Introduction
Prior studies have shown that computed tomography (CT)-based lumbar bone density and strength measurements predict incident vertebral fracture. This study investigated whether CT-based bone density and strength measurements from the thoracic spine predict incident vertebral fracture and compared the performance of thoracic and lumbar bone measurements to predict incident vertebral fracture.
Methods
This case-control study of community-based men and women (age 74.6 ± 6.6) included 135 cases with incident vertebral fracture at any level and 266 age- and sex-matched controls. We used baseline CT scans to measure integral and trabecular volumetric bone mineral density (vBMD) and vertebral strength (via finite element analysis, FEA) at the T8 and L2 levels. Association between these measurements and vertebral fracture was determined by using conditional logistic regression. Sensitivity and specificity for predicting incident vertebral fracture were determined for lumbar spine and thoracic bone measurements.
Results
Bone measurements from T8 and L2 predicted incident vertebral fracture equally well, regardless of fracture location. Specifically, for predicting vertebral fracture at any level, the odds ratio (per 1-SD decrease) for the vBMD and strength measurements at L2 and T8 ranged from 2.0 to 2.7 (

< 0.0001) and 1.8 to 2.8 (
p
< 0.0001), respectively. Results were similar when predicting fracture only in the thoracic versus the thoracolumbar spine. Lumbar and thoracic spine bone measurements had similar sensitivity and specificity for predicting incident vertebral fracture.
Conclusion
These findings indicated that like those from the lumbar spine, CT-based bone density and strength measurements from the thoracic spine may be useful for identifying individuals at high risk for vertebral fracture.
ABSTRACT
We report the discovery of HD 110113 b (TESS object of interest-755.01), a transiting mini-Neptune exoplanet on a 2.5-d orbit around the solar-analogue HD 110113 (Teff = 5730 K). Using TESS ...photometry and High Accuracy Radial velocity Planet Searcher (HARPS) radial velocities gathered by the NCORES program, we find that HD 110113 b has a radius of 2.05 ± 0.12 R⊕ and a mass of 4.55 ± 0.62 M⊕. The resulting density of $2.90^{+0.75}_{-0.59}$ g cm−3 is significantly lower than would be expected from a pure-rock world; therefore HD 110113 b must be a mini-Neptune with a significant volatile atmosphere. The high incident flux places it within the so-called radius valley; however, HD 110113 b was able to hold on to a substantial (0.1–1 per cent) H–He atmosphere over its ∼4 Gyr lifetime. Through a novel simultaneous Gaussian process fit to multiple activity indicators, we were also able to fit for the strong stellar rotation signal with period 20.8 ± 1.2 d from the RVs and confirm an additional non-transiting planet, HD 110113 c, which has a mass of 10.5 ± 1.2 M⊕ and a period of $6.744^{+0.008}_{-0.009}$ d.
ABSTRACT We present Hubble Space Telescope (HST) Wide Field Camera 3 (WFC3) observations of the source and lens stars for planetary microlensing event OGLE-2005-BLG-169, which confirm the relative ...proper motion prediction due to the planetary light curve signal observed for this event. This (and the companion Keck result) provide the first confirmation of a planetary microlensing signal, for which the deviation was only 2%. The follow-up observations determine the flux of the planetary host star in multiple passbands and remove light curve model ambiguity caused by sparse sampling of part of the light curve. This leads to a precise determination of the properties of the OGLE-2005-BLG-169Lb planetary system. Combining the constraints from the microlensing light curve with the photometry and astrometry of the HST/WFC3 data, we find star and planet masses of and . The planetary microlens system is located toward the Galactic bulge at a distance of kpc and the projected star-planet separation is AU, corresponding to a semimajor axis of AU.
Renal impairment is associated with poor prognosis in multiple myeloma (MM). This subgroup analysis of the phase 3 Assessment of Proteasome Inhibition for Extending Remissions (APEX) study of ...bortezomib vs high-dose dexamethasone assessed efficacy and safety in patients with relapsed MM with varying degrees of renal impairment (creatinine clearance (CrCl) <30, 30-50, 51-80 and >80 ml min(-1)). Time to progression (TTP), overall survival (OS) and safety were compared between subgroups with CrCl < or =50 ml min(-1) (severe-to-moderate) and >50 ml min(-1) (no/mild impairment). Response rates with bortezomib were similar (36-47%) and time to response rapid (0.7-1.6 months) across subgroups. Although the trend was toward shorter TTP/OS in bortezomib patients with severe-to-moderate vs no/mild impairment, differences were not significant. OS was significantly shorter in dexamethasone patients with CrCl < or =50 vs >50 ml min(-1) (P=0.003), indicating that bortezomib is more effective than dexamethasone in overcoming the detrimental effect of renal impairment. Safety profile of bortezomib was comparable between subgroups. With dexamethasone, grade 3/4 adverse events (AEs), serious AEs and discontinuations for AEs were significantly elevated in patients with CrCl < or =50 vs >50 ml min(-1). These results indicate that bortezomib is active and well tolerated in patients with relapsed MM with varying degrees of renal insufficiency. Efficacy/safety were not substantially affected by severe-to-moderate vs no/mild impairment.
