Povezava med polimorfizmom gena za angiotenzinsko konvertazo, za angiotenzinogen in za atrijski natriuretični faktor ter esencialno arterijsko hipertenzijo pri mladostnikih : [magistrsko delo]Bidovec, MatjažEssential arterial hypertension (EAH) is a complex disease trait, which appears to be due to interaction of multiple genetic and environmental factors. Advances in molecular genetics and development ... of new techniques haveled to identification of numerous genetic polymorphisms in candidate genesfor EAH.The renin-angiotensin system (RAS) and natriuretic factor (NAF) play an important role in the process of EAH. The genes of these systems are candidate genes for EAH. In our study we analysed the influence of angiotenzinogen genes, angiotensin converting enzyme (AK) genes, angiotensin II type-1 receptor (AT1R) and NAF on development of arterial hypertension (AH)in children. In our association study we included 57 children aged 8-19, with the diagnosis essential arterial hypertension (EAH) and they were compared to a control group with normal blood pressure. It has been confirmed that positive family history and overweight are higher risk factors for development of EAH. Multiple logistic regression analysis showed that the positive family history of EAH confers an independent risk for EAH. In the association study we confirmed that positive family history for EAH confers even a higher risk for EAH. Different combination of risk genotypes were more often found in patientd than in the control group and consequently we may speak of the interaction of different risk genotypes. Patinets with combination AA polymorphism AT1R and A2A2 polymorphism of NAF gene have a 3.2 higher risk for development of EAH (95% confidence interval = 0.9-10.6č p =0.04). At the same time had a higher risk for the EAH the persons with a genotype combination TT of polymorphism for angiotensinogen and A2A2 genotype polimorphism or A2A2 genotype of NAF gene and genotype II for AK gene.
