As basic knowledge of reproductive physiology is currently lacking in many species, increasing ...the understanding of reproductive physiology is imperative and includes the development of novel artificial reproduction technologies. Despite the relatively comprehensive knowledge on molecular mechanisms underlying reproduction in livestock species such as cattle, pregnancy failures are likewise far from understood. Contrary to other wildlife species, the European roe deer (Capreolus capreolus) displays a remarkably high pregnancy rate. In parts, cattle and roe deer exhibit comparable features of preimplantation embryo development. Therefore, understanding the high fertility rate in the roe deer holds a great potential for cross-species knowledge gain. As the only known species among the artiodactylae, the roe deer displays a long period of embryonic diapause. The preimplantation blastocyst reaches a diameter of 1 mm only at around 4 months compared to around 13 days post estrus in cattle. The expanded blastocyst survives in a uterine microenvironment that contains a unique set of yet unidentified factors that allow embryonic stem cells to proliferate at low pace without impairing their developmental potential. Upon reactivation, intimate embryo-maternal communication comparable to those reported in cattle is thought to occur. In this review, current knowledge, parallels and differences of reproductive physiology in cattle and roe deer are reviewed. The roe deer is proposed as a unique model species to (1) enhance our knowledge of fertility processes, (2) define factors that support embryo survival for an extended period, (3) advance knowledge on embryonic stem cells, and (4) unravel potential implications for the development of novel strategies for artificial reproductive technologies.

European roe deer (Capreolus capreolus L.) are the most common game species in Europe, hunted for meat and trophies. Forensic investigations involving roe deer poaching may often benefit from an individual identification method to link a suspect to a specific incident. The current paper presents a forensically validated DNA profiling system for European roe deer called “STRoe deer”. This DNA profiling system consists of 12 novel unlinked tetra-nucleotide short tandem repeat (STR) loci and two sexing markers, with an allelic ladder to facilitate accurate genotyping. Validation results using 513 European roe deer samples collected from a single population from the Swiss Plateau demonstrated successful amplification of all 14 loci with as little as 0.05 ng of European roe deer DNA. Species-specificity tests showed that other members of the Cervidae family exhibited partial profiles and non-specific peaks, whereas most members of the Bovidae family showed just non-specific cross-species amplification products. Three different methods to calculate match probabilities for randomly sampled European roe deer genotypes resulted in median match probabilities ranging from 1.4 × 10−13 to 2.5 × 10−5. These methods accounted for possible population structure, occurrence of null alleles and individual relatedness. Based on these results, we conclude that STRoe deer is a robust genotyping system that should prove a valuable tool for individual identification and sexing of European roe deer to support criminal investigations.
To counteract the negative effects of forest fragmentation on wildlife, it is crucial to maintain functional ecological networks. We identified the ecological networks for 2 mammals with very ...different degrees of forest specialization, the European badger Meles meles and the Roe deer Capreolus capreolus, by differentiating 4 agroforestry elements as either nodes or connectivity elements, and by defining the distance that provides the functional connectivity between fragments. Species occurrence data were collected in a wide agroecosystem in northern Italy. To test the role of hedgerows, traditional poplar cultivations, short rotation coppices, and reforestations as ecological network elements for the 2 species we applied the method of simulated species perceptions of the landscape (SSPL), comparing the ability of different SSPLs to explain the observed species distribution. All analyses were repeated considering different scenarios of species movement ability through the matrix. Model outputs seem to show that the specialist and highly mobile Roe deer has the same movement ability throughout the matrix (2 km) as the European badger, a smaller, but generalist species. The ecological network identified for the European badger was widespread throughout the area and was composed of woodlands, poplar cultivations and hedgerows as nodes and short rotation coppices as connectivity elements. Conversely, the ecological network of the Roe deer was mostly limited to the main forest areas and was composed of woodlands, poplar cultivations and reforestations as nodes and short rotation coppices and hedgerows as connectivity elements. The degree of forest specialization strongly affects both species perception of habitat and movement ability throughout the matrix, regardless of species size. Embryonic diapause in mammals leads to a reversible developmental arrest. While completely halted in many species, European roe deer (
) embryos display a continuous deceleration of proliferation. ...During a 4-mo period, the cell doubling time is 2 to 3 wk. During this period, the preimplantation blastocyst reaches a diameter of 4 mm, after which it resumes a fast developmental pace to subsequently implant. The mechanisms regulating this notable deceleration and reacceleration upon developmental resumption are unclear. We propose that amino acids of maternal origin drive the embryonic developmental pace. A pronounced change in the abundance of uterine fluid mTORC1-activating amino acids coincided with an increase in embryonic mTORC1 activity prior to the resumption of development. Concurrently, genes related to the glycolytic and phosphate pentose pathway, the TCA cycle, and one carbon metabolism were up-regulated. Furthermore, the uterine luminal epithelial transcriptome indicated increased estradiol-17β signaling, which likely regulates the endometrial secretions adapting to the embryonic needs. While mTORC1 was predicted to be inactive during diapause, the residual embryonic mTORC2 activity may indicate its involvement in maintaining the low yet continuous proliferation rate during diapause. Collectively, we emphasize the role of nutrient signaling in preimplantation embryo development. We propose selective mTORC1 inhibition via uterine catecholestrogens and let-7 as a mechanism regulating slow stem cell cycle progression.
