Lymphocyte cytosolic protein 2 (LCP2) is one of the SLP-76 family of adapters, which are critical intermediates in signal cascades downstream of several receptors. LCP2 regulates immunoreceptor ...signaling (such as T-cell receptors) and is also required for integrin signaling in neutrophils and platelets. However, the role of LCP2 in the tumor microenvironment is still unknown. In this study, we found a significant increase of mRNA and protein expression of LCP2 in metastatic skin cutaneous melanoma compared to normal skin. The upregulation of LCP2 was associated with good overall survival of patients with metastatic skin cutaneous melanoma, who received pharmacotherapy and radiation. GSEA signaling pathways analysis showed that LCP2 was involved in multiple pathways of immune response and correlation analysis revealed LCP2 was positively correlated with molecules in TCR signaling and 11 immune checkpoints, while LCP2 negatively correlated with 2 immune checkpoints in the metastatic skin cutaneous melanoma. According to the different expressions of LCP2, high LCP2 expression was positively correlated with more tumor-infiltrating CD8
T cells. Furthermore, Kaplan-Meier plot indicated that LCP2 acted as a prognostic biomarker for progression-free survival of patients with metastatic skin cutaneous melanoma receiving anti-PD1 immunotherapy. In conclusion, our results integrated both the expression and function of LCP2 in melanoma using multiple tools, shedding light on the potential role of LCP2 in melanoma, and suggesting LCP2 serves as a prognostic biomarker and therapeutic target in anti-tumor immunity.
Perovskite-type oxide (PTO) of LaFeO3 supported Ni–Co bimetallic catalysts were prepared by citric acid complexation-impregnation method and were used for the steam reforming of ethanol (SRE) to ...produce hydrogen. The anti-sintering and anti-coking properties of the catalysts for the reaction have been investigated and compared with the monometal catalysts. The catalysts were characterized by using temperature programmed reduction, X-ray diffraction, transmission electron microscopy and thermal analysis techniques. The results indicate that the catalyst was both highly selective to hydrogen and very stable for SRE reaction. Characterization results indicated that Ni–Co was in the state of solid solution alloy. Comparing with corresponding monometal catalysts, the bimetal catalyst exhibited better anti-sintering ability and similar anti-carbon deposition ability. The valuable information indicated in this work is that based on the special characters of PTO, bimetal nano-particles can be supported on metal oxides, which should be a new and promising method for preparing supported bimetal catalysts.

Shanghai composite index reflects the changes of stock prices, and the methods for various models to predict the stock index emerge one after another, and artificial intelligence is also widely used ...in various fields due to its stability and accuracy. In this paper, artificial intelligence is applied to Shanghai composite index to predict the stock index. A total of 3422 Shanghai composite indexes from January 1, 2005 to January 1, 2019 were collected, including five indexes: opening price, maximum price, closing price, minimum price and trading volume. Then MA, KDJ and MACD were selected as technical indexes, and their application methods and advantages in Shanghai composite index were analyzed in detail. In addition, in this paper, logistic regression and support vector machine (SVM) in artificial intelligence model were adopted to predict the ups and downs. Finally, it indicates that the support vector basis method based on radial basis is more suitable for stock index prediction model. In this paper, a framework of index prediction is provided by combining technical indicators with artificial intelligence.

. However, neutralization by 14 of the 17 most-potent monoclonal antibodies that we tested was reduced or abolished by the K417N, E484K or N501Y mutation. Notably, these mutations were selected when we cultured recombinant vesicular stomatitis virus expressing SARS-CoV-2 S in the presence of the monoclonal antibodies elicited by the vaccines. Together, these results suggest that the monoclonal antibodies in clinical use should be tested against newly arising variants, and that mRNA vaccines may need to be updated periodically to avoid a potential loss of clinical efficacy.
Luminescent solar concentrators (LSCs) have attracted extensive interest due to their potential application in building integrated photovoltaics. As for the emitters, organic dyes and semiconductor ...quantum dots are generally used for LSC fabrication. However, carbon-dots as a new class of quantum dots used in LSC study are rare. Here, N-doped carbon-dots (N-CDs) are used as phosphors to be embedded in polyvinylpyrrolidone (PVP) thin films for LSC fabrication because of the good compatibility between them. The optical properties and performance of N-CDs/PVP thin film based LSCs with different layer numbers are investigated. The high performance of N-CDs/PVP thin film based LSCs with a power conversion efficiency of 4.97% has demonstrated that N-CDs embedded in PVP thin films is an effective approach for LSC fabrication which makes the industrial applications of LSCs more feasible.

