The ranges of wolves (Canis lupus) and bears (Ursus arctos) across Europe have expanded recently, and it is important to assess public attitudes toward this expansion because responses toward these ...species vary widely. General attitudes toward an object are good predictors of broad behavioral patterns; thus, attitudes toward wolves and bears can be used as indicators to assess the social foundation for future conservation efforts. However, most attitude surveys toward bears and wolves are limited in scope, both temporally and spatially, and provide only a snapshot of attitudes. To extend the results of individual surveys over a much larger temporal and geographical range so as to identify transnational patterns and changes in attitudes toward bears and wolves over time, we conducted a meta‐analysis. Our analysis included 105 quantitative surveys conducted in 24 countries from 1976 to 2012. Across Europe, people's attitudes were more positive toward bears than wolves. Attitudes toward bears became more positive over time, but attitudes toward wolves seemed to become less favorable the longer people coexisted with them. Younger and more educated people had more positive attitudes toward wolves and bears than people who had experienced damage from these species, and farmers and hunters had less positive attitudes toward wolves than the general public. For bears attitudes among social groups did not differ. To inform conservation of large carnivores, we recommend that standardized longitudinal surveys be established to monitor changes in attitudes over time relative to carnivore population development. Our results emphasize the need for interdisciplinary research in this field and more advanced explanatory models capable of capturing individual and societal responses to changes in large carnivore policy and management.

The acknowledgement of uncertainty and complexity in social-ecological systems has increased the implementation of collaborative governance regimes for environmental issues. The performance of these new regimes to deliver favourable social and ecological outcomes must therefore be evaluated. We focus on the case of Swedish wildlife governance, which has a tradition of using collaborative elements. In relation to moose (Alces alces), these collaborative aspects were recently formalized in an amended policy. We aim to assess some aspects of this new regime’s performance with respect to intermediate ecological outcomes (i.e. quota fulfilment). We use path analysis to test the causal effects of system context and collaboration dynamics on governance outcomes. Collaboration dynamics were assessed using a web-based survey sent to all stakeholders in Moose Management Groups (response rate = 82 %). Our originally specified model yielded a good fit (SRMR of .030 and robust TLI of .996) and explained 20 % of the variation in outcomes. Context variables revealed significant direct effects on collaboration dynamics and outcomes. Larger Moose Management Areas and fluctuations in forage availability required more time investment from actors, while high land use diversity and density of other ungulate species negatively affected moose quota fulfilment. Moose Management Groups that invested more time and perceived to have a good knowledge base achieved better quota fulfilment. Collaboration dynamics thus had a positive direct effect on outcomes. From a policy perspective, our results raise questions regarding institutional fit because context factors had significant negative effects on collaboration dynamics and the outcomes of the collaborative process.
Hyperthermia is a form of a cancer treatment which is frequently applied in combination with radiotherapy (RT) to improve therapy responses and radiosensitivity. The mode of action of hyperthermia is ...multifactorial; the one hand by altering the amount of the blood circulation in the treated tissue, on the other hand by modulating molecular pathways involved in cell survival processes and immunogenic interactions. One of the most dominant proteins induced by hyperthermia is the major stress‐inducible heat shock protein 70 (Hsp70). Hsp70 can be found in the blood either as a free‐protein (free HSP70) derived from necrotic cells, or lipid‐bound (liposomal Hsp70) when it is actively released in extracellular vesicles (EVs) by living cells. The aim of the study was to evaluate the levels of free and liposomal Hsp70 before and after treatment with RT alone or hyperthermia combined with radiotherapy (HTRT) in dogs and cats to evaluate therapy responses. Peripheral blood was collected from feline and canine patients before and at 2, 4, 6 and 24 h after treatment with RT or HTRT. Hsp70 enzyme‐linked immunosorbent assays (ELISAs) were performed to determine the free and liposomal Hsp70 concentrations in the serum. The levels were analysed after the first fraction of radiation to study immediate effects and after all applied fractions to study cumulative effects. The levels of free and liposomal Hsp70 levels in the circulation were not affected by the first singular treatment and cumulative effects of RT in cats however, after finalizing all treatment cycles with HTRT free and liposomal Hsp70 levels significantly increased. In dogs, HTRT, but not treatment with RT alone, significantly affected liposomal Hsp70 levels during the first fraction. Free Hsp70 levels were significantly increased after RT, but not HTRT, during the first fraction in dogs. In dogs, on the other hand, RT alone resulted in a significant increase in liposomal Hsp70, but HTRT did not significantly affect the liposomal Hsp70 when cumulative effects were analysed. Free Hsp70 was significantly induced in dogs after both, RT and HTRT when cumulative effects were analysed. RT and HTRT treatments differentially affect the levels of free and liposomal Hsp70 in dogs and cats. Both forms of Hsp70 could potentially be further investigated as potential liquid biopsy markers to study responses to RT and HTRT treatment in companion animals.
