In the current work, we conducted an immunocytochemical search for markers of ongoing neurogenesis (e.g. nestin) in auditory cortex from postmortem sections of autism spectrum disorder (ASD) and ...age-matched control donors. We found nestin labeling in cells of the vascular system, indicating blood vessels plasticity. Evidence of angiogenesis was seen throughout superior temporal cortex (primary auditory cortex), fusiform cortex (face recognition center), pons/midbrain and cerebellum in postmortem brains from ASD patients but not control brains. We found significant increases in both nestin and CD34, which are markers of angiogenesis localized to pericyte cells and endothelial cells, respectively. This labeling profile is indicative of splitting (intussusceptive), rather than sprouting, angiogenesis indicating the blood vessels are in constant flux rather than continually expanding.

(ED) leaves in five streams for 75, 56, 46, 26, 14, or 7 days. In all of the streams, leaf litter breakdown was higher for EC and lower in ED. This result may be related to different chemical characteristics among the five studied species. We observed higher frequency of shredders in MS and stream 4, and scrapers in streams 1 and 2. The concentration of ergosterol was higher in MS and lower in EC when adenosine triphosphate differed significantly among streams. Our data indicated that leaf litter quality (chemical and structural components) is a main factor affecting leaf breakdown in Amazonian streams.
Awareness and research on epilepsy‐related deaths (ERD), in particular Sudden Unexpected Death in Epilepsy (SUDEP), have exponentially increased over the last two decades. Most publications have ...focused on guidelines that inform clinicians dealing with these deaths, educating patients, potential risk factors and mechanisms. There is a relative paucity of information available for pathologists who conduct these autopsies regarding appropriate post mortem practice and investigations. As we move from recognizing SUDEP as the most common form of ERD toward in‐depth investigations into its causes and prevention, health professionals involved with these autopsies and post mortem procedure must remain fully informed. Systematizing a more comprehensive and consistent practice of examining these cases will facilitate (i) more precise determination of cause of death, (ii) identification of SUDEP for improved epidemiological surveillance (the first step for an intervention study), and (iii) biobanking and cell‐based research. This article reviews how pathologists and healthcare professionals have approached ERD, current practices, logistical problems and areas to improve and harmonize. The main neuropathology, cardiac and genetic findings in SUDEP are outlined, providing a framework for best practices, integration of clinical, pathological and molecular genetic investigations in SUDEP, and ultimately prevention.
Mesenchymal stromal cells (MSCs) can self-renew, differentiate into specialised cells and have different embryonic origins-ectodermal for dental pulp-derived MSCs (DPSCs) and mesodermal for adipose ...tissue-derived MSCs (ADSCs). Data on DPSCs adipogenic differentiation potential and timing vary, and the lack of molecular and genetic information prompted us to gain a better understanding of DPSCs adipogenic differentiation potential and gene expression profile. While DPSCs differentiated readily along osteogenic and chondrogenic pathways, after 21 days in two different types of adipogenic induction media, DPSCs cultures did not contain lipid vacuoles and had low expression levels of the adipogenic genes proliferator-activated receptor gamma (PPARG), lipoprotein lipase (LPL) and CCAAT/enhancer-binding protein alpha (CEBPA). To better understand this limitation in adipogenesis, transcriptome analysis in undifferentiated DPSCs was carried out, with the ADSC transcriptome used as a positive control. In total, 14,871 transcripts were common to DPSCs and ADSCs, some were unique (DPSCs: 471, ADSCs: 1032), and 510 were differentially expressed genes. Detailed analyses of overrepresented transcripts showed that DPSCs express genes that inhibit adipogenic differentiation, revealing the possible mechanism for their limited adipogenesis.
A finite volume based Eulerian-Lagrangian model has been created within OpenFOAM® in order to predict the behavior of particle clouds as well as particle deposition thicknesses on substrates under ...the influence of electro-static effects. The model resolves close to electrode effects as well as phenomena within the entire deposition chamber. It considers fluid dynamic effects, particle inertia, gravity, electric- as well as mechanic particle-particle interaction, corona formation, dynamic particle charging mechanisms, and coupling of particle motion to Reynolds-Averaged Navier-Stokes (RANS) based flow simulations. Resulting deposition pattern predictions were experimentally validated. It is demonstrated qualitatively and quantitatively that the measured deposition thicknesses and patterns vary by; i) applied voltage, ii) airflow rate, pistol-substrate iii) distance and iv) angle. Furthermore, the software has been prepared such that it works on the cloud computing software KaleidoSim®, which enables the simultaneous browser-based running of hundreds of cases for large parameter studies.
