A general consensus exists that IBD is associated with compositional and metabolic changes in the intestinal microbiota (dysbiosis). However, a direct causal relationship between dysbiosis and IBD ...has not been definitively established in humans. Findings from animal models have revealed diverse and context-specific roles of the gut microbiota in health and disease, ranging from protective to pro-inflammatory actions. Moreover, evidence from these experimental models suggest that although gut bacteria often drive immune activation, chronic inflammation in turn shapes the gut microbiota and contributes to dysbiosis. The purpose of this Review is to summarize current associations between IBD and dysbiosis, describe the role of the gut microbiota in the context of specific animal models of colitis, and discuss the potential role of microbiota-focused interventions in the treatment of human IBD. Ultimately, more studies will be needed to define host-microbial relationships relevant to human disease and amenable to therapeutic interventions.
Optical spectroscopy study of the recent AG Pegasi (AG Peg) outburst observed during the second half of 2015 is presented. Considerable variations of the intensity and the shape of the spectral ...features as well as the changes of the hot component parameters, caused by the outburst, are discussed and certain similarities between the outburst of AG Peg and the outburst of a classical symbiotic stars are shown. It seems that after the end of the symbiotic nova phase, AG Peg became a member of the classical symbiotic stars group.
The mammalian intestine is colonized by beneficial commensal bacteria and is a site of infection by pathogens, including helminth parasites. Helminths induce potent immunomodulatory effects, but ...whether these effects are mediated by direct regulation of host immunity or indirectly through eliciting changes in the microbiota is unknown. We tested this in the context of virus-helminth coinfection. Helminth coinfection resulted in impaired antiviral immunity and was associated with changes in the microbiota and STAT6-dependent helminth-induced alternative activation of macrophages. Notably, helminth-induced impairment of antiviral immunity was evident in germ-free mice, but neutralization of Ym1, a chitinase-like molecule that is associated with alternatively activated macrophages, could partially restore antiviral immunity. These data indicate that helminth-induced immunomodulation occurs independently of changes in the microbiota but is dependent on Ym1.
In order for a virus to persist, there must be a balance between viral replication and immune clearance. It is commonly believed that adaptive immunity drives clearance of viral infections and, thus, ...dysfunction or viral evasion of adaptive immunity is required for a virus to persist. Type I interferons (IFNs) play pleiotropic roles in the antiviral response, including through innate control of viral replication. Murine norovirus (MNoV) replicates in dendritic cells (DCs) and type I IFN signaling in DCs is important for early control of MNoV replication. We show here that the non-persistent MNoV strain CW3 persists systemically when CD11c positive DCs are unable to respond to type I IFN. Persistence in this setting is associated with increased early viral titers, maintenance of DC numbers, increased expression of DC activation markers and an increase in CD8 T cell and antibody responses. Furthermore, CD8 T cell function is maintained during the persistent phase of infection and adaptive immune cells from persistently infected mice are functional when transferred to Rag1-/- recipients. Finally, increased early replication and persistence are also observed in mixed bone marrow chimeras where only half of the CD11c positive DCs are unable to respond to type I IFN. These findings demonstrate that increased early viral replication due to a cell-intrinsic innate immune deficiency is sufficient for persistence and a functional adaptive immune response is not sufficient for viral clearance.

Aims. We report on recent optical observations of the stellar and the nebular remnants of 22 southern post-novae. Methods. In this study, for each of our targets, we obtained and analysed long-slit ...spectra in the spectral range 3500–6600 Å and in Hα+N ii narrow-band images. Results. The changes in the emission lines’ equivalent widths with the time since the outburst agree with earlier published results of other authors. We estimated an average value α = 2.37 for the exponent of the power law fitted to the post-novae continua. Our observations clearly show the two-component structure of the V842 Cen expanding nebulae, owing to the different velocities of the ejected matter. We discovered an expanding shell around V382 Vel with an outer diameter of about 12″.
The erosive-atrophic form of oral lichen planus (OLP) is associated with severe pain and burning sensation and is often unresponsive to treatment. Topical corticosteroids are considered as a ...medication of first choice but they can produce adverse effects. Therefore, new therapeutic approaches are required.
The aim of this study was to investigate the effectiveness of biomodulation with diode laser in patients presenting with long-standing erosive-atrophic OLP.
