Hoy en dia, los avances tecnologicos e innovadores contribuyen significativamente al progreso de las sociedades, empresas y Estados. Sin embargo, las solicitudes de patentes que se dan a raiz de las ...invenciones en materia de biotecnologia pueden ser objeto de rechazo por las oficinas de patentes, debido a que su explotacion comercial puede ser contraria al orden publico, la moral y las buenas costumbres. Mediante el presente escrito, se realiza un estudio de la excepcion de patentabilidad del orden publico y moral en relacion con la tecnologia de edicion genetica CRISPR-Cas9 a traves del estudio comparado de las legislaciones de Estados Unidos, la Union Europea y de Colombia. Adicionalmente, se analiza el otorgamiento parcial de la patente CRISPR-Cas9, donde la Superintendencia de Industria y Comercio de Colombia solicito modificar el alcance de las reivindicaciones con el fin de conceder la patente, por considerar que la explotacion comercial de la invencion vulnera el orden publico y moral. Finalmente, se menciona brevemente la relacion de COVID-19 con dicha tecnica de edicion genetica.
Background Hemophilia B is a rare inherited genetic bleeding disorder caused by a deficiency or lack of coagulation factor IX, the gene for which (F9) is located on the X chromosome. Hemophilia B is ...currently incurable and the standard treatment is coagulation factor replacement therapy. Although gene therapy has the potential to cure hemophilia, significant barriers are still needed to be overcome, e.g., off-target effects and immunoreactivity, so new approaches must be explored. Nonsense mutations account for 8% of all the hemophilia B mutation types and can result in the development of coagulation factor inhibitors. In this study, CRISPR/Cas9 technology was used to construct a mouse embryonic stem cell model with a hemophilia B nonsense mutation (F9 c.223C > T) in humans to investigate the pathogenesis and treatment of nonsense mutations in hemophilia B. Methods First, a donor plasmid with a mutation (F9 c.223 C > T) and sgRNAs were constructed. Second, both the donor plasmid and the px330-sgRNA were electroporated into mouse embryonic stem cell, and the mutant cells were then screened using puromycin and red fluorescence. Third, the mutant cell lines were tested for pluripotency and the ability to differentiate into three layers. Finally, the effect of mutation on gene function was studied in the differentiation system. Results The mutant vector and effective sgRNA were constructed, and the mutant cell line was screened. This mutant cell line exhibited pluripotency and the ability to differentiate into three layers. This point mutation affects F9 expression at both the RNA and protein levels in the differentiation system. Conclusions The mutant cell line obtained in the current study had a single-base mutation rather than a base deletion or insertion in the exon, which is more similar to clinical cases. In addition, the mutant has the characteristics of mouse embryonic stem cells, and this point mutation affects F9 gene transcription and translation, which can be used as a disease model for studying the pathogenesis and treatment of hemophilia at the stem cell level. The eighth exon of Dip2a gene was replaced with PGK-Puro-P2A-mCherry. This 46C-Dip2a KO cell line offers a useful resource to investigate the molecular mechanisms of DIP2A in the process of cell fate determination, as well as a potential source of building disease mouse model.

This cell line has contributed to further investigation of molecular mechanism of Dip2c during cell differentiation, as well as a cell model for screening for neurogenic drug and cancer clinical cure.
La enfermedad de Parkinson (EP) es el segundo trastorno neurodegenerativo más común, caracterizado por la pérdida selectiva de neuronas dopaminérgicas en la substancia nigra pars compacta, ...produciendo depleción en los niveles de dopamina y dando como resultado las manifestaciones clínicas de la enfermedad que se pueden clasificar como síntomas motrices y no motrices.
En los últimos años, la generación de nuevos modelos animales basados en los sistemas de edición genética por nucleasas: ZFN, TALEN, CRISPR/Cas9, permiten la realización de modificaciones personalizadas en el genoma, replicando características clave que definen a la EP y consecuentemente avances significativos en la comprensión del proceso fisiopatológico de este trastorno.
En esta revisión recopilamos los estudios más novedosos de esta nueva generación de modelos in vitro e in vivo de la EP que permiten emular síntomas clave y tener una mayor comprensión de la etiología o los mecanismos involucrados en el proceso de iniciación o desarrollo de la enfermedad y la futura prueba de terapias realmente eficaces para detener o ralentizar su progresión.
Parkinson's disease (PD) is the second most common neurodegenerative disorder. It is characterised by selective loss of dopaminergic neurons in the substantia nigra pars compacta, which results in dopamine depletion, leading to a number of motor and non-motor symptoms.
In recent years, the development of new animal models using nuclease-based genome-editing technology (ZFN, TALEN, and CRISPR/Cas9 nucleases) has enabled the introduction of custom-made modifications into the genome to replicate key features of PD, leading to significant advances in our understanding of the pathophysiology of the disease.
We review the most recent studies on this new generation of in vitro and in vivo PD models, which replicate the most relevant symptoms of the disease and enable better understanding of the aetiology and mechanisms of PD. This may be helpful in the future development of effective treatments to halt or slow disease progression.

It focuses on the ...synthesis and characterization of the mesoporous silica nanparticles capped with the CRISPR/Cas9 plasmid. The work is focused on the optimization of the CRISPR/Cas9 plasmid construction, which targets green fluorescent protein (GFP), as well as on the optimization of the GFP editing conditions in human cell lines using the MSNs as a carrier for plasmid delivery.
El objetivo de este trabajo es la puesta a punto de un nuevo nanodispositivo, basado en nanopartículas mesoporosas de sílice (MSNs), para la liberación en células humanas del plásmido CRISPR/Cas9 para la edición génica. El trabajo se centrará en la síntesis y caracterización de las nanopartículas mesoporosas de sílice funcionalizadas con el plásmido CRISPR/Cas9. La biotecnología CRISPR/Cas9 brinda el potencial de editar el genoma de células eucariotas y procariotas, abriendo la oportunidad de que cada individuo modifique su propio genoma, basado en su propia decisión. Sin embargo, este tema es altamente controversial a pesar de la evidencia científica que muestra los beneficios potenciales de CRISPR/Cas9 para el tratamiento de algunas enfermedades y para el desarrollo de alternativas biotecnológicas. Esta nota expone de manera básica cómo funciona CRISPR/Cas9 y aborda algunas implicaciones que tiene la toma de decisiones sobre el manejo de CRISPR/Cas9 para editar las células humanas.
