Several billion tonnes of mineral dust is emitted, and transported through winds every year from arid-semiarid areas. North African dust hot spots located in the Sahara are responsible for 50–70% of the global mineral dust budget. Dust-loaded air-masses originated from these sources can be transported over long distances and can also affect remote areas, such as North and South Americas, Europe, and the Middle East.
In this study, we analysed 218 identified Saharan dust events (SDEs) in the Carpathian Basin (Central Europe) during 1979 to 2018. Systematic identification of SDEs and analyses of dust emission, dust source area activity, dust transporting wind systems, and transport routes revealed that different synoptic meteorological patterns are responsible for SDEs, and these are occurring mostly in spring and summer. The characteristic synoptic meteorological background of episodes was also identified, and three major types of atmospheric pressure-system patterns were distinguished.
In recent years, several intense wintertime dust deposition events have been recorded in Central Europe. All of the identified unusual episodes were characterised by severe washout of mineral dust material and were related to very similar synoptic meteorological situations. Enhanced southward propagation of a high-latitude upper-level atmospheric trough to north-western Africa and orographic blocking of Atlas Mountains played an essential role in the formation of severe dust storms, whereas the long-range transport was associated with the northward branch of the meandering jet. The occurrence and southerly penetration of high-latitude upper-level atmospheric trough to low-latitudes and the increased meridionality of the dominant flow patterns may be associated with enhanced warming of the Arctic, leading to more meandering jet streams.

Mineral dust emissions from Saharan sources have an impact on the atmospheric environment and sedimentary units in distant regions. Here, we present the first systematic observations of long-range ...Saharan dust transport towards Iceland. Fifteen Saharan dust episodes were identified to have occurred between 2008 and 2020 based on aerosol optical depth data, backward trajectories and numerical models. Icelandic samples from the local dust sources were compared with deposited dust from two severe Saharan dust events in terms of their granulometric and mineralogical characteristics. The episodes were associated with enhanced meridional atmospheric flow patterns driven by unusual meandering jets. Strong winds were able to carry large Saharan quartz particles (> 100 µm) towards Iceland. Our results confirm the atmospheric pathways of Saharan dust towards the Arctic, and identify new northward meridional long-ranged transport of giant dust particles from the Sahara, including the first evidence of their deposition in Iceland as previously predicted by models.
Loess deposits are recorders of aeolian activity during past glaciations. Since the size distribution of loess deposits depends on distance to the dust source, and environmental conditions at the ...source, during transport, and at deposition, loess particle size distributions and derived statistical measures are widely used proxies in Quaternary paleoenvironmental studies. However, the interpretation of these proxies often only considers dust transport processes. To move beyond such overly simplistic proxy interpretations, and toward proxy interpretations that consider the range of environmental processes that determine loess particle size distribution variations we provide a comprehensive review on the physics of dust particle mobilization and deposition. Furthermore, using high-resolution bulk loess and quartz grain size datasets from a last glacial/interglacial sequence, we show that, because grain size distributions are affected by multiple, often stochastic processes, changes in these distributions over time allow multiple interpretations for the driving processes. Consequently, simplistic interpretations of proxy variations in terms of only one factor (e.g. wind speed) are likely to be inaccurate. Nonetheless using loess proxies to understand temporal changes in the dust cycle and environmental parameters requires (i) a careful site selection, to minimize the effects of topography and source distance, and (ii) the joint use of bulk and quartz grain size proxies, together with high resolution mass accumulation rate calculations if possible.
Monitoring of low-molecular weight cancer biomarkers, such as tryptophan (Trp) and its derivative kynurenine (Kyn), might be advantageous to non-invasive skin cancer detection. Thus, we assessed ...several approaches of topical sampling of Trp and Kyn, in relation to phenylalanine (Phe) and tyrosine (Tyr), on the volar forearm of six healthy volunteers. The sampling was performed with three hydrogels (made of agarose or/and chitosan), hydrated starch films, cotton swabs, and tape stripping. The biomarkers were successfully sampled by all approaches, but the amount of collected Kyn was low, 20 ± 10 pmol/cm2. Kyn quantification was below LOQ, and thus, it was detected only in 20% of topical samples. To mitigate variability problems of absolute amounts of sampled amino acids, Tyr/Trp, Phe/Trp, and Phe/Tyr ratios were assessed, proving reduced inter-individual variation from 79 to 45% and intra-individual variation from 42 to 21%. Strong positive correlation was found between Phe and Trp, pointing to the Phe/Trp ratio (being in the 1.0–2.0 range, at 95% confidence) being least dependent on sampling materials, approaches, and sweating. This study leads to conclusion that due to the difficulty in quantifying less abundant Kyn, and thus the Trp/Kyn ratio, the Phe/Trp ratio might be a possible, alternative biomarker for detecting skin cancers.
Well-characterized archival formalin-fixed paraffin-embedded (FFPE) tissues are of much value for prospective biomarker discovery studies, and protocols that offer high throughput and good ...reproducibility are essential in proteomics. Therefore, we implemented efficient paraffin removal and protein extraction from FFPE tissues followed by an optimized two-enzyme digestion using suspension trapping (S-Trap). The protocol was then combined with TMTpro 16plex labeling and applied to lung adenocarcinoma patient samples. In total, 9585 proteins were identified, and proteins related to the clinical outcome were detected. Because acetylation is known to play a major role in cancer development, a fast on-trap acetylation protocol was developed for studying endogenous lysine acetylation, which allows identification and localization of the lysine acetylation together with quantitative comparison between samples. We demonstrated that FFPE tissues are equivalent to frozen tissues to study the degree of acetylation between patients. In summary, we present a reproducible sample preparation workflow optimized for FFPE tissues that resolves known proteomic-related challenges. We demonstrate compatibility of the S-Trap with isobaric labeling and for the first time, we prove that it is feasible to study endogenous lysine acetylation stoichiometry in FFPE tissues, contributing to better utility of the existing global tissue archives. Studies on atmospheric dust and long-range transport of mineral dust have been a focus of atmospheric science in recent years. With its wide range of direct and indirect effects, mineral dust is one of the most uncertain elements in the mechanisms of climate change, and a deeper understanding of its role is essential for understanding future processes.