Even though biomass gasification remains a promising technology regarding de-centralized sustainable energy supply, its main limitations, namely the issues of unsteady operation, tar formation in ...after-treatment systems, and consequential high maintenance requirements, have never been fully overcome. In order to tackle the latter two deficiencies and to increase the understanding of thermodynamic and thermokinetic producer gas phase phenomena within the after-treatment zones, a numerical system dynamic model has been created. Thereby, naphthalene has been chosen to represent the behavior of tars. The model has been validated against a wide variety of measured and simulated producer gas compositions. This work particularly focuses on the investigation and minimization of tar formation within after-treatment systems at low pressures and decreasing temperatures. Model-based analysis has led to a range of recommended measures, which could reduce the formation tendency and thus the condensation of tars in those zones. These recommendations are (i) to decrease gas residence time within pipes and producer gas purification devices, (ii) to increase temperatures in low-pressure zones, (iii) to increase hydrogen to carbon ratio, and (iv) to increase oxygen to carbon ratio in the producer gas. Furthermore, the numerical model has been included in the cloud-computing platform KaleidoSim. Thus, a wider range of process parameter combinations could be investigated in reasonable time. Consequentially, a simulation-based sensitivity analysis of producer gas composition with respect to process parameter changes was conducted and the validity basis of the above recommendations was enlarged.
The G/C single-nucleotide polymorphism in the serotonin 1a receptor promoter, rs6295, has previously been linked with depression, suicide and antidepressant responsiveness. In vitro studies suggest ...that rs6295 may have functional effects on the expression of the serotonin 1a receptor gene (HTR1A) through altered binding of a number of transcription factors. To further explore the relationship between rs6295, mental illness and gene expression, we performed dual epidemiological and biological studies. First, we genotyped a cohort of 1412 individuals, randomly split into discovery and replication cohorts, to examine the relationship between rs6295 and five psychiatric outcomes: history of psychiatric hospitalization, history of suicide attempts, history of substance or alcohol abuse, current posttraumatic stress disorder (PTSD), current depression. We found that the rs6295G allele is associated with increased risk for substance abuse, psychiatric hospitalization and suicide attempts. Overall, exposure to either childhood or non-childhood trauma resulted in increased risk for all psychiatric outcomes, but we did not observe a significant interaction between rs6295 and trauma in modulating psychiatric outcomes. In conjunction, we also investigated the potential impact of rs6295 on HTR1A expression in postmortem human brain tissue using relative allelic expression assays. We found more mRNA produced from the C versus the G-allele of rs6295 in the prefrontal cortex (PFC), but not in the midbrain of nonpsychiatric control subjects. Further, in the fetal cortex, rs6295C allele exhibited increased relative expression as early as gestational week 18 in humans. Finally, we found that the C:G allelic expression ratio was significantly neutralized in the PFC of subjects with major depressive disorder (MDD) who committed suicide as compared with controls, indicating that normal patterns of transcription may be disrupted in MDD/suicide. These data provide a putative biological mechanism underlying the association between rs6295, trauma and mental illness. Moreover, our results suggest that rs6295 may affect transcription during both gestational development and adulthood in a region-specific manner, acting as a risk factor for psychiatric illness. These findings provide a critical framework for conceptualizing the effects of a common functional genetic variant, trauma exposure and their impact on mental health.

High purity copper, suitable for electrical applications, can only be obtained by electro-winning. The hallmark of this process is its self-induced natural convection through density variations of ...the electrolyte at both anode and cathode. In order to do this, first the full dynamic complexity of the process needs to be understood. Thus an OpenFoam®-based 2D model of the process has been created. This finite-volume multiphysics approach solves the laminar momentum and copper-ion species conservation equations, as well as local copper-ion conversion kinetics. It uses a Boussinesq approximation to simulate the species-momentum coupling, namely natural draft forces induced by variations of the spatial copper concentration within the fluid. The model shows good agreement with benchmark-cases of real-life electrochemical cells found in literature. An additional flow was imposed at the bottom of a small scale electrochemical cell in order to increase the ionic transport and thereby increase the overall performance of the cell. In a small scale electrochemical cell in strictly laminar flow, the overall performance could be increased and stratification decreased.
In the scope of this study an experimental powder coating setup is designed and the method to extract statistically significant trends from the data generated is developed. The ultimate goals are to ...i) validate a previously developed 3D Euler-LaGrangian numerical solver 1 and to ii) characterize the essential parameters for industrial powder coating processes in subsequent phases. The experiments involved coating a flat plate substrate with a corona spraying pistol. The resulting coating thickness has been quantified through the state of the art Coatmaster technology 2,3. The raw data generated from the Coatmaster has been filtered and rigorously analyzed to identify statistically significant trends. Furthermore, characteristic variables have been constructed for subsequent comparison to the numerical solver. This study reveals the challenges involved in assessing experimental data to extract meaningful comparisons for numerical solver validation.
