Abstract
Background
Insecticide-based vector control is responsible for reducing malaria mortality and morbidity. Its success depends on a better knowledge of the vector, its distribution, and ...resistance status to the insecticides used. In this paper, we assessed Anopheles gambiae sensu lato (A gambiae s.l.) population resistance to pyrethroids in different ecological settings.
Methods
The World Health Organization standard bioassay test was used to assess F0A gambiae s.l. susceptibility to pyrethroids. Biochemical Synergist assays were conducted with piperonyl butoxide (PBO), S,S,S-tributyl phosphotritioate, and diethyl maleate. L1014F, L1014S, and N1575Y knockdown resistance (kdr) mutations were investigated using TaqMan genotyping.
Results
Anopheles gambiae sensu lato was composed of Anopheles arabienisis, Anopheles coluzzii, and A gambiae in all study sites. Anopheles gambiae sensu lato showed a strong phenotypic resistance to deltamethrin and permethrin in all sites (13% to 41% mortality). In many sites, pre-exposure to synergists partially improved the mortality rate suggesting the presence of detoxifying enzymes. The 3 kdr (L1014F, L1014S, and N1575Y) mutations were found, with a predominance of L1014F, in all species.
Conclusions
Multiple resistance mechanisms to pyrethroids were observed in A gambiae s.l. in Mali. The PBO provided a better partial restoration of susceptibility to pyrethroids, suggesting that the efficacy of long-lasting insecticidal nets may be improved with PBO.

The primary aims of this study are to ...determine the vector composition, biting rates, trophic preference, and the overall importance of distinguishing outdoor versus indoor malaria transmission through a study at two communities in rural Mali.
Mosquito collection was carried out between July 2012 and June 2016 at two rural Mali communities (Dangassa and Koïla Bamanan) using pyrethrum spray-catch and human landing catch approaches at both indoor and outdoor locations. Species of Anopheles gambiae complex were identified by polymerase chain reaction (PCR). Enzyme-Linked -Immuno-Sorbent Assay (ELISA) were used to determine the origin of mosquito blood meals and presence of Plasmodium falciparum sporozoite infections.
A total of 11,237 An. gambiae sensu lato (s.l.) were collected during the study period (5239 and 5998 from the Dangassa and Koïla Bamanan sites, respectively). Of the 679 identified by PCR in Dangassa, Anopheles coluzzii was the predominant species with 91.4% of the catch followed by An. gambiae (8.0%) and Anopheles arabiensis (0.6%). At the same time in Koïla Bamanan, of the 623 An. gambiae s.l., An. coluzzii accounted for 99% of the catch, An. arabiensis 0.8% and An. gambiae 0.2%. Human Blood Index (HBI) measures were significantly higher in Dangassa (79.4%; 95% Bayesian credible interval (BCI) 77.4, 81.4) than in Koïla Bamanan (15.9%; 95% BCI 14.7, 17.1). The human biting rates were higher during the second half of the night at both sites. In Dangassa, the sporozoite rate was comparable between outdoor and indoor mosquito collections. For outdoor collections, the sporozoite positive rate was 3.6% (95% BCI 2.1-4.3) and indoor collections were 3.1% (95% BCI 2.4-5.0). In Koïla Bamanan, the sporozoite rate was higher indoors at 4.3% (95% BCI 2.7-6.3) compared with outdoors at 2.4% (95% BCI 1.1-4.2). In Dangassa, corrected entomological inoculation rates (cEIRs) using HBI were 13.74 95% BCI 9.21-19.14 infective bites/person/month (ib/p/m) at indoor, and 18.66 95% BCI 12.55-25.81 ib/p/m at outdoor. For Koïla Bamanan, cEIRs were 1.57 95% BCI 2.34-2.72 ib/p/m and 0.94 95% BCI 0.43-1.64 ib/p/m for indoor and outdoor, respectively. EIRs were significantly higher at the Dangassa site than the Koïla Bamanan site.
The findings in this work may indicate the occurrence of active, outdoor residual malaria transmission is comparable to indoor transmission in some geographic settings. The high outdoor transmission patterns observed here highlight the need for additional strategies to combat outdoor malaria transmission to complement traditional indoor preventive approaches such as long-lasting insecticidal nets (LLINs) and indoor residual spraying (IRS) which typically focus on resting mosquitoes.
The long-lasting insecticidal nets (LLINs) and indoor residual spraying of insecticide (IRS) are major malaria vector control strategies in Mali. The success of control strategies depends on a better ...understanding of the status of malaria vectors with respect to the insecticides used. In this study we evaluate the level of resistance of Anopheles gambiae (sensu lato) to bendiocarb and the molecular mechanism that underlies it.
Larvae of An. gambiae (s.l.) were collected from breeding habitats encountered in the three study sites and bioassayed with bendiocarb. The ace-1 target site substitution G119S was genotyped using a TaqMan assay.
