Hemicellulolytic microorganisms play a significant role in nature by recycling hemicellulose, one of the main components of plant polysaccharides. Xylanases (EC 3.2.1.8) catalyze the hydrolysis of ...xylan, the major constituent of hemicellulose. The use of these enzymes could greatly improve the overall economics of processing lignocellulosic materials for the generation of liquid fuels and chemicals. Recently cellulase-free xylanases have received great attention in the development of environmentally friendly technologies in the paper and pulp industry. In microorganisms that produce xylanases low molecular mass fragments of xylan and their positional isomers play a key role in regulating its biosynthesis. Xylanase and cellulase production appear to be regulated separately, although the pleiotropy of mutations, which causes the elimination of both genes, suggests some linkage in the synthesis of the two enzymes. Xylanases are found in a cornucopia of organisms and the genes encoding them have been cloned in homologous and heterologous hosts with the objectives of overproducing the enzyme and altering its properties to suit commercial applications. Sequence analyses of xylanases have revealed distinct catalytic and cellulose binding domains, with a separate non-catalytic domain that has been reported to confer enhanced thermostability in some xylanases. Analyses of three-dimensional structures and the properties of mutants have revealed the involvement of specific tyrosine and tryptophan residues in the substrate binding site and of glutamate and aspartate residues in the catalytic mechanism. Many lines of evidence suggest that xylanases operate via a double displacement mechanism in which the anomeric configuration is retained, although some of the enzymes catalyze single displacement reactions with inversion of configuration. Based on a dendrogram obtained from amino acid sequence similarities the evolutionary relationship between xylanases is assessed. In addition the properties of xylanases from extremophilic organisms have been evaluated in terms of biotechnological applications.
Plant defensins form one of the classes of a large family of PR-proteins (pathogenesis-related proteins), which have different structure-functional properties and provide plant survival and ...adaptation under stress conditions. The defensin synthesis is activated at certain stages of the plant ontogeny, as well as during infection and under the influence of abiotic stress factors. The plant defensins exhibit a broad spectrum of biological activities, including inhibition of a growth of pathogenic microorganisms and parasitic plants, protein biosynthesis, proliferation of tumor cells, proteolytic enzymes, α-amylase, and the HIV-1 reverse transcriptase and blockage of the ion channels. Some of these peptides exhibit allergenic properties. Plant defensins are not only multifunctional peptides which participate in various processes in plants, but also important components of the immune system. This review summarizes data on the structural features of plant defensins and defensin-like proteins, signaling pathways of an activation of their biosynthesis in a plant cell, their biological functions, and their role in the immune response. Possible areas of practical application of these peptides are discussed.

The phenolic compounds of Inonotus rheades (Pers.) Bondartsev and Singer (Hymenochaetaceae), a typical xylotrophic basidiomycete, and accumulation of styrylpyrones in mycelium under the influence of ...light of different wavelengths were investigated. Six styrylpyrones (cis- and trans-hispidin, cis- and trans-bisnoryangonin, and phellinins A1 and A2) and 5 bis(styrylpyrones) (3,14'-bishispidinyl, hypholomin B, 3-bisnoryangonyl-14'-hispidin, 1,1-distyrylpyrylethane, and rheadinin) were detected in the extract of I. rheades mycelium using reversed phase ultra-performance liquid chromatography with diode array detection and electrospray ionization mass spectrometry (RP-UPLC-DAD-ESI/MS). The results showed that the maximal content of styrylpyrones was observed under the influence of blue light (8.10 mg/g of dry mycelium weight). Moreover, hispidin was the dominant compound in all experimental groups. Pigmentation intensity gradually decreased after shifting the light spectrum into darkness. It can be concluded that cultivation of I. rheades mycelium under the blue part of the light spectrum leads to the accumulation of styrylpyrones that have nutraceutical and medicinal significance.