The aim of our research was to provide the first systematic data on the so far episodically documented northward transport mineral dust from arid-semiarid areas. So, in this paper, we present dust storm events from lower latitudes reaching the Finnish atmosphere, based on the MERRA-2 model Dust Column Mass Density data and after a multistep verification procedure using independent data source. In total, 86 long-range dust storm events were identified between 1980 and 2022, when air masses loaded with dust reached Finland. Based on backward-trajectories different sources were identified: 59 were Saharan, 22 were Aral-Caspian, and five were associated with Middle Eastern source areas. Considerable variation in inter-annual frequencies was observed among the source areas, which may be due to changes in circulation conditions and the effects of human activity (agriculture and land use changes in Aral Sea region).
There is a clear maximum of dust events in spring (60%), followed by summer and autumn (where 10 of the 11 autumn episodes were from the Sahara). However, the number and proportion of scarce winter events have more than doubled since 2010 compared to the preceding 30 years, but no autumn events were registered during this period. This clear temporal variation coincides with changes in dust transport observed in other regions of Europe, driven by greater atmospheric meridionality associated with climate change and driven by reduced temperature difference between low and high latitudes due to enhanced temperature increases at Arctic regions.
Although current LC-MS technology permits scientists to efficiently screen clinical samples in translational research, e.g., steroids, biogenic amines, and even plasma or serum proteomes, in a daily ...routine, maintaining the balance between throughput and analytical depth is still a limiting factor. A typical approach to enhance the proteome depth is employing offline two-dimensional (2D) fractionation techniques before reversed-phase nanoLC-MS/MS analysis (1D-nanoLC-MS). These additional sample preparation steps usually require extensive sample manipulation, which could result in sample alteration and sample loss. Here, we present and compare 1D-nanoLC-MS with an automated online-2D high-pH RP × low pH RP separation method for deep proteome profiling using a nanoLC system coupled to a high-resolution accurate-mass mass spectrometer. The proof-of-principle study permitted the identification of ca. 500 proteins with ∼10,000 peptides in 15 enzymatically digested crude serum samples collected from healthy donors in 3 laboratories across Europe. The developed method identified 60% more peptides in comparison with conventional 1D nanoLC-MS/MS analysis with ca. 4 times lower throughput while retaining the quantitative information. Serum sample preparation related changes were revealed by applying unsupervised classification techniques and, therefore, must be taken into account while planning multicentric biomarker discovery and validation studies. Overall, this novel method reduces sample complexity and boosts the number of peptide and protein identifications without the need for extra sample handling procedures for samples equivalent to less than 1 μL of blood, which expands the space for potential biomarker discovery by looking deeper into the composition of biofluids.
The generation of high quality plasma from whole blood is of major interest for many biomedical analyses and clinical diagnostic methods. However, it has proven to be a major challenge to make use of ...microfluidic separation devices to process fluids with high cell content, such as whole blood. Here, we report on an acoustophoresis based separation chip that prepares diagnostic plasma from whole blood linked to a clinical application. This acoustic separator has the capacity to sequentially remove enriched blood cells in multiple steps to yield high quality plasma of low cellular content. The generated plasma fulfills the standard requirements (<6.0 × 109 erythrocytes/L) recommended by the Council of Europe. Further, we successfully linked the plasmapheresis microchip to our previously developed porous silicon sandwich antibody microarray chip for prostate specific antigen (PSA) detection. PSA was detected by good linearity (R2 > 0.99) in the generated plasma via fluorescence readout without any signal amplification at clinically relevant levels (0.19−21.8 ng/mL).
Injectable bioelectronics could become an alternative or a complement to traditional drug treatments. To this end, a new self-doped p-type conducting PEDOT-S copolymer (A5) was synthesized. This ...copolymer formed highly water-dispersed nanoparticles and aggregated into a mixed ion–electron conducting hydrogel when injected into a tissue model. First, we synthetically repeated most of the published methods for PEDOT-S at the lab scale. Surprisingly, analysis using high-resolution matrix-assisted laser desorption ionization-mass spectroscopy showed that almost all the methods generated PEDOT-S derivatives with the same polymer lengths (i.e., oligomers, seven to eight monomers in average); thus, the polymer length cannot account for the differences in the conductivities reported earlier. The main difference, however, was that some methods generated an unintentional copolymer P(EDOT-S/EDOT-OH) that is more prone to aggregate and display higher conductivities in general than the PEDOT-S homopolymer. Based on this, we synthesized the PEDOT-S derivative A5, that displayed the highest film conductivity (33 S cm–1) among all PEDOT-S derivatives synthesized. Injecting A5 nanoparticles into the agarose gel cast with a physiological buffer generated a stable and highly conductive hydrogel (1–5 S cm–1), where no conductive structures were seen in agarose with the other PEDOT-S derivatives. Furthermore, the ion-treated A5 hydrogel remained stable and maintained initial conductivities for 7 months (the longest period tested) in pure water, and A5 mixed with Fe3O4 nanoparticles generated a magnetoconductive relay device in water. Thus, we have successfully synthesized a water-processable, syringe-injectable, and self-doped PEDOT-S polymer capable of forming a conductive hydrogel in tissue mimics, thereby paving a way for future applications within in vivo electronics.