The three species of the An. gambiae complex in Mali, i.e. An. arabiensis, An. coluzzii and An. gambiae (s.s.) were found in sympatry in the three surveyed localities with different frequencies. We observed a resistance and suspicious resistance of the three species to bendiocarb with a mortality rate ranging from 37% to 86%. The allelic frequency of the G119S mutation was higher in An. gambiae (s.s.) compared to the other two species; 42.86%, 25.61% and 16.67% respectively in Dangassa, Koula, and Karadié. The allelic frequency of G119S in An. coluzzii ranged from 4.5% to 8.33% and from 1.43% to 21.15% for An. arabiensis. After exposure to bendiocarb, the G119S mutation was found only in survivors. The survival of Anopheles gambiae (s.l) populations from the three surveyed localities was associated with the presence of the mutation.
The study highlights the implication of G119S mutation in bendiocarb resistance in An. gambiae (s.s.), An. arabiensis and An. coluzzii populations from the three surveyed localities.
Introduction: Le paludisme demeure encore un problème majeur de santé public au Mali. Les mécanismes de transmission du paludisme en zones rurales sont bien connus mais restent encore tres mal connus ...en conditions urbaine. La présente étude a pour but de tester la susceptibilité d’adaptation d’An. coluzzii aux conditions écologiques de ponte et de développements larvaires des Culex et Aedes.
Méthodes : Trois répétitions ont été conduites en conditions de laboratoire dans un insectarium ayant 26-27ºC comme temperature et 80-95% d’humidité relative.
Résultats : Les taux d’éclosion étaient d’environ 90% pour le gîte témoins, 73% et 7% respectivement pour les gîtes Aedes et Culex avec un apport alimentaire. Mais sans apport de nourriture, ces taux sont de 22% pour les gîtes d’Aedes et 2% pour les gîtes de Culex. Les larves d’An. coluzzii étaient incapables de survivre dans les gîtes de Culex, avec ou sans apport alimentaire. Par contre dans les gîtes d’Aedes, leur taux de survie à varié de 53% au 1er jour à seulement 4% au 10e jour sans apport de nourriture. Mais avec un apport alimentaire, le taux de survie dans les gîtes d’Aedes a varié de 63% le 4e jour jusqu’à 21% le 10e jour. Les Mâles et les femelles d’Aedes adultes ont pu survivre jusqu’à 25 jours sans apport alimentaire. Par contre les adultes provenant des gîtes d’Aedes avec apport alimentaire peuvent survivre jusqu’à 39 jours.
Conclusion : Cette étude a montré qu’An. coluzzii n’a pas la capacité de survivre dans les gîtes d’eau de Culex, mais il est susceptible de se développer dans les gîtes d’eau d’Aedes.
Introduction: Malaria still remains a major public health problem in Mali. The mechanisms of malaria transmission in rural areas are well known but still remain very poorly understood in urban conditions. The current study aims to test the adaptive susceptibility of An. coluzzii to the ecological conditions of egg-laying and larval development of Culex and Aedes. Methods: Three replicates were conducted under laboratory conditions in an insectary with 26-27°C temperature and 80-95% relative humidity. Results: The hatching rates were about 90% for the control breeding site, 73% and 7% respectively for the Aedes and Culex breeding site with food supply. But without food, these rates are 22% for Aedes breeding site and 2% for Culex breeding site. The larvae of An. coluzzii were unable to survive in Culex breeding site, with or without food supply. On the other hand, in Aedes breeding sites, their survival rate varied from 53% on the 1st day to only 4% on the 10th day without food supply. But with a food supply, the survival rate in Aedes breeding site varied from 63% on the 4th day to 21% on the 10th day. Males and females of adult Aedes were able to survive up to 25 days without food supply. On the other hand, adults from Aedes breeding sites with food can survive up to 39 days. Conclusion: This study showed that An. coluzzii does not have the ability to survive in Culex breeding site, but it is likely to grow very well in Aedes breeding site.
Au Mali, la schistosomiase est un problème de santé publique comme dans tous les 42 paysafricains sur 76 concernés dans le monde, avec 230 millions de personnes infectées sur 800 millions de ...personnes exposées et plus de 800 000 décès annuels. L’objectif de cette étude était de tester l’efficacité d’une dose unique de Plaziquantel pour traiter les enfants d’âge scolaire de 11 villages maliens situés en zones endémiques de la schistosomiase urinaire. Après assentiment ou consentement, l’enregistrement des volontaires âgés de cinq ans ou plus a été fait. Après dépistage systématique de l’ensemble des volontaires pour savoir la prévalence de l’infection avant traitement, une dose unique de Praziquantel a été donnée aux sujets infectés. Un mois après la prise du médicament, un second dépistage a été fait chez les mêmes volontaires afin de mesurer l’effet du traitement. La technique de filtration de l'urine pour la détection de l'excrétion des œufs de schistosomiase a été utilisée comme méthode de diagnostic. Sur 549 volontaires testés à l’enregistrement (397 féminins et 152 masculins), 9,1% (51/549) étaient infectés par S. haematobium. Un mois après le traitement à la dose unique de Praziquantel, un taux de réduction significatif de 3,5% (P= 0,03) a été constaté. Cette étude a montré que le Praziquantel reste toujours efficace pour le traitement de la schistosomiase urinaire. Le maintien de ce produit comme molécule distribuée pour le traitement de masse du programme national de lutte contre la schistosomiase est justifié.
