Errors in the simulation of clouds in general circulation models (GCMs) remain a long-standing issue in climate projections, as discussed in the Intergovernmental Panel on Climate Change (IPCC) ...Fourth Assessment Report. This highlights the need for developing new analysis techniques to improve our knowledge of the physical processes at the root of these errors. The Cloud Feedback Model Intercomparison Project (CFMIP) pursues this objective, and under that framework the CFMIP Observation Simulator Package (COSP) has been developed. COSP is a flexible software tool that enables the simulation of several satellite-borne active and passive sensor observations from model variables. The flexibility of COSP and a common interface for all sensors facilitates its use in any type of numerical model, from high-resolution cloud-resolving models to the coarser-resolution GCMs assessed by the IPCC, and the scales in between used in weather forecast and regional models. The diversity of model parameterization techniques makes the comparison between model and observations difficult, as some parameterized variables (e.g., cloud fraction) do not have the same meaning in all models. The approach followed in COSP permits models to be evaluated against observations and compared against each other in a more consistent manner. This permits a more detailed diagnosis of the physical processes that govern the behavior of clouds and precipitation in numerical models. The World Climate Research Programme (WCRP) Working Group on Coupled Modelling has recommended the use of COSP in a subset of climate experiments that will be assessed by the next IPCC report. In this article we describe COSP, present some results from its application to numerical models, and discuss future work that will expand its capabilities.
Global and local feedback analysis techniques have been applied to two ensembles of mixed layer equilibrium CO.sub.2 doubling climate change experiments, from the CFMIP (Cloud Feedback Model ...Intercomparison Project) and QUMP (Quantifying Uncertainty in Model Predictions) projects. Neither of these new ensembles shows evidence of a statistically significant change in the ensemble mean or variance in global mean climate sensitivity when compared with the results from the mixed layer models quoted in the Third Assessment Report of the IPCC. Global mean feedback analysis of these two ensembles confirms the large contribution made by inter-model differences in cloud feedbacks to those in climate sensitivity in earlier studies; net cloud feedbacks are responsible for 66% of the inter-model variance in the total feedback in the CFMIP ensemble and 85% in the QUMP ensemble. The ensemble mean global feedback components are all statistically indistinguishable between the two ensembles, except for the clear-sky shortwave feedback which is stronger in the CFMIP ensemble. While ensemble variances of the shortwave cloud feedback and both clear-sky feedback terms are larger in CFMIP, there is considerable overlap in the cloud feedback ranges; QUMP spans 80% or more of the CFMIP ranges in longwave and shortwave cloud feedback. We introduce a local cloud feedback classification system which distinguishes different types of cloud feedbacks on the basis of the relative strengths of their longwave and shortwave components, and interpret these in terms of responses of different cloud types diagnosed by the International Satellite Cloud Climatology Project simulator. In the CFMIP ensemble, areas where low-top cloud changes constitute the largest cloud response are responsible for 59% of the contribution from cloud feedback to the variance in the total feedback. A similar figure is found for the QUMP ensemble. Areas of positive low cloud feedback (associated with reductions in low level cloud amount) contribute most to this figure in the CFMIP ensemble, while areas of negative cloud feedback (associated with increases in low level cloud amount and optical thickness) contribute most in QUMP. Classes associated with high-top cloud feedbacks are responsible for 33 and 20% of the cloud feedback contribution in CFMIP and QUMP, respectively, while classes where no particular cloud type stands out are responsible for 8 and 21%.

elemental mapping. Sample preparation methods that accurately preserve the
in vivo
elemental distribution are essential if one wishes to translate the knowledge of elemental distributions measured
ex vivo
toward increased understanding of chemical and physiological pathways of brain disease. The choice of sample preparation is particularly important for metal ions that exist in a labile or mobile form, for which the
in vivo
distribution could be easily distorted by inappropriate sample preparation. One of the most widely studied brain structures, the hippocampus, contains a large pool of labile and mobile Zn. Herein, we describe how sucrose cryoprotection, the gold standard method of preparing tissues for immuno-histochemistry or immuno-fluorescence, which is also often used as a sample preparation method for elemental mapping studies, drastically alters hippocampal Zn distribution. Based on the results of this study, in combination with a comparison against the strong body of published literature that has used either rapid plunge freezing of brain tissue, or sucrose cryo-protection, we strongly urge investigators in the future to cease using sucrose cryoprotection as a method of sample preparation for elemental mapping, especially if Zn is an analyte of interest.

