid polypeptide (IAPP) in pancreatic β ...cells—is limited. However, amyloid-mediated membrane damage is known to play a key role in IAPP cytotoxicity, and therefore the effects of lipid composition on modulating IAPP-membrane interactions have been the focus of intense research. In particular, membrane cholesterol content varies with aging and consequently with adverse environmental factors such as diet and lifestyle, but its role in the development of the disease is controversial. In this study, we employ a combination of experimental techniques and in silico molecular simulations to shed light on the role of cholesterol in IAPP aggregation and the related membrane disruption. We show that if anionic POPC/POPS vesicles are used as model membranes, cholesterol has a negligible effect on the kinetics of IAPP fibril growth on the surface of the bilayer. In addition, cholesterol inhibits membrane damage by amyloid-induced poration on membranes, but enhances leakage through fiber growth on the membrane surface. Conversely, if 1:2 DOPC/DPPC raft-like model membranes are used, cholesterol accelerates fiber growth. Next, it enhances pore formation and suppresses fiber growth on the membrane surface, leading to leakage. Our results highlight a twofold effect of cholesterol on the amyloidogenicity of IAPP and help explain its debated role in type 2 diabetes mellitus.
In this study, a novel multifunctional nanoplatform based on core-shell nanoparticles of spherical gold nanoparticles (AuNPs) capped with low and high molecular weight (200 and 700 kDa) hyaluronic ...acid (HA), was assembled via a green, one-pot redox synthesis method at room temperature. A multitechnique characterization approach by UV-visible spectroscopy, dynamic light scattering and atomic force microscopy pointed to the effective 'surface decoration' of the gold nanoparticles by HA, resulting in different grafting densities of the biopolymer chains at the surface of the metal nanoparticle, which in turn affected the physicochemical properties of the nanoparticles. Specifically, the spectral features of the gold plasmonic peak (and the related calculated optical size), the hydrodynamic diameter and the nanoparticle stability were found to depend on the molecular weight of the HA. The CD44-targeting capability of HA-functionalized gold nanoparticles was tested in terms of antibacterial activity and cytotoxicity. An enhanced inhibitory activity against both Gram-negative
and Gram-positive
was found, with a HA molecular weight (MW)-dependent trend for the HA-capped AuNPs compared to the bare, glucose-capped AuNPs. Cell viability assays performed on two CD44-positive cell models, namely normal human umbilical vein endothelial (HUVEC) and prostate tumor (PC-3) cells, in comparison with neuroblastoma cells (SH-SY5Y), which do not express the CD44 receptor, demonstrated an increased cytotoxicity in neuroblastoma compared to prostate cancer cells upon the cellular treatments by HA-AuNP compared to the bare AuNP, but a receptor-dependent perturbation effect on cytoskeleton actin and lysosomal organelles, as detected by confocal microscopy. These results highlighted the promising potentialities of the HA-decorated gold nanoparticles for selective cytotoxicity in cancer therapy. Confocal microscopy imaging of the two human tumor cell models demonstrated a membrane-confined uptake of HA-capped AuNP in the cancer cells that express CD44 receptors and the different perturbation effects related to molecular weight of HA wrapping the metallic core of the plasmonic nanoparticles on cellular organelles and membrane mobility.
Over the last few years, the study of the SARS-CoV-2 spike protein and its mutations has become essential in understanding how it interacts with human host receptors. Since the crystallized structure ...of the spike protein bound to the angiotensin-converting enzyme 2 (ACE2) receptor was released (PDB code 6M0J), in silico studies have been performed to understand the interactions between these two proteins. Specifically, in this study, heterocyclic compounds with different chemical characteristics were examined to highlight the possibility of interaction with the spike protein and the disruption of the interaction between ACE2 and the spike protein. Our results showed that these compounds interacted with the spike protein and interposed in the interaction zone with ACE2. Although further studies are needed, this work points to these heterocyclic push-pull compounds as possible agents capable of interacting with the spike protein, with the potential for the inhibition of spike protein-ACE2 binding.
Nanostructured noble metal thin films are highly studied for their interesting plasmonic properties. The latter can be effectively used for the detection of small and highly diluted molecules by the ...surface-enhanced Raman scattering (SERS) effect. Regardless of impressive detection limits achieved, synthesis complexity and the high cost of gold restrict its use in devices. Here, we report on a novel two-step approach that combines the deposition of a silver-aluminum thin film with dealloying to design and fabricate efficient SERS platforms. The magnetron sputtering technique was used for the deposition of the alloy thin film to be dealloyed. After dealloying, the resulting silver nanoporous structures revealed two degrees of porosity: macroporosity, associated to the initial alloy morphology, and nanoporosity, related to the dealloying step. The resulting nanoporous columnar structure was finely optimized by tuning deposition (i.e., the alloy chemical composition) and dealloying (i.e., dealloying media) parameters to reach the best SERS properties. These are reported for samples dealloyed in HCl and with 30 atom % of silver at the initial state with a detection limit down to 10
