Železo je důležitý biogenní stopový prvek (ve formě hemu je obsaženo v hemoglobinu, myoglobinu či cytochromech, některé sloučeniny obsahují železo vázané na atom síry, např. akonitáza, uplatňuje se i ...v imunitním systému). Patří mezi přechodné kovy, a proto se může ve formě Fe2+ účastnit tzv. Fentonovy reakce, při které dochází ke vzniku nebezpečného a pro organismus toxického hydroxylového radikálu. Metabolismus železa proto musí být velmi přísně regulován – oxidací Fe2+ na Fe3+ ceruloplasminem a jeho inaktivací vazbou na specifi cké proteiny (transferrin, ferritin nebo laktoferrin) nebo jeho chelatací. Přebytečné železo nemůže být z organismu vyloučeno, proto je regulován především jeho příjem. Regulace metabolismu železa probíhá jak na buněčné, tak na systémové úrovni. Na buněčné úrovni je zapojen systém Iron regulation proteins (IRPs)-Iron responsive elements (IREs), který řídí tvorbu proteinů, které se v metabolismu železa uplatňují, a to ovlivněním transkripce, změnou stability mRNA pro daný protein i změnou translace daného proteinu. Na systémové úrovni se v metabolismu železa uplatňuje nedávno objevený peptid hepcidin produkovaný především v hepatocytech, který má v organismu charakter hormonu. Cílovou molekulou hepcidinu je pravděpodobně ferroportin – exportér železa z buňky. Výsledkem působení hepcidinu je snížený export železa z enterocytů duodena a z makrofágů a tedy i snížený přísun železa transferrinem erytroblastům. Tím dojde ke snížení erytropoezy a k anémii. Syntéza hepcidinu je regulována na úrovni transkripce (ovlivnění promotoru genu pro hepcidin a tedy tvorby mRNA). Jeho sekreci tlumí snížená zásoba železa, anémie a hypoxie, stimuluje ji nadbytek železa a především zánět (prostřednictvím IL-6). Zvýšená hladina hepcidinu při zánětu je tak důležitým faktorem pro vznik anémie chronických chorob.
Iron is an important biogenic trace element (in the heme-form is contained in the hemoglobin, myoglobin or cytochromes, some compounds contain iron bound to sulphur atom (aconitase), iron also plays an important role in the immune system). Iron belongs to transition elements and thereby it can be involved in its ferrous form in Fenton reaction, which generates dangerous and toxic hydroxyl radical. Hence metabolism of iron must be strictly regulated – by ceruloplasmin oxidation of Fe2+ on Fe3+ and by inactivation of Fe2+ by its bond to specifi c proteins (transferrin, ferritin or lactoferrin) or its chelation. Iron excess cannot be eliminated thereby especially its uptake is regulated. Iron metabolism is regulated both on cellular and systemic level. System Iron regulation proteins (IRPs) – Iron responsive elements (IREs) participates on cellular level. It controls the production of proteins involved in iron metabolism by interference of transcription, change in mRNA stability and change in translation. Recently discovered peptide hepcidin produced especially in hepatocytes participates on systemic level. The target molecule for hepcidin is probably ferroportin – iron cell exporter. The impact of hepcidin is the decrease of iron uptake from duodenal enterocytes and macrophages and thereby the decrease of iron distribution in transferrin-form to erythroblasts. The result of hepcidin action is the decrease of erythropoiesis and anemia. The hepcidin synthesis is regulated on transcription level (hepcidin gene promotor and production of mRNA is infl uenced). Hepcidin synthesis is decreased by low iron store, anemia and hypoxia, increased by high iron stores and especially in the state of infl ammation (through IL-6). The higher level of hepcidin during infl ammatory state is one of the important factors in anemia of chronic disease genesis.

