In this study, the cross-talk between gama-aminobutyric acid (GABA) and calcium ion (Ca2+) signalling in the regulation of lipid production and cell growth in microalgae under fulvic acid and salinity stress (FA-salinity treatment) was investigated. GABA enhanced the lipid content and lipid productivity rate considerably, which were 1.27 and 1.29 times higher than those of the control, respectively. The levels of biosynthetic gene transcription, GSH, Ca2+ and cellular GABA were promoted by GABA addition, but decreased the ROS levels. Furthermore, the application of Ca2+ also increased lipid synthesis by regulating ROS and GABA signalling and lipogenesis-related genes. These results indicated that cytosolic GABA and Ca2+ levels exert crucial cross-talk in the modulation of cell growth and lipid accumulation induced by FA-salinity treatment. Collectively, this study demonstrated the beneficial effects caused by induction of the combination of chemical compounds on lipid production and provided new insights into lipid synthesis in microalgae.
We have here elucidated the crystal structures of five nostoclide analogues. A common feature in all compounds is a substituent at the para-position of the benzylidene group. Compounds with either ...bromine (3) or hydroxyl (4) as para-substituent crystallizes with Z’ = 2 as result of conformerism. It was also observed that Z' > 1 in the compound with a para-dimethylamino substituent (1). However, its four crystallographically independent molecules are conformationally similar. They are not related by crystallographic symmetry due to the offset packing of their C–H … OC nonclassical hydrogen bonded double chains. This compound (1) has also crystallized in a chiral space group (P21) despite the lack of a stereocenter. Such enantiomorphism phenomenon is related to the presence of only one of the two mirror benzyl conformations with phenyl ring at the equatorial position opposite the lactone oxygen atom. The molecular mean plane of nostoclide analogues has been featured by high level of planarity, except in the brominated compound where two twisted conformations occurred due to rotations on the single bond axis into benzylidene group. The benzyl conformation has been the greatest difference between the two crystallographically independent molecules of the para-hydroxylated compound (4). The crystal packing of the compounds is marked by double catemer motif assembled through C–H … OC non-classical hydrogen bonds, although C–H … π interactions do play an important role in stabilizing the crystal packing of some compounds of the series.

3D-printed geopolymer lattices (3DGPLs) have recently been considered as promising candidates for hazardous waste management due to their low cost, porous nature, and excellent environmental ...stability. However, the ion adsorption capacity influenced by usage conditions such as multi-ion systems and radiation remains largely unexplored. Furthermore, effective strategies to immobilize adsorbed ions for long-term disposal are still lacking. This work tackles these questions by studying the Cs+ adsorption and immobilization properties of 3DGPLs in the presence of competing ions and gamma-ray radiation. Our results reveal the inhibitory intensity of Cs+ adsorption capacity by five common metal cations in seawater, ranked in the following order: Na+ > K+ > Ca2+ > Mg2+ > Sr2+. After exposure to different doses of gamma-ray radiation, the Cs+ adsorption capacity of 3DGPLs shows no significant change, demonstrating their outstanding radiation resistance. By encapsulating the Cs-containing 3DGPL with a homologous GP coating, the Cs+ immobilization rate reaches 98.545% after leaching in NaCl solution at 60 °C for 5 days. The already-high Cs+ immobilization rate can be further increased to 99.228% by sintering treatment due to the densification effect. This study not only provides microscopic insights into the adsorption of hazardous ions by 3DGPLs under usage conditions, but also offers a feasible approach to prevent the leakage of loaded ions through GP coating technology.

