The plant growth-promoting bacterium (PGPB) Pseudomonas sp. UW4, previously isolated from the rhizosphere of common reeds growing on the campus of the University of Waterloo, promotes plant growth in ...the presence of different environmental stresses, such as flooding, high concentrations of salt, cold, heavy metals, drought and phytopathogens. In this work, the genome sequence of UW4 was obtained by pyrosequencing and the gaps between the contigs were closed by directed PCR. The P. sp. UW4 genome contains a single circular chromosome that is 6,183,388 bp with a 60.05% G+C content. The bacterial genome contains 5,423 predicted protein-coding sequences that occupy 87.2% of the genome. Nineteen genomic islands (GIs) were predicted and thirty one complete putative insertion sequences were identified. Genes potentially involved in plant growth promotion such as indole-3-acetic acid (IAA) biosynthesis, trehalose production, siderophore production, acetoin synthesis, and phosphate solubilization were determined. Moreover, genes that contribute to the environmental fitness of UW4 were also observed including genes responsible for heavy metal resistance such as nickel, copper, cadmium, zinc, molybdate, cobalt, arsenate, and chromate. Whole-genome comparison with other completely sequenced Pseudomonas strains and phylogeny of four concatenated "housekeeping" genes (16S rRNA, gyrB, rpoB and rpoD) of 128 Pseudomonas strains revealed that UW4 belongs to the fluorescens group, jessenii subgroup.
Modern CCD cameras are usually capable of a spatial accuracy greater than 1/50 of the pixel size. However, such accuracy is not easily attained due to various error sources that can affect the image ...formation process. Current calibration methods typically assume that the observations are unbiased, the only error is the zero-mean independent and identically distributed random noise in the observed image coordinates, and the camera model completely explains the mapping between the 3D coordinates and the image coordinates. In general, these conditions are not met, causing the calibration results to be less accurate than expected. In the paper, a calibration procedure for precise 3D computer vision applications is described. It introduces bias correction for circular control points and a nonrecursive method for reversing the distortion model. The accuracy analysis is presented and the error sources that can reduce the theoretical accuracy are discussed. The tests with synthetic images indicate improvements in the calibration results in limited error conditions. In real images, the suppression of external error sources becomes a prerequisite for successful calibration.
Persistent urbanization is challenging planners to reimagine the absorptive capacities of cities. While more and more city dwellers respond to the challenge with informal solutions, policy and ...planning responses have fallen short of providing compelling answers. We propose redirecting attention to the use of land. This paper develops a model that centers on the role of land use regulation in conditioning and sustaining housing informality. Taking variations in income, location, and regulation into account, our framework uncovers the effective, household-level regulatory burden. Using this model to audit existing land use regimes gives planners the tools to plan effectively for targeted adjustments.
To mitigate the diesel particle pollution problem, diesel vehicles are fitted with modern exhaust after-treatment systems (ATS), which efficiently remove engine-generated primary particles (soot and ...ash) and gaseous hydrocarbons. Unfortunately, ATS can promote formation of low-vapor-pressure gases, which may undergo nucleation and condensation leading to formation of nucleation particles (NUP). The chemical nature and formation mechanism of these particles are only poorly explored. Using a novel mass spectrometric method, online measurements of low-vapor-pressure gases were performed for exhaust of a modern heavy-duty diesel engine operated with modern ATS and combusting low and ultralow sulfur fuels and also biofuel. It was observed that the gaseous sulfuric acid (GSA) concentration varied strongly, although engine operation was stable. However, the exhaust GSA was observed to be affected by fuel sulfur level, exhaust after-treatment, and driving conditions. Significant GSA concentrations were measured also when biofuel was used, indicating that GSA can be originated also from lubricant oil sulfur. Furthermore, accompanying NUP measurements and NUP model simulations were performed. We found that the exhaust GSA promotes NUP formation, but also organic (acidic) precursor gases can have a role. The model results indicate that that the measured GSA concentration alone is not high enough to grow the particles to the detected sizes.