Every year, there are millions of documented vehicle collisions involving cervids across Europe and North America. While temporal patterns in collision occurrence are relatively well described, few ...studies have targeted deer behaviour as a critical component of collision prevention. In this study, we investigated weekly and daily patterns in road crossing behaviour in roe deer. Using road crossing events and movement data obtained from GPS telemetry, we employed mixed-effect models to explain frequency and timing of crossings at five road segments by a number of predictors including traffic volume, deer movement activity and the presence of wildlife warning reflectors. We analysed 13,689 road crossing events by 32 study animals. Individual variation in crossing frequency was high but daily patterns in crossing events were highly consistent among animals. Variation in the intensity of movement activity on a daily and seasonal scale was the main driver of road crossing behaviour. The seasonal variation in crossing frequency reflected differences in movement activity throughout the reproductive cycle, while daily variation in the probability to cross exhibited a clear nocturnal emphasis and reflected crepuscular activity peaks. The frequency of road crossings increased as a function of road density in the home-range, while traffic volume only exerted marginal effects. Movement activity of roe deer in our study coincided with commuter traffic mainly in the early morning and late afternoon during winter and during periods of high spatial activity such as the rut. Both timing and frequency of crossing events remained unchanged in the presence of reflectors. Our results emphasise the importance of behavioural studies for understanding roe deer vehicle-collision patterns and thus provide important information for collision prevention. We suggest that mitigation of collision risk should focus on strategic seasonal measures and animal warning systems targeting drivers.
Roe deer (Capreolus capreolus) is the most abundant cervid in Europe and, as such, has a considerable impact over several human activities. Accurate roe deer population size estimates are useful to ...ensure their proper management. We tested 3 methods for estimating roe deer abundance (drive counts, pellet-group counts, and camera trapping) during two consecutive years (2012 and 2013) in the Apennines (Italy) in order to assess their precision and applicability. During the study period, population density estimates were: drive counts 21.89±12.74 roe deer/km2 and pellet-group counts 18.74±2.31 roe deer/km2 in 2012; drive counts 19.32±11.12 roe deer/km2 and camera trapping 29.05±7.48 roe deer/km2 in 2013. Precision of the density estimates differed widely among the 3 methods, with coefficients of variation ranging from 12% (pellet-group counts) to 58% (drive counts). Drive counts represented the most demanding method on account of the higher number of operators involved. Pellet-group counts yielded the most precise results and required a smaller number of operators, though the sampling effort was considerable. When compared to the other two methods, camera trapping resulted in an intermediate level of precision and required the lowest sampling effort. We also discussed field protocols of each method, considering that volunteers, rather than technicians, will more likely be appointed for these tasks in the near future. For this reason, we strongly suggest that for each method managers of population density monitoring projects take into account ease of use as well as the quality of the results obtained and the resources required.

The best means of assessing the living conditions of animals is the method of vegetation analysis. The productivity of hunting grounds is determined by the feeding and protective conditions of the ...territory. The classification of hunting grounds by a set of environmental conditions, productivity, and respective capacity is a necessary basis for a rational game husbandry, so they are grouped by types. In the practice of hunting organization of Ukraine, a classification based on phytocoenotic approach is applied. The objects of the study were hunting grounds of forest and forestry farms ruled by the State Agency of Forest Resources of Ukraine and located in the Forest-steppe (right-bank) forestry zone of Ukraine. The European roe deer exhibits the highest behavioural plasticity among representatives of ungulates, as it can successfully reside in both mosaic forest lands and in the field, forming respective forest and field ecotypes. The typological structure of hunting grounds of farms is analysed, which has an influence on the qualitative estimation of the living conditions for the European roe deer. From the northwest to the southeast of the Forest-steppe (right-bank) forest zone of Ukraine, the average hunting ground for the European roe deer varies from 2.7 to 2.4. In areas with predominance of arable land or coniferous forests, medium-quality conditions are formed, and deciduous forests provide good protective and feeding conditions for the European roe deer. It is revealed that good living conditions in the lands do not guarantee a high number of roe deer. Thus, in areas with predominance of arable lands, the number of roe deer was below the optimum by only 10%, and in areas with predominance of deciduous forests it was from 20 to 31.7 %. For practical purposes, it is necessary to determine the approximate level of optimal numbers, taking into account the density of animals in the best game farms in the region, as well as the known density at which reproductive indices and quality of populations deteriorate, and damage to forestry and agriculture is palpable.
We analysed effects of females' body mass and age on reproductive capacity of European roe deer (Capreolus capreolus) in a large sample set of 1312 females (305 yearlings and 1007 adults), hunted ...throughout Slovenia, central Europe, in the period 2013-2015. Body mass positively affected probability of ovulation and potential litter size (number of corpora lutea), although its effect was more pronounced in yearlings than in adults. Between age groups, we found clear differences in responses of both reproductive parameters to body mass which influences primarily reproductive performance of younger, and in particular, lighter individuals: at the same body mass yearlings would at average have smaller litters than adults, and at lower body mass also young to middle-aged adults would have smaller litters than old ones. In addition, while yearlings have to reach a critical threshold body mass to attain reproductive maturity, adult females are fertile (produce ova) even at low body mass. However, at higher body mass also younger individuals shift their efforts into the reproduction, and after reaching an age-specific threshold the body mass does not have any further effects on the reproductive output of roe deer females. Increased reproductive capacity at more advanced age, combined with declining body mass suggests that old does allocate more of their resources in reproduction than in body condition.