Hyperthermia (HT) as an adjuvant to radiation therapy (RT) is a multimodality treatment method to enhance therapeutic efficacy in different tumours. High demands are placed on the hardware and ...treatment planning software to guarantee adequately planned and applied HT treatments. The aim of this prospective study was to determine the effectiveness and safety of the novel HT system in tumour‐bearing dogs and cats in terms of local response and toxicity as well as to compare planned with actual achieved data during heating. A novel applicator with a flexible number of elements and integrated closed‐loop temperature feedback control system, and a tool for patient‐specific treatment planning were used in a combined thermoradiotherapy protocol. Good agreement between predictions from planning and clinical outcome was found in 7 of 8 cases. Effective HT treatments were planned and verified with the novel system and provided improved quality of life in all but 1 patient. This individualized treatment planning and controlled heat exposure allows adaptive, flexible and safe HT treatments in palliatively treated animal patients.
The intact nasal barrier is a prerequisite for a functioning defense of the upper airway system, in particular the permanent threat by inhaled potentially harmful microorganisms. Antimicrobial ...peptides (AMP) play an important role in maintaining barrier function. There is few data about AMP in respect of nasal mucosa. This study is addressed to gain further insight into the differential AMP expression and secretion pattern according to defined anatomical regions of the vestibulum nasi and turbinates.
ELISA was applied to quantify concentrations of AMP RNase-7, psoriasin, hBD-2, hBD-3 and LL-37 in nasal secretions of 20 healthy volunteers. Immunohistochemistry was used to detect the local cellular sources of AMP in the vestibulum nasi (squamous epithelium) and compared to the mucosa of the turbinates (pseudostratified epithelium) in 10 healthy volunteers.
Expression of RNase 7 and psoriasin was detected in all nasal secretion specimens, whereas LL-37 was detected in 16, hBD-2 in 5 and hBD-3 in 6 specimens. In the vestibulum nasi, luminal cell layers were demonstrated as local cellular sources for hBD-3 and RNase 7, whereas psoriasin was found in all layers of the stratified squamous epithelium. LL-37 was detected in 1 stroma cells sample, whereas hBD-2 was not detected at all. In turbinate biopsie,s hBD-3 and LL-37 were detectable in the epithelium, stroma cells and submucosal glands. RNase 7 was only present in submucosal glands. HBD-2 and psoriasin were not detected.
These data demonstrate that the nasal epithelium contains a chemical defense shield through the expression and secretion of various AMP.
Cladophialophora boppii is a black yeast-like fungus that up to now has been only rarely described as a cause of human infection and whose role as a pathogen was not established despite its repeated ...isolation and genetic identification in these reports. Here we report the first case of a verified toenail infection caused by this fungus in a woman without any systemic disease or evidence of immunodeficiency. Identical dark molds were isolated from the same toenail at three points of time. Species identification was performed by scrutinizing the isolates morphologic, physiologic and genetic characteristics which resulted in their identification as Cladophialophora boppii. Oral treatment with terbinafin plus topical ciclopiroxolamine was effective.
Collaborative governance regimes may be vulnerable because of dependency on stakeholders??? voluntary engagement and efforts. This study focuses on the Swedish moose management system, a multi-level ...collaborative governance regime inspired by the ecosystem approach. Self-determination theory is used to explore perceived prerequisites of basic needs for intrinsic motivation across sub-groups of stakeholder representatives who are engaged across different social-ecological contexts. Questionnaire data collected among representatives at two governance levels, moose management groups (n = 624) and moose management units (n = 979), were subjected to two-step cluster analysis. The analyses revealed two sub-groups of representatives, characterized by differences in species composition and land ownership structure: managers of multi-ungulate areas and managers of large-carnivore areas. In several respects, these groups significantly differed in how they perceived the prerequisites. This included prerequisites of perceived competence with regard to their need for knowledge of topics and usefulness of monitoring methods, perceived autonomy operationalized as possibilities to perform their tasks with sufficient time, resources, and support from their organizations, and perceived relatedness to different groups of actors. Further efforts should be made to understand the conditions required for representatives to energize and direct their behavior. The institutional system must better fit the needs of stakeholder representatives across various local contexts, otherwise the space for local voluntary engagement might be hampered.
