•We described the establishment of the first biological models for Xia-Gibbs syndrome (XGS) study: three induced pluripotent stem cell (iPSC) lines derived from cells of XGS patients and a zebrafish ...strain with ahdc1 LoF variant obtained through CRISPR/Cas9-mediated editing.•The iPSC lines showed expression of pluripotency factors and have the capacity to differentiate into the three germ layers.•The zebrafish line is fertile and breeding between heterozygous and wild-type animals generated offspring in a genotypic proportion in agreement with Mendelian law.•These models may be relevant to investigate the gibbin protein functions, its role in neurodevelopment and XGS pathophysiological mechanisms. In addition, the detailed description of the methodology to establish these models will be of value to the community interested in genetic disease modeling.
Xia-Gibbs syndrome (XGS) is a syndromic form of intellectual disability caused by heterozygous AHDC1 variants, but the pathophysiological mechanisms underlying this syndrome are still unclear. In this manuscript, we describe the development of two different functional models: three induced pluripotent stem cell (iPSC) lines with different loss-of-function (LoF) AHDC1 variants, derived by reprogramming peripheral blood mononuclear cells from XGS patients, and a zebrafish strain with a LoF variant in the ortholog gene (ahdc1) obtained through CRISPR/Cas9-mediated editing. The three iPSC lines showed expression of pluripotency factors (SOX2, SSEA-4, OCT3/4, and NANOG). To verify the capacity of iPSC to differentiate into the three germ layers, we obtained embryoid bodies (EBs), induced their differentiation, and confirmed the mRNA expression of ectodermal, mesodermal, and endodermal markers using the TaqMan hPSC Scorecard. The iPSC lines were also approved for the following quality tests: chromosomal microarray analysis (CMA), mycoplasma testing, and short tandem repeat (STR) DNA profiling. The zebrafish model has an insertion of four base pairs in the ahdc1 gene, is fertile, and breeding between heterozygous and wild-type (WT) animals generated offspring in a genotypic proportion in agreement with Mendelian law. The established iPSC and zebrafish lines were deposited on the hpscreg.eu and zfin.org platforms, respectively. These biological models are the first for XGS and will be used in future studies that investigate the pathophysiology of this syndrome, unraveling its underlying molecular mechanisms.
O atual estudo objetiva realizar uma análise de espécie fitoindicadora de alterações ambientais rápidas em área urbana, observando-se a atividade fenológica da espécie Clitoria fairchildiana e, a ...partir disso, propor medidas de conservação e manejo de áreas verdes, tendo em vista seu contributo ao monitoramento ambiental no espaço citadino. Foram escolhidas três áreas de estudos localizadas na Av. Adélia Franco (zona sul - área 1); Praça Fausto Cardoso (zona centro – área 2) e Praça Princesa Isabel (zona norte - área 3) de Aracaju – SE, onde foram obtidos o índice de atividade fenológica e o índice de intensidade de Fournier dos indivíduos analisados em cada área, assim como os dados dos indicadores abióticos (temperatura, umidade relativa do ar e luminosidade) durante um período de oito meses de levantamentos in situ. Os dados dos indicadores tanto bióticos como abióticos foram submetidos ao teste estatístico de Spearman, a fim de estabelecer o nível de correlação entre eles. Observou-se que a Clitoria fairchildiana é fitoindicadora de mudanças ambientais rápidas em áreas urbanas, devido demonstrar nítidas alterações em suas fenofases correlacionadas diretamente às mudanças de temperatura e de luminosidade, sendo o conjunto arbóreo da Praça Fausto Cardoso o que melhor evidenciou correlações diretas entre as fenofases e os fatores abióticos, sobretudo a diminuição da floração ante o aumento da temperatura no ambiente urbano aracajuano. Use of Phytoindicator Species as Subside for Monitoring Environmental Changes in Urban Areas ABSTRACT The current study aims to conduct an analysis of a phytoindicator species for rapid environmental changes in an urban area, observing the phonological activity of the species Clitoria fairchildiana, and from this, to propose conservation and management of green areas in view of its contribution to environmental monitoring in city space. We chose three study areas located in Adélia Franco Avenue (south zone – Area 1); Fausto Cardoso Square (downtown area - Area 2) and Princesa Isabel Square (north zone - Area 3) in Aracaju - SE, where were obtained the phenological activity index and the Fournier Intensity Index from the analyzed individuals in each area, as well as abiotic indicator data (temperature, relative humidity and light) over a period of eight months of in situ surveys. Both abiotic and biotic indicator data were tested using Spearman rank test, in order to establish the level of correlation between them. It was observed that Clitoria fairchildiana is phytoindicator of rapid environmental changes in urban areas due to show sharp changes on its phenophases directly correlated to changes in temperature and luminosity, with all trees of the Fausto Cardoso Square that better evidenced direct correlations between phenology and abiotic factors, particularly the reduction of flowering compared with the temperature increase in the urban environment of Aracaju.Keywords: Phytoindication; Phenology; Green Areas; Urban Climate; Aracaju.