Motnje sinteze žolčnih kislin so skupina redkih dednih pre-snovnih bolezni, ki se kažejo s holestazo, odpovedjo ali cirozo jeter ter občasno s steatorejo in/ali nevrološkimi simptomi. Prepoznavanje ...ni vedno enostavno. Najpomembnejši korak k postavitvi diagnoze je njihova vključitev med diferencialno-diagnostične možnosti. V laboratorijskih izvidih je običajno prisotna holestaza z nizko vrednostjo gama-GT in nižjo ali normalno koncentracijo žolčnih kislin. Posamezne encimske motnje ugotavljamo z masno spektrometrijo urina, potrdimo pa jih z genetskim diagnosticiranjem. Večino motenj sinteze žolčnih kislin lahko uspešno dolgoročno zdravimo oralno s holno kislino.
Background The intimal hyperplasia (IH) and vascular remodelling that follows endovascular injury, for instance after post-angioplasty re-stenosis, results in downstream ischaemia and progressive end ...organ damage. Interferon gamma (IFNγ) is known to play a critical role in this process. In mouse models we have previously shown that fibrocytes expressing tissue factor (TF) are recruited early to the site of injury. Through thrombin generation and protease activated receptor-1 (PAR-1) activation, fibrocytes secrete angiopoietin-2, stimulate neointimal cell proliferation, inhibit apoptosis and induce CXCL-12 production, all of which contribute to the progressive IH that then develops. In this study we investigated the relationship between TF, angiopoietin-2 and IFNγ. Methods and results IH developing in carotid arteries of wild-type mice 4 weeks after endoluminal injury contained a significant proportion of IFNγ+ fibrocytes and macrophages, which we show, using a previously defined adoptive transfer model, were derived from circulating CD34+ cells. IH did not develop after injury in IFNγ-deficient mice, except after transplantation of WT bone marrow or adoptive transfer of WT CD34+ cells. In vitro , CD34+ cells isolated from post-injury mice did not express IFNγ, but this was induced when provided with FVIIa and FX, and enhanced when prothrombin was also provided: In both cases IFNγ secretion was TF-dependent and mediated mainly through protease activated PAR-1. IFNγ was predominantly expressed by fibrocytes. In vivo , all IFNγ+ neointimal cells in WT mice co-expressed angiopoietin-2, as did the small numbers of neointimal cells recruited in IFNγ-/- mice. Adoptively transferred WT CD34+ cells treated with either an anti-TIE-2 antibody, or with siRNA against angiopoetin-2 inhibited the expression of IFNγ and the development of IH. Conclusion TF-dependent angiopoietin-2 production by newly recruited fibrocytes, and to a lesser extent macrophages, switches on IFNγ expression, and this is necessary for the IH to develop. These novel findings enhance our understanding of the pathophysiology of IH and expose potential targets for therapeutic intervention.
Objetivando contribuir com o estudo da história e cultura afro-brasileiras e, consequentemente, opondo-se à colonialidade do poder e do saber, neste artigo visamos comparar os projetos de abolição de ...Joaquim Nabuco e de Luís Gama, realizando um levantamento parcial de suas respectivas características e pressupostos. Assim, verificamos diferenças e semelhanças entre as suas propostas e seus posicionamentos políticos e ideológicos para concretizá-las. Sustenta-se o argumento de que o projeto de abolição assumido por Nabuco, entendido aqui como um representante das elites dirigentes daquele contexto histórico, é diametralmente oposto ao projeto de Luís Gama, ex-escravizado que logrou ser poeta e advogado antiescravista. Ambos agem como intelectuais orgânicos de suas classes de origem. O primeiro defendia que a abolição fosse conduzida pelos homens livres, mas deliberada no Parlamento; portanto, sem a participação dos/as escravizados/as e com o objetivo último de embranquecer o Brasil. O segundo defendia que a conquista da liberdade tivesse participação efetiva dos/as escravizados/as, inclusive com uso da violência, como, por exemplo, o assassinato dos senhores pelos/as escravizados/as.

The purpose of this activity is to empower the Gebang ...hamlet farmer women's group, Kemiri village, Tanjungsari District Gunungkidul Regency in optimizing mocaf products so that they have time keep the old and not undergo physical and chemical changes, practical and easy to process in the form of Instant Mocaf Flour Pancake Mocaf “Gama pantelo” as a snack or dessert that can be eaten at any time. The method of activity is done by demonstration and hands-on practice. “Gama Pantelo” Instant Premix Pancake Flour is made by pouring mocaf flour, refined sugar, baking powder, milk powder, and vanilli in a basin by passing it on a sieve to get a homogeneous powder size, then stirring it evenly and then mixing the instant mocaf pancake flour in aluminum foil zipperlock packaging. Pancakes are made with instant premix mocaf pancakes mixed with 1 egg and 180 ml of water then stirred evenly using a mixer. Furthermore, after mixing evenly, the dough is cooked on a small teflon that has been previously smeared with a little oil / butter, then baked until a little cavities arise on the surface of the dough and then the dough is turned over and baked until the pancakes turn golden brown. After that it is appointed and can be served. This activity succeeded in arousing the motivation of farmer women's groups in making variations of cassava preparations.
Adoptive transfer of T cells genetically engineered to express chimeric antigen receptors (CAR) has demonstrated striking efficacy for the treatment of several hematological malignancies, including ...B-cell lymphoma, leukemia, and multiple myeloma. However, CAR T-cell efficacy has been very limited in most solid tumors. In this context, it is of paramount importance to understand the determinants that condition CAR T-cell success versus failure. To control tumor growth, CAR T cells need to form conjugates with their targets
via
the assembly of an immunological synapse. Here, we review recent advances showing that the adhesion between CAR T cells and cancer cells from solid tumors strengthens over time in an IFNγ- and ICAM-1-dependent manner, resulting in CAR T cell-mediated killing. We discuss how these findings can be exploited to increase the efficacy of the CAR T-cell strategy against solid tumors.
