Epigenetic signatures in germ cells, capable of both responding to the parental environment and shaping offspring neurodevelopment, are uniquely positioned to mediate transgenerational outcomes. ...However, molecular mechanisms by which these marks may communicate experience-dependent information across generations are currently unknown. In our model of chronic paternal stress, we previously identified nine microRNAs (miRs) that were increased in the sperm of stressed sires and associated with reduced hypothalamic–pituitary–adrenal (HPA) stress axis reactivity in offspring. In the current study, we rigorously examine the hypothesis that these sperm miRs function postfertilization to alter offspring stress responsivity and, using zygote microinjection of the nine specific miRs, demonstrated a remarkable recapitulation of the offspring stress dysregulation phenotype. Further,we associated long-term reprogramming of the hypothalamic transcriptome with HPA axis dysfunction, noting a marked decreased in the expression of extracellular matrix and collagen gene sets that may reflect an underlying change in blood–brain barrier permeability. We conclude by investigating the developmental impact of sperm miRs in early zygotes with single-cell amplification technology, identifying the targeted degradation of stored maternal mRNA transcripts including sirtuin 1 and ubiquitin protein ligase E3a, two genes with established function in chromatin remodeling, and this potent regulatory function of miRs postfertilization likely initiates a cascade of molecular events that eventually alters stress reactivity. Overall, these findings demonstrate a clear mechanistic role for sperm miRs in the transgenerational transmission of paternal lifetime experiences.
This survey analyzes two national pharmacovigilance databases in order to determine the major adverse reactions observed with the use of cholinesterase inhibitors in dementia. We conducted a ...statistical analysis of the Food and Drug Administration Adverse Event Reporting System (FAERS) and the Canada Vigilance Adverse Reaction Database (CVARD) concerning the side effects of cholinesterase inhibitors. The statistics calculated for each adverse event were the frequency and the reporting odds ratios (ROR). A total of 9877 and 2247 reports were extracted from the FAERS and CVARD databases, respectively. A disproportionately higher frequency of reports of death as an adverse event for rivastigmine, compared to the other acetylcholinesterase inhibiting drugs, was observed in both the FAERS (ROR = 3.42; CI95% = 2.94-3.98; P<0.0001) and CVARD (ROR = 3.67; CI95% = 1.92-7.00; P = 0.001) databases. While cholinesterase inhibitors remain to be an important therapeutic tool against Alzheimer's disease, the disproportionate prevalence of fatal outcomes with rivastigmine compared with alternatives should be taken into consideration.
Neuropsychiatric disease frequently presents with an underlying hyporeactivity or hyperreactivity of the HPA stress axis, suggesting an exceptional vulnerability of this circuitry to external ...perturbations. Parental lifetime exposures to environmental challenges are associated with increased offspring neuropsychiatric disease risk, and likely contribute to stress dysregulation. While maternal influences have been extensively examined, much less is known regarding the specific role of paternal factors. To investigate the potential mechanisms by which paternal stress may contribute to offspring hypothalamic-pituitary-adrenal (HPA) axis dysregulation, we exposed mice to 6 weeks of chronic stress before breeding. As epidemiological studies support variation in paternal germ cell susceptibility to reprogramming across the lifespan, male stress exposure occurred either throughout puberty or in adulthood. Remarkably, offspring of sires from both paternal stress groups displayed significantly reduced HPA stress axis responsivity. Gene set enrichment analyses in offspring stress regulating brain regions, the paraventricular nucleus (PVN) and the bed nucleus of stria terminalis, revealed global pattern changes in transcription suggestive of epigenetic reprogramming and consistent with altered offspring stress responsivity, including increased expression of glucocorticoid-responsive genes in the PVN. In examining potential epigenetic mechanisms of germ cell transmission, we found robust changes in sperm microRNA (miR) content, where nine specific miRs were significantly increased in both paternal stress groups. Overall, these results demonstrate that paternal experience across the lifespan can induce germ cell epigenetic reprogramming and impact offspring HPA stress axis regulation, and may therefore offer novel insight into factors influencing neuropsychiatric disease risk.
