To evaluate the difference and efficacy of two donor liver procurement methods for treatment of pediatric acute liver failure (PALF) by living donor liver transplantation (LDLT).
A total of 17 ...patients (12 men, 5 women) with PALF who underwent LDLT in our hospital between October 2016 and October 2020, and prognostic efficacy of donors and recipients using two donor liver procurement methods were analyzed.
The donors and recipients were both divided into laparoscopic (7 cases) and open (10 cases) donor liver procurement groups. In the recipients, two deaths occurred in the laparoscopic group and one in the open group, and there were three postoperative complications in the laparoscopic group and six in the open group. The cumulative 1-year and 3-year survival rates in the laparoscopic group and the open group were 80.0% and 85.7% separately. There was no difference in the postoperative survival and complications rates between the two groups. In the donors, the operation time, postoperative hospital stay, and blood loss of the laparoscopic group was significantly reduced compared with the open group (
≤ 0.01). No death or serious complication occurred in either donor group.
Laparoscopic donor liver procurement is worth recommending than open donor liver procurement for treatment of PALF combined with LDLT in qualified transplant centers.
This paper investigates a gene expression model, which is mediated by sRNAs (small RNAs) and includes discrete and distributed delays. We take both the strong and weak kernel forms of distributed ...delay into consideration. The discrete time delay is chosen as the bifurcation parameter. By analyzing the distribution of characteristic values, we obtain the sufficient conditions of stability and examine the existence of periodic oscillations. When the discrete time delay is small and not greater than the threshold, the equilibrium of the gene expression model is asymptotically stable. When the bifurcation parameter exceeds the critical value, the model can produce limit cycles. Finally, numerical simulations are implemented to verify the correctness of our theoretical results.
Protein arginine methyltransferase (PRMT) enzymes catalyze posttranslational modifications of target proteins and are often upregulated in human cancers. In this study, we purified two chemical ...compounds from seeds of Foeniculum vulgare based on their ability to inhibit the enzymatic activity of PRMT5. These two compounds were identified as Pheophorbide a (PPBa) and Pheophorbide b (PPBb), two breakdown products of chlorophyll. PPBa and PPBb inhibited the enzymatic activity of both Type I and Type II PRMTs with IC

values at sub micromole concentrations, inhibited the arginine methylation of histones in cells, and suppressed proliferation of prostate cancer cells. Molecular docking results predicted that PPBa binds to an allosteric site in the PRMT5 structure with a high affinity (ΔG = -9.0 kcal/mol) via hydrogen bond, ionic, and π-π stacking interactions with amino acid residues in PRMT5. Another group of natural compounds referred to as protoporphyrins and sharing structural similarity with pheophorbide also inhibited the PRMT enzymatic activity. This study is the first report on the PRMT-inhibitory activity of the tetrapyrrole macrocycles and provides useful information regarding the application of these compounds as natural therapeutic reagents for cancer prevention and treatment.
Objective. To explore the effects of pramipexole combined with nerve growth factor (NGF) on cognitive impairment and urinary Alzheimer-associated neural thread protein (AD7c-NTP) expression in ...patients with Parkinson’s disease (PD). Methods. Fifty patients with PD treated in our hospital from February 2020 to April 2021 were enrolled. The patients were arbitrarily assigned into control group and study group. The former was treated with pramipexole, and the latter was treated with pramipexole combined with NGF. The efficacy, cognitive function, serum inflammatory factors, cortisol levels, serum macrophage migration inhibitory factor (MIF), brain-derived neurotrophic factor (BDNF), urine AD7c-NTP levels, and the incidence of adverse reactions were compared. Results. First of all, the effective rate in the study group was higher compared to the control group (P<0.05). After treatment, the cognitive function was enhanced, and the scores of Montreal cognitive assessment (MoCA) in the study group were higher compared to the control group (P<0.05). The levels of serum IL-6, CRP, and TNF-α decreased after treatment, and the levels of serum IL-6, CRP, and TNF-α in the study group were remarkably lower compared to the control group (P<0.05). In addition, the levels of serum DA, NE, and 5-HT increased after treatment, and the levels of serum DA, NE, and 5-HT in the study group were remarkably higher compared to the control group (P<0.05). Then, the levels of serum MIF and urine AD7c-NTP decreased and BDNF increased after treatment, and the level of BDNF in the study group was higher compared to the control group, while the levels of serum MIF and urine AD7c-NTP in the study group were lower compared to the control group (P<0.05). Finally, the adverse reactions were compared. The incidence of adverse reactions in the study group was lower compared to the control group, and the difference exhibited not statistically significant (16.00% vs. 24.00%, P>0.05). Conclusion.Pramipexole combined with NGF therapy not only can effectively strengthen the cognitive impairment of patients with PD and promote clinical efficacy and high safety but also can inhibit inflammatory state, regulate brain neurotransmitters, and reduce urinary AD7c-NTP levels.
