Background and aimsAcute respiratory tract infections are a major cause of morbidity and mortality worldwide, particularly in infants and young children. High-throughput, accurate, broad etiologic ...diagnosis tools were critical for effective epidemic control. In this study, the diagnostic capacities of an Ibis platform based on PCR/ESI-MS assay were evaluated on clinical samples.MethodsA total of 120 NPAs were collected from 120 children ( < 5 years old) hospitalised with lower respiratory infections between November 2010 and October 2011. The respiratory virus detection assay was performed with PCR/ESI-MS assay and DFA assay respectively. The all discordant PCR/ESI-MS and DFA results were resolved with RT-PCR plus sequencing.ResultsThe overall agreement for PCR/ESI-MS and DFA was 98.3% (118/120). Compared to the results from DFA, the sensitivity and specificity of the PCR/ESI-MS assay were 100% and 97.5%, respectively. The PCR/ESI-MS assay also detected more multi-viruses infection and demonstrated more detailed types information than DFA. Among the 12 original specimens with disagreement results between PCR/ESI-MS and DFA, 9 have confirmed PCR/ESI-MS results.ConclusionThis assay is a high-throughput, sensitive, specific and promising method to detect and subtype the conventional viruses for respiratory tract infections, and allowed rapid identification of mixed pathogens.

Quantitative real-time PCR (qRT-PCR) was employed to identify the expression level of miR-613. CCK8 and colony formation assays were incorporated to assess cell viability and proliferation capacity. Cell migration and invasion assays were performed to investigate the metastasis capacity of Wilms' tumor cells. Flow cytometry was used to detect cell cycle distribution and cell apoptosis. Protein levels were assessed by western blotting assay. The target gene was predicted and verified by bioinformatics analysis and luciferase assay.
The expression of miR-613 was downregulated in Wilms' tumor tissues compared with adjacent normal tissues (n=32). Overexpression of miR-613 could attenuate Wilms' tumor cell viability, proliferation, invasion, and migration capacity, as well as induce cell cycle arrest at the G0/G1 phase. FRS2 was chosen as the target of miR-613 by bioinformatics analysis and a luciferase reporter assay. MiR-613 expression was inversely correlated with FRS2 in Wilms' tumor tissues. Moreover, restoration of FRS2 rescued the tumor suppressive role of miR-613 in Wilms' tumor cell growth and metastasis.
MiR-613 had a tumor-suppressive effect on Wilms' tumor progression and metastasis via targeting FRS2 in vitro, which provided an innovative and candidate target for the diagnosis and treatment of Wilms' tumor.
The fish-borne clonorchiasis caused by the oriental liver fluke Clonorchis sinensis is endemic in a number of countries with over 35 million people being infected globally. Rapid and accurate ...detection of C. sinensis in its intermediate host fish is important for the control and prevention of clonorchiasis in areas where the disease is endemic. In the present study, we established a loop-mediated isothermal amplification (LAMP) approach for the sensitive and rapid detection of C. sinensis metacercariae in fish. The specificity and sensitivity of primers designed from the C. sinensis cathepsins B3 gene were evaluated, and specific amplification products were obtained with C. sinensis, while no amplification products were detected with DNA of related trematodes, demonstrating the specificity of the assay. The LAMP assay was proved to be 100 times more sensitive than a conventional polymerase chain reaction for detection of C. sinensis. The established LAMP assay provides a useful tool for the rapid and sensitive detection of C. sinensis in fish, which has important implications for the effective control of human clonorchiasis.

The study prepares glass fiber reinforced composites through Vacuum Assisted Resin Infusion (VARI) process, selects glass fibers of different specifications, and studies the effects of ...lay-up mode, thickness, simulated normal incident angle, and temperature of fiber composites s on their ballistic performance. Results show that with slimmer fiber monofilament, reinforced materials perform better in ballistics; the lay-up mode has limited influence on ballistic performance; the relationship between thickness and ballistic performance is positively linear; and the relationship between normal incident angle and ballistic performance is parabolic. Compared with room temperature conditions, the ballistic performance of composites does not decrease and even increases slightly under low temperatures. Yet, the performance decreases sharply at high temperatures.
Abstract
The effect of bubbles and void defects on the properties of structural armor composite was investigated, and the properties of the composites repaired following the secondary infusion scheme ...were characterized. The results showed that bubbles could significantly decrease the mechanical properties of composites, the tensile strength and bending strength of the composites with bubbles were decreased by about 11% and 5% respectively, and the bending modulus of elasticity was 9% less. The ballistic performance of the composites with void defects was decreased. For V50, it was decreased by about 6%, and the specific energy absorption was reduced by about 9%. The void defect has little effect on the static mechanical properties of the composites, but will significantly reduce the anti-aging ability of the composites in hygrothermal environment. The mechanical properties and ballistic performance of the composites do not decrease significantly after the secondary infusion in the dry spot area.
Abstract
A hybrid composite reinforced by aramid, glass and carbon fibers and processed with integrated vacuum infusion is studied in this paper. Reasons why the composite suffers from warpage after ...post-curing are analyzed. Such deformation is restrained by adjusting the thickness ratio of the fibers. Results indicate that with a constant areal density, the minimum deformation is 0.28 when the thickness ratio of the surface layer to the back layer is 1.46.
The stochastic averaging is applied to derive the random responses of the SDOF vibro-impact system with one-side wall under additive and multiplicative broadband noise, where the impact is described ...by the modified Hertzian contact model. Compared with the popular simplified “classical model” (i.e., x˙−=rx˙+), the whole impact process is captured by the modified Hertzian contact model without any assumptions. One example is given to illustrate the proposed technique. The comparison between the analytical results and those from Monte Carlo simulations manifests the accuracy of the proposed technique. It should be pointed out that the technique proposed in this paper is powerful for the cases of weak excitation and big enough bandwidth of the broadband noise. Finally, the influences of various parameters are also discussed in detail.
