B. besnoiti is thought to be transmitted mainly by hematophagous insects. This paper presents a review of what is currently known of the epidemiology of bovine besnoitiosis and the key manifestations of the disease. It highlights the urgent need for research programs to improve efficient and sustainable control methods.
Johne's disease or paratuberculosis, caused by Mycobacterium avium subsp. paratuberculosis (MAP), occurs in domestic and wild animals worldwide, causing a significant economic loss to livestock ...industries. After a prolonged incubation time, infected cattle shed MAP bacilli into feces and spread the disease to an uninfected animal population. It is largely unknown how (or whether) the interplay between the pathogen and the host immunity determines timing of shedding after the long incubation time. Such information would provide an understanding of pathogenesis in individual animals and the epidemiology of MAP infection in animal populations. In this review, we summarize current knowledge of bovine Johne's disease pathology, pathogenesis, immunology and genetics. We discuss knowledge gaps that direly need to be addressed to provide a science-based approach to diagnostics and (immuno)prophylaxis. These knowledge gaps are related to anatomical/clinical manifestation of MAP invasion, interaction of bacteria with phagocytes, granuloma formation, shedding, establishment and kinetics of adaptive immune responses in the pathogenesis of the disease. These topics are discussed at the molecular, cellular and tissue levels with special attention to the within host dynamics including the temporal and the spatial context relevant for the various host-pathogen interactions.
A retrospective cohort study was performed with the objective of determining whether the serum concentrations of nonesterified fatty acids (NEFA), β-hydroxybutyric acid (BHBA), or calcium were ...associated with the risk of culling within 60 d in milk (DIM) in Holstein cows, and to establish thresholds for each metabolite that were predictive of increased culling risk. Data from 5,979 cows in Ontario (Canada) and several US states were obtained from 4 previously reported studies. For each metabolite and each of 3 sampling weeks (−1, +1, and +2 relative to calving), an optimal threshold was calculated based on having the maximum combined sensitivity (Se) and specificity (Sp) and used to categorize the serum concentrations into high and low risk groups. Logistic regression models were built for each metabolite and each week of sampling, as well as considering together all metabolites in wk −1 and wk +1 relative to calving. Cow was considered the experimental unit and herd as a random effect. Considered separately, precalving NEFA ≥0.4mmol/L odds ratio (OR)=1.8; 95% confidence interval (CI)=1.4 to 2.2, NEFA ≥0.8mmol/L in wk +1 relative to calving (OR=2.0; 95% CI=1.5 to 2.6) and NEFA ≥0.8mmol/L in wk +2 (OR=4.2; 95% CI=1.9 to 9.4 for cows in lactation 2; OR=2.1; 95% CI=1.4 to 3.3 for cows in lactation ≥3) were each associated with an increased risk of culling within the first 60 DIM. Similarly, BHBA ≥0.7mmol/L in wk −1 (OR=1.8; 95% CI=1.3 to 2.5), BHBA ≥1.2mmol/L in wk +1 (OR=1.8; 95% CI=1.4 to 2.2), and BHBA ≥1.6mmol/L in wk +2 (OR=3.2; 95% CI=1.6 to 6.4 for cows in lactation 2; OR=2.3; 95% CI=1.6 to 3.3 for cows in lactation ≥3) were each associated with an increased risk of culling within the first 60 DIM. Likewise, calcium ≤2.3mmol/L in wk −1 (OR=1.6; 95% CI=1.2 to 2.2), calcium ≤2.2mmol/L in wk +1 (OR=1.5; 95% CI=1.2 to 1.9), and calcium ≤2.3mmol/L in wk +2 (OR=2.3; 95% CI=1.1 to 3.1) were each associated with an increased risk of culling within the first 60 DIM. When all metabolites were analyzed together, serum NEFA and calcium concentrations in wk −1 and serum NEFA concentration in wk +1 remained in the models. In conclusion, elevated serum NEFA and BHBA concentrations and lower serum calcium concentrations within 1 wk before calving through 2 wk after calving were associated with an increased risk of culling in early lactation. Measuring the concentration of selected metabolites around parturition may help to develop monitoring and intervention strategies to prevent early culling in transition dairy cows.