The non-linear grey Bernoulli model NGBM(1,1) is a novel grey model for forecasting non-linear time series. In this study, the first order weighted accumulative generation operation (1-WAGO) and ...first order weighted inverse accumulative generation operation (1-WIAGO) are defined and introduced into NGBM(1,1) model to increase its prediction precision. An optimization model for solving unknown parameters is designed based on a Nash equilibrium. Two case studies with non-linear characteristics are analyzed to prove the feasibility and efficacy of the improved model. The results indicated that the new method further enhanced the adaptability of NGBM to non-linear raw data sequences, and notably increased the prediction precision. We previously demonstrated that the Groucho protein AES (amino-terminal enhancer of split) functions as a co-repressor of the AR (androgen receptor). It physically interacts with the N-terminal ...domain of AR and inhibits AR-driven transcription, but the molecular mechanism of its action remained unclear. In the present paper we report that the AES protein contains one inhibitory domain, and one positive and one negative regulatory domain. The negative regulatory domain inhibits AES dimerization and AES-mediated inhibition of AR-driven transcription through an interaction with the inhibitory domain. The positive regulatory domain blocked this interaction and relieved the inhibitory effect. In addition, we discovered mechanisms by which AES regulates AR transcriptional activity, which included disruption of the interaction between the AR N-terminal and C-terminal domains, and inhibition of AR-DNA interaction. Although AES broadly inhibited the activity of androgen-dependent luciferase reporters in a transient transfection assay, it selectively regulated the expression of endogenous androgen-dependent genes in prostate cancer cells.
It is an efficient way to deal with the time series prediction problem in grey perturbed systems by constructing effective grey operators. Due to the fixed structures of existing strengthening ...operators, the action intensity of these operators cannot be effectively controlled. This study aims to propose a kind of nonlinear strengthening operators with flexible structures to effectively control the action intensity of operators to raw data and obtain the optimum prediction accuracy. Based on the axiom system of grey operators, power average strengthening operator (PASO), geometric power average strengthening operator (GPASO), weighted power merage strengthening operator (WPASO), weighted geometric power average strengthening operator (WGPASO) in turn are constructed by introducing variable parameters into the construction of strengthening operators. Moreover, the properties of these operators, the relationship between different variable parameters and the action intensity of these operators are studied. Finally, with the prediction of China's marine electric power industry output value as an example, the effectiveness and superiority of these strengthening operators are verified. Abstract
The development and advancement of prostate cancer is supported by a plethora of genetic and proteomic abnormalities, including events of post-translational modifications. The protein ...arginine methyltransferase 5 (PRMT5) enzyme regulates epigenetic events of histone modifications and of post-translational modifications within protein signaling pathways. Under abnormal conditions of overexpression and/or upregulation, PRMT5 functions by constitutively driving the growth and/or proliferation of dysregulated cells. Overexpression and/or upregulation of PRMT5 correlates with disease progression as observed among numerous cancer types, including breast, colorectal, leukemia, lung, melanoma and prostate cancers. We demonstrated previously that PRMT5 localized to the cytoplasm, functions to drive both growth and proliferation of prostatic tumors. Plants naturally produce chemical toxins as mechanisms of defense against microbial and other biological threats. Human exploitation, consumption and/or application of agents isolated from plants for therapeutic intervention dates back throughout the millennia. In this study, we extracted, purified and evaluated natural, small, chemical compounds from plant products that antagonize PRMT5 activity in prostate cancer cells. We found that crude and purified extracts of Dendrobium, Fennel Seed and Magnolia plants attenuated prostate tumor growth and proliferation by selective inhibition of PRMT5 methyltransferase activity. These findings establish the first set of natural PRMT5-specific inhibitors reported.

This paper expatiates the stability and bifurcation for a fractional-order neural network (FONN) with double leakage delays. Firstly, the characteristic equation of the developed FONN is ...circumspectly researched by employing inequable delays as bifurcation parameters. Simultaneously the bifurcation criteria are correspondingly extrapolated. Then, unequal delays-spurred-bifurcation diagrams are primarily delineated to confirm the precision and correctness for the values of bifurcation points. Furthermore, it lavishly illustrates from the evidence that the stability performance of the proposed FONN can be demolished with the presence of leakage delays in accordance with comparative studies. Eventually, two numerical examples are exploited to underpin the feasibility of the developed theory. The results derived in this paper have perfected the retrievable outcomes on bifurcations of FONNs embodying unique leakage delay, which can nicely serve a benchmark deliberation and provide a comparatively credible guidance for the influence of multiple leakage delays on bifurcations of FONNs.
