Silicon-substituted hydroxyapatite (SiHA) macroporous scaffolds have been prepared by robocasting. In order to optimize their bone regeneration properties, we have manufactured these scaffolds ...presenting different microstructures: nanocrystalline and crystalline. Moreover, their surfaces have been decorated with vascular endothelial growth factor (VEGF) to evaluate the potential coupling between vascularization and bone regeneration. In vitro cell culture tests evidence that nanocrystalline SiHA hinders pre-osteblast proliferation, whereas the presence of VEGF enhances the biological functions of both endothelial cells and pre-osteoblasts. The bone regeneration capability has been evaluated using an osteoporotic sheep model. In vivo observations strongly correlate with in vitro cell culture tests. Those scaffolds made of nanocrystalline SiHA were colonized by fibrous tissue, promoted inflammatory response and fostered osteoclast recruitment. These observations discard nanocystalline SiHA as a suitable material for bone regeneration purposes. On the contrary, those scaffolds made of crystalline SiHA and decorated with VEGF exhibited bone regeneration properties, with high ossification degree, thicker trabeculae and higher presence of osteoblasts and blood vessels. Considering these results, macroporous scaffolds made of SiHA and decorated with VEGF are suitable bone grafts for regeneration purposes, even in adverse pathological scenarios such as osteoporosis.
For the first time, the in vivo behavior of scaffolds made of silicon substituted hydroxyapatites (SiHA) has been evaluated under osteoporosis conditions. In order to optimize the bone regeneration properties of these bioceramics, 3D macroporous scaffolds have been manufactured by robocasting and implanted in osteoporotic sheep. Our experimental design shed light on the important issue of the biological response of nano-sized bioceramics vs highly crystalline bioceramics, as well as on the importance of coupling vascularization and bone growth processes by decorating SiHA scaffolds with vascular endothelial growth factor.

In this letter, a new strategy to enhance the discrimination of high energy gamma rays from the huge charged cosmic rays background in large cosmic rays ground arrays is presented. This strategy is ...based on the introduction of a new simple variable, Pγhα, which combines the probability of tagging muons and/or very energetic particles in each single array station. The discrimination power of this new variable, particularly important for and above multi-TeV energies, is illustrated for a few specific examples in the case of a hypothetical water Cherenkov detector cosmic ray array, both in the case of low and high particle stations occupancy. The results are very encouraging and hopefully will be demonstrated in the present and future gamma-ray Observatories.

The concept of a small, single-layer water Cherenkov detector, with three photomultiplier tubes (PMTs), placed at its bottom in a

Mercedes
Water Cherenkov Detector) is ...presented. The PMTs are placed near the lateral walls of the stations with an adjustable inclination and may be installed inside or outside the water volume. To illustrate the technical viability of this concept and obtain a first-order estimation of its cost, an engineering design was elaborated. The sensitivity of these stations to low energy Extensive Air Shower (EAS) electrons, photons and muons is discussed, both in compact and sparse array configurations. It is shown that the analysis of the intensity and time patterns of the PMT signals, using machine learning techniques, enables the tagging of muons, achieving an excellent gamma/hadron discrimination for TeV showers. This concept minimises the station production and maintenance costs, allowing for a highly flexible and fast installation. Mercedes Water Cherenkov Detectors (WCDs) are thus well-suited for use in high-altitude large gamma-ray observatories covering an extended energy range from the low energies, closing the gap between satellite and ground-based measurements, to very high energy regions, beyond the PeV scale.

The osteogenic and angiogenic responses to metal macroporous scaffolds coated with silicon substituted hydroxyapatite (SiHA) and decorated with vascular endothelial growth factor ...(VEGF) have been evaluated in vitro and in vivo. Ti6Al4V-ELI scaffolds were prepared by electron beam melting and subsequently coated with Ca10(PO4)5.6(SiO4)0.4(OH)1.6 following a dip coating method. In vitro studies demonstrated that SiHA stimulates the proliferation of MC3T3-E1 pre-osteoblastic cells, whereas the adsorption of VEGF stimulates the proliferation of EC2 mature endothelial cells. In vivo studies were carried out in an osteoporotic sheep model, evidencing that only the simultaneous presence of both components led to a significant increase of new tissue formation in osteoporotic bone.
Reconstruction of bones after severe trauma or tumors extirpation is one of the most challenging tasks in the field of orthopedic surgery. This scenario is even more complicated in the case of osteoporotic patients, since their bone regeneration capability is decreased. In this work we present a porous implant that promotes bone regeneration even in osteoporotic bone. By coating the implant with osteogenic bioceramics such as silicon substituted hydroxyapatite and subsequent adsorption of vascular endothelial growth factor, these implants stimulate the bone ingrowth when they are implanted in osteoporotic sheep.