Prediction of postpartum hyperketonemia (HYK) using Fourier transform infrared (FTIR) spectrometry analysis could be a practical diagnostic option for farms because these data are now available from ...routine milk analysis during Dairy Herd Improvement testing. The objectives of this study were to (1) develop and evaluate blood β-hydroxybutyrate (BHB) prediction models using multivariate linear regression (MLR), partial least squares regression (PLS), and artificial neural network (ANN) methods and (2) evaluate whether milk FTIR spectrum (mFTIR)–based models are improved with the inclusion of test-day variables (mTest; milk composition and producer-reported data). Paired blood and milk samples were collected from multiparous cows 5 to 18 d postpartum at 3 Wisconsin farms (3,629 observations from 1,013 cows). Blood BHB concentration was determined by a Precision Xtra meter (Abbot Diabetes Care, Alameda, CA), and milk samples were analyzed by a privately owned laboratory (AgSource, Menomonie, WI) for components and FTIR spectrum absorbance. Producer-recorded variables were extracted from farm management software. A blood BHB ≥1.2 mmol/L was considered HYK. The data set was divided into a training set (n = 3,020) and an external testing set (n = 609). Model fitting was implemented with JMP 12 (SAS Institute, Cary, NC). A 5-fold cross-validation was performed on the training data set for the MLR, PLS, and ANN prediction methods, with square root of blood BHB as the dependent variable. Each method was fitted using 3 combinations of variables: mFTIR, mTest, or mTest + mFTIR variables. Models were evaluated based on coefficient of determination, root mean squared error, and area under the receiver operating characteristic curve. Four models (PLS–mTest + mFTIR, ANN–mFTIR, ANN–mTest, and ANN–mTest + mFTIR) were chosen for further evaluation in the testing set after fitting to the full training set. In the cross-validation analysis, model fit was greatest for ANN, followed by PLS and MLR. Diagnostic strength after cross-validation was poorest for MLR and was similar for ANN and PLS. Models that used mTest + mFTIR variables performed marginally better than models that used only mFTIR or mTest variables. These results suggest that blood BHB prediction models that use mFTIR + mTest variables may be useful additions to existing HYK diagnostic and management programs.
Neospora caninum is a protozoan parasite of animals. Until 1988, it was misidentified as Toxoplasma gondii. Since its first recognition in dogs in 1984 and the description of the new genus and ...species Neospora caninum in 1988, neosporosis has emerged as a serious disease of cattle and dogs worldwide. Abortions and neonatal mortality are a major problem in livestock operations, and neosporosis is a major cause of abortion in cattle. Although antibodies to N. caninum have been reported, the parasite has not been detected in human tissues. Thus, the zoonotic potential is uncertain. This review is focused mainly on the epidemiology and control of neosporosis in cattle, but worldwide seroprevalences of N. caninum in animals and humans are tabulated. The role of wildlife in the life cycle of N. caninum and strategies for the control of neosporosis in cattle are discussed.
Our objectives were to describe and compare the uterine bacterial composition of postpartum Holstein cows diagnosed as healthy (n = 8), subclinical endometritis (SCE; n = 8), or clinical endometritis ...(CE; n = 5) in the fifth week postpartum. We did metagenomic analyses of 16S rRNA gene sequences from endometrial cytobrush samples at 10, 21, and 35 days in milk (DIM), and endometrial bacterial culture at 35 DIM. Uterine bacterial composition in healthy, SCE, and CE was stable at 10, 21, and 35 DIM. Alpha and beta diversities showed a different uterine microbiome from CE compared to healthy or SCE, but no differences were found between healthy and SCE cows. At the phylum level, the relative abundance of Bacteroidetes and Fusobacteria, and at genera level, of Trueperella was greater in CE than healthy or SCE cows. Trueperella pyogenes was the predominant bacteria cultured in cows with CE, and a wide variety of bacterial growth was found in healthy and SCE cows. Bacteria that grew in culture were represented within the most abundant bacterial genera based on metagenomic sequencing. The uterine microbiota was similar between SCE and healthy, but the microbiome in cows with CE had a loss of bacterial diversity.