Biomass could be a key source of renewable energy. Agricultural waste products, such as corn stover, provide a convenient means to replace fossil fuels, such as coal, and a large amount of feedstock ...is currently available for energy consumption in the U.S. This study has two main objectives: (1) to understand the impact of corn stover moisture content and grinder speed on grind physical properties; and (2) develop response surface models and optimize these models using a hybrid genetic algorithm. The response surface models developed were used to draw surface plots to understand the interaction effects of the corn stover grind moisture content and grinder speed on the grind physical properties and specific energy consumption. The surface plots indicated that a higher corn stover grind moisture content and grinder speed had a positive effect on the bulk and tapped density. The final grind moisture content was highly influenced by the initial moisture content of the corn stover grind. Optimization of the response surface models using the hybrid genetic algorithm indicated that moisture content in the range of 17 to 19% (w.b.) and a grinder speed of 47 to 49 Hz maximized the bulk and tapped density and minimized the geomantic mean particle length. The specific energy consumption was minimized when the grinder speed was about 20 Hz and the corn stover grind moisture content was about 10% (w.b.).
In the present study, withaferin A (WA), a steroidal lactone with anti-inflammatory and anti-tumor properties, inhibited proteasome activity and induced endoplasmic reticulum (ER) and cytoplasmic HSP ...accumulation in Xenopus laevis A6 kidney epithelial cells. Proteasomal inhibition by WA was indicated by an accumulation of ubiquitinated protein and a decrease in chymotrypsin-like activity. Additionally, immunoblot analysis revealed that treatment of cells with WA induced the accumulation of HSPs including ER chaperones, BiP and GRP94, as well as cytoplasmic/nuclear HSPs, HSP70 and HSP30. Furthermore, WA-induced an increase in the relative levels of the protein kinase, Akt, while the levels of actin were unchanged compared to control. Northern blot experiments determined that WA induced an accumulation in bip, hsp70 and hsp30 mRNA but not eIF-1α mRNA. Interestingly, WA acted synergistically with mild heat shock to enhance HSP70 and HSP30 accumulation to a greater extent than the sum of both stressors individually. This latter phenomenon was not observed with BiP or GRP94. Immunocytochemical analysis indicated that WA-induced BiP accumulation occurred mainly in the perinuclear region in a punctate pattern, while HSP30 accumulation occurred primarily in a granular pattern in the cytoplasm with some staining in the nucleus. Prolonged exposure to WA resulted in disorganization of the F-actin cytoskeleton as well as the production of relatively large HSP30 staining structures that co-localized with F-actin. Finally, prior exposure of cells to WA treatment, which induced the accumulation of HSPs conferred a state of thermal protection since it protected the F-actin cytoskeleton against a subsequent cytotoxic thermal challenge.
Numerous studies have elucidated the health benefits of organosulfur compounds, known as isothiocyanates (ITCs), derived from cruciferous vegetables. As electrophiles, ITCs have the ability to ...directly bind and modify thiol-containing compounds such as glutathione and cellular protein, including tubulin. While the biochemical effects of ITCs have been well characterized, less information is available regarding their effects on the accumulation of stress-inducible heme oxygenase-1 (HO-1), heat shock proteins (HSPs) and the possible formation of aggregated protein due to thiol modification. The present study has examined the effect of the ITCs, benzyl isothiocyanate (BITC) and phenethyl isothiocyanate (PEITC), on the accumulation of HO-1, HSP70 and HSP30 in Xenopus laevis A6 kidney epithelial cells. Immunoblot analysis revealed that both BITC and PEITC induced the accumulation of HO-1 and HSP70 whereas HSP30 levels were enhanced only in cells treated with BITC. Immunocytochemistry determined that ITC treatment induced F-actin disorganization and membrane ruffling and enhanced accumulation of HO-1 in the cytoplasm. Additionally, BITC induced enhanced levels of ubiquitinated protein, aggregated protein, and the collapse and fragmentation of microtubules. In comparison, treatment of cells with the proteasomal inhibitor, MG132, induced the accumulation of all three stress proteins, aggregated protein and aggresome-like structures. Finally, cells pretreated with BITC inhibited the formation of MG132-induced aggresome-like structures in the perinuclear region. This latter finding suggests that BITC-induced microtubule fragmentation may impede the movement of aggregated protein via microtubules and their subsequent coalescence into aggresome-like structures in the perinuclear region.