Mosquitoes are potentially harmful and vectors of pathogens. They compromise the rest and well-being of humans and animals. The main goal of this study is to determine the composition of mosquitoes ...responsible of human biting at the Faculty of Sciences and Technics of Bamako-Mali. Longitudinal monitoring with monthly cross-sectional visit was carried out from September to December 2019, in order to collect the endophilic and endophagic mosquitoes. The spray-catch was used as a collection methodin 21 rooms selected randomly at the FST. Mosquitoes were identified morphologically andthen by PCR. ELISA-CSP test was used for Plasmodium infection index and the ELISA blood-meal test was to determine mosquitoes blood origin. In total, 802 mosquitoes were collected: 794Culex and 8Anopheles. There were 200 males and 602 females. Female mosquitoes were separated by gonotrophic stages: 231 unfed, 223 fed, 80 semi-gravid and 68 gravid. Up to 34% of Culex and 67% of Anopheles had a preference for human blood, but no female tested positive for Plasmodium infection. This result would be due to the small number of Anopheles captured. An. coluzzii is the only Anopheles species collected.This study shows that mosquitoes are linked to serious problems of nuisance and risk of pathogens transmission in the university. They highly prefer to feed on human host.
Introduction: Les Arthropodes sont vecteurs de nombreuses maladies chez les hommes, les animaux et les plantes. Cest pourquoi lidentification des vecteurs devient necessaire afin de mener une lutte a ...travers des programmes nationaux de lutte. Methodes: Cette etude a ete conduite dans trois differentes zones eco-climatiques du Mali afin de voir la variation saisonniere des Arthropodes dinteret medical, veterinaire et Agricole au Mali. La collecte des arthropodes a ete faite a laide de deux types de piege (Piege demergence et piege en T) dans les villages de Bia, de Kenieroba et de Thierola. Les identifications ont ete faites a la loupe pour les Arthropodes, et par PCR pour les moustiques. Limportance des Arthropodes collectes a ete evaluee a travers leur dominance et leur frequence en fonction des zones et des saisons. Resultats: Letude des arthropodes des trois zones a permis davoir un apercu de lensemble de variations qualitatives et quantitatives des differents Arthropodes. Un total de 20 especes dArthropodes appartenant a trois classes a ete identifie (Arachnides, Insectes Myriapodes), 13 ordres (Araneides, Coleopteres, Dermapteres, Dictyopteres, Dipteres, Hemipteres, Hymenopteres, Lepidopteres, Nevropteres, Odonates, Orthopteres, Plecopteres, Trichopteres). Les Dipteres sont mieux representes a Bia et a Thierola et les Coleopteres frequents a Kenieroba. Conclusion: Il ressort de cette etude que les Arthropodes etaient plus frequents pendant la saison des pluies sur les sites. Les Arthropodes dinteret agricole constituaient le groupe dominant sur chacun des sites, suivi de ceux dinteret medical.
Les Aedes et les Culex sont des moustiques impliques dans la transmission de plusieurs agents pathogenes responsables des maladies telles que les arboviroses et des parasitoses. ...L’importance de la lutte biologique/genetique a ete demontre avec succes dans la lutte contre les glossines et les anopheles par certaines etudes. Le but de cette etude est de tester la possibilite d’utiliser les microbiotes comme source alimentaire principale des larves de moustiques des genres Culex et Aedes en vue d’utiliser ces microbiotes comme methode de lutte biologique/genetique contre la transmission des pathogenes. Quatre repetitions de gites de Culex et d’Aedes accompagnees de gites temoins ont ete utilises pour elever des larves de moustique, soumis a une alimentation a base des microbiotes naturelles cultivees au laboratoire a Bamako. Les microbiotes identifies etaient de bacteries et de protozoaires dans les cultures, avec une presence particuliere des algues dans les gites d’Aedes. Le taux d’emergence des imagos etait de 83% pour le temoin 63%, 61%, 84% et 84% respectivement pour les repetitions 1 a 4 dans les gites Culex. Dans les gites Aedes, le taux etait de 100% pour le temoin et les repetitions 3 et 4, (4%) et (15%) dans 1 et 2. La culture au laboratoire des microbiotes est donc possible. L’alimentation exclusive des larves de Culex avec les microbiotes s’est revelee possible, mais les larves d’Aedes ont besoin d’un apport alimentaire additif aux microbiotes pour leur bon developpement. L’option d’utilisation des microbiotes naturelles dans les perspectives de luttes biologiques/genetiques est donc possible.