The search for materials with better performance, longer service life, lower environmental impact, and lower overall cost is at the forefront of polymer science and material engineering. This has led ...to the development of self‐healing polymers with a range of healing mechanisms including capsular‐based, vascular, and intrinsic self‐healing polymers. The development of self‐healable systems has been inspired by the healing of biological systems such as skin wound healing and broken bone reconstruction. The goal of using self‐healing polymers in various applications is to extend the service life of polymers without the need for replacement or human intervention especially in restricted access areas such as underwater/underground piping where inspection, intervention, and maintenance are very difficult. Through an industrial and scholarly lens, this paper provides: a) an overview of self‐healing polymers; b) classification of different self‐healing polymers and polymer‐based composites; c) mechanical, thermal, and electrical analysis characterization; d) applications in coating, composites, and electronics; e) modeling and simulation; and f) recent development in the past 20 years. This review highlights the importance of healable polymers for an economically and environmentally sustainable future, the most recent advances in the field, and current limitations in fabrication, manufacturing, and performance.

Emission released associated with the burning of fuels have been presented in this paper. Different sources of emissions in a cement industry has been identified and presented in this study. Different techniques to reduce CO
2 emissions from the cement manufacturing industries are reviewed and presented in this paper. The major techniques are: capture and storage CO
2 emissions, reducing clinker/cement ratio by replacing clinker with different of additives and using alternative fuels instead of fossil fuels. Apart from these techniques, various energy savings measures in cement industries expected to reduce indirect emissions released to the atmosphere. Based on review results it was found that sizeable amount of emission can be mitigated using different techniques and energy savings measures.
Protein rich wastes from aquaculture systems result in total ammonia nitrogen (TAN), total organic carbon (TOC) and biochemical oxygen demand (BOD). A number of conventional approaches have been ...adopted for the removal of these wastes in aquaculture ponds and hatcheries with varying degrees of success but they face critical problems such as membrane fouling, high cost or the generation of toxic by-products. To overcome such issues, electrochemical technology is commonly employed. The advantages of electrochemical treatment include high efficiency, ambient operating conditions, small equipment sizes, minimal sludge generation and rapid start-up. An even better system involves bio-electrochemical reactors (BERs), which have the potential to generate energy from wastewater (by means of microbial fuel cells) or a valuable product such as hydrogen (using microbial electrolysis cells). Mechanisms of cathodic nitrate reduction and anodic oxidation in electrochemical and bio-electrochemical technology are reported in this review. Also some work on the simultaneous removal of nitrate and organic matter by Electro-Fenton and microbial fuel cells are elaborated upon. It is apparent that BERs can remove contaminants at high efficiencies (≈
99%) whilst giving least impact upon the environment.

We study the microstructure, optical, thermal, dielectric, and mechanical properties of flexible oxidized cellulose (OC) films loaded with different mass fractions of cubic structure ...Bi0.5Na0.25K0.25TiO3 by blending solution technique and casting method. The films were characterized using infrared, X-ray diffraction, scanning electron microscopy, and UV–visible spectroscopy. The optical results confirmed the formation of crystalline bismuth sodium titanate/OC semiconductor films with a direct energy bandgap (3.002–3.276 eV) and have the ability for optoelectronic applications. The dielectric constant and dielectric loss of OC /BNKT film decreased obviously with increasing frequency. However, the ferroelectric state of the BNKT and its correlation with structure verified the existence of a relaxer behavior to this ratio. OC film with 20% dopped BNKT displayed energetic bactericidal activity against Gram-positive and Gram-negative bacteria. The presented study suggested that this film can be used as an antibacterial packaging material to diminish packaging's adverse environmental impacts with non-cytotoxicity.