ABSTRACT
The observation of very-high-energy γ-rays (VHE γ-rays, $E \gt 100\,$ GeV) has ledto the identification of extremely energetic processes and particle-acceleration sites both within our ...Galaxy and beyond. It is expected that VHE facilities, such as the Cherenkov Telescope Array, will explore these sources with an unprecedented level of detail. However, the transient and unpredictable nature of many important processes means that their observation requires the development of proper monitoring strategies. In this study, we estimate the properties of VHE transients that can be effectively detected by monitoring facilities. We use data collected by the Fermi-LAT instrument during its monitoring campaign to select events that are probably associated with VHE emission. We use this sample to estimate the frequency, the luminosity and the time-scales of various transients, focusing on blazar flares and gamma-ray bursts. We discuss how the balance between the field of view, sensitivity and duty cycle of an observatory affects the likelihood of detecting transients that occur at the inferred rates, and we conclude by describing the contribution that current and near-future monitoring facilities can make to the identification and study of VHE transient emission.
ABSTRACT
Gamma-ray Bursts (GRB) were discovered by satellite-based detectors as powerful sources of transient γ-ray emission. The Fermi satellite detected an increasing number of these events with ...its dedicated Gamma-ray Burst Monitor (GBM), some of which were associated with high energy photons ($E \gt 10$ GeV), by the Large Area Telescope (LAT). More recently, follow-up observations by Cherenkov telescopes detected very high energy emission ($E \gt 100$ GeV) from GRBs, opening up a new observational window with implications on the interpretation of their central engines and on the propagation of very energetic photons across the Universe. Here, we use the data published in the 2nd Fermi-LAT Gamma Ray Burst Catalogue to characterize the duration, luminosity, redshift, and light curve of the high energy GRB emission. We extrapolate these properties to the very high energy domain, comparing the results with available observations and with the potential of future instruments. We use observed and simulated GRB populations to estimate the chances of detection with wide-field ground-based γ-ray instruments. Our analysis aims to evaluate the opportunities of the Southern Wide-field-of-view Gamma-ray Observatory (SWGO), to be installed in the Southern Hemisphere, to complement CTA. We show that a low-energy observing threshold ($E_{low} \lt 200$ GeV), with good point source sensitivity ($F_{lim} \approx 10^{-11} \,\mathrm{erg\, cm^{-2}\, s^{-1}}$ in $1$ yr), are optimal requirements to work as a GRB trigger facility and to probe the burst spectral properties down to time-scales as short as $10$ s, accessing a time domain that will not be available to Imaging Atmospheric Cherenkov Telescopes instruments.
The incorporation of cobalt ions into the composition of bioactive glasses has emerged as a strategy of interest for bone regeneration purposes. In the present work, we have designed a set of ...bioactive mesoporous glasses SiO2-CaO-P2O5-CoO (Co-MBGs) with different amounts of cobalt. The physicochemical changes introduced by the Co2+ ion, the in vitro effects of Co-MBGs on preosteoblasts and endothelial cells and their in vivo behaviour using them as bone grafts in a sheep model were studied. The results show that Co2+ ions neither destroy mesoporous ordering nor inhibit in vitro bioactive behaviour, exerting a dual role as network former and modifier for CoO concentrations above 3 % mol. On the other hand, the activity of Co-MBGs on MC3T3-E1 preosteoblasts and HUVEC vascular endothelial cells is dependent on the concentration of CoO present in the glass. For low Co-MBGs concentrations (1mg/ml) cell viability is not affected, while the expression of osteogenic (ALP, RUNX2 and OC) and angiogenic (VEGF) genes is stimulated. For Co-MBGs concentration of 5 mg/ml, cell viability decreases as a function of the CoO content. In vivo studies show that the incorporation of Co2+ ions to the MBGs improves the bone regeneration activity of these materials, despite the deleterious effect that this ion has on bone-forming cells for any of the Co-MBG compositions studied. This contradictory effect is explained by the marked increase in angiogenesis that takes place inside the bone defect, leading to an angiogenesis-osteogenesis coupling that compensates for the partial decrease in osteoblast cells.
The development of new bone grafts implies to address the need for osteogenesis-angiogenesis coupling that allows bone regeneration with viable tissue in the long term. In this sense the incorporation of cobalt ions into the composition of bioactive glasses has emerged as a strategy of great interest in this field. Due to the potential cytotoxic effect of cobalt ions, there is an important controversy regarding the suitability of their incorporation in bone grafts. In this work, we address this controversy after the implantation of cobalt-doped mesoporous bioactive glasses in a sheep model. The incorporation of cobalt ions in bioactive glasses improves the bone regeneration ability of these bone grafts, due to enhancement of the angiogenesis-osteogenesis coupling.
