Fluorescence lifetime imaging microscopy of a fluorescence probe, 3,6-bis(1-methyl-2-vinylpyridinium) carbazole diiodide (o-BMVC), provides an objective method for preoperative diagnosis of ...fine-needle aspiration (FNA) of thyroid nodules. The key of this o-BMVC test of FNA smears is the measurement of the digital number of o-BMVC foci in the nucleus. Thus, there are three categories classified in the o-BMVC test, which are nondiagnostic for unsatisfactory samples, benign for less numbers of o-BMVC foci, and malignant for more numbers of o-BMVC foci. The discrimination of indeterminate (including atypia, follicular neoplasm, suspicious) cytology into benign or malignant cases can reduce diagnostic uncertainty and benefit clinical decision making. This pilot study strongly suggests that the o-BMVC test is an invaluable method for diagnosing FNA samples. Particularly, the combination of FNA cytology and the o-BMVC test holds great promise to improve the efficacy of diagnosis and reduce the healthcare costs.
As typical glucose oxidase nanozymes, gold nanoparticles (Au NPs) have attracted much attention due to their wide-ranging applications. Ligand caps, as the "cure-all solution" for NPs, not only play ...important roles in the size and shape control of Au NPs but also influence their catalytic activity and selectivity. A deep understanding of the catalytic mechanism and precise description of the important role of ligands can provide possible ways to design functional Au NPs. Here, with the specific example of Au(111) capped with chiral phenylalanine (Phe), the chiral selective oxidation mechanism of glucose and the important role of the ligands were studied via first-principles calculations. All results show that the dehydrogenation of glucose to form glucono delta-lactone (GDL) is favored on clean Au(111), while the subsequent hydrolysis of GDL is the rate-limiting step for glucose oxidation. The flat and nonchiral Au(111) surface shows negligible selectivity in relation to the oxidation of d- and l-glucose, while chiral Phe-Au(111) shows selective adsorption towards d- and l-glucose. l-Phe-capped Au(111) prefers to adsorb d-glucose, while d-Phe-capped Au(111) prefers to adsorb l-glucose. Considering the three steps in the capped ligand catalysis (adsorption, replacement and reaction), we propose that the ligands play key roles in selectively adsorbing reactants before the subsequent exchange and reaction steps.
Genetic susceptibility is likely involved in nasopharyngeal carcinoma (NPC), a cancer caused by Epstein-Barr virus (EBV) infection. Understanding of genetic factors involved in NPC and how they ...contribute to EBV-induced carcinogenesis is limited. We conducted whole-exome capture/sequencing among 251 individuals from 97 multiplex families from Taiwan (205 affected, 21 obligate carriers, and 25 unaffected) using SeqCap EZ Human Exome Library v3.0 and Illumina HiSeq. Aligned sequences were filtered to identify likely-to-be-functional deleterious variants that co-segregated with disease. Ingenuity Pathway analysis was performed. Circulating magnesium levels were measured in 13 individuals in 2 families with NIPAL1 mutations and in 197 sporadic NPC cases and 237 controls. We identified variants in 12 genes likely involved in cancer pathogenesis, viral infection or immune responses to infection. These included genes postulated to be involved in magnesium transport (NIPAL1), EBV cell entry (ITGB6), modulation of EBV infection (BCL2L12, NEDD4L), telomere biology (CLPTM1L, BRD2, HNRNPU), modulation of cAMP signaling (RAPGEF3), DNA repair (PRKDC, MLH1), and Notch signaling (NOTCH1, DLL3). Pathway based analysis demonstrated enrichment for Notch signaling genes (p-value = 0.0006). Evaluation of individuals within NIPAL1 families suggested lower serum magnesium in NPC compared to unaffected members. A significant reduction in serum magnesium levels was observed among sporadic NPC cases compared to controls (7.1% NPC/1.7% controls below normal range; OR = 4.5; 95% CI = 1.4,14) and is consistent with findings demonstrating a role for magnesium channeling in T-cell responses to EBV. We identified novel genes associated with NPC that point to new areas of inquiry to better understand genetic factors that determine the fate of viral infections and/or otherwise predisposes to NPC.

Based on the engineering background in which the rock surrounding a wellbore is affected by a thermal shock, impact disturbances from drilling vibration, cyclic heat extraction and high temperature ...during hydrothermal geothermal energy mining, the environmental conditions in the shaft wall rock are simulated by means of high temperature, cooling, immersing granite in water with different curing temperatures and applying impact loads. Additionally, an experimental study on the mechanical characteristics of circular granite specimens under radial impact loads and in the heat treatment and water curing conditions is carried out. The results show that the inner diameters of the rings, heating temperatures, curing water temperatures and cycle heating times are less affected than other parameters by the impact load-strain curves of circular granite, which can generally be divided into three sections, i.e., the initial straight stage, nonlinear ascent yield stage and post-peak nonlinear decline stage. The factors in the test weaken the capacity of the circular granite to resist the impact, but the sizes of the inner diameters of the rings play a leading role. Dynamic tensile strain is generated in the inner wall along the impact direction during the impact, while compressive strain is produced on the inner wall in the vertical impact loading direction. By analysing the crack propagation and final failure mode of circular granite, it is found that dynamic tensile failures are generated, crack initiation starts from the inner wall along the impact loading direction, and the outer circle in the vertical direction lags behind. The crack starts early and develops quickly on one side of the transmission bar. Finally, the failure criterion is established on the basis of some assumptions and circular-granite deformation failure characteristics, and the parameters, measured by the Brazilian disk test, are reasonably verified via substitution into the failure criterion equation.
