•A steroids-specific target library was constructed and was manually prepared.•The library enriches the potential targets of steroids out of target space.•Ranking by name instead of ...PDB ID makes target screening more efficiency and precise.•Protein flexibility was considered partially leading to more accurate prediction.
Steroids exist universally and play critical roles in various biological processes. Identifying potential targets of steroids is of great significance in studying their physiological and biochemical activities, the side effects and for drug repurposing. Herein, aiming at more precise steroids targets prediction, a steroids-specific target library integrating 3325 PDB or homology modeling structures categorized into 196 proteins was built by considering chemical similarity from DrugBank and biological processes from KEGG. The main properties of this library include: (1) It was manually prepared and checked to eliminate mistakes. (2) The library enriched the possible steroids targets and could decrease the false positives of structure-based target screening for steroids. (3) The ranking by protein name instead of PDB ID could make the screening more efficiency and precise. (4) Protein flexibility was taken into account partially by the different active conformations through the structural redundancy of each category of protein, which leads to more accurate prediction. The case studies of glycocholic acid and 24-epibrassinolide proved its powerful predictive accuracy. In summary, our strategy to build the steroids-specific protein library for steroids target prediction is a promising approach and it provides a novel idea for the target prediction of small molecules.
Infrared thermal imaging is a passive non-contact detection and identification technology, which is not subject to electromagnetic infection and good concealment, is widely used in military and ...commercial fields. However, due to the limitations of the existing infrared imaging system mechanisms, the spatial resolution of the acquired infrared images is low and the edge details are blurred, which in turn leads to poor performance in downstream missions based on infrared images. In this paper, in order to better solve the above problems, we propose a new super-resolution reconstruction framework for infrared images, called DBFE, which extracts and retains abundant structure and textual information for robust infrared image high-resolution reconstruction with a novel structure-textual encoder module. Extensive experiment demonstrates that our proposed method achieves significantly superior contraband high-resolution reconstruction results on the multiple dataset compared to progressive methods for high resolution infrared image reconstruction, effectively proving the practicability of the method proposed in this paper.
Selenized glucose can be easily prepared via the selenization reaction of glucose using in situ generated NaHSe as the selenization reagent. The technique has been industrialized to produce the ...chemical in kilogram scale, making it an easily available material in laboratory presently. The selenized glucose may be widely used as the starting material for the preparation of selenium-containing catalysts, as the organoselenium additive for feeds, and as the efficient selenium-enriched foliar fertilizers. In this work, we found that the treating Fusarium graminearum, a fungal pathogen inciting wheat scab disease, with selenium glucose could significantly inhibit the generation of the deoxynivalenol (DON) toxin, which might be a breakthrough for reducing the detriment of wheat scab disease.

•Identification of endogenous peptides in P. cyrtonema for the first time.•Explored the effect of alkaline extraction and acid precipitation on P. cyrtonema endogenous ...peptides.•Prediction of antioxidant peptide activity in P. cyrtonema base on bioinformatics analysis combine with molecular docking.
Polygonatum cyrtonema Hua (P. cyrtonema), a medicinal and edible, has a long-standing tradition of treating respiratory diseases, regulating immune function, and enhancing liver health. Although polysaccharides, saponins, and flavonoids have been identified as its major bioactive components, the presence and properties of bioactive peptides in P. cyrtonema are relatively understudied. This study utilized an alkali extraction-acid precipitation method to extract crude protein from P. cyrtonema. Subsequently, Nano-LC-Q Exactive MS was utilized to characterize successfully the sequences and abundances of endogenous peptides from different sources. A total of 2571 M1(endogenous) peptides and 2122 M2(crude protein) peptides were identified using a combination of database searches and de novo strategies, with significantly higher confidence levels observed for M2 peptides identified through the database search approach. Further analysis of subsequent overall differences reveals that, based on the chemical properties and bioinformatics analyses of peptides from different sources, the M1 peptide primarily consists of 4–6 amino acids (62.1 %), while the M2 peptide is mainly composed of more than 8 amino acids (60.8 %). Molecular docking technology was then applied to verify the antioxidant prediction results and to evaluate the differences in antioxidant potential between M1 peptides and M2 peptides, with seven of the top ten ranked peptides in terms of docking energy being M1 peptides. These findings notably advance our understanding of endogenous peptides in P. cyrtonema and provide valuable reference information for identifying potential candidate bioactive compounds. This information will support future comprehensive validation of the biological activities of these peptides in P. cyrtonema and contribute to drug discovery research.
