DNA analysis of different forms of gestational trophoblastic disease.

In the period of September ...1993 to April 2003, eighty-nine cases of gestational trophoblastic disease were analysed. There were 22 cases of partial hydatidiform moles, 58 cases of complete hydatidiform mole, 5 cases of invasive mole and 4 cases of gestational choriocarcinomas. Southern hybridization and polymerase chain reaction were used for DNA analysis.
From 22 analyzed cases of partial hydatidiform moles 19 (86.4%) were triploid and 3 (13.6%) diploid ones. There were 58 cases of complete hydatidiform mole and out of them 29 (50%) were homozygous, 28 (48.3%) heterozygous, and in one case (1.7%) both paternal and maternal genome was detected. In 8 cases of heterozygous and in one case of homozygous complete hydatidiform mole occurred a malignant transformation to gestational choriocarcinoma.
Molecular analysis can determine the nuclear DNA origin of complete hydatidiform mole and allow us to define the patients with higher risk of malignant transformation usually to gestational choriocarcinoma.

To evaluate subsequent pregnancy experience in patients following chemotherapy for malignant form of gestational trophoblastic disease.

There were evaluated subsequent pregnancy experiences in 38 patients after chemotherapy for malignant form of gestational trophoblastic disease registered in Slovak center of gestational trophoblastic disease. Histological and cytogenetical analysis of all placentas after deliveries and material from curettage specimens after miscarriages, abortions and ectopic pregnancies were performed.
11 women conceived following successful chemotherapy of gestational trophoblastic disease became pregnant a total 19 times. Out of them there were 9 full-term deliveries, 2 spontaneous abortion, 1 ended in ectopic pregnancy and 7 pregnancies were terminated in therapeutic abortion. Cytogenetical analysis was successful in 7 to 10 reproductive losses with normal karyotype in all analysed cases.
Patients after successful chemotherapy of gestational trophoblastic disease have a normal reproductive outcome.
The research was carried out with the aim to investigate oversown grasses, legumes and mixtures under different climatic conditions. At the research sites, yield of legume treatments was higher than ...the control. The production of oversown grass-clover mixture treatments was also higher than the control. Mean production of oversown grass treatments was lower than the control at the Ľubiša site. At Liptovská Teplička site, yields of grass-clover mixture treatments were lower than the control but the yield at the oversown grass treatments was higher than the control. The legumes oversown into seminatural grassland showed good results, especially when red clover (Trifolium pratense L.) was used - the red clover was very flexible. The oversown grasses, legumes and grass-clover mixtures improved yield and enhanced sward canopy.

