In this work, an adaptive generalized cross-correlation (AGCC) method is proposed that focuses on the problem of the conventional cross-correlation method not effectively realizing the time delay ...estimation of signals with strong periodicity. With the proposed method, the periodicity of signals is judged and the center frequencies of the strongly periodical components are determined through the spectral analysis of the input signals. Band-stop filters that are used to suppress the strongly periodical components are designed and the mutual power spectral density of the input signals that is processed by the band-stop filters is calculated. Then, the cross-correlation function that is processed is the inverse Fourier transform of the mutual power spectral density. Finally, the time delay is estimated by seeking the peak position of the processed cross-correlation function. Simulation experiments and practical velocity measurement experiments were carried out to verify the effectiveness of the proposed AGCC method. The experimental results showed that the new AGCC method could effectively realize the time delay estimation of signals with strong periodicity. In the simulation experiments, the new method could realize the effective time delay estimation of signals with strong periodicity when the energy ratio of the strongly periodical component to the aperiodic component was under 150. Meanwhile, the cross-correlation method and other generalized cross-correlation methods fail in time delay estimation when the energy ratio is higher than 30. In the practical experiments, the velocity measurement of slug flow with strong periodicity was implemented in small channels with inner diameters of 2.0 mm, 2.5 mm and 3.0 mm. With the proposed method, the relative errors of the velocity measurement were less than 4.50%.

Circular RNAs (circRNAs) are a special class of endogenous non-coding RNAs. Numerous studies have shown that circRNAs have shown great potential in regulating the biological functions of tumors, including proliferation, migration, invasion, and treatment resistance, and that differences in their expression levels are closely related to the clinical prognosis of tumor patients. This review systematically compares the mechanisms of circRNAs in the process of tumor development and radiosensitivity and provides insight into the clinical translation of circRNAs in radiotherapy.
Microalgae are competitive and commercial sources for health-benefit carotenoids. In this study, a Chromochloris zofingiensis mutant (Cz-pkg), which does not shut off its photosystem and stays green ...upon glucose treatment, was generated and characterized. Cz-pkg was developed by treating the algal cells with a chemical mutagen as N-methyl-N’-nitro-N-nitrosoguanidine and followed by a color-based colony screening approach. Cz-pkg was found to contain a dysfunctional cGMP-dependent protein kinase (PKG). By cultivated with CO2 aeration under mixotrophy, the mutant accumulated lutein up to 31.93 ± 1.91 mg L−1 with a productivity of 10.57 ± 0.73 mg L−1 day−1, which were about 2.5- and 8.5-fold of its mother strain. Besides, the lutein content of Cz-pkg could reach 7.73 ± 0.52 mg g−1 of dry weight, which is much higher than that of marigold flower, the most common commercial source of lutein. Transcriptomic analysis revealed that in the mutant Cz-pkg, most of the genes involved in the biosynthesis of lutein and chlorophylls were not down-regulated upon glucose addition, suggesting that PKG may regulate the metabolisms of photosynthetic pigments. This study demonstrated that Cz-pkg could serve as a promising strain for both lutein production and glucose sensing study.
RNA modification, like DNA methylation, histone modification, non-coding RNA modification and chromatin rearrangement, plays an important role in tumours. N6-methyladenosine (m6A) is the most ...abundant RNA modification in cells, and it regulates RNA transcription, processing, splicing, degradation, and translation. m6A-associated proteins have been used as new biomarkers and therapeutic targets for tumour prediction and monitoring. There are three main types of proteins involved in m6A methylation: methyltransferases (METTL3, METTL14, WTAP, RBM15, ZC3H13 and KIAA1429), demethylases (FTO, ALKBH5 and ALKBH3) and RNA-binding proteins (YTHDF1-3, YTHDC1-2, IGF2BPs and HNRNPs). This article reviews the origins, characteristics and functions of m6A and its relationship with digestive system tumours based on recent research. The expression of m6A regulators can be used as an evaluation indicator of tumour growth and progression and as a prognostic indicator. In-depth research on m6A methylation in digestive system tumours may provide new directions for clinical prediction and further treatment.
Electrodes are basic components of C4D (capacitively coupled contactless conductivity detection) sensors, and different electrode structures (the configuration pattern or the electrode geometry) can ...lead to different measurement results. In this work, the effects of electrode geometry of radial configuration on the measurement performance of C4D sensors are investigated. Two geometrical parameters, the electrode length and the electrode angle, are considered. A FEM (finite element method) model based on the C4D method is developed. With the FEM model, corresponding simulation results of conductivity measurement with different electrode geometry are obtained. Meanwhile, practical experiments of conductivity measurement are also conducted. According to the simulation results and experimental results, the optimal electrode geometry of the C4D sensor with radial configuration is discussed and proposed. The recommended electrode length is 5–10 times of the pipe inner diameter and the recommended electrode angle is 120–160°.