Ferritin and increased iron stores first appea red on the list of cardiovascular risk factors more than 30 years ago and their causal role in the pathogenesis of ...atherosclerosis has been heavily discussed since the early 1990s. It seems that besides traditional factors such as hyperlipoprotein emia, hyp ertension, diabetes mellitus, obesity, physical inactivity, smoking and family history, high iron stores represent an additional parameter that could modify individual cardiovascular risk. The role of iron in the pathogenesis of atherosclerosis was origina lly primarily associated with its ability to cataly ze the formation of highly reactive free oxygen radicals and the oxidation of atherogenic lipoproteins. Later, it became clear that the mechanism is more complex. Atherosclerosis is a chronic fibroprolife rative inflammatory process and iron, through increased oxidation stress as well as directly, can control both native and adaptive immune responses. Within the arterial wall, iron affects all of the cell types that participate in the atherosclerotic proces s (monocytes/macrophages, endothelial cells, vascular smooth muscle cells and platelets). Most intracellular iron is bound in ferritin, whereas redox-active iron forms labile iron pool. Pro-inflammatory and anti-inflammatory macrophages within arterial plaque differ with regard to the amount of intracellular iron and most probably with regard to their labile iron pool. Yet, the relation between plasma ferritin and intracellular labile iro n pool has not been fully clarified. Data from population studies document that the consumption of meat and lack of physical activity contribute to increased iron stores. Patients with hereditary h emochromatosis, despite extreme iron storage, do not show i ncreased manifestation of atherosclerosis probably due to the low expression of hepcidin in macrophages.

Hepcidin, a key regulator of iron metabolism, plays a crucial role in the pathogenesis of anemia of chronic disease. Although it ...is produced mainly in the liver, its recently described expression in adipose tissue has been shown to be enhanced in massive obesity due to chronic low-grade inflammation. Our objective was to study the changes in hepcidin expression in adipose tissue during acute-phase reaction. We measured hepcidin mRNA expression from isolated subcutaneous and epicardial adipose tissue at the beginning and at the end of the surgery. The expression of mRNAs for hepcidin and other iron-related genes (transferrin receptor 1, divalent metal transporter 1, ferritin, ferroportin) were measured by real-time RT-PCR. Hepcidin expression significantly increased at the end of the surgery in subcutaneous but not in epicardial adipose tissue. Apart from the increased levels of cytokines, the parameters of iron metabolism showed typical inflammation-induced changes. We suggest that acute inflammatory changes could affect the regulation of hepcidin expression in subcutaneous adipose tissue and thus possibly contribute to inflammation-induced systemic changes of iron metabolism.
Dariusz Rozmus a jeho spolupracovníci dospěli v posledních letech k názoru, že hutníci ve Slezsku v 11. až 13. stol. používali k hutnění olověných rud originální, „později zapomenutou“ metodu, kterou ...nazývají „redukce olova železem“. Olověné rudy skutečně lze, při dodržení určitých podmínek, redukovat železem a/nebo kysličníkem železnatým. Základní podmínkou takové technologie je oddělení vsázky od paliva, neboť uhlík a kysličník uhelnatý jsou podstatně silnějšími reduktanty než železo a jeho sloučeniny. Oddělení vsázky a paliva nebylo na studovaných objektech prokázáno. Co si však z výzkumu zaslouží pozornost (a co autoři opomněli podrobněji studovat), je prokazatelné použití železářských strusek jako součásti vyzdívky či výmazu olovářských pecí.
Dariusz Rozmus and his co-workers have come to the conclusion in recent
years that metallurgists in Silesia between the eleventh and thirteenth centuries used an original and ‘later forgotten’ method to smelt lead ores in a process they describe as the ‘reduction of lead by iron’. When certain conditions are maintained, lead ore truly can be reduced by iron and/or iron oxide. The basic condition for using this method is the separation of the charge from the carbon-containing fuel (wood, charcoal) in the furnace – for example by placing the charge in crucibles. Carbon and carbon monoxide are stronger reductants than iron and its compounds. The division of the ore charge and the fuel were not proven in the studied features. Meriting attention from the research, however (something the authors neglected to study in great detail), is the demonstrable use of iron-making slag as part of the lining of the lead smelting furnace.