In the last decade, several new aspects of the inflammation of the bovine endometrium have been investigated and described, including a new definition of subclinical endometritis. This review ...summarizes the recent discussion about the definition, diagnosis and pathomechanism of subclinical endometritis. Subclinical endometritis also referred to as cytological endometritis is defined by findings of endometrial cytology, which is usually performed with the cytobrush-technique or by low-volume flushing of the uterus. The sampling procedure is minimally invasive and has no negative impact on subsequent conception rate. The suggested threshold value for polymorphonuclear cells (PMN) as diagnostic for subclinical endometritis depends on the time postpartum and varies from 5 to 18%. It has also been shown that a general threshold of 5% PMN is eligible for all cows between 21 and 62 days postpartum. Accuracy and repeatability of counting PMN under the microscope have been evaluated and can be regarded as reliable. The impact of subclinical endometritis on reproductive performance is characterized by decreased conception rates, and prolonged days to first service and days open. In addition, it has been demonstrated that subclinical endometritis has an impact on survival and quality of the embryo. Some studies, however, did not confirm this negative effect of subclinical endometritis on fertility. More detailed analyses of the cytobrush samples revealed higher mRNA expression of several cytokines in cows with subclinical endometritis compared with healthy cows, and contributed to the understanding of detrimental effects of subclinical endometritis on fertility. In contrast to clinical endometritis, there are no predominant bacteria related to subclinical endometritis, but associations between the presence of α-hemolytic streptococci and Trueperella pyogenes and subclinical endometritis have been found. For the treatment of subclinical endometritis, intrauterine infusions with cephapirin as well as the administration of PGF2α have been recommended. Other studies, however, did not confirm the efficiency of these treatments.

Johne’s disease is the clinical manifestation of Mycobacterium avium subsp. paratuberculosis (MAP) infection and has become widespread since it was first observed in the United States in the early ...1900s. MAP is primarily spread through the fecal-oral route, and herds generally become infected by unknowingly purchasing infected animals. The economic losses from the disease are primarily due to decreased milk production, decreased weaning weights in nursing young stock, increased replacement costs, and decreased slaughter value.

Epizootic haemorrhagic disease (EHD) is an infectious non-contagious viral disease transmitted by insects of the genus Culicoides which affects wild and domestic ruminants. The causative agent, the ...epizootic haemorrhagic disease virus (EHDV), belongs to the family Reoviridae, genus Orbivirus and shares many morphological and structural characteristics with the other members of the genus such as bluetongue, African horse sickness and equine encephalosis viruses. In recent years EHD outbreaks have been reported in countries bordering the European Union. They caused disease in cattle and severe repercussion on the livestock industry of the affected countries. In the light of recent European bluetongue epizootic these events pose an increasing threat to the European Union. This review includes the most recent information regarding the virus and the disease as well as tools for its diagnosis and control. It is our conviction that more attention should be drawn to both EHDV and the disease itself in order to fulfil all these gaps and not to be unprepared in case future possible incursions.
Ketosis is associated with many transition cow diseases and the subclinical form has been found to be a common condition in high-producing dairy cows. The objectives of this field study in the ...Netherlands were (1) to determine risk factors for subclinical ketosis SCK; 1.2–2.9mmol of β-hydroxybutyrate (BHBA)/L of serum and clinical ketosis (CK: ≥3.0mmol of BHBA/L of serum) at 7 to 14 d in milk and (2) to assess the association of SCK and CK with production parameters at the first dairy herd improvement (DHI) testing. Twenty-three dairies were enrolled by a local veterinary practice from 2009 to 2010, and 1,715 cows were screened for ketosis by measuring serum BHBA concentrations at 7 to 14 d in milk. Overall, 47.2% of cows had SCK and 11.6% had CK. Mixed generalized logit models with a random effect of herd were used to evaluate cow level factors associated with SCK and CK. The associations of SCK and CK with milk production parameters were tested using mixed linear models with a random effect of herd. Cows at a moderate (3.25–3.75) or fat (≥4) body condition score before calving were more likely to develop SCK and CK than thin (body condition score≤3.0) cows. The risk for developing SCK was higher in parity 2 and older cows compared with heifers, whereas for CK only, parity ≥3 cows had a higher risk. The quarter of the year in which a cow calved was associated with the risk for SCK and CK. For SCK quarter 1 (January–March) and quarter 2 (April–June), and for CK quarter 1, quarter 2, and quarter 3 (July–September) all increased the risk of development of the condition compared with quarter 4 (October–December). An increased yield of colostrum at first milking was associated with increasing risk for SCK and CK. Prolonged previous lactation length and dry period length were both associated with increased odds for SCK and CK. Subclinical ketosis and CK were associated with a higher milk yield, a higher milk fat percentage, and a lower milk protein percentage at first DHI test day. Overall the study reinforces previous findings that the major risk factors for both SCK and CK are increasing parity, overconditioning of animals prepartum, season of calving, and dry period length. In addition, previous lactation length and liters of colostrum have been identified as additional risk factors for the development of ketosis.