ABSTRACT Objective The aim of this study was to evaluate, in vitro, the union stability of resin cements to the dental substract through microtensile bond strength (µTBS) analysis and scanning ...electron microscopy (SEM). Methods Fifty-four third human molars, stored in water for a short (24 hours) and long period of time (1 year) presented a flat oclusal superficial dentin. The teeth were distributed in six different groups: G1- Panavia F2.0/Kuraray; G2- RelyXUnicem/3M ESPE; G3- G-Cem/GC; G4- Biscem/ Bisco; G5- Panavia F2.0/Kuraray without pre-treatment and G6- Multilink Sprint/Ivoclar-Vivadent which were adhered to its respective indirect resin composite restoration, (G1- Clearfil AP-X/Kuraray; G2- Filtek Z350/3M ESPE; G3- Gradia Direct X(tm)/GC; G4- Aelite(tm)/ Bisco; G5- Clearfil AP-X/Kuraray; G6- Tetric Ceram/ Ivoclar-Vivadent). The resin blocks were cemented and the sticks were obtained by tooth, with an area of adhesive interface of 0,8mm² (±0,2). Results The mean values, submitted to Mann-Whitney and Kruskal-Wallis tests (α = 5%) were in MPa after 24 hours: G1 = 9.66 (A), G2 = 13.37 (A); G3 = 15.89 (A); G5 = 4.18 (B); G6 = 11.01 (A) and after 1 year: G1 = 9.75 (A), G2 = 11.73 (A); G3 = 20.10 (B); G5 = 6.80 (A); G6 = 21.09 (B). All G4 group presented pretest failures. Conclusion During the one year period, with the exception of BisCem, the self-adhesive resin cements were a favorable alternative for the adhesive cementation, standing out among these, the G-Cem and Multilink Sprint.
RESUMO Objetivo Avaliar, in vitro, a estabilidade da união de cimentos resinosos ao substrato dental através da resistência de união e análise em microscopia eletrônica de varredura (MEV). Métodos Cinquenta e quatro terceiros molares humanos, armazenados em água por um curto (24 horas) e longo período de tempo (1 ano), tiveram a face oclusal removida expondo a superfície dentinária. Os dentes foram distribuídos em seis grupos distintos: G1- Panavia F2.0 / Kuraray; G2- RelyX Unicem / 3M ESPE; G3- G-Cem / GC; G4- BisCem / Bisco; G5- Panavia F2.0 / Kuraray sem pré-tratamento e G6- Multilink Sprint / Ivoclar-Vivadent que foram aderidas ao seu respectivo restaurações de resina composta indireta, (G1- Clearfil AP-X / Kuraray; G2- Filtek Z350 / 3M ESPE; G3- Gradia Direct X (tm) / GC; G4- Aelite (tm) / Bisco; G5- Clearfil AP-X / Kuraray; G6- Tetric Ceram / Vivadent Ivoclar-). Os blocos de resina foram cimentados e foram obtidos palitos, com área de interface adesiva de 0,8mm² (± 0,2). Resultados Os valores médios, submetidos à Mann-Whitney e Kruskal-Wallis testes (α = 5%) foram em MPa após 24 horas: G1 = 9,66 (A), G2 = 13,37 (A); G3 = 15,89 (A); G5 = 4,18 (B); G6 = 11,01 (A) e depois de 1 ano: G1 = 9,75 (A), G2 = 11,73 (A); G3 = 20,10 (B); G5 = 6,80 (A); G6 = 21,09 (B). O grupo todo G4 apresentou falhas pré-teste. Conclusão Durante o período de um ano de investigação, os cimentos de resinosos auto-adesivos, exceto o Bis Cem, eram uma alternativa favorável para a cimentação adesiva, destacando-se entre estes o G-Cem e Multilink Sprint.
