According to the previousstudies, there is an unsolved question on the contribution of the sympathetic nervous system acting thru the alpha1 and alpha2 adrenergic receptors. Our aim was to clarify the role of these receptors in the origin ofboth microcirculatory responses with the use of the laser-Doppler method forskin flow measurements. In the first part of the study we compared responses to the direct and indirect (the cooling of contralateral hand) localcooling before and after intradermal injection of the alpha-adrenoceptor antagonists and agonists with the response in the denervated replanted fingers. In the second part we compared cutaneous postocclusive reactive hyperaemia (PRH before and after intradermal injection of the alpha-adrenoceptor antagonists and agonists. METHODSČ Laser-Doppler (LD) flow measurements were performed on the fingertips of healthy volunteers and of patients with replanted digits. LD flow was monitored 2 minutes before and 6 minutes during local cooling of one hand at 15 degrees Celsius and before and after an 8-minute occlusion of digital arteries. In healthy fingers, we studied the effect of intradermal microinjection (1 micro l) of alpha antagonists (prazosine, yohimbine) and alfa agonists (phenilephrine, clonidine) on local cooling and PRH responses. We evaluated the degree of the LD flow decrease in response to the direct and indirect local cooling with respect to the values obtained before cooling (ANOVA, Dunnet t-test).