Lignification, which occurs in many horticultural fruit and vegetables, brings about undesirable texture and unfavorable consumer preference. However, this problem has rarely been studied. In this ...work, three pumelo cultivars cvs "Hongroumiyou" (HR), "Bairoumiyou" (BR), and "Huangroumiyou" (HuR) were stored at 25 °C for 90 days, and juice sacs were sampled to explore the lignin accumulation and its relationship to sucrose and energy depletion were investigated. The results displayed that HuR contained lower sucrose content, lower ATP level, but higher lignin content compared to BR and HR during postharvest storage, indicating that the sequence according to storage resistance on the basis of lignin content is as follows: HuR < BR < HR. Furthermore, sucrose degradation attributed to enhanced activities of neutral invertase (NI), soluble acid invertase (S-AI), cell wall-bound invertase (B-AI), and energy deficit on account of declined ATP level, showed significantly negative correlation with lignin accumulation, suggesting that lignin accumulation occurrence could induce sucrose degradation and energy deficit during postharvest storage. Additionally, higher activities of phenylalanine ammonia-lyase (PAL), polyphenol oxidase (PPO), peroxidase (POD) could accelerate lignin synthesis and resulted in lignin accumulation during postharvest pumelo storage.
Memory formation for a weak passive avoidance task in the dark-incubated chicks is facilitated by light exposure or corticosterone administration at optimally pre-hatch time points. To explore the ...potential mechanisms underlying activation of brain memory function development by light or corticosterone exposure during late embryo, steroid receptor antagonist RU486, or protein synthetic inhibitor anisomycin, was administered intraembryonically to the embryos of either only 24-h light-exposure or complete darkhatched on embryonic day 20 (E20). The results showed that RU486 and anisomycin significantly retarded the facilitated retention both by light and corticosterone exposure in the dark-incubated chicks. They also suggest that the act of corticosterone or light exposure on the development of brain memory function is mediated by the effect of steroid receptor, or afterward on related protein syntheses that is involved in memory formation of post-hatched performance of day-old chicks.
The effect of Batroxobin expression of neural cell adhesion molecule (NCAM) in left temporal ischemic rats with spatial memory disorder was investigated by means of Morri's water maze and ...immunohistochemical methods. The results showed that the mean reaction time and distance of temporal ischemic rats for searching a goal were significantly longer than those of sham-operated rats and at the same time NCAM expression of left temporal ischemic region was significantly increased. However, the mean reaction time and distance of Batroxobin-treated rats were shorter and they used normal strategies more often and earlier than those of ischemic rats. The number of NCAM immune reactive cells of Batroxobin-treated rats was more than that of ischemic group. In conclusion, Batroxobin can improve spatial memory disorder of temporal ischemic rats and the regulation of the expression of NCAM is probably related to the neuroprotective mechanism.
The effect of Batroxobin on expression of c-Jun in left temporal ischemic rats with spatial memory disorder was investigated by means of Morri's water maze and immunohistochemistry methods. The ...results showed that the mean reaction time and distance of temporal ischemic rats for searching a goal were significantly longer than those of sham-operated rats, and at the same time c-Jun expression of left temporal ischemic region was significantly increased. However, the mean reaction time and distance of Batroxobin-treated rats were shorter and they used normal strategies more often and earlier than those of ischemic rats. The number of c-Jun immune reactive cells of Batroxobin-treated rats was also less than that of ischemic group. In conclusion, Batroxobin can improve spatial memory disorder in temporal ischemic rats, and the down-regulation of the expression of c-Jun is probably related to the neuroprotective mechanism.