the aim of this study was to analyze the influence of ozone therapy (OZN) on peri-implant bone repair in critical bones by installing osseointegrated implants in the tibia of ovariectomized rats.
...ovariectomy was performed on 30 Wistar rats, aged six months (Rattus novergicus), and, after 90 days, osseointegrated implants were installed in each tibial metaphysis. The study groups were divided into the animals that received intraperitoneal ozone at a concentration of 700 mcg/kg - OZ Group (n=15) - and a control group that received an intraperitoneal saline solution and, for this reason, was named the SAL group (n=15). The applications for both groups occurred during the immediate post-operative period on the 2nd, 4th, 6th, 8th, 10th, and 12th day post-surgery. At various stages (14, 42, and 60 days), the animals were euthanized, and tests were performed on their tibiae. These tests include histomorphometric and immunohistochemical analyses, computerized microtomography, sampling in light-cured resin for calcified sections, and confocal microscopy. The obtained data were then analyzed using One-way ANOVA and the Shapiro-Wilk, Kruskal-Wallis, and student t-tests (P<0.05).
our findings indicate that the OZ group (3.26±0.20 mm) showed better cellular organization and bone neoformation at 14 days (SAL group, 0.90±1.42 mm) (P=0.001). Immunohistochemistry revealed that osteocalcin labeling was moderate in the OZ group and mild in the SAL group at 14 and 42 days post-surgery. The data from the analysis of calcified tissues (microtomography, histometric, and bone dynamism analysis) at 60 days showed no statistically significant differences between the groups (P=0.32).
it was concluded that ozone therapy anticipated the initial phases of the peri-implant bone repair process.

O enorme crescimento das cidades brasileiras verificado nas últimas décadas, aliado à dificuldade das camadas populares para a aquisição da moradia, tem contribuído para a constante ocupação de áreas inadequadas à urbanização, sujeitas aos riscos de movimentos de massa. Este grave problema social, potencializado pela falta de planejamento público, expõe, a cada verão, grande número de famílias a graves acidentes nas encostas. Por sua vez, os movimentos de massa, especialmente os escorregamentos, são fenômenos deflagrados pela precipitação. Dessa forma, o entendimento do ritmo climático e da dinâmica pluvial representa importante passo para a previsão e controle desses acidentes. A Região Noroeste da cidade de Juiz de Fora - MG constitui um local de intensa expansão urbana, onde a ocupação desordenada vem causando uma série de transtornos ambientais, inclusive a ocorrência de escorregamentos. Este trabalho tem como objetivo central o reconhecimento da dinâmica e da gênese pluvial em correlação com a ocorrência de escorregamentos na Região Noroeste da Área Urbana de Juiz de Fora. A abordagem parte da escala regional, através do entendimento das principais características climáticas do entorno da cidade de Juiz de Fora, passando-se a escala local, por intermédio da técnica de análise rítmica.
