The cellular origin of gastric cancer remains elusive. Leucine-rich repeat-containing G-protein-coupled receptor 5 (Lgr5) is the first identified marker of gastric stem cells. However, the role of ...Lgr5+ stem cells in driving malignant gastric cancer is not fully validated. Here, we deleted Smad4 and PTEN in murine gastric Lgr5+ stem cells by the inducible Cre-LoxP system and marked mutant Lgr5+ stem cells and their progeny with Cre-reporter Rosa26tdTomato. Rapid onset and progression from microadenoma and macroscopic adenoma to invasive intestinal-type gastric can- cer (IGC) were found in the gastric antrum with the loss of Smad4 and PTEN. In addition, invasive IGC developed at the murine gastro-forestomach junction, where a few Lgr5+ stem cells reside. In contrast, Smad4 and PTEN deletions in differentiated cells, including antral parietal cells, pit cells and corpus Lgr5+ chief cells, failed to initiate tumor growth. Furthermore, mutant Lgr5+ cells were involved in IGC growth and progression. In the TCGA (The Cancer Genome Atlas) database, an increase in LGR5 expression was manifested in the human IGC that occurred at the gastric antrum and gastro-esophageal junction. In addition, the concurrent deletion of SMAD4 and PTEN, as well as their reduced expression and deregulated downstream pathways, were associated with human IGC. Thus, we demon- strated that gastric Lgr5+ stem cells were cancer-initiating cells and might act as cancer-propagating cells to contrib- ute to malignant progression.
Previous studies have shown that Dickkopf-3 (Dkk3) is inactivated in lung cancer cells, while the inactivation of the Wnt/beta-catenin signaling pathway by Dkk3 inhibits lung cancer progression. In ...the present study, we investigated whether Dkk3 enhances the sensitivity of lung cancer cells to cisplatin. A549, Calu1 and H460 lung adenocarcinoma cell lines were transfected with DKK3 siRNA, while the cisplatin-resistant subline A549cis was transfected with DKK3. DKK3 expression was attenuated in A549cis, Calu1cis and H460cis compared to A549, Calu1 and H460, respectively. Lung adenocarcinoma cell growth, proliferation, apoptosis, cell cycle in vitro and in vivo were then analyzed. DKK3 knockdown by siRNA transfection rendered A549, Calu1 and H460 resistant to cisplatin. As a result of DKK3 transfection, the expression of DKK3 and E-cadherin was significantly upregulated, while that of MMP7, survivin, c-myc and cyclin D1 was downregulated. DKK3 overexpression retarded cell proliferation, induced cell cycle arrest and apoptosis, and reduced cell invasive ability in the A549 and A549cis cells. In addition, the proportions of apoptotic cells and the PARP level were significantly increased in A549cis- and H460cis-DKK3 cells treated with cisplatin. Moreover, tumor growth was retarded more in cisplatin-treated nude mice seeded with A549cis-DKK3 cells than with A549cis cells. Cell viability increased with the pretreatment of SB216763 for 2 h in A549cis and A549cis-DKK3 cells incubated with cisplatin (1 microM) for 72 h. In conclusion, the re-activation of Dkk3 enhances the chemosensitivity to cisplatin in cisplatin-resistant lung adenocarcinoma cell lines, which requires additional studies to realize this potential in clinical use. The objective of this study was to determine the efficiency of different plant systems in capturing deep soil nitrate (NO3-) to reduce NO3- leaching in a field plot experiment using 15N labelling. ...The study was conducted on a calcareous alluvial soil on the North China Plains and the plant systems evaluated included alfalfa (Medicago sativa), American black poplar (Populus nigra) and cocksfoot (Dactylis). ^15N-labelled N fertilizer was injected to 90 cm depth to determine the recovery of ^15N by the plants. With conventional water and nutrient management, the total recovery of ^15N-labeled NO3--N was 23.4% by alfalfa after two consecutive growth years. The recovery was significantly higher than those by American black poplar (12.3%) and cocksfoot (11.4%). The highest proportion of soil residual ~SN from the labeled fertilizer N (%Ndff) was detected around 90 cm soil depth at the time of the 1st year harvest and at 110-130 cm soil depth at time of the 2nd year harvest. Soil %Ndff in 0-80 cm depth was significantly higher in the alfalfa treatment than those in all the other treatments. The soil %Ndff below 100 cm depth was much lower in the alfalfa than those in all the other treatments. These results indicated that ^15N leaching losses in the alfalfa treatment were significantly lower than by those in the black poplar and cocksfoot treatments, due to the higher root density located in nitrate labeling zone of soil profile. In conclusion, alfalfa may be used as a plant to capture deep soil NO3- left from previous crops to reduce NO3- leaching in high intensity crop cultivation systems of North China Plain.
