Despite the considerable research efforts being made to learn more about COVID-19, little is known about the presence of SARS-CoV-2 genetic material in biological fluids other than respiratory ...droplets, blood, and feces. The aim of this post-mortem study was to assess the presence of SARS-CoV-2 RNA in the knee synovial fluid, synovial tissue, and bone tissue of COVID-19 patients in order to discover whether the joint is a possible route of transmission during orthopaedic surgical procedures, and clarify the possible role of SARS-CoV-2 as a directly arthritogenic virus.
Post-mortem synovial fluid, synovial tissue and bone tissue samples were collected from the knees of five patients who died of COVID-19 in our hospital between September and October 2020, and analysed for the presence of SARS-CoV-2 using a commercial real-time polymerase chain reaction (RT-PCR) panel. Quantitative RT-PCR was used to test post-mortem nasopharyngeal swabs of all of the patients.
No SARS-CoV-2 RNA was detected in any of the knee samples, despite the positivity of the throat swab.
Our findings indicate that SARS-CoV-2 was not detected in knee synovial fluid, synovial membrane or bone. This makes it unlikely that these are potential sources of contagion, and suggests that SARS-CoV-2 is not directly arthritogenic.
The preparation and performance of bimetallic Ni(1-x)Cox-YSZ and Ni(1-x)Cux-YSZ anodes were tested to overcome common deficiencies of carbon and sulphur poisoning in SOFCs. Ni1-xCoxO-YSZ and ...Ni(1-x)CuxO-YSZ precursors were synthesised via co-precipitation of their respective chlorides. Single cell solid oxide fuel cells of these bimetallic anodes were tested in H2, CH4, and H2S/CH4 fuel mixtures. Addition of Cu2+ into the NiO lattice resulted in large metal particle sizes and decreased SOFC performance. Addition of Co2+ into the NiO lattice to form Ni0.92Co0.08O-YSZ anode precursor produced a cermet with a large BET surface area and active metal surface area, thus increasing the rate of hydrogen oxidation for this sample. The performance of both bimetallics was found to quickly degrade in dry CH4 due to carbon deposition and lifting of the anode from the electrolyte. Ni0.69Co0.31-YSZ showed superior activity in a 10 vol% H2S/CH4 fuel mixture, surpassing the performance with H2 fuel, thereby demonstrating the exciting prospect of using sulphidated Ni(1-x)Cox-YSZ as SOFC anodes in sulphur containing methane streams. The active anode became a sulphidated alloy (Ni-Co-S) under operating conditions. This anode showed enhanced performance, which surpassed those of sulphidated Ni and Co anodes, thereby suggesting a synergistic behaviour in the Ni-Co-S anode. 50 refs.
Because of their dependence on water, natural and human systems are highly sensitive to changes in the hydrologic cycle. The authors introduce a new measure of hydroclimatic intensity (HY-INT), which ...integrates metrics of precipitation intensity and dry spell length, viewing the response of these two metrics to global warming as deeply interconnected. Using a suite of global and regional climate model experiments, it is found that increasing HY-INT is a consistent and ubiquitous signature of twenty-first-century, greenhouse gas–induced global warming. Depending on the region, the increase in HY-INT is due to an increase in precipitation intensity, dry spell length, or both. Late twentieth-century observations also exhibit dominant positive HY-INT trends, providing a hydroclimatic signature of late twentieth-century warming. The authors find that increasing HY-INT is physically consistent with the response of both precipitation intensity and dry spell length to global warming. Precipitation intensity increases because of increased atmospheric water holding capacity. However, increases in mean precipitation are tied to increases in surface evaporation rates, which are lower than for atmospheric moisture. This leads to a reduction in the number of wet days and an increase in dry spell length. This analysis identifies increasing hydroclimatic intensity as a robust integrated response to global warming, implying increasing risks for systems that are sensitive to wet and dry extremes and providing a potential target for detection and attribution of hydroclimatic changes.
Microcavity polaritons are two-dimensional bosonic fluids with strong nonlinearities, composed of coupled photonic and electronic excitations. In their condensed form, they display quantum ...hydrodynamic features similar to atomic Bose-Einstein condensates, such as long-range coherence, superfluidity and quantized vorticity. Here we report the unique phenomenology that is observed when a pulse of light impacts the polariton vacuum: the fluid which is suddenly created does not splash but instead coheres into a very bright spot. The real-space collapse into a sharp peak is at odd with the repulsive interactions of polaritons and their positive mass, suggesting that an unconventional mechanism is at play. Our modelling devises a possible explanation in the self-trapping due to a local heating of the crystal lattice, that can be described as a collective polaron formed by a polariton condensate. These observations hint at the polariton fluid dynamics in conditions of extreme intensities and ultrafast times.
Aspartate-glutamate carrier isoform 1 (AGC1) is a carrier responsible for the export of mitochondrial aspartate in exchange for cytosolic glutamate and is part of the malate-aspartate shuttle, ...essential for the balance of reducing equivalents in the cells. In the brain, mutations in SLC25A12 gene, encoding for AGC1, cause an ultra-rare genetic disease, reported as a neurodevelopmental encephalopathy, whose symptoms include global hypomyelination, arrested psychomotor development, hypotonia and seizures. Among the biological components most affected by AGC1 deficiency are oligodendrocytes, glial cells responsible for myelination processes, and their precursors oligodendrocyte progenitor cells (OPCs). The AGC1 silencing in an in vitro model of OPCs was documented to cause defects of proliferation and differentiation, mediated by alterations of histone acetylation/deacetylation. Disrupting AGC1 activity could possibly reduce the availability of acetyl groups, leading to perturbation of many biological pathways, such as histone modifications and fatty acids formation for myelin production. Here, we explore the transcriptome of mouse OPCs partially silenced for AGC1, reporting results of canonical analyses (differential expression) and pathway enrichment analyses, which highlight a disruption in fatty acids synthesis from both a regulatory and enzymatic stand. We further investigate the cellular effects of AGC1 deficiency through the identification of most affected transcriptional networks and altered alternative splicing. Transcriptional data were integrated with differential metabolite abundance analysis, showing downregulation of several amino acids, including glutamine and aspartate. Taken together, our results provide a molecular foundation for the effects of AGC1 deficiency in OPCs, highlighting the molecular mechanisms affected and providing a list of actionable targets to mitigate the effects of this pathology.