Angelman syndrome (AS) is a rare neurodevelopmental disorder characterized by developmental delay, impaired communication, motor deficits and ataxia, intellectual disabilities, microcephaly, and ...seizures. The genetic cause of AS is the loss of expression of UBE3A (ubiquitin protein ligase E6-AP) in the brain, typically due to a deletion of the maternal 15q11-q13 region. Previous studies have been performed using a mouse model with a deletion of a single exon of Ube3a. Since three splice variants of Ube3a exist, this has led to a lack of consistent reports and the theory that perhaps not all mouse studies were assessing the effects of an absence of all functional UBE3A. Herein, we report the generation and functional characterization of a novel model of Angelman syndrome by deleting the entire Ube3a gene in the rat. We validated that this resulted in the first comprehensive gene deletion rodent model. Ultrasonic vocalizations from newborn Ube3a
were reduced in the maternal inherited deletion group with no observable change in the Ube3a
paternal transmission cohort. We also discovered Ube3a
exhibited delayed reflex development, motor deficits in rearing and fine motor skills, aberrant social communication, and impaired touchscreen learning and memory in young adults. These behavioral deficits were large in effect size and easily apparent in the larger rodent species. Low social communication was detected using a playback task that is unique to rats. Structural imaging illustrated decreased brain volume in Ube3a
and a variety of intriguing neuroanatomical phenotypes while Ube3a
did not exhibit altered neuroanatomy. Our report identifies, for the first time, unique AS relevant functional phenotypes and anatomical markers as preclinical outcomes to test various strategies for gene and molecular therapies in AS.
We report the discovery and tomographic detection of WASP-174b, a planet with a near-grazing transit on a 4.23-d orbit around a V= 11.9, F6V star with Fe/H = 0.09 ± 0.09. The planet is in a ...moderately misaligned orbit with a sky-projected spin-orbit angle of λ = 31° ± 1°. This is in agreement with the known tendency for orbits around hotter stars to be misaligned. Owing to the grazing transit, the planet's radius is uncertain with a possible range of 0.8-1.8 RSUBJup/SUB. The planet's mass has an upper limit of 1.3 MSUBJup/SUB. WASP-174 is the faintest hot-Jupiter system so far confirmed by tomographic means.
New observations with Herschel allow accurate measurement of the equilibrium temperature of large dust grains heated by the interstellar radiation field (ISRF), which is critical in deriving dust ...column density and masses. We present temperature maps derived from the Herschel SPIRE and PACS data in two fields along the Galactic plane, obtained as part of the Hi-GAL survey during the Herschel science demonstration phase (SDP). We analyze the distribution of the dust temperature spatially, as well as along the two lines-of-sight (LOS) through the Galaxy. The zero-level offsets in the Herschel maps were established by comparison with the IRAS and Planck data at comparable wavelengths. We derive maps of the dust temperature and optical depth by adjusting a detailed model for dust emission at each pixel. The dust temperature maps show variations in the ISRF intensity and reveal the intricate mixture of the warm dust heated by massive stars and the cold filamentary structures of embedded molecular clouds. The dust optical depth at 250μm is well correlated with the gas column density, but with a significantly higher dust emissivity than in the solar neighborhood. We correlate the optical depth with 3-D cubes of the dust extinction to investigate variations in the ISRF strength and dust abundance along the line of sight through the spiral structure of the Galaxy. We show that the warmest dust along the LOS is located in the spiral arms of the Galaxy, and we quantify their respective IR contribution.
Vitamin D recently has been proposed to play an important role in a broad range of organ functions, including cardiovascular (CV) health; however, the CV evidence-base is limited. We prospectively ...analyzed a large electronic medical records database to determine the prevalence of vitamin D deficiency and the relation of vitamin D levels to prevalent and incident CV risk factors and diseases, including mortality. The database contained 41,504 patient records with at least one measured vitamin D level. The prevalence of vitamin D deficiency (≤30 ng/ml) was 63.6%, with only minor differences by gender or age. Vitamin D deficiency was associated with highly significant (p <0.0001) increases in the prevalence of diabetes, hypertension, hyperlipidemia, and peripheral vascular disease. Also, those without risk factors but with severe deficiency had an increased likelihood of developing diabetes, hypertension, and hyperlipidemia. The vitamin D levels were also highly associated with coronary artery disease, myocardial infarction, heart failure, and stroke (all p <0.0001), as well as with incident death, heart failure, coronary artery disease/myocardial infarction (all p <0.0001), stroke (p = 0.003), and their composite (p <0.0001). In conclusion, we have confirmed a high prevalence of vitamin D deficiency in the general healthcare population and an association between vitamin D levels and prevalent and incident CV risk factors and outcomes. These observations lend strong support to the hypothesis that vitamin D might play a primary role in CV risk factors and disease. Given the ease of vitamin D measurement and replacement, prospective studies of vitamin D supplementation to prevent and treat CV disease are urgently needed.