Multifunctional nanocarriers with multilayer core-shell architecture were prepared by coating superparamagnetic Fe3O4 nanoparticles with diblock copolymer folate-poly(ethylene glycol)-b-poly(glycerol ...monomethacrylate) (FA-PEG-b- PGMA), and triblock copolymer methoxy poly(ethylene glycol)-b-poly(2-(dimethylamino) ethyl methacrylate)-b- poly(glycerol monomethacrylate) (MPEG-b-PDMA-b-PGMA). The PGMA segment was attached to the surfaces of Fe304 nanoparticles, and the outer PEG shell imparted biocompatibility. In addition, folate was conjugated onto the surfaces of the nanocarriers. Cisplatin was then loaded into the nanocarrier by coordination between the Pt atom in cisplatin and the amine groups in the inner shell of the multilayer architecture. The loaded cisplatin showed pH-responsive release: slower release at pH 7.4 (i.e. mimicking the blood environment) and faster release at more acidic pH (i.e. mimicking endosome/lysosome conditions). All of the cisplatin-loaded nanoparticles showed concentration-dependent cytotoxicity in HeLa cells. However, the folate-conjugated cisplatin-loaded carriers exhibited higher cytotoxicity in HeLa cells than non-folate conjugated cisplatin-loaded carriers.
The novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the cause of a rapidly spreading illness,coronavirus disease 2019 (COVID-19),affecting more than seventeen million people ...around the world.Diagnosis and treatment guidelines for clinicians caring for patients are needed.In the early stage,we have issued "A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus (2019-nCoV) infected pneumonia (standard version)";now there are many direct evidences emerged and may change some of previous recommendations and it is ripe for develop an evidence-based guideline.We formed a working group of clinical experts and methodologists.The steering group members proposed 29 questions that are relevant to the management of COVID-19 covering the following areas: chemoprophylaxis,diagnosis,treatments,and discharge management.We searched the literature for direct evidence on the management of COVID-19,and assessed its certainty generated recommendations using the Grading of Recommendations,Assessment,Development and Evaluation (GRADE) approach.Recommendations were either strong or weak,or in the form of ungraded consensus-based statement.Finally,we issued 34 statements.Among them,6 were strong recommendations for,14 were weak recommendations for,3 were weak recommendations against and 11 were ungraded consensus-based statement.They covered topics of chemoprophylaxis (including agents and Traditional Chinese Medicine (TCM) agents),diagnosis (including clinical manifestations,reverse transcription-polymerase chain reaction (RT-PCR),respiratory tract specimens,IgM and IgG antibody tests,chest computed tomography,chest X-ray,and CT features of asymptomatic infections),treatments (including lopinavirritonavir,umifenovir,favipiravir,interferon,remdesivir,combination of antiviral drugs,hydroxychloroquine/chloroquine,interleukin-6 inhibitors,interleukin-1 inhibitors,glucocorticoid,qingfei paidu decoction,lianhua qingwen granules/capsules,convalescent plasma,lung transplantation,invasive or noninvasive ventilation,and extracorporeal membrane oxygenation (ECMO)),and discharge management (including discharge criteria and management plan in patients whose RT-PCR retesting shows SARS-CoV-2 positive after discharge).We also created two figures of these recommendations for the implementation purpose.We hope these recommendations can help support healthcare workers caring for COVID-19 patients.
Three generation systems, namely, steam Rankine cycle (SRC), organic Rankine cycle (ORC), and steam-organic combined Rankine cycle (S-ORC), were simulated using the Engineering Equation Solver fEES) ...to efficiently utilize flue gas emissions from 200 to 450 ℃ in iron and steel plants. Based on the simulation results for thermal efficiency, exergy efficiency, and power generation, the performances of the three power generation systems were compared and analyzed. To further utilize waste heat from the turbine exhaust steam of the ORC system, cas- cade ()RC (CORC) was designed for heat sources above 300 ℃. Based on a comprehensive performance comparison, the application of the ORC using R141b is preferable for 200 to 300 ℃ flue gas. For 300 to 450 ℃ flue gas, CORC is an alternative technology to improve the efficiency and quality of waste heat utilization. For flue gas above 450 ℃, S-ORC can achieve higher efficiency and power generation than conventional SRC, with a relatively small negative pressure and high dryness of the turbine outlet steam. Hence, S-ORC can be considered as a substitute for SRC.