Approximately 12% of apparently previously cognitively well patients undergoing anaesthesia and noncardiac surgery will develop symptoms of cognitive dysfunction after their procedure. Recent ...articles in this Journal have highlighted the difficulties of confirming any clear links between anaesthesia and cognitive dysfunction, in part because of the lack of consistency regarding definition and diagnosis. Postoperative cognitive dysfunction (POCD) is usually self-limiting and rarely persists in the longer term, although plausible biological mechanisms for an impact on brain protein deposition do exist. Clinical research studies are frequently confounded by a lack of agreed definitions and consistency of testing. Preoperative assessment of neurocognitive function and risk factor identification is imperative in order to ascertain the true extent of POCD and any causative link to anaesthesia and surgery. At present a multidisciplinary care bundle approach to risk factor stratification and reduction is the most attractive management plan based on evidence of slight benefit from individual interventions. As yet no individual anaesthetic technique, drug or mode of monitoring has been proved to reduce the incidence of POCD. Providing patients with appropriate and accurate information can be difficult because of conflicting evidence. The Royal College of Anaesthetists’ patient liaison group has produced a useful patient information leaflet that is designed to provide guidance in discussions of individual risks whilst considerable uncertainties remain.
Longitudinal evidence on the association between air pollution and blood pressure (BP) in adolescence is scarce. We explored this association in an ethnically diverse cohort of schoolchildren. ...Sex-stratified, linear random-effects modelling was used to examine how modelled residential exposure to annual average nitrogen dioxide (NO2), particulate matter (PM2.5, PM10) and ozone (O3), measures in μg/m3, associated with blood pressure. Estimates were based on 3,284 adolescents; 80% from ethnic minority groups, recruited from 51 schools, and followed up from 11-13 to 14-16 years old. Ethnic minorities were exposed to higher modelled annual average concentrations of pollution at residential postcode level than their White UK peers. A two-pollutant model (NO2 & PM2.5), adjusted for ethnicity, age, anthropometry, and pubertal status, highlighted associations with systolic, but not diastolic BP. A μg/m3 increase in NO2 was associated with a 0.30 mmHg (95% CI 0.18 to 0.40) decrease in systolic BP for girls and 0.19 mmHg (95% CI 0.07 to 0.31) decrease in systolic BP for boys. In contrast, a 1 μg/m3 increase in PM2.5 was associated with 1.34 mmHg (95% CI 0.85 to 1.82) increase in systolic BP for girls and 0.57 mmHg (95% CI 0.04 to 1.03) increase in systolic BP for boys. Associations did not vary by ethnicity, body size or socio-economic advantage. Associations were robust to adjustments for noise levels and lung function at 11-13 years. In summary, higher ambient levels of NO2 were associated with lower and PM2.5 with higher systolic BP across adolescence, with stronger associations for girls.
Optical approaches for observing the dynamics of single molecules have required pico- to nanomolar concentrations of fluorophore in order to isolate individual molecules. However, many biologically ...relevant processes occur at micromolar ligand concentrations, necessitating a reduction in the conventional observation volume by three orders of magnitude. We show that arrays of zero-mode waveguides consisting of subwavelength holes in a metal film provide a simple and highly parallel means for studying single-molecule dynamics at micromolar concentrations with microsecond temporal resolution. We present observations of DNA polymerase activity as an example of the effectiveness of zero-mode waveguides for performing single-molecule experiments at high concentrations.