A research was carried out in the Nízke Tatry (Low Tatras) mountain range region investigating cultivars of four intergeneric grass hybrids (IGHs Lofa, Bečva, Hykor and the newly-bred HŽ 13 DK), ...three tetraploid red clover (Trifolium pratense) cultivars (Beskyd, Amos, Dolina) and three simple grass/clover mixtures composed of HŽ 13 DK and the red clover cultivars. The results recorded in the first harvest year showed that the monocultures of intergeneric grass hybrids were the most advantageous ones when the condition of sward was considered (low weed infestation, very consistent sward). Their ground cover was ca 90 percent at the first cut and ranged between 94 and 100 percent at the second and third cuts and consequently the yield of dry matter (DM) was from 9.392 to 11.292 t per ha. The sward condition at red clover and at simple grass/clover mixtures equalled that of IGHs, however, the weed proportion was higher (10 to 20 percent of ground cover at the first cut). This situation changed in favour of the sown species after the first cut as shown by the botanical composition (the proportion of sown species ranged from 87 to 90 percent). Such a condition of sward influenced also DM yield which was from 8.197 to 8.481 t per ha at the red clover monocultures and from 9.034 to 9.374 t per ha at the simple grass/clover mixtures. Considering the species and varieties used in the research, the significant differences in DM yields were found only between the intergeneric hybrid cv. Hykor (festucoid type) and the other IGHs, namely lolioid ones. The differences in DM yield between the red clover cultivars were not significant thus indicating their versatile use in upland and mountain regions. Similar conclusions were drawn for the simple grass/clover mixtures because high DM yields were recorded also here. Moreover, the importance of swards with the above-mentioned composition is rising also in relation to the assessments of their dietary and nutritive value. The following parameters were recorded at IGHs: PDI between 48.1 and 57.3 g kg per ha of dry matter and NEL from 5.37 to 5.94 MJ per kg. The differences found between the red clover monocultures and the simple grass/clover mixtures were not notable. The values of PDI ranged between 76.2 and 80.0 g per kg DM and NEL was 5.34 - 5.66.
to evaluate the evolution of diabetic retinopathy (DR) after cataract surgery on eyes previously treated by photocoagulation for DR. The compensation of glycaemia (glycosylated haemoglobin, HbA1c) ...and prevalence of attendant diseases (hypertension, ischemic heart disease and alteration of lipid metabolisms) were evaluated.
Forty eyes (40 patients) were evaluated, of which ten (25 %) eyes with DR were treated without laser photocoagulation (LK), and 30 (75 %) eyes were previously treated by LK. Two criteria for evaluating the stabilisation were used, particularly the best corrected visual acuity (BCVA) and the necessity of additional laser photocoagulation.
As many as 100 % of eyes without LK and 86.7 % of eyes with LK had BCVA improved on the first day after surgery (p ≤ 0.05). The BCVA of 0.5 or better was achieved in 31 eyes (77.5 %). The mean level of HbA1c in group without LK was 7.09 ± 1.21 %; in group after LK it was 7.36 ± 1.64 %. The mean level of HbA1c in the group with stable BCVA was 7.14 ± 0.46 % and the duration of DM was 18.6 ± 2.93 years. HbA1c, in group of eyes with worsened BCVA, was 7.86 ± 2.23 %, and the duration of DM was 21.0 ± 8.54 years. HbA1c in the group of stable eyes (without LK after surgery) was 6.71 ± 0.1 %, and the duration of DM was 17.84 ± 8.97 years. HbA1c, in group, where additional LK was applied, was 7.56 ± 0.69 %, and the duration of DM was 19.54 ± 3.26 years. The difference in levels of HbA1c was statistically significant (p ≤ 0.1). The prevalence of attendant disease was highest in the group of eyes with worsened BCVA and necessity of additional LK.
The results of study confirm an improvement in BCVA after cataract surgery by phacoemulsification in the most of patients. Better stabilisation of glycaemia levels and lower frequency of internal diseases were in group with stable BCVA and without necessity of additional LK after cataract surgery (Tab. 3, Fig. 1, Ref. 28).

This cross‐sectional study was designed to investigate the extent of genetic susceptibility by targeting variants in interleukin (IL)−4/IL‐13 signalling pathways leading to atopic disease in ...early childhood. We evaluated involvement of five single nucleotide polymorphisms IL4 C‐590T, IL13 C‐1055T, IL13 Arg130Gln, IL4RA Ile50Val and IL4RA Gln576Arg, in the control of serum total and antigen‐specific immunoglobulin (Ig)E levels. Furthermore, we analysed their association with changes in gene expression of five cytokines having key roles in inflammatory and anti‐inflammatory immune response IL‐4, IL‐13, interferon (IFN)‐γ, IL‐8 and IL‐10. Total and antigen‐specific IgE levels in serum and gene expression of selected cytokines in peripheral blood were measured in 386 children aged 1–8 years. TaqMan allelic discrimination, amplification refractory mutation system–polymerase chain reaction (ARMS–PCR) and restriction fragment length polymorphisms (RFLP) methods validated by sequencing were used for genotyping. All genotypes for children with total and antigen‐specific IgE levels in the normal range were in Hardy–Weinberg equilibrium. Gene expression analyses were carried out using TaqMan gene expression assays. We found elevated total IgE levels in carriers of IL13 Arg130Gln variant allele odds ratio (OR) = 1·84; 95% confidence interval (CI) = 1·16‐2·93. This effect was more apparent for boys (OR = 2·31; 95% CI = 1·25‐4·28). However, no significant association was observed for the other four variants examined. We found up‐regulation of IFN‐γ in children with elevated serum total IgE levels carrying the Arg130 allele (P = 0·005). No differences were found for IL4, IL8 or IL10, while IL13 gene expression was under the detection limit. IL13 Arg130Gln genotypes can play a role in genetic susceptibility to allergy via regulation of serum total IgE levels and affecting IFN‐γ gene expression.
