European guidelines recommend the routine offer of an HIV test in patients with a number of AIDS-defining and non-AIDS conditions believed to share an association with HIV; so called indicator ...conditions (IC). Adherence with this guidance across Europe is not known. We audited HIV testing behaviour in patients accessing care for a number of ICs. Participating centres reviewed the case notes of either 100 patients or of all consecutive patients in one year, presenting for each of the following ICs: tuberculosis, non-Hodgkins lymphoma, anal and cervical cancer, hepatitis B and C and oesophageal candidiasis. Observed HIV-positive rates were applied by region and IC to estimate the number of HIV diagnoses potentially missed. Outcomes examined were: HIV test rate (% of total patients with IC), HIV test accepted (% of tests performed/% of tests offered) and new HIV diagnosis rate (%). There were 49 audits from 23 centres, representing 7037 patients. The median test rate across audits was 72% (IQR 32-97), lowest in Northern Europe (median 44%, IQR 22-68%) and highest in Eastern Europe (median 99%, IQR 86-100). Uptake of testing was close to 100% in all regions. The median HIV+ rate was 0.9% (IQR 0.0-4.9), with 29 audits (60.4%) having an HIV+ rate >0.1%. After adjustment, there were no differences between regions of Europe in the proportion with >0.1% testing positive (global p = 0.14). A total of 113 patients tested HIV+. Applying the observed rates of testing HIV+ within individual ICs and regions to all persons presenting with an IC suggested that 105 diagnoses were potentially missed. Testing rates in well-established HIV ICs remained low across Europe, despite high prevalence rates, reflecting missed opportunities for earlier HIV diagnosis and care. Significant numbers may have had an opportunity for HIV diagnosis if all persons included in IC audits had been tested.
In an ecological survey of nitrogen-fixing bacteria isolated from the rhizosphere and as endophytes of sugarcane, maize and teosinte plants in Brazil, Mexico and South Africa, a new phylogenetically ...homogeneous group of N(2)-fixing bacteria was identified within the genus Burkholderia. This polyphasic taxonomic study included microscopic and colony morphology, API 20NE tests and growth on different culture media at different pH and temperatures, as well as carbon source assimilation tests and whole-cell protein pattern analysis. Analysis of 16S rRNA gene sequences showed 99.2-99.9 % similarity within the novel species and 97.2 % similarity to the closest related species, Burkholderia sacchari. The novel species was composed of four distinct amplified 16S rDNA restriction analysis groups. The DNA-DNA reassociation values within the novel species were greater than 70 % and less than 42 % for the closest related species, B. sacchari. Based on these results and on many phenotypic characteristics, a novel N(2)-fixing species is proposed for the genus Burkholderia, Burkholderia tropica sp. nov., with the type strain Ppe8(T) (=ATCC BAA-831(T)=LMG 22274(T)=DSM 15359(T)). B. tropica was isolated from plants grown in geographical regions with climates ranging from temperate subhumid to hot humid.
We compare the effects of three different approved sources of supplemental zilpaterol on growth-performance responses and carcass characteristics of finishing lambs.
Twenty four Pelibuey×Katahdin ...lambs (46.75±2.43 kg) were used in a 33-day feeding trial. Lambs were fed a dry rolled corn-based finishing diet. Treatments consisted of the non-supplemental basal diet (Control) versus the basal diet supplemented with 125 mg zilpaterol/kg of diet (as fed basis) from three commercial sources marketed in Mexico: Zilmax (ZIL), Grofactor, and Zipamix.
Compared to controls, zilpaterol (ZH) supplementation did not affect dry matter intake (DMI), but increased carcass adjusted daily weight gain (ADG, 36.7%), gain efficiency (34.2%), and dietary net energy (26.0%), and decreased (23.4%) the ratio of observed:expected DMI. Compared to controls, supplemental ZH increased hot carcass weight (6.4%), dressing percentage (3.2%), m. longissimus thoracis (LM) area (15.6%), and shoulder muscle:fat ratio (28.7%), but decreased kidney-pelvic-heart fat, and fat thickness. Supplemental ZH increased 10.9% and 14.3% whole cut weight of loin and leg, respectively, and the proportion (as percentage of cold carcass weight) of leg (4.3%). These increases were reflected in greater forequarter and hindquarter weights. Lambs fed ZH increased (4.6%) empty body weight (EBW) and reduced (14.7%) liver/spleen weight (as g/kg EBW). Likewise, ZH supplementation tended (p = 0.08) to lower (8.9%) visceral fat. Growth performance, energetic efficiency, hot carcass weight, dressing percentage, LM area and whole cuts were not different across supplemental ZH sources. However, compared with non-supplemented controls, only ZIL appreciably decreased carcass fat distribution, including fat thickness, percentage kidney pelvic and heart fat, shoulder fat, and visceral fat.
Supplemental ZH increases ADG, gain efficiency, carcass dressing percentage, and LM area. The magnitude of these responses was similar among ZH sources. Nevertheless, compared with non-supplemented controls, only ZIL appreciably decreases carcass fat. The basis for this is uncertain, but indicative that some practical differences in zilpaterol bio-equivalency may exist across commercial sources tested.