Během posledního desetiletí došlo k výraznému nárůstu počtu projektů a objevů souvisejících se železem doby železné ve Velké Británii. Patří mezi ně objev jedné z nejranějších tavicích pecí v ...Messinghamu v severním Lincolnshiru, rozsáhlá výroba tradičních nástrojů podél údolí Temže a nálezy železných předmětů včetně mečů a hrotů kopí v hrobech arrasské kultury východního Yorkshiru, například u Pocklingtonu. Došlo také k povzbudivému nárůstu počtu doktorských prací realizovaných na britských univerzitách (některé pod autorovým vedením), zabývajících se nejstaršími železnými předměty a jejich výrobou a deponováním. Příspěvek se zabývá počátky výroby železa ve Velké Británii a vztahy mezi výrobou železa, jeho použitím a ukládáním železných artefaktů v krajině ve světle těchto nejnovějších objevů.
Over the last decade, there has been a noticeable increase in the number of projects and discoveries relating to Iron Age iron in the UK. These include the discovery of one of the earliest smelting furnaces at Messingham, North Lincolnshire, an extensive industry along the Thames Valley and finds of iron objects including swords and spearheads within the graves of the Arras culture of Eastern Yorkshire, for example at Pocklington. There has also been an encouraging increase of the number of PhD theses being undertaken in UK universities on early iron objects and their production and deposition, including those supervised and examined by the writer. This contribution will consider the origins of iron production in Iron Age Britain and the relationship between iron production, its uses and the deposition of iron artefacts within the landscape in the light of these recent discoveries.

t would be desirable to expand the existing general knowledge concerning direct action of metals on the ovary. Nevertheless, ...the results of testing of iron compound on porcine ovarian cells should be interpreted carefully because iron is an essential element which could also induce changes in cellular processes. The aim of this in vitro study was 1) to examine dose-dependent effects of iron on the secretory activity of porcine ovarian granulosa cells, and 2) to outline the potential intracellular mediators mediating these effects. Specifically, we evaluated the effect of iron sulphate on the release of insulin-like growth factor I (IGF-I) and progesterone, as well as the expression of markers of proliferation (cyclin B1) and apoptosis (caspase-3) in porcine ovarian granulosa cells. Concentrations of IGF-I and progesterone were determined by RIA, cyclin B1 and caspase-3 expression by immunocytochemistry (ICC). Our results show a significantly decreased IGF-I secretion by ovarian granulosa cells after iron sulphate addition at the doses 0.5 and 1.0 mg/ml. The iron sulphate additions at do ses 0.17 and 1.0 mg/ml had no effect on progesterone secretion. In contrast, iron sulphate addition at doses 0.17-1.0 mg/ml resulted in stimulation of cyclin B1 and caspase-3 expression. In conclusion, the present results indicate a direct effect of iron on 1) secretion of growth factor IGF-I but not steroid hormone progesterone, 2) expression of markers of proliferation (cyclin B1), or 3) apoptosis (caspase-3) of porcine ovarian granulosa cells. These results support an idea that iron could play a regulatory role in porcine ovarian function: hormone release, prolif eration and apoptosis.
