Background. An experimental study of the antitumor effect of nanocomplexes (NCs) consisting of GdYEuVO4 nanoparticles and cholesterol indicates their potential use in oncological practice. The ...mechanism of the antitumor effect of NCs may be associated with the formation of reactive oxygen species (ROS), leading to subsequent tumor cell death. Objective. To study the pro-oxidant and antitumor properties of NCs consisting of GdYEuVO4 nanoparticles and cholesterol in an in vitro system. Methods. Experiments were performed on Ehrlich ascites carcinoma (EAC) cells, which were introduced intraperitoneally into BALB/c mice. On the 7th day of EAC development, cells were isolated and treated with NCs for 3 hours in vitro. Untreated cells served as the control. The formation of intracellular ROS was quantified by flow cytometry using the Fluorometric Intracellular ROS Kit. The metabolic activity of EAC cells was assessed using the colorimetric MTT test. The number of cells undergoing apoptosis or necrosis was evaluated using flow cytometry and the FITC Annexin V Apoptosis Detection Kit I. Results. Incubation of EAC cells with NCs resulted in more than a 3-fold increase in ROS formation compared to the control. NCs also caused almost a two-fold inhibition of the metabolic activity of EAC cells, accompanied by a 25% decrease in the number of viable EAC cells. It was shown that NCs are unique compounds capable of simultaneously inducing several types of cell death, with necrosis being the predominant mode of tumor cell death after NC treatment. Conclusions. The cytotoxic effect of NCs on tumor cells is mediated through their pro-oxidant properties. These results can be considered when developing new cancer therapy strategies.
Проблематика. Експериментальне вивчення протипухлинної дії нанокомплексів (НК), що складаються з наночастинок GdYEuVO4 і холестерину, свідчить про доцільність їх використання в онкологічній практиці. Механізм реалізації протипухлинної дії НК може бути пов’язаний з утворенням під їхнім впливом активних форм кисню (АФК), які зумовлюють загибель пухлинних клітин. Мета. Вивчення прооксидантних і протипухлинних властивостей НК, що складаються з наночастинок GdYEuVO4 і холестерину, в системі in vitro. Методика реалізації. Експерименти проводили на клітинах асцитної аденокарциноми Ерліха (АКЕ), які перещеплювали внутрішньочеревно мишам лінії BALB/c. На 7-му добу розвитку АКЕ клітини виділяли та in vitro обробляли НК протягом 3 год. Контролем були клітини, не оброблені НК. Формування внутрішньоклітинних АФК визначали методом проточної цитометрії з використанням набору Fluorometric Intracellular ROS Kit, рівень метаболічної активності клітин АКЕ – за допомогою МТТ-тесту колориметричним методом, кількість клітин у стані апоптозу або некрозу оцінювали з використанням проточної цитометрії та набору реагентів FITC Annexin V Apoptosis Detection Kit I. Результати. Інкубація клітин АКЕ з НК приводить до більш ніж трикратного посилення порівняно з контролем формування АФК. Під впливом НК також відбувається майже двократне пригнічення метаболічної активності клітин АКЕ, що супроводжувалося зменшенням на 25 % кількості життєздатних клітин АКЕ. Показано, що НК належать до тих унікальних сполук, які одночасно можуть викликати кілька видів загибелі клітин, але загибель пухлинних клітин після обробки НК відбувалася переважно унаслідок некрозу. Висновки. Цитотоксична дія НК відносно клітин АКЕ реалізується за рахунок їхніх прооксидантних властивостей. Отримані результати можуть бути враховані при створенні нових стратегій терапії злоякісних новоутворень.

Dendritic cells (DCs) initiate and shape both innate and adaptive immune responses. They specialize in presenting antigens to naïve T cells, thereby directing T cell immune responses and contributing ...significantly to the maintenance of antitumor immunity. In both human and animal bodies, these cells are present in limited quantities, posing challenges in their procurement. Hence, the quest for obtaining DCs with antitumor properties in vitro from progenitor cells for clinical or experimental use remains pertinent. This research aims to consolidate existing studies on deriving immune DCs from progenitor cells for application in anticancer therapy. Analysis of published reports reveals that monocytes from peripheral blood, mononuclear cells from bone marrow, and cord blood can serve as precursor cells of immune DCs. Protocols for generating immature DCs from progenitor cells involve the addition of various combinations of cytokines to the culture, including granulocyte-macrophage colony-stimulating factor, interleukin-4, and other cytokines. The extensive range of cytokines and conditions influencing the differentiation and functional activity of DCs results in considerable heterogeneity in the phenotypic and functional characteristics of these cells. Sources of tumor antigen for DC-based vaccines encompass tumor lysates, individual tumor proteins, peptides, and tumor cells in a state of immunogenic apoptosis. This paper delves into the use of maturation factors and cryopreservation as integral stages in obtaining immune DCs. A comprehensive understanding of the parameters involved in obtaining immune DCs is imperative for the development of DC-based vaccines to unleash their full antitumor potential.