Establishing a transgenic plant largely relies on a selectable marker gene that can confer antibiotic or herbicide resistance to plant cells. The existence of such selectable marker genes in ...genetically modified foods has long been criticized. Plant cells generally exhibit too low an activity of phosphomannose isomerase (PMI) to grow with mannose as a sole carbon source. In this study, we characterized PMI from the green microalga Chlorococcum sp. and assessed its feasibility as a selectable marker for plant biotechnology. Chlorococcum sp. PMI (ChlPMI) was shown to be closely related to higher plants but more distant to bacterial counterparts. Overexpression of ChlPMI in tomato induced callus and shoot formation in media containing mannose (6 g/L) and had an average transformation rate of 3.9%. Based on this transformation system, a polycistronic gene cluster containing crtB, HpBHY, CrBKT and SlLCYB (BBBB) was co-expressed in a different tomato cultivar. Six putative transformants were achieved with a transformation rate of 1.4%, which produced significant amounts of astaxanthin due to the expression of the BBBB genes. Taken together, these findings indicate that we have established an additional tool for plant biotechnology that may be suitable for genetically modifying foods safely.
Tomato is a major source of dietary carotenoids that play an important role in human health. Improving the carotenoid profiles of tomato fruit has become a primary aspect in tomato breeding. The ...tomato ‘Huang Song’ landrace grows vigorously but with low contents of carotenoids in this yellowish fruit. To overcome this problem and extend lycopene to high-value astaxanthin, we co-expressed four genes encoding for phytoene synthase (crtB), β-carotene hydroxylase (BHY), β-carotene ketolase (BKT), and lycopene β-cyclase (LCYB), respectively, in the fruit of the landrace via linking the genes with 2A-sequences and driving their expression with the E8 promoter. The transgenic lines showed similar phenotypes to WT except for the fruit colors. The transgenic fruit exhibited deep red color due to the accumulation of novel ketocarotenoids including astaxanthin, canthaxanthin, and ketolutein and enhanced production of native carotenoids which were 20 ~ 44-fold of the control. The T0, T1, and T2 plants exhibited similar growing status and carotenoid profiles, indicating that the transformants were genetically stable. The transgenes resulted in upregulation of most of the endogenous carotenogenic genes in fruit at mature green stage and more plastoglobules in chromoplast. This study provides insights into metabolic engineering of tomato for enhanced production of value-added carotenoids.
Failure of antipsychotic medications to resolve symptoms in patients with schizophrenia creates a clinical challenge that is known as treatment resistance. The causes of treatment resistance are ...unknown, but it is associated with earlier age at onset and more severe cognitive deficits. The authors tested the hypothesis that white matter deficits that are involved in both neurodevelopment and severity of cognitive deficits in schizophrenia are associated with a higher risk of treatment resistance.
The study sample (N=122; mean age, 38.2 years) included schizophrenia patients at treatment initiation (N=45), patients whose symptoms were treatment responsive (N=40), and patients whose symptoms were treatment resistant (N=37), as well as healthy control subjects (N=78; mean age, 39.2 years). White matter regional vulnerability index (RVI) was tested as a predictor of treatment resistance and cognitive deficits. Higher RVI is indicative of better agreement between diffusion tensor imaging fractional anisotropy across the brain in an individual and the pattern identified by the largest-to-date meta-analysis of white matter deficits in schizophrenia.
Patients with treatment-resistant symptoms showed the highest white matter RVI (mean=0.38 SD=0.2), which was significantly higher than the RVI among patients with treatment-responsive symptoms (mean=0.30 SD=0.02). At the onset of treatment, schizophrenia patients showed significantly higher RVI than healthy control subjects (mean=0.18 SD=0.03 and mean=0.13 SD=0.02, respectively). RVIs were significantly correlated with performance on processing speed and negative symptoms.
Schizophrenia affects white matter microstructure in specific regional patterns. Susceptibility to white matter regional deficits is associated with an increased likelihood of treatment resistance. Developments to overcome schizophrenia treatment resistance should consider white matter as an important target.
Abstract Background Clozapine is an off-label drug used in most countries to prevent suicide in individuals with schizophrenia. However, few studies have reported real-world prescription practices. ...This study aimed to explore the association between a history of suicidal behavior and clozapine prescribing during eight weeks of hospitalization for individuals with early-stage schizophrenia. Methods This observational cohort study used routine health data collected from a mental health hospital in Beijing, China. The study included 1057 inpatients who had schizophrenia onset within 3 years. History of suicidal behavior was coded from reviewing medical notes according to the Columbia Suicide Severity Rating Scale. Information on antipsychotic use during hospitalization was extracted from the prescription records. Time to clozapine use was analyzed using Cox regression models adjusted for sociodemographic and clinical covariates. Results The prevalence rates of self-harm, suicidal behavior, and suicide attempt were 12.3%, 7.5%, and 5.4%, respectively. A history of self-harm history was positively associated with clozapine uses upon admission (4.1% vs. 0.8%, exact p = 0.009). Among those who had not used clozapine and had no clozapine contraindication, A history of suicidal behavior increased the possibility of switch to clozapine within 56 days after admission (Hazard Ratio95% CI, 6.092.08–17.83) or during hospitalization (4.181.62–10.78). Conclusion The use of clozapine for early-stage schizophrenia was more frequent among those with suicidal behavior than among those without suicidal behavior in China, although the drug instructions do not label its use for suicide risk.