The emergence of SARS-CoV-2 and the ensuing explosive epidemic of COVID-19 disease has generated a need for assays to rapidly and conveniently measure the antiviral activity of SARS-CoV-2-specific ...antibodies. Here, we describe a collection of approaches based on SARS-CoV-2 spike-pseudotyped, single-cycle, replication-defective human immunodeficiency virus type-1 (HIV-1), and vesicular stomatitis virus (VSV), as well as a replication-competent VSV/SARS-CoV-2 chimeric virus. While each surrogate virus exhibited subtle differences in the sensitivity with which neutralizing activity was detected, the neutralizing activity of both convalescent plasma and human monoclonal antibodies measured using each virus correlated quantitatively with neutralizing activity measured using an authentic SARS-CoV-2 neutralization assay. The assays described herein are adaptable to high throughput and are useful tools in the evaluation of serologic immunity conferred by vaccination or prior SARS-CoV-2 infection, as well as the potency of convalescent plasma or human monoclonal antibodies.
Immune imbalance of T lymphocyte subsets is a hallmark of psoriasis, but the molecular mechanisms underlying this aspect of psoriasis pathology are poorly understood. Here, we report that ...microRNA-210 (miR-210), a miR that is highly expressed in both psoriasis patients and mouse models, induces helper T (Th) 17 and Th1 cell differentiation but inhibits Th2 differentiation through repressing STAT6 and LYN expression, contributing to several aspects of the immune imbalance in psoriasis. Both miR-210 ablation in mice and inhibition of miR-210 by intradermal injection of antagomir-210 blocked the immune imbalance and the development of psoriasis-like inflammation in an imiquimod-induced or IL-23-induced psoriasis-like mouse model. We further showed that TGF-β and IL-23 enhance miR-210 expression by inducing HIF-1α, which recruits P300 and promotes histone H3 acetylation in the miR-210 promoter region. Our results reveal a crucial role for miR-210 in the immune imbalance of T lymphocyte subsets in psoriasis and suggest a potential therapeutic avenue.

In the context of global urbanization, more and more people are attracted to these cities with superior geographical conditions and strategic positions, resulting in the emergence of world super ...cities. However, with the increasing of urban development, the underlying surface of the city has changed, the soil originally covered with vegetation has been substituted by hardened pavement such as asphalt and cement roads. Therefore, the infiltration capacity of urban rainwater is greatly limited, and waterlogging is becoming more and more serious. In addition, the suburbs of the main urban areas of super cities are usually villages and mountains, and frequent flash floods seriously threaten the life and property safety of people in there. Flood sensitivity assessment is an effective method to predict and mitigate flood disasters. Accordingly, this study aimed at identifying the areas vulnerable to flood by using Geographic Information System (GIS) and Remote Sensing (RS) and apply Logistic Regression (LR) model to create a flood sensitivity map of Beijing. 260 flood points in history and 12 predictors elevation, slope, aspect, distance to rivers, Topographic Wetness Index (TWI), Stream Power Index (SPI), Sediment Transport Index (STI), curvature, plan curvature, Land Use/Land Cover (LULC), soil, and rainfall were used in this study. Even more noteworthy is that most of the previous studies discussed flash flood and waterlogging separately. However, flash flood points and waterlogging points were included together in this study. We evaluated the sensitivity of flash flood and waterlogging as a whole and obtained different results from previous studies. In addition, most of the previous studies focused on a certain river basin or small towns as the study area. Beijing is the world's ninth largest super cities, which was unusual in previous studies and has important reference significance for the flood sensitivity analysis of other super cities. The flood inventory data were randomly subdivided into training (70%) and test (30%) sets for model construction and testing using the Area Under Curve (AUC), respectively. The results turn out that: (1) elevation, slope, rainfall, LULC, soil and TWI were highly important among these elements, and were the most influential variables in the assessment of flood sensitivity. (2) The AUC of the test dataset revealed a prediction rate of 81.0%. The AUC was greater than 0.8, indicating that the model assessment accuracy was high. (3) The proportion of high risk and extremely high risk areas was 27.44%, including 69.26% of the flood events in this study, indicating that the flood distribution in these areas was relatively dense and the susceptibility was high. Super cities have a high population density, and once flood disasters occur, the losses brought by them are immeasurable. Thus, flood sensitivity map can provide meaningful information for policy makers to enact appropriate policies to reduce future damage.