Background
Radiation‐induced sarcoma of the head and neck (RISHN) is a rare yet devastating potential complication of radiotherapy treatment. We aimed to evaluate the clinicopathological ...characteristics and molecular signatures of RISHN in patients who underwent radiotherapy for head and neck cancer (HNC) to identify high‐risk patients and enable earlier cancer detection.
Methods
This study retrospectively evaluated 24 sarcoma patients who received radiotherapy for HNC between 1994 and 2019. Patients were divided into two groups based on RISHN latency period. Patient demographics, initial tumor staging, risk factors, and survival between groups were analyzed, and whole‐exome sequencing (WES) of selected samples was performed.
Results
The median age at diagnosis of RISHN was 54 years, and the male‐to‐female ratio was 2:1. The latency period ranged from 0.8 to 64.4 years (median 6.5 years), with a median survival of 21.5 months. Primary cancer in the oral cavity, treatment with alkylating agents, alcohol consumption, betel nut chewing, and smoking were identified as risk factors for short (<5 years) latency periods. The majority of RISHN cases occurred in the oral cavity (58.3%). WES analysis showed that tumor necrosis factor and cell cycle checkpoint pathways were differentially involved in both patient groups.
Conclusions
Although case numbers were small, our cohort represents the largest case series of RISHN from a single institution to date. Clinicians must be aware of factors affecting RISHN development and latency, and risk factor identification may lead to earlier detection and prevention in the future.
Osteoarthritis (OA) is one of the most common diseases leading to physical disability, with age being the main risk factor, and degeneration of articular cartilage is the main focus for the ...pathogenesis of OA. Autophagy is a crucial intracellular homeostasis system recycling flawed macromolecules and cellular organelles to sustain the metabolism of cells. Growing evidences have revealed that autophagy is chondroprotective by regulating apoptosis and repairing the function of damaged chondrocytes. Then, OA is related to autophagy depending on different stages and models. In this review, we discuss the character of autophagy in OA and the process of the autophagy pathway, which can be modulated by some drugs, key molecules and non-coding RNAs (microRNAs, long non-coding RNAs and circular RNAs). More in-depth investigations of autophagy are needed to find therapeutic targets or diagnostic biomarkers through in vitro and in vivo situations, making autophagy a more effective way for OA treatment in the future. The aim of this review is to introduce the concept of autophagy and make readers realize its impact on OA. This study investigated the diagnostic accuracy and affecting factors of ultrasound (US)-guided core-needle biopsy (CNB) in patients with treated head and neck squamous cell carcinoma (HNSCC). We ...retrospectively reviewed patients with treated HNSCC who received US-guided CNB from January 2011 to December 2018 with corresponding imaging. Pathological necrosis and fibrosis of targeted lymph nodes (LNs) were evaluated. We analyzed the correlation between CNB accuracy and clinical and pathological characteristics. In total, 260 patients were included. The overall sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and accuracy of CNB for nodal recurrence were 84.47%, 100%, 100%, 54.67%, and 86.92%, respectively. CNB of fibrotic LNs had significantly worse sensitivity, NPV, and accuracy than that of non-fibrotic LNs. Similarly, CNB of necrotic LNs had significantly worse sensitivity, NPV, and accuracy than non-necrotic LNs. Multivariate regression revealed that fibrotic LN was the only independent factor for a true positive rate, whereas both necrotic LN and fibrotic LN were independent factors for a false negative rate. The diagnostic accuracy of CNB in treated HNSCC patients is affected by LN necrosis and fibrosis. Therefore, CNB results, particularly for necrotic or fibrotic LNs, should be interpreted carefully.
Computational thinking (CT), a way to think about solving problems, is recognized as a basic skill for the 21st century. Although game-based learning (GBL) has been shown to improve student's CT ...skills, students may fail to self-evaluate or debate as they play games, thus failing to develop higher-order thinking. Also not widely explored is how teachers are to practice the CT core competencies during the learning process to cultivate students' problem-solving thinking. As the adoption of student-generated questions (SGQ) is an instructional strategy to promote higher-order thinking, we develop a GBL platform that uses this strategy to enhance students' CT skills, learning motivation, and learning confidence. For this purpose, fifty-three primary school students were recruited for this study, and data were collected during four classes of experimental CT activities. This study shows that the experimental group using the SGQ strategy with a GBL platform exhibited significantly higher CT skills than the control group. In addition, with respect to learning motivation and learning confidence it is found that the experimental group also outperformed the control group. This confirms that integrating the SGQ strategy and CT core competencies fosters higher-order cognitive thinking in students in problem-solving through gamification of the learning environment, resulting in higher motivation and confidence to complete experimental activities.

As important pollinators, honey bees play a crucial role in both maintaining the ecological balance and providing products for humans. Although several versions of the western honey bee genome have ...already been published, its transcriptome information still needs to be refined. In this study, PacBio single-molecule sequencing technology was used to sequence the full-length transcriptome of mixed samples from many developmental time points and tissues of
queens, workers and drones. A total of 116,535 transcripts corresponding to 30,045 genes were obtained. Of these, 92,477 transcripts were annotated. Compared to the annotated genes and transcripts on the reference genome, 18,915 gene loci and 96,176 transcripts were newly identified. From these transcripts, 136,554 alternative splicing (AS) events, 23,376 alternative polyadenylation (APA) sites and 21,813 lncRNAs were detected. In addition, based on the full-length transcripts, we identified many differentially expressed transcripts (DETs) between queen, worker and drone. Our results provide a complete set of reference transcripts for
that dramatically expand our understanding of the complexity and diversity of the honey bee transcriptome.
