We report clinical and biochemical finding from three unrelated patients presenting ONCE (Optic Neuropathy, Cardiomyopathy and Encephalopathy with lactic acidosis and combined oxidative ...phosphorylation deficiency) syndrome. Whole‐exome sequencing (WES) of the three patients and the healthy sister of one of them was used to identify the carry gene. Clinical and biochemical findings were used to filter variants, and molecular, in silico and genetic studies were performed to characterize the candidate variants. Mitochondrial DNA (mtDNA) defects involving mutations, deletions or depletion were discarded, whereas WES uncovered a double homozygous mutation in the MTO1 gene (NM_001123226:c.1510C>T, p.R504C, and c.1669G>A, p.V557M) in two of the patients and the homozygous mutation p.R504C in the other. Therefore, our data confirm p.R504C as pathogenic mutation responsible of ONCE syndrome, and p.V557M as a rare polymorphic variant.
The R504C mutation in MTO1 gene is the responsible of ONCE syndrome (Optic Neuropathy, Cardiomyopathy and Encephalopathy with lactic acidosis and combined OXPHOS deficiency) and V557M is a rare polymorphic variant.
Blind equalizers avoid the transmission of pilot sequences, allowing a more efficient use of the channel bandwidth. Normally, after a first rough equalization is achieved, it is necessary to switch ...these equalizers to a decision-directed (DD) mode to reduce the steady-state mean-square error (MSE) to acceptable levels. The selection of an appropriate MSE threshold for switching between the blind and the DD modes is critical to obtain a good overall performance; however, this is not an easy task, since it depends on several factors such as the signal constellation, the communication channel, or the signal-to-noise ratio. In this paper, we propose an equalization scheme that adaptively combines a blind and a DD equalizers running in parallel. The combination is itself adapted in a blind manner, and as a result the overall scheme can automatically switch between the component filters, avoiding the need to set the transition MSE level a priori. The performance of our proposal is illustrated both analytically and through an extensive set of simulations, where we show its advantages with respect to existing hard- and soft-switching equalization schemes.

Analysis of the potentiostatic current density transients allowed to establish that the zinc electrodeposition mechanism occurs via the simultaneous presence of a Langmuir-type adsorption–desorption equilibrium, an instantaneous nucleation process with two-dimensional (2D) growth limited by the rate of lattice incorporation, and a diffusion-controlled three-dimensional nucleation and growth contribution (3D), which is particularly notorious at 348 K. This is the first time that a 2D-3D nucleation transition has been observed during the electrochemical deposition of metals from deep eutectic solvents (DES). From the 2D nucleation process contribution to the total current density, it was possible to determine the surface roughness factor and the electrochemically active surface area of the glassy carbon electrode (GCE). The time evolution of the Zn monolayer formation onto the GCE was also reported.
Niemann–Pick disease type C (NP-C) is an inherited neurovisceral lysosomal lipid storage disease characterized by progressive neurological deterioration. Different clinical forms have been defined ...based on patient age at onset: perinatal, early-infantile (EI), late-infantile (Li), juvenile and adult. We evaluated the efficacy and tolerability of miglustat in 16 symptomatic NP-C patients, with comparative reference to one neurologically asymptomatic, untreated patient. All patients were categorized according to age at neurological disease onset, and were assessed using a standardized clinical assessment protocol: disability and cognitive function scales, positron emission tomography (PET), and biochemical markers. PET and disability scale evaluations indicated that cerebral hypometabolism and neurological symptoms were stabilized during treatment in juvenile-onset NP-C patients. EI and Li NP-C patients, who had higher disease severity at baseline (treatment start), showed increased disability scores and progressive cerebral hypometabolism during follow up. Similarly, while cognitive scale scores remained relatively stable in patients with juvenile NP-C, cognition deteriorated in EI and Li patients. Plasma chitotriosidase (ChT) activity was lower in the juvenile NP-C subgroup than in EI and Li patients, and generally increased in patients who discontinued treatment. Plasma CCL18/PARC and ChT activities indicated greater macrophagic activity in EI and Li patients
versus juveniles. Miglustat was generally well tolerated; frequent adverse events included diarrhea and flatulence, which were managed effectively by dietary modification and loperamide. Overall, miglustat appeared to stabilize neurological status in juvenile-onset NP-C patients, but therapeutic benefits appeared smaller among younger patients who were at a more advanced stage of disease at baseline.
Summary Background Conradi–Hünermann–Happle syndrome (CDPX2, OMIM 302960) is an inherited X‐linked dominant variant of chondrodysplasia punctata which primarily affects the skin, bones and eyes. ...CDPX2 results from mutations in EBP (emopamil binding protein), and presents with increased levels of sterol precursors 8(9)‐cholestenol and 8‐dehydrocholesterol.
Objectives To expand the understanding of CDPX2, clinically, biochemically and genetically.
Methods We present one of the largest series reported to date, including 13 female patients belonging to nine Spanish families. Patients were studied biochemically using gas chromatography–mass spectrometry, genetically using polymerase chain reaction and in their methylation status using the HUMARA assay.
Results In our cases, there was a clear relationship between abnormal sterol profile and the EBP gene mutation. We describe three novel mutations in the EBP gene. EBP mutations were inherited in three out of nine families and were sporadic in the remaining cases.
Conclusions No clear genotype–phenotype correlation was found. Patients’ biochemical profiles did not reveal a relationship between sterol profiles and severity of disease. A skewed X‐chromosome inactivation may explain the clinical phenotype in CDPX2 in some familial cases.

In October 2015, Zika virus (ZIKV) outbreak the Brazilian Ministry of Health (MoH). In response, the Brazilian Society of Medical Genetics established a task force (SBGM‐ZETF) to study the phenotype ...of infants born with microcephaly due to ZIKV congenital infection and delineate the phenotypic spectrum of this newly recognized teratogen. This study was based on the clinical evaluation and neuroimaging of 83 infants born during the period from July, 2015 to March, 2016 and registered by the SBGM‐ZETF. All 83 infants had significant findings on neuroimaging consistent with ZIKV congenital infection and 12 had confirmed ZIKV IgM in CSF. A recognizable phenotype of microcephaly, anomalies of the shape of skull and redundancy of the scalp consistent with the Fetal Brain Disruption Sequence (FBDS) was present in 70% of infants, but was most often subtle. In addition, features consistent with fetal immobility, ranging from dimples (30.1%), distal hand/finger contractures (20.5%), and feet malpositions (15.7%), to generalized arthrogryposis (9.6%), were present in these infants. Some cases had milder microcephaly or even a normal head circumference (HC), and other less distinctive findings. The detailed observation of the dysmorphic and neurologic features in these infants provides insight into the mechanisms and timings of the brain disruption and the sequence of developmental anomalies that may occur after prenatal infection by the ZIKV.
The electronic spectroscopy of isolated furfural (2-furaldehyde) in the gas phase has been investigated using high-resolution photoabsorption spectroscopy in the 3.5-10.8 eV energy-range, with ...absolute cross section measurements derived. Electron energy loss spectra are also measured over a range of kinematical conditions. Those energy loss spectra are used to derive differential cross sections and in turn generalised oscillator strengths. These experiments are supported by ab initio calculations in order to assign the excited states of the neutral molecule. The good agreement between the theoretical results and the measurements allows us to provide the first quantitative assignment of the electronic state spectroscopy of furfural over an extended energy range.