Microbial xylanases have a promising biotechnological potential to be used in industries. In this study, regulation of xylanase production was examined in
Bacillus cereus
BSA1. Xylanase production ...was induced by xylan. The enzyme production further increased in the presence of xylose and arabinose in very low concentration with addition of xylan (0.5% up to 6.02 U/ml). Addition of glucose (about 0.1%) to the media supplemented with xylan repressed xylanase production. Even higher concentration (>0.1%) of xylose and arabinose repressed xylanase biosynthesis. Glucose-mediated repression was partially relived by addition of cyclic adenosine monophosphate. Chemical like 2-4-dinitrophenol, which can inhibit adenosine triphosphate synthesis in cell, repressed xylanase synthesis and it suggested xylanase synthesis to be an energy dependent process.
The plasma membrane H+ V-ATPase from the midgut of larval Manduca sexta, commonly called the tobacco hornworm, is the sole energizer of epithelial ion transport in this tissue, being responsible for ...the alkalinization of the gut lumen up to a pH of more than 11 and for any active ion movement across the epithelium. This minireview deals with those topics of our recent research on this enzyme that may contribute novel aspects to the biochemistry and physiology of V-ATPases. Our research approaches include intramolecular aspects such as subunit topology and the inhibition by macrolide antibiotics, intermolecular aspects such as the hormonal regulation of V-ATPase biosynthesis and the interaction of the V-ATPase with the actin cytoskeleton, and supramolecular aspects such as the interactions of V-ATPase, K+/H+ antiporter, and ion channels, which all function as an ensemble in the transepithelial movement of potassium ions.
Guanyl-specific ribonucleases from Bacillus intermedius and Bacillus pumilus are actively secreted under phosphate starvation by recombinant strains of Bacillus subtilis with native regulatory ...systems and by strains defective in some proteins of the Spo0A phosphorylation pathway. The level of expression of ribonuclease genes has been shown to increase approximately sixfold in recombinant strains with mutation in the spo0A gene and threefold in the spo0A/abrB mutants, as compared with native strains. These results demonstrate that the Spo0A protein regulates the production of ribonucleases and thus acts as a repressor, while the AbrB protein is an activator of expression of the genes encoding ribonucleases from Bacillus intermedius and Bacillus pumilus in Bacillus subtilis cells.
We studied the biosynthesis of Bacillus intermedius glutamyl endopeptidase in the recombinant Bacillus subtilis strain AJ73 Delta 58.21 during the stationary growth phase. We optimized the ...composition of the culture medium to favor effective enzyme production during the stationary growth phase and found that the nutritional requirements for glutamyl endopeptidase synthesis were different in the stationary phase and the growth retardation phase. Proteinase accumulation was activated by complex organic substrates (casein and gelatin). During the final stages of the culture growth, the enzyme production was stimulated by Ca super(2+), Mn super(2+), and Co super(2+) and inhibited by Zn super(2+), Fe super(2+), and Cu super(2+). The synthesis of glutamyl endopeptidase in the late stationary phase was not inhibited by glucose, unlike that in the trophophase during proliferation. We conclude that the regulatory mechanisms of proteinase synthesis during vegetative growth and sporulation are different.
The growth of the recombinant Bacillus subtilis strain AJ73 carrying the Bacillus intermedius 3-19 glutamyl endopeptidase gene on a multicopy plasmid and the effect of some nutrients on the ...efficiency of extracellular glutamyl endopeptidase production in the stationary growth phase were studied. In this phase, the concentration of glutamyl endopeptidase in the culture liquid peaked at the 48th and 78th hours of cultivation and depended on the composition of the cultivation medium. Unlike the synthesis of glutamyl endopeptidase in the trophophase (i.e., during vegetative growth), which was suppressed by glucose, the synthesis of this enzyme during sporulation was resistant to glucose present in the cultivation medium. A multifactorial experimental design allowed optimal proportions between the concentrations of major nutrients (peptone and inorganic phosphate) to be determined. Inorganic phosphate and ammonium ions augmented the production of glutamyl endopeptidase by 30-150%, and complex organic substrates, such as casein and gelatin, enhanced the production of glutamyl endopeptidase by 50-100%. During sporulation, the production of glutamyl endopeptidase was stimulated by some bivalent cations (Ca super(2+), Mg super(2+), and Co super(2+)) and inhibited by others (Zn super(2+), Fe super(2+), and Cu super(2+)). The inference is drawn that the regulatory mechanisms of glutamyl endopeptidase synthesis during vegetative growth and sporulation are different.