This non-interventional, retrospective registry study (GSK ID: HO-17-17558) utilized data from the specialist care hospital register. Data extraction was from first hospital visit with a COPD diagnosis (index date) from January 1, 2004 until December 31, 2015 or death. Patients (aged >18 years with ≥1 report of post-bronchodilation forced expiratory volume in 1 s (FEV
)/forced vital capacity (FVC) ratio <0.7) were categorized as having non-severe or severe COPD (FEV
>50% or ≤50% of reference, respectively). Patients who were initially non-severe but progressed to severe were classified as having progressing COPD. Patients without spirometry registry data were classified as having clinically verified COPD. Patients were grouped according to BEC (≥300 cells/μL, <300 cells/μL or BEC unknown). HCRU, estimated associated costs and mortality were evaluated according to COPD severity and BEC.
There were 9042 patients with COPD; 340 non-severe, 326 progressing, 394 severe, and 7982 clinically verified. BEC was available for 31.8% of patients. The mean follow-up time was 3.7-6.5 years in the classified patient-groups. All-cause mortality was 46% during follow-up. Severe COPD was associated with more COPD-related HCRU and higher mortality than non-severe COPD. Patients with BEC ≥300 cells/μL had higher overall HCRU but improved survival compared with those with BEC <300 cells/μL. Overall direct costs were similar across COPD severity categories, 3300-3900€/patient-year, although COPD-related costs were higher in patients with severe versus non-severe COPD.

Growing concern over the environmental impacts and other credence characteristics of food has resulted in increasing interest in the quality attributes of meat products in Finland. The aim of this study was to provide information on the relative preferences of consumers for minced meat attributes. Using a choice experiment, we examined whether the meat type, method of production, fat content, price and presence of carbon footprint information have an impact on consumer choice. A low fat content was found to have a particularly positive effect on the choice of a minced meat product. The carbon footprint information had a significant impact on the meat type-specific consumer preferences: the popularity of beef products decreased and of pork products increased when the footprint information was presented to the consumers. Six heterogeneous consumer classes were identified with latent class analysis: price-conscious (23% of the respondents), fat content-conscious (23%), ideological but passive (17%), content with conventional (14%), beef-preferring (12%) and method of production -conscious consumers (11%). Consumers were generally willing to pay more for a low fat content, but the relative willingness to pay estimates were largely dependent on the consumer groups: premiums for organic and animal welfare-oriented production methods also existed. These attributes could thus represent good means for differentiating minced meat products. The impact of carbon footprint information on the willingness to pay estimates was relatively low.
International organizations such as the United Nations, the World Bank, the Organization for Economic Cooperation and Development, and the International Monetary Fund routinely organize ...cross-learning programs on specific topics for their member governments. Likewise, many national governments organize offers little theoretical or practical guidance on how best to organize such cross-learning activities. One fundamental question is whether to proceed on the basis of cohort- or task-oriented programs, where a cohort-based approach would emphasize shared, institutionalized learning over time amongst local governments with shared planning priorities. To assess this question, we use a case study comparing 286 cities and their avowed priorities for China's 11th and 12th Five-Year Plans. The evidence from our case study supports a task-rather than a cohort-oriented approach. Moreover, because of China's unique administrative structure, with an integrated approach entailing proactive national level guidance and directives, we conclude that for most other countries a cohort-oriented approach would be even less effective. The practical implication of these results is that a task-oriented approach to cross-learning is more advisable.