Natural vanilla extract (NVE) and synthetic vanilla extract (SVE) were microencapsulated by spray-drying, using whey protein concentrate (WPC) as wall material. Physicochemical properties, volatility ...profile, vanillin concentration and antioxidant activity were evaluated by measuring total polyphenols, the inhibition of radical scavenging activity by DPPH, chain-breaking activity and reducing power. Both microparticles exhibited high vanillin retention and antioxidant activity during spray dried process, but microparticles of SVE-WPC showed higher vanillin retention than microparticles of NVE-WPC during storage. The micrographs of the microcapsules showed morphological characteristics that were ideal for ensuring stability and integrity during storage to different water activities. WPC was found a suitable wall material to produce powder with vanilla extracts. Vanilla powder represents an interesting food additive for incorporation into functional foods.

In this paper, the Ramon starch was isolated and its chemical composition and physical and microscopic characteristics were determined. Corn starch was used as reference. In general, the proximal composition was similar between starches studied. Ramon starch granules were oval–spherical and rounded with sizes between 6.5 and 15μm. Starch purity was high (92.57%) with amylose content of 25.36%. The gelatinization temperature was 83.05°C and transition enthalpy was 21.423J/g. At 90°C, solubility was 20.42%, swelling power 17.64gwater/gstarch and water absorption capacity was 13gwater/gstarch. The pH, clarity and color (Hue angle) of Ramon starch were higher to those reported for corn starch. The results achieved suggest that Ramon starch has potential for application in food systems requiring high processing temperatures and it is also a promising option for use in the manufacture of biodegradable materials.
Choline is a precursor of acetylcholine, phosphatidylcholine, and the methyl-donor betaine. Reports indicate that supplementation with rumen-protected choline improves postpartum reproductive ...function of dairy cows. The objective was to determine whether addition of choline to culture medium of in vitro-produced embryos alters the phenotype of the resultant blastocysts. Treatments were choline chloride (ChCl; 0.004, 1.3, 1.8, and 6.37 mM) and phosphatidylcholine (1.3 mM). Treatment with 0.004 mM ChCl improved development to the blastocyst stage, increased blastocyst cell number, and increased the percentage of blastocysts that were hatching or hatched. Development was not affected by higher concentrations of ChCl but was reduced by 1.3 mM phosphatidylcholine. Treatment of embryos with 1.3 mM ChCl (but not other concentrations) increased expression in blastocysts of 11 of 165 genes examined (AMOT, NANOG, HDAC8, HNF4A, STAT1, MBNL3, SOX2, STAT3, KDM2B, SAV1, and GPAM) and decreased expression of one gene (ASS1). Treatment with 1.3 mM ChCl decreased global DNA methylation at d 3.5 of development and increased DNA methylation at d 7.5 in blastocysts. Treatment with 1.8 mM ChCl also increased methylation in blastocysts. In conclusion, addition of choline to the culture medium alters the phenotype of preimplantation bovine embryos produced in vitro. Choline chloride can act in a concentration-dependent manner to alter development, expression of specific genes, and DNA methylation.
Assessment of tumor-infiltrating lymphocytes (TILs) in histopathologic specimens can provide important prognostic information in diverse solid tumor types, and may also be of value in predicting ...response to treatments. However, implementation as a routine clinical biomarker has not yet been achieved. As successful use of immune checkpoint inhibitors and other forms of immunotherapy become a clinical reality, the need for widely applicable, accessible, and reliable immunooncology biomarkers is clear. In part 1 of this review we briefly discuss the host immune response to tumors and different approaches to TIL assessment. We propose a standardized methodology to assess TILs in solid tumors on hematoxylin and eosin sections, in both primary and metastatic settings, based on the International Immuno-Oncology Biomarker Working Group guidelines for TIL assessment in invasive breast carcinoma. A review of the literature regarding the value of TIL assessment in different solid tumor types follows in part 2. The method we propose is reproducible, affordable, easily applied, and has demonstrated prognostic and predictive significance in invasive breast carcinoma. This standardized methodology may be used as a reference against which other methods are compared, and should be evaluated for clinical validity and utility. Standardization of TIL assessment will help to improve consistency and reproducibility in this field, enrich both the quality and quantity of comparable evidence, and help to thoroughly evaluate the utility of TILs assessment in this era of immunotherapy.
Starch sources with different physicochemical properties are demanded by new applications that require specific properties. Ramon starch was characterized by X-ray diffraction, thermogravimetric ...analysis, fourier transform infrared spectroscopy (FTIR) analysis and rheology, and compared to corn starch. Ramon starch exhibited a C-type X-ray diffraction pattern. According to thermogravimetric analysis, degradation of corn and Ramon starches occurred in the temperature range between 292 and 369 °C, which coincides with the degradation of other starches studied. Pasting properties varied among the corn and Ramon starches. Ramon and corn starches were characterized as viscoelastic systems with G′ > G″, during the stages of heating-cooling kinetics. These results support the potential use of Ramon starch as a thickening and gelling agent in food, an excipient in pharmaceutical solid forms, and as biodegradable polymers for food packaging.