Tissue injury is a common sequela of acute virus infection localized to a specific organ such as the lung. Tissue injury is an immediate consequence of infection with lytic viruses. It can also ...result from the direct destruction of infected cells by effector CD8(+) T lymphocytes and indirectly through the action of the T cell-derived proinflammatory cytokines and recruited inflammatory cells on infected and uninfected tissue. We have examined CD8(+) T cell-mediated pulmonary injury in a transgenic model in which adoptively transferred, virus-specific cytotoxic T lymphocytes (CTLs) produce lethal, progressive pulmonary injury in recipient mice expressing the viral target transgene exclusively in the lungs. We have found that over the 4-5 day course of the development of lethal pulmonary injury, the effector CTLs, while necessary for the induction of injury, are present only transiently (24-48 h) in the lung. We provide evidence that the target of the antiviral CD8(+) T cells, the transgene expressing type II alveolar cells, are not immediately destroyed by the effector T cells. Rather, after T cell-target interaction, the type II alveolar cells are stimulated to produce the chemokine monocyte chemoattractant protein 1. These results reinforce the concept that, in vivo, the cellular targets of specific CTLs may participate directly in the development of progressive tissue injury by activating in response to interaction with the T cells and producing proinflammatory mediators without sustained in vivo activation of CD8(+) T cell effectors.
A basic step in understanding the dynamics of a fish population is to quantify movement and mortality rates. Conventional mark-recapture experiments have provided the foundation for studies on animal ...movement, particularly for fish. Previous studies have shown rapid mixing of sablefish (Anoplopoma fimbria) among fishery regulatory areas, with the pattern of movement related to fish size. Over 300 000 tag releases in Alaska and over 27 000 tag recoveries from 1979 to 2009 were analyzed. We used a Markov model to quantify annual movement probabilities among areas for three size groups of sablefish. The negative-binomial likelihood was used to model the tag-recovery data because of significant overdispersion. Annual movement probabilities were high, ranging from 10% to 88% depending on area of occupancy at each time step and size group. Overall, movement probabilities were very different between areas of occupancy and moderately different between size groups. Estimated annual movement of small sablefish from the central Gulf of Alaska had the reverse pattern of a previous study, with 29% moving westward and 39% moving eastward. Movement probabilities also varied annually, with decreasing movement until the late 1990s and increasing movement until 2009. Year-specific magnitude in movement probability of large fish was highly negatively correlated with female spawning biomass estimates from the federal stock assessment. Mean mortality estimates from time at liberty were similar to the federal stock assessment. Incorporating these tag-recovery and movement data into a fully age-structured spatial stock assessment model will inform harvest apportionment strategies to conserve spawning biomass and maximize future yields.
Population assessments seldom incorporate habitat information or use previously observed distributions of fish density. Because habitat affects the spatial distribution of fish density and overall ...abundance, the use of habitat information and previous estimates of fish density can produce more precise and less biased population estimates. In this study, we describe how poststratification can be applied as an unbiased estimator to data sets that were collected under a probability sampling design, typical of many multispecies trawl surveys. With data from a multispecies survey of juvenile flatfish, we show how poststratification can be applied to a data set that was not collected under a probability sampling design, where both the precision and the bias are unknown. For each of four species, three estimates of total abundance were compared: 1) unstratified; 2) poststratified by habitat; and 3) poststratified by habitat and fish density (high fish density and low fish density) in nearby years. Poststratification by habitat gave more precise and (or) less design-biased estimates than an unstratified estimator for all species in all years. Poststratification by habitat and fish density produced the most precise and representative estimates when the sample size in the high fish-density and low fish-density strata were sufficient (in this study, n greater than or equal to 20 in the high fish-density stratum, n greater than or equal to 9 in the low fish-density stratum). Because of the complexities of statistically testing the annual stratified data, we compared three indices of abundance for determining statistically significant changes in annual abundance. Each of the indices closely approximated the annual differences of the poststratified estimates. Selection of the most appropriate index was dependent upon the species' density distribution within habitat and the sample size in the different habitat areas. The methods used in this study are particularly useful for estimating individual species abundance from multispecies surveys and for retrospective studies.