The enormous growth of brazilian cities verified in the last decades, allied to the difficulty of the popular classes for the acquirement of the domicile, has contributed to the constant occupation of inappropriate areas for urbanization, liable to the risks of mass movements. This serious social problem, increased by the lack of public planning, exposes, every summer, a great number of families to serious accidents on the hillsides. Thus, mass movements, specially landslides, are phenomenons deflagrated by precipitation. Therefore, understanting the climatic rhythm and pluvial dynamics, represents an important step to prevent and control these accidents. The Nortwest Region of the city of Juiz de Fora - MG, constitutes a location of intense urban expansion, where disordered occupation has been provoking several enviromental disturbances, inclusively the occurence of landslides. This work has as a main purpose , the admission of the dynamics and pluvial genesis in the Nortwest Region of Juiz de Fora Urban Area. The approach begins from the regional scale, through the understanting of the main climatic charactiristics around the city of Juiz de Fora, passing to local scale, through the technique of the rhythmic analysis.

Em meio tropical, os escorregamentos figuram entre os fenômenos que mais geram acidentes em áreas de risco, causando uma série de perdas econômicas e humanas a cada ano. Várias cidades brasileiras congregam a ameaça dos escorregamentos e a vulnerabilidade de parte significativa de sua população, mas, apesar disso, os trabalhos preventivos são iniciativas raras ou até mesmo inexistentes. Nos casos em que essa atividade é desenvolvida, geralmente pode-se verificar a negligência sobre certos aspectos fundamentais do risco, dentre eles a percepção e a conduta dos moradores envolvidos. Neste trabalho, os riscos de escorregamentos na Vila Mello Reis, em Juiz de Fora (MG), foram pesquisados por intermédio da abordagem perceptiva. Mais especificamente, foram investigadas as seguintes variáveis: percepção dos elementos condicionantes e deflagradores dos escorregamentos, da causalidade dos acidentes e da responsabilidade sobre os riscos; avaliação e escolha do local de moradia; limiares de segurança; ajustamentos à situação de risco; e tomada de decisão e participação na escala do bairro. Logo, a partir dos resultados obtidos, procurou-se construir um conjunto de propostas visando subsidiar o trabalho de planejamento e de gestão urbanos no cenário da pesquisa, sobretudo no que diz respeito à prevenção de acidentes envolvendo escorregamentos.
In a tropical environment, the landslide appear among the phenomena that generate more accidents in risk areas, causing a series of economic and human losses each year. Several Brazilian cities gather the landslides threatment and the vulnerability of a significative part of their population, but nonethless, the preventive works are rare initiative or either nonexistants. In cases when this activity is carried out generally it can be verified the neglicency about some fundamental aspects of risk among them the perception and the behave of some community people engaged in it. In this work, the landslides risks in Mello Reis village, in Juiz de Fora city (MG), were researched through the perceivity approach. It were investigated specifically the following variables: perception of the conditioning elements and landslides startes, the accidents causes and the risks responsability; evaluation and the dweling site choicing; security threshold; adjustment to the risk situation; decision making and participation at the neighbourhood area. So, through the reached outcomes, it was tried to construct a set of propositions aiming to subsidize the planning and urban management on the research setting, above all concerning to accidents prevention due to landslides.
Avaliaram-se as variáveis técnicas e econômicas do sistema de produção em confinamento de 88 animais superprecoces (sete meses de idade) pertencentes a três grupos genéticos, com 23 animais Charolês ...x Nelore, 26 Red Angus x Nelore e 39 Abeerden Angus x Nelore. Foram registrados pesos no inicio do confinamento e ao abate, duração do confinamento, ganhos de peso no período e diário dos animais e, por meio de conceitos econômicos e operacionais, foram calculados os custos de produção. O modelo estatístico utilizado na análise das variáveis incluiu efeitos do grupo genético e do peso inicial como covariável. Não houve diferença entre grupos genéticos para peso ao abate e ganho de peso diário. A duração do confinamento foi menor para animais Charolês x Nelore (159 dias), intermediária para Red Angus x Nelore (170 dias) e maior para Abeerden Angus x Nelore (178 dias). Os custos total, operacional e variável da diária foram 1,02; 0,85 e 0,89 US$/dia, respectivamente. O confinador obteve lucro de 0,01 US$/kg e rentabilidade de 1,8%/mês. Não houve diferenças (P>0,05) entre grupos genéticos para custos, receita, margens bruta e líquida, lucro e rentabilidade.