Робота присвячена узагальненню досліджень щодо отримання імунних ДК із клітин-попередників для застосування в протипухлинній терапії. Аналіз літературних джерел показав, що як клітини-попередники імунних ДК можуть виступати моноцити периферичної крові, мононуклеари кісткового мозку, кордової крові. Протоколи, які використовуються для отримання незрілих ДК із клітин-попередників, передбачають додавання в культуру різних комбінацій цитокінів, включаючи гранулоцитарно-макрофагальний колонієстимулювальний фактор, інтерлейкін-4 тощо. Широке розмаїття цитокінів і умов, здатних впливати на диференціацію та функціональну активність ДК, обумовлює надзвичайну їх гетерогенність за фенотиповими і функціональними характеристиками. Як джерела пухлинного антигена для виробництва вакцини на основі ДК використовують пухлинні лізати, окремі пухлинні білки, пептиди, пухлинні клітини в стані імуногенного апоптозу. An important component of the pathogenesis of autoimmune diseases is the immune system deregulation as an impaired tolerance to its own antigens by reducing the content of T-regulatory cells. Their ...formation is closely related to the function of dendritic cells (DCs), so in autoimmune diseases the use of DCs with tolerogenic potential is promising for the restoration of antigen-specific tolerance. Recently, the issue of establishing the banks of tolerogenic DCs for clinical use, which involves their cryopreservation, has been actively discussed. To date, there is no common protocol for DCs freezing, which would take into account the different sources of their obtaining, the initial structural and functional state before freezing, composition of cryopreservation media and other factors. The review summarizes experimental data on cryopreservation of peripheral blood and bone marrow mononuclear cells or monocytes. The potential for their further ex vivo differentiation into DCs to ensure the stability of immature phenotype and tolerogenic function has been studied.
The objective of this research is to predict the level of cognitive load of various degrees, using the physiological measurement from wearables and the subject's psychological and demographic traits. ...This paper focuses on combining machine learning and machine reasoning techniques that capture joint distributions of both the objective and subjective predictors of the level of cognitive load. We used the CogLoad dataset, which contains both the physiological signals (heart rate, intervals between successive heartbeats, galvanic-skin response, and temperature), demographics, and psychological traits such as personality type. The four physiological signals were the source of the objective features and the input to the classifiers such as the feed-forward neural network. The latter showed the accuracy at 68.19%, F1-score at 72.6% in detecting the level of cognitive load. The distributions of the classifier results, as well as the demographic information (age, gender, education), and psychological personality indicators, were used to create a causal graph and demonstrate the inference of several scenarios for the advanced predictive decision support.

The importance of the video data resource in various practical domains is justified. This encompasses control systems, video surveillance, monitoring, and video conferencing. It is evident that there ...is an imbalance between the intensity of the video stream and the existing characteristics that ensure the bandwidth of information and telecommunication systems. This discrepancy results in a conflict with the specified requirements for the quality attributes of video services. Consequently, there arises a necessity to elevate the accessibility of video data re-sources while preserving their integrity through the application of information and telecommunication technologies. These tech-nologies are employed at various stages of video data delivery. Consequently, a contradiction arises with modern coding tech-nologies, primarily due to the inverse relationship between the volume of encoded segments and their semantic integrity. It is substantiated that less informative segments can facilitate the implementation of novel processing methods, resulting in a reduction in the bit volume of segments. This, in turn, creates favorable conditions for augmenting the overall integrity of video resources. Hence, the objective of this article is to devise a methodology for encoding low-informative segments within a video data resource to enhance their accessibility. This method involves crafting a syntactic description of the condensed repre-sentation of areas. Nonetheless, it is essential to note that this approach does not compromise the semantic integrity of the video resource. The method comprises the following procedural steps: firstly, reducing psychovisual redundancy in the temporal dimension by identifying areas suitable for local correction and rounding the fundamental elements of these areas to six upper bits.