Twelve patients, clinically and histologically diagnosed with OLP, participated in this study. The level of pain and the clinical scores of total 59 lesions were recorded before treatment using visual analog scale and Thongprasom sign scoring system respectively. All patients received low level laser therapy (LLLT) with diode laser (810 nm) with parameters (0.5 W, 30 s, 1.2 J/cm2) three times weekly for a month. The response rate was assessed according to the decrease in pain and sign scores. Treatment efficacy index was calculated.
There was a significant reduction in pain after LLLT (p<0.0001). Improvement in clinical signs was achieved in 59.3% of the lesions. At the end of the treatment 5.1% of the lesions exhibited score 5; 6.8% - score 4, 11.9% of the lesions were scored 3 and 8.5% and 30.5% showed score 2 and score 1, respectively. Complete resolution was revealed in 37.3% of the lesions. All patients experienced some degree of improvement. Most of the cases showed moderate recovery.
The present results indicate that LLLT is an effective and harmless modality for management of erosive-atrophic OLP.
The radiation risk radiometer‐dosimeter (R3D)‐R2 solid‐state detector performed radiation measurements at the European Space Agency EXPOSE‐R2 platform outside of the Russian “Zvezda” module at the ...International Space Station (ISS) from 24 October 2014 to 11 January 2016. The ISS orbital parameters were average altitude of 415 km and 51.6° inclination. We developed special software and used experimentally obtained formulas to determine the radiation flux‐to‐dose ratio from the R3DR2 Liulin‐type deposited‐energy spectrometer. We provide for the first time simultaneous, long‐term estimates of radiation dose external to the ISS for four source categories: (i) galactic cosmic ray particles and their secondary products; (ii) protons in the South Atlantic Anomaly region of the inner radiation belt (IRB); (iii) relativistic electrons and/or bremsstrahlung in the outer radiation belt (ORB); and (iv) solar energetic particle (SEP) events. The latter category is new in this study. Additionally, in this study, secondary particles (SP) resulting from energetic particle interaction with the detector and nearby materials are identified. These are observed continuously at high latitudes. The detected SPs are identified using the same sorting requirements as SEP protons. The IRB protons provide the highest consistent hourly dose, while the ORB electrons and SEPs provide the most extreme hourly doses. SEPs were observed 11 times during the study interval. The R3DR2 data support calculation of average equivalent doses. The 30 day and 1 year average equivalent doses are much smaller than the skin and eyes doses recommendations by the National Council on Radiation Protection (Report 132), which provides radiation protection guidance for Low Earth Orbit.

Context. M31N 2015-01a (or M31LRN 2015) is a red nova that erupted in January 2015 – the first event of this kind observed in M 31 since 1988. Very few similar events have been confirmed as of 2015. ...Most of them are considered to be products of stellar mergers. Aims. Results of an extensive optical monitoring of the transient in the period January–March 2015 are presented. Methods. Eight optical telescopes were used for imaging. Spectra were obtained on the Large Altazimuth Telescope (BTA), the Gran Telecsopio Canarias (GTC) and the Rozhen 2 m telescope. Results. We present a highly accurate 70 d light curve and astrometry with a 0.05′′uncertainty. The colour indices reached a minimum of 2−3 d before peak brightness and rapidly increased afterwards. The spectral type changed from F5I to F0I in 6 d before the maximum and then to K3I in the next 30 d. The luminosity of the transient was estimated to be 8.7+3.3-2.2 × 105 L⊙ during the optical maximum. Conclusions. Both the photometric and the spectroscopic results confirm that the object is a red nova, similar to V838 Monocerotis.
The intestinal epithelium is a key physical interface that integrates dietary and microbial signals to regulate nutrient uptake and mucosal immune cell function. The transcriptional programs that ...regulate intestinal epithelial cell (IEC) quiescence, proliferation, and differentiation have been well characterized. However, how gene expression networks critical for IECs are posttranscriptionally regulated during homeostasis or inflammatory disease remains poorly understood. Herein, we show that a conserved family of microRNAs, miR-181, is significantly downregulated in IECs from patients with inflammatory bowel disease and mice with chemical-induced colitis. Strikingly, we showed that miR-181 expression within IECs, but not the hematopoietic system, is required for protection against severe colonic inflammation in response to epithelial injury in mice. Mechanistically, we showed that miR-181 expression increases the proliferative capacity of IECs, likely through the regulation of Wnt signaling, independently of the gut microbiota composition. As epithelial reconstitution is crucial to restore intestinal homeostasis after injury, the miR-181 family represents a potential therapeutic target against severe intestinal inflammation.