Cíl studie: Metabolismus železa musí být velmi pečlivě regulován. Klíčovým systémovým regulátorem je peptidový hormon hepcidin, který způsobuje “uzamčení” železa uvnitř buňky; to pak nemůže být ...utilizováno. Dialyzovaní pacienti často trpí anémií, která je způsobena mnoha faktory, mj. stavem mikrozánětu a také zřejmě zvýšenou retencí hepcidinu v důsledku snížené glomerulární filtrace. Naším cílem bylo popsat vztahy mezi hepcidinem a dalšími parametry metabolismu železa, zánětu a erytropoezy u těchto pacientů. Typ studie: Observační Název a sídlo pracoviště: Ústav klinické biochemie a hematologie LF UK a FN v Plzni, Alej Svobody 80, 304 60 Plzeň Materiál a metody: Do studie bylo zařazeno 164 chronicky dialyzovaných pacientů (věk 66 ± 13, 25 - 92 let; 101 mužů a 63 žen) a 37 zdravých kontrol (věk 55 ± 20, 21 - 92 let; 16 mužů a 21 žen), u kterých byl stanoven jejich kompletní krevní obraz, parametry metabolismu železa, zánětu a výživy. Výsledky: Koncentrace železa, transferrinu a hemoglobinu byla významně nižší u dialyzovaných pacientů (p<0,0001), naopak ferritin (p<0,0001), solubilní transferrinové receptory(p<0,05), hepcidin (p=0,0003), CRP a IL-6 (p<0,0001) byly u těchto pacientů významně vyšší než u zdravých kontrol. Hepcidin slabě koreloval pozitivně s CRP a ferritinem a negativně s transferrinem u dialyzovaných pacientů, u zdravých kontrol nebyla nalezena žádná korelace hepcidinu s ostatními parametry. Závěr: Zdá se, že vliv zánětu na hladiny hepcidinu není u hemodialyzovaných pacientů zcela zásadní a na jeho koncentraci se podílí i jiné faktory, např. jeho retence.
Objective: Peptide hormone hepcidin is a key systemic regulator of the iron metabolism. Hepcidin binds to the iron cell exporter ferroportin so iron is kept in the cells unavailable for erythropoiesis. The synthesis of hepcidin is up-regulated by high iron stores and inflammation. Dialyzed patients have very often impaired iron management – they suffer from anemia, which is caused by many factors including the state of micro inflammation and hepcidin retention due to decreased glomerular filtration rate. Our aim was to describe the relationship of hepcidin and other parameters of iron metabolism, erythropoiesis and inflammation. Design: Observation Settings: Institute of Clinical Biochemistry and Hematology, Charles University Hospital in Pilsen, Alej Svobody 80, 304 60 Plzeň Material and Methods: Complete blood cell count, iron, ferritin, transferrin, CRP, albumin, creatinine, hepcidin, soluble transferrin receptors (sTfR) and IL-6 were measured in samples from 164 patients included in chronic hemodialysis program (age 66 ± 13, 25 - 92 years), 63 women and 101 men and 37 control healthy volunteers (age 55 ± 20, 21 - 92 years), 21 women and 16 men. Results: Iron, transferrin and hemoglobin were significantly lower in the patients group (p<0.0001) while ferritin (p<0.0001), sTfR (p<0.05), hepcidin (p=0.0003), CRP and IL-6 (p<0.0001) were significantly higher in the patients group. Hepcidin weakly positively correlated with CRP and ferritin and weakly negatively correlated with transferrin in hemodialyzed patients. No correlation of hepcidin with other biochemical parameters in controls was found. Conclusion: It seems that the influence of inflammation on hepcidin levels in hemodialyzed patients is not crucial and other factors including its retention in end-stage renal disease participate.