This experiment was carried out to evaluate technical and economic parameters for feedlot production system of 88 seven-month-old animals of three genetic groups (23 Charolais x Nellore, 26 Red Angus x Nellore, and 39 Aberdeen Angus x Nellore). Production traits (initial and slaughtering weights, number of days in feedlot, and total and daily weight gains) were recorded and costs were estimated using operational and economic concepts. The statistical model used to analyze the traits included the fix effect of genetic group and initial body weight as a covariate. No differences were observed among the genetic groups for slaughtering weight and daily weight gain. Number of days in feedlot was low for Charolais x Nellore (159 days), intermediate for Red Angus x Nellore (170 days), and high for Aberdeen Angus x Nellore (178 days) animals. Total, operational, and variable costs were 1.02; 0.85; and 0.89 US$/day. Farmer profit was 0.01 US$/kg and profitability/month was 1.78%. Significant differences among the genetic groups were not observed for cost, income, gross and net margins, profit, and profitability.
As medidas do AMSU-A para os canais que são sensíveis à superfície terrestre sobre os continentes não tem sido amplamente utilizadas para ajustar a previsão numérica de tempo de curto prazo (PNTs), ...devido à complexidade das características da superfície terrestre. Nesse sentido, o presente artigo utiliza o Sistema de Assimilação de Dados (G3DVAR) do Centro de Previsão de Tempo e Estudos Climáticos do Instituto Nacional de Pesquisas Espaciais (CPTEC/INPE), que inclui tais medidas de radiâncias feitas pelo sensor que está a bordo dos satélites da série NOAA. A versão operacional do sistema G3DVAR contempla apenas o satélite NOAA-15. Adicionalmente, foram realizados experimentos numéricos que incluíram os satélites NOAA-18 e NOAA-19. É feita uma avaliação sobre a variável temperatura de brilho simulada para os canais sensíveis à superfície terrestre (i), através de uma comparação com observações, e (ii) através da avaliação da equação de transferência radiativa, para os três satélites. Os resultados indicaram que o modelo de transferência radiativa em média superestima a temperatura de brilho nos canais sensíveis à superfície terrestre para os três satélites na região da América do Sul para os meses de verão. Além disso, as observações dos satélites incorporadas no sistema tiveram um aceite superior ao do satélite NOAA-15, de maneira que os satélites NOAA-18 e NOAA-19 podem ser incorporados no modo operacional do Sistema G3DVAR.
Channels sensitive to the terrestrial surface have not been widely used in Numerical Weather Prediction (NWP) due to the inherent complexities of the land surface. Thus, the present paper tested the tridimensional variational data assimilation framework implemented in the Global Model (G3DVAR) at the Center for Weather Forecast and Climate Studies (CPTEC) from the National Institute for Space Research (INPE) with near surface radiances from the NOAA-15 satellite. Since the operational version of the G3DVAR uses only NOAA-15, other experiments including the NOAA-18 and NOAA-19 satellites were performed. An assessment of the simulated brightness temperature for the near surface channels was made (i) through a comparison against observations and (ii) through a validation of the radiative transfer equation for the three satellites. The results indicate that the radiative transfer model overestimate, on average, the brightness temperature from the channels sensitive to the earth surface for all the considered satellites over South America during the austral summer. Furthermore, the number of observations accepted by the assimilation system increased substantially when NOAA-18 and NOAA-19 satellites were included, suggesting that incorporating these two new satellites should bring a positive impact to the G3DVAR operational system at CPTEC/INPE.