Four Holstein steers with ruminal and duodenal cannulas were used in a 4 × 4 Latin square design to examine the effect of daily intake of 0, 2, 4 or 6 g/steer of standardized plant extract containing ...a mixture of quaternary benzophenanthridine alkaloids and protopine alkaloids (QBA+PA) on the characteristics of ruminal fermentation and characteristics of digestion. The basal diet consisted of a steam-flaked corn-based finishing diet that contained 62% corn and 12% sudangrass hay and the rest of diet was composed of mainly dried distillers grains, molasses, fat, and minerals. The source of QBA+PA used was Sangrovit-RS (Phytobiotics Futterzusatzstoffe GmbH, Eltville, Germany) and supplementation levels of 2, 4, and 6 g Sangrovit-RS∙steer∙d, which represented a net daily ingestion of approximately 6, 12, and 18 mg of QBA+PA compounds, respectively. Inclusion of QBA+PA linearly increased ( = 0.04) flow to the duodenum of nonammonia N and linearly decreased ( < 0.01) duodenal flows of ammonia N. Ruminal microbial efficiency (duodenal microbial N; g/kg OM fermented in the rumen) and protein efficiency (duodenal nonammonia N; g/g N intake) were increased ( < 0.05) as the level of QBA+PA increased. There were no effects of QBA+PA supplementation on ruminal, postruminal, and total tract digestion of OM, starch, and NDF, but postruminal and total tract digestion of N increased ( < 0.01) as the level of QBA+PA increased. Digestible energy of the diet tended to increase (linear affect, = 0.09) with QBA+PA supplementation. Ruminal pH and total VFA molar concentrations were not different between treatments. Ruminal NH-N concentration linearly decreased ( = 0.02) with QBA+PA supplementation. Ruminal molar proportion of acetate increased ( = 0.04) as the supplementation level of QBA+PA increased. It is concluded that QBA+PA supplementation enhances efficiency of N utilization in feedlot steers fed a steam-flaked corn-based finishing diet. This effect was due, in part, to enhanced ruminal microbial efficiency, decreased ruminal degradation of dietary nonammonia N, and enhanced postruminal N digestion.

Probiotics (beneficial living microorganisms) and prebiotics (fiber, cell wall material, mannan polysaccharides derived through hydrolysis of yeast cell walls) are feed additives that may have beneficial extra-nutritional pharmaceutical and/or metabolic effects on livestock health and growth performance. Due to differences in modes of action, their combination may have additive effects on digestion and fermentation in ruminants. For this reason, four male lambs (Dorper; 45.1 ± 2.7 kg initial weight) with “T” cannulas in the rumen were used in a 4 × 4 Latin square experiment to evaluate the effects of single or combined supplementation of probiotics and prebiotics on ruminal fermentation and total tract digestion. Dietary treatments consisted of a cracked corn-based basal finishing diet supplemented with: 1) no eubiotics (Control); 2) 3 g of live saccharomyces cerevisiae /lamb/day (2 × 1010 cfu/g, SC), 3) 3 g of mannan oligosaccharide (30 % w/w) plus b-glucans (20 % w/w) /lamb/day (MOS), and 4) combination of 1.5 g/day SC and 1.5 g/day MOS (SCMOS). Compared to controls, SC supplementation tended (P = 0.09) to increase total tract digestion of DM and OM, and increased (P < 0.05) total tract digestion of N, starch, and digestible energy (DE). Compared with Control, MOS increased total tract NDF (7.9 %, P < 0.01) and starch digestion, and tended to improve total tract digestion of (P = 0.09), N (P = 0.07), and DE diet (P = 0.07). Compared to Control, SCMOS increased (P ≤ 0.03) total tract digestion of all fractions evaluated, including a 4.2 % (P < 0.01) increase in DE. Although lambs fed MOS had greater (6.7 %, P = 0.02) NDF digestion than those fed SC, differences in total tract digestion of DM, OM, N, starch and DE diet were not appreciable (P ≥ 0.24). Compared with SC and MOS fed separately, SCMOS increased (P < 0.05) total tract digestion of N and NDF. Supplemental MOS and SCMOS tended (P = 0.09) to promote greater ruminal pH than the Control. Combining supplementation decreased (P ≤ 0.03) the molar proportion of butyrate and ruminal ammonia, consistent with decreased of ruminal concentration of C. aminophilum. Probiotic/prebiotic supplementation of high-energy lamb finishing diets enhances total tract digestion and digestible energy. Reduction hyper-ammonia producing ruminal bacteria with the combination probiotic plus prebiotic may contribute to improved dietary N economy. The combination of probiotics with prebiotics potentiate positive effects on digestion and ruminal fermentation in lambs fed a high-energy diets.