The first deep blank-field 450 μm map (1σ 1.3 mJy) from the Submillimetre Common-User Bolometer Array-2 SCUBA-2 Cosmology Legacy Survey (S2CLS), conducted with the James Clerk Maxwell Telescope ...(JCMT) is presented. Our map covers 140 arcmin2 of the Cosmological Evolution Survey field, in the footprint of the Hubble Space Telescope (HST) Cosmic Assembly Near-Infrared Deep Extragalactic Legacy Survey. Using 60 submillimetre galaxies detected at ≥3.75σ, we evaluate the number counts of 450-μm-selected galaxies with flux densities S
450 > 5 mJy. The 8 arcsec JCMT beam and high sensitivity of SCUBA-2 now make it possible to directly resolve a larger fraction of the cosmic infrared background (CIB, peaking at λ ∼ 200 μm) into the individual galaxies responsible for its emission than has previously been possible at this wavelength. At S
450 > 5 mJy, we resolve (7.4 ± 0.7) × 10−2 MJy sr−1 of the CIB at 450 μm (equivalent to 16 ± 7 per cent of the absolute brightness measured by the Cosmic Background Explorer at this wavelength) into point sources. A further ∼40 per cent of the CIB can be recovered through a statistical stack of 24 μm emitters in this field, indicating that the majority ( 60 per cent) of the CIB at 450 μm is emitted by galaxies with S
450 > 2 mJy. The average redshift of 450 μm emitters identified with an optical/near-infrared counterpart is estimated to be 〈z〉 = 1.3, implying that the galaxies in the sample are in the ultraluminous class (
). If the galaxies contributing to the statistical stack lie at similar redshifts, then the majority of the CIB at 450 μm is emitted by galaxies in the luminous infrared galaxy (LIRG) class with L
IR > 3.6 × 1011 L.
Periprosthetic infection (PI) causes significant morbidity and mortality after fixation and joint arthroplasty and has been extensively linked to the formation of bacterial biofilms. Poly(methyl ...methacrylate) (PMMA), as a cement or as beads, is commonly used for antibiotic release to the site of infection but displays variable elution kinetics and also represents a potential nidus for infection, therefore requiring surgical removal once antibiotics have eluted. Absorbable cements have shown improved elution of a wider range of antibiotics and, crucially, complete biodegradation, but limited data exist as to their antimicrobial and antibiofilm efficacy. Synthetic calcium sulfate beads loaded with tobramycin, vancomycin, or vancomycin-tobramycin dual treatment (in a 1:0.24 wt/wt ratio) were assessed for their abilities to eradicate planktonic methicillin-resistant Staphylococcus aureus (MRSA) and Staphylococcus epidermidis relative to that of PMMA beads. The ability of the calcium sulfate beads to prevent biofilm formation over multiple days and to eradicate preformed biofilms was studied using a combination of viable cell counts, confocal microscopy, and scanning electron microscopy of the bead surface. Biofilm bacteria displayed a greater tolerance to the antibiotics than their planktonic counterparts. Antibiotic-loaded beads were able to kill planktonic cultures of 10(6) CFU/ml, prevent bacterial colonization, and significantly reduce biofilm formation over multiple days. However, established biofilms were harder to eradicate. These data further demonstrate the difficulty in clearing established biofilms; therefore, early preventive measures are key to reducing the risk of PI. Synthetic calcium sulfate loaded with antibiotics has the potential to reduce or eliminate biofilm formation on adjacent periprosthetic tissue and prosthesis material and, thus, to reduce the rates of periprosthetic infection.

${>}10^{23}~\text{W}/\text{cm}^{2}$
). Technologies are being developed to upgrade the OMEGA EP Laser System with the goal to pump an optical parametric amplifier line (EP OPAL) with two of the OMEGA EP beamlines. The resulting ultraintense pulses (1.5 kJ, 20 fs,
$10^{24}~\text{W}/\text{cm}^{2}$
) would be used jointly with picosecond and nanosecond pulses produced by the other two beamlines. A midscale OPAL pumped by the Multi-Terawatt (MTW) laser is being constructed to produce 7.5-J, 15-fs pulses and demonstrate scalable technologies suitable for the upgrade. MTW OPAL will share a target area with the MTW laser (50 J, 1 to 100 ps), enabling several joint-shot configurations. We report on the status of the MTW OPAL system, and the technology development required for this class of all-OPCPA laser system for ultraintense pulses.