Cíl studie: Sledovat vliv opakovaných krevních odběrů a diety obohacené o železo na kostní metabolismus u samců potkanů kmene Wistar. Typ studie: Základní výzkum. Název a sídlo pracoviště: Vivárium a ...radioizotopové laboratoře, LF UK Hradec Králové, Ústav klinické biochemie a diagnostiky, Fakultní nemocnice Hradec Králové. Materiál a metody: Potkani byli po dobu 8 týdnů krmeni standardní laboratorní dietou (SLD, 27 mg Fe/kg diety) nebo dietou obohacenou o železo (FE, 400 mg Fe/kg diety). Týdně byl proveden odběr (w) 0,5 ml krve/100 g tělesné hmotnosti, celkem 8krát. Skupiny: 1. kontrolní skupina SLD; 2. kontrolní skupina FE; 3. SLD-w; 4. FE-w. V krvi byl stanoven krevní obraz, respiratorní vzplanutí (RB), v séru koncentrace prohepcidinu a železa, v játrech koncentrace železa. Z kostních ukazatelů jsme stanovili: osteokalcin (OC), N-terminální propeptid prokolagenu typu I (PINP), C-terminální telopeptid kolagenu typu I (CTx) a tartát-rezistentní kyselou fosfatázu (TRACP). Byla změřena kostní minerální hustota (BMD). Výsledky: U skupin s krevními odběry bylo vyšší spontánní RB a železo v séru, naopak došlo k poklesu sérového prohepcidinu, hemoglobinu i železa v játrech ve srovnání s SLD a FE, u FE-w bylo také vyšší stimulované RB. Hodnoty PINP (p < 0,05), CTx (p < 0,05) a TRAP (p < 0,05) vzrostly u SLD-w ve 3. týdnu a u FE-w v 1. týdnu, hodnoty OC (p < 0,05) vzrostly pouze u FE-w v 1. týdnu, poté všechny hodnoty poklesly na hodnoty SLD a FE. BMD vzrostla po odběrech v bederní a ocasní oblasti (p < 0,01). Závěr: Opakované krevní odběry a dieta obohacená o železo vedly k vyšší reaktivitě buněk makrofágového systému, k vyšší aktivitě osteoklastů, ke stimulaci osteoblastů s následným pozitivním vlivem na kvalitu kostní tkáně. Objective: We studied the infl uence of repeated blood withdrawals and iron enriched diet on biochemical markers of bone metabolism in male Wistar rats. Design: Basic research. Settings: Radioisotope Laboratories and Vivarium, Charles University, Medical Faculty, Hradec Kralove, Institute of Clinical Biochemistry and Diagnostics, University Hospital, Hradec Kralove. Material and Methods: Rats were fed for 8 weeks with standard laboratory diet (SLD, 27mg Fe/kg diet) or iron enriched diet (FE, 400 mg Fe/kg diet) and had blood withdrawals (w) 0.5 ml/100 g body weight, 8 times. Animals were divided into 4 groups: 1. Control group SLD; 2. Control group FE; 3. SLD-w; 4. FE-w. The following items were assessed in blood; haemoglobin concentration and respiratory burst (RB), iron stores in liver tissue. In serum were evaluated prohepcidin, iron and bone metabolism markers: osteocalcin (OC), N-terminal propeptid of procollagen I (PINP), C-terminal telopeptide of collagen I (CTx) and tartrate resistant acid phosphatase (TRACP). Bone mineral density (BMD) was measured. Results: Spontaneous RB and iron in serum increased in animals with repeated blood withdrawals, but serum prohepcidin, haemoglobin concentration and iron in liver decreased vs. SLD and FE, but in FE-w animals increased stimulated RB, too. Values of PINP (p < 0.05), CTx (p < 0.05) and TRAP (p < 0.05) increased by SLD-w in 1st week and by FE-w in 3rd week, values of OC (p < 0.05) increased only by FE-w, but then all these values decreased to values of SLD and FE. BMD increased by blood withdrawals in femur (p < 0.01) and lumbar part (p < 0.01). Conclusion: Repeated blood withdrawals and iron enriched diet contributed to stimulation reactivity of scavenger cells, elevation activity of osteoclast, stimulation of osteoblast with subsequent positive effect on quality of bone tissue. The protective role of nutrition factors such as calcium, vitamin D and vitamin K for the integrity of the skeleton is well understood. In ...addition, integrity of the skeleton is positively influenced by certain trace elements (e.g. zinc, copper, manganese, magnesium, iron, selenium, boron and fluoride) and negatively by others (lead, cadmium, cobalt). Deficiency or excess of these elements influence bone mass and bone quality in adulthood as well as in childhood and adolescence. However, some protective elements may become toxic under certain condition s, depending on dosage (serum concentration), duration of treatment and interactions among individual elements. We review the beneficial and toxic effects of key elements on bone homeostasis.