The Mediterranean is expected to be one of the most prominent and vulnerable climate change “hotspots” of the twenty-first century, and the physical mechanisms underlying this finding are still not ...clear. Furthermore, complex interactions and feedbacks involving ocean–atmosphere–land–biogeochemical processes play a prominent role in modulating the climate and environment of the Mediterranean region on a range of spatial and temporal scales. Therefore, it is critical to provide robust climate change information for use in vulnerability–impact–adaptation assessment studies considering the Mediterranean as a fully coupled environmental system. The Mediterranean Coordinated Regional Downscaling Experiment (Med-CORDEX) initiative aims at coordinating the Mediterranean climate modeling community toward the development of fully coupled regional climate simulations, improving all relevant components of the system from atmosphere and ocean dynamics to land surface, hydrology, and biogeochemical processes. The primary goals of Med-CORDEX are to improve understanding of past climate variability and trends and to provide more accurate and reliable future projections, assessing in a quantitative and robust way the added value of using high-resolution and coupled regional climate models. The coordination activities and the scientific outcomes of Med-CORDEX can produce an important framework to foster the development of regional Earth system models in several key regions worldwide.
Contaminants of emerging concern (CEC) are still under research given the vast diversity of compounds reaching freshwater ecosystems and adverse effects they might cause. In this study, the ...environmental fate of 73 CEC, comprising sweeteners, stimulants and several pharmaceutical therapeutic classes, and changes in fluvial biofilm photosynthetic parameters were evaluated in a semi-arid urban river receiving diffuse and point sources of pollution (Suquía river, Argentina). Out of the 37 CEC detected, 30 were quantified in surface water (n.d. – 9826 ng/L), 10 in biofilm (n.d. – 204 ng/gd.w.) and 9 in the clay fraction of sediments (n.d. – 64 ng/gd.w.). CEC distribute differently among the 3 matrices: water phase presents the biggest diversity of compounds (14 CEC families), being analgesic/anti-inflammatories the most abundant family. Antibiotics largely predominated in biofilms (7 CEC families), while the stimulant caffeine and some antibiotics where the most abundant in sediments (6 CEC families). Different CEC accumulated in biofilms and sediments upstream and downstream the city, and big shifts of biofilm community occurred downstream WWTP. The shift of biofilm community upstream (F0 > 0) and downstream the WWTP (F0 = 0) shows a sensitive response of F0 to the impact of WWTP. Biofilm photosynthetic parameters responded in less impacted urban sites (sites 1, 2 and 3), where significant correlations were found between ketoprofen and some antibiotics and biofilm parameters. The diversity and amount of CEC found in the urban section of Suquía river alert to the magnitude of point and non-point sources of pollution.

BACKGROUNDMosaic and consensus HIV-1 immunogens provide two distinct approaches to elicit greater breadth of coverage against globally circulating HIV-1 and have shown improved immunologic breadth in ...nonhuman primate models.METHODSThis double-blind randomized trial enrolled 105 healthy HIV-uninfected adults who received 3 doses of either a trivalent global mosaic, a group M consensus (CON-S), or a natural clade B (Nat-B) gp160 env DNA vaccine followed by 2 doses of a heterologous modified vaccinia Ankara-vectored HIV-1 vaccine or placebo. We performed prespecified blinded immunogenicity analyses at day 70 and day 238 after the first immunization. T cell responses to vaccine antigens and 5 heterologous Env variants were fully mapped.RESULTSEnv-specific CD4+ T cell responses were induced in 71% of the mosaic vaccine recipients versus 48% of the CON-S recipients and 48% of the natural Env recipients. The mean number of T cell epitopes recognized was 2.5 (95% CI, 1.2-4.2) for mosaic recipients, 1.6 (95% CI, 0.82-2.6) for CON-S recipients, and 1.1 (95% CI, 0.62-1.71) for Nat-B recipients. Mean breadth was significantly greater in the mosaic group than in the Nat-B group using overall (P = 0.014), prime-matched (P = 0.002), heterologous (P = 0.046), and boost-matched (P = 0.009) measures. Overall T cell breadth was largely due to Env-specific CD4+ T cell responses.CONCLUSIONPriming with a mosaic antigen significantly increased the number of epitopes recognized by Env-specific T cells and enabled more, albeit still limited, cross-recognition of heterologous variants. The study of income inequality is important for predicting the wealth of a country. There is an increasing number of publications where the authors call for the use of several indices simultaneously ...to better account for the wealth distribution. Due to the fact that income data are usually collected through sample surveys, the sampling properties of income inequality measures should not be overlooked. The most widely used inequality measure is the Gini index, and its inferential aspects have been deeply investigated. An alternative inequality index could be the Bonferroni inequality index, although less attention on its inference has been paid in the literature. The aim of this paper is to address the inference of the Bonferroni index in a finite population framework. The Bonferroni index is linearized by differentiation with respect to the sample indicators which allows for conducting a valid inference. Furthermore, the linearized variables are used to evaluate the effects of the different observations on the Bonferroni and Gini indices. The result demonstrates once for all that the former is more sensitive to the lowest incomes in the distribution than the latter.
