The glycosylated minor hemoglobin components Hb A1a+b and HbA1c are elevated in insulin-dependent juvenile diabetic patients, 3.2+/-0.7 (+/-1 SD) and 10.0+/-1.9% of total hemoglobin respectively, ...versus 2.1+/-0.4 and 4.9+/-0.7% in a normal non-diabetic control population. Total glycosylated hemoglobin components, Hb A1a+b+c, correlated with the degree of diabetic blood glucose regulation as measured by antecedent 24-h urinary glucose excretion determined in 220 diabetic patients immediately before, 1, 2, and 3 months prior to the HB A1a+b+c measurement. This assay for long-term blood glucose regulation was utilized to determine the effect of hyperglycemia on plasma cholesterol levels in 112 diabetic patients. Hb A1a+b+c levels correlated with plasma cholesterol levels, suggesting that long-term hyperglycemia is associated with hypercholesterolemia. It is suggested that glycosylated hemoglobin measurement is a good index of long-term blood glucose levels in diabetic patients.
This study compares the ethical perceptions of White and Hispanic consumers (n=259) in two different urban areas in two different states in the Southwestern United States with regard to 14 ...questionable retail practices. In addition to examining the culture of the subjects and cultural proximity to the merchant, demographics of income and gender were included. ANOV A routines indicate that Hispanics, women, and those with lower incomes are less tolerant of the questionable practices Studiesd (p=.05 or lower). Cultural proximity to the merchant had no effect.
The role of adaptive shifts in species formation has been the subject of considerable controversy for many years. Here we examine the phylogeny of a large radiation of Hawaiian spiders in the genus ...Tetragnatha to determine the extent to which species splitting is associated with shifts in ecological affinity. We use molecular data from ribosomal 12S and cytochrome oxidase mitochondrial DNA, and allozymes to assess phylogenetic affinity. Ecological associations were recorded for all species under study, and shifts are considered in the context of the phylogeny. Results indicate that there are two major clades of Hawaiian Tetragnatha, one of which has abandoned web building (spiny-leg clade), while the other retains the ancestral condition of web building. Within the spiny-leg clade, the molecular information suggests that the species on any one island are generally most closely related to each other. Preliminary results for the web-building "complex" of species indicate that there may be groups of web builders that have speciated in a similar manner. Results of the study suggest that, at least within the spiny-leg clade, matching sets of taxa have evolved independently on the different Hawaiian islands. There appears to have been a one-to-one convergence of the same set of "ecomorph" types on each island in a manner similar to that of lizards of the Caribbean.
Articular cartilage from arthritic joints of rats immunized with type II collagen is severely depleted of proteoglycans. Depletion begins within 48 hours after the onset of inflammation, prior to ...extensive pannus formation, and may represent a critical first step in cartilage destruction. We have immunolocalized stromelysin, an enzyme that is believed to play a major role in the pathologic degradation of proteoglycans, in the joints of rats with collagen-induced arthritis. Immunoperoxidase staining of frozen tissue sections demonstrated the presence of stromelysin in both the synovium and chondrocytes. In contrast, collagenase was localized primarily to the pannus-cartilage junction. Neither enzyme was detectable in joints from normal animals. To test the hypothesis that chondrocytes respond directly to inflammatory mediators by increasing the production of stromelysin, isolated chrondrocytes were incubated with various concentrations of interleukin-1. The culture media were also assayed for the presence of stromelysin by immunoreactivity on Western blots and by analysis of enzymatic activity on casein substrate gels. A 3-fold increase in a doublet of proteins synthesized in response to 10 units/ml of interleukin-1 was observed. These proteins also immunoreacted with the stromelysin antibody and degraded casein. Northern blotting results established that the increased levels of stromelysin were accompanied by increases in stromelysin-specific messenger RNA levels. These results suggest that stromelysin is responsible for proteoglycan degradation in early inflammatory arthritis, and that chondrocytes may play a direct role in the earliest stages of the degradation of their own matrices.
The ability of chondrocytes to synthesize chondroitin-4-sulfate (C4S) as opposed to chondroitin-6-sulfate (C6S) is a phylogenetically related phenomenon seen among adult higher vertebrates and ...developmentally during the embryogenesis of these vertebrates. While the embryonic cartilage may be initially a C6S matrix, C4S synthesis is seen to develop with time. We have histochemically localized these differences in sulfation with the cationic carbocyanine dye, Stains-all, in a spectrum of cartilages that vary in the sulfation position of their chondroitin sulfate. Cartilages from the rat and rabbit that are predominantly C4S stained magenta at pH 4.3, while the C6S-rich cartilage matrices from the regenerating rabbit ear and lamprey cranium stained blue. Embryonic chicken cartilages develop a gradient of magenta matrix with age, with increased concentration toward the articular surface. Both magenta and blue matrices were absent after pretreatment with chondroitinase ABC but were present after Streptomyces hyaluronidase digestion. The magenta staining was a property of the cartilage matrix as a whole, since isolated C4S and C6S stained blue. The differential staining was seen at pH 4.3, but not at pH 8.8, suggesting an interaction between the chondroitin sulfate and the adjacent tissue proteins.
SWR mice are resistant to collagen-induced arthritis but produce antibodies to type II collagen. To determine if these antibodies have arthritogenic potential, serum from collagen-immunized mice was ...concentrated and passively transferred to DBA/1 mice. The recipients developed severe arthritis within 72 hours. To evaluate the role of complement, SWR mice were bred with congenic inbred B10.D2/oSn (complement deficient) and B10.D2/nSn (complement normal) mice. Collagen-immunized (SWR x B10.D2/nSn)F1 mice had high levels of C5 and were susceptible to arthritis, while (SWR x B10.D2/oSn)F1 mice were deficient in C5 and were resistant to arthritis.
High-efficiency performance of GaAs Schottky-Read IMPATT diodes has been observed at X-band frequencies. The highest efficiency measured was 26.1 percent with 2.5-W continuous-wave (CW) output power ...at 8.8 GHz for a single-mesa diode while multiple-mesa diodes have delivered more than 7 W at X band. The diodes were fabricated from multiple-layer epitaxial material with gold-plated heat sinks. Details of materials preparation and diode fabrication are presented. Theoretical calculations of diode breakdown voltage and efficiencies have been made as a function of the structural properties of the diodes. Good agreement has been obtained between the experimental microwave oscillator performance and the theoretical calculations.
