Aims
This research assessed the safety of aqueous ozone (AO) on human skin after multiple exposures for up to 40 hours.
Methods and Results
Full thickness recombinant human skin (EpiDerm FT, EFT‐400) ...was exposed to AO for 7 seconds per minute for the first 6 minutes of each hour, repeated hourly over four time periods (4, 10, 20 and 40 hours). An MTT assay assessed viability of skin cells after exposure, compared to incubator control, negative control and vehicle control (distilled water). No significant difference in tissue viability was found between the AO condition and any of the control conditions through 20 hours of exposures. At 40 hours of exposure, tissue viability was lower in the AO group when compared with negative control (p = 0.030) but not the other controls.
Conclusions
The current study supports further consideration of repeated application of AO on human skin, such as for hand hygiene.
Impact Statement
The present research is the first well‐controlled in vitro study assessing the cytotoxicity of repeated exposures of AO on a full‐thickness human skin model. This information helps to inform the evaluation of AO as a potential alternative for hand and wound antisepsis.
This study investigated the perceptual relationship between acoustic and electric stimuli presented to CI users with functional contralateral hearing.
Fourteen subjects with unilateral profound ...deafness implanted with a MED-EL CI scaled the perceptual differences between pure tones presented to the acoustic hearing ear and electric biphasic pulse trains presented to the implanted ear. The differences were analyzed with a multidimensional scaling (MDS) analysis. Additionally, speech performance in noise was tested using sentence material presented in different spatial configurations while patients listened with both their acoustic hearing and implanted ears.
Results of alternating least squares scaling (ALSCAL) analysis consistently demonstrate that a change in place of stimulation is in the same perceptual dimension as a change in acoustic frequency. However, the relative perceptual differences between the acoustic and the electric stimuli varied greatly across subjects. A degree of perceptual separation between acoustic and electric stimulation (quantified by relative dimensional weightings from an INDSCAL analysis) was hypothesized that would indicate a change in perceptual quality, but also be predictive of performance with combined acoustic and electric hearing. Perceptual separation between acoustic and electric stimuli was observed for some subjects. However, no relationship between the degree of perceptual separation and performance was found.
•Multi-dimensional scaling was used to explore acoustic and electric hearing.•Changes in place of stimulation corresponded to a changes in acoustic frequency.•Variability was observed in differences between electric and acoustic stimulation.•Observed acoustic and electric differences were not related to speech performance.
We previously identified the bisbenzimide Hoechst 33342 (H42) as a potent multi-stage inhibitor of the prototypic poxvirus, the vaccinia virus (VACV), and several parapoxviruses. A recent report ...showed that novel bisbenzimide compounds similar in structure to H42 could prevent human cytomegalovirus replication. Here, we assessed whether these compounds could also serve as poxvirus inhibitors. Using virological assays, we show that these bisbenzimide compounds inhibit VACV spread, plaque formation, and the production of infectious progeny VACV with relatively low cell toxicity. Further analysis of the VACV lifecycle indicated that the effective bisbenzimide compounds had little impact on VACV early gene expression but inhibited VACV late gene expression and truncated the formation of VACV replication sites. Additionally, we found that bisbenzimide compounds, including H42, can inhibit both monkeypox and a VACV mutant resistant to the widely used anti-poxvirus drug TPOXX (Tecovirimat). Therefore, the tested bisbenzimide compounds were inhibitors of both prototypic and pandemic potential poxviruses and could be developed for use in situations where anti-poxvirus drug resistance may occur. Additionally, these data suggest that bisbenzimide compounds may serve as broad-activity antiviral compounds, targeting diverse DNA viruses such as poxviruses and betaherpesviruses.IMPORTANCEThe 2022 mpox (monkeypox) outbreak served as a stark reminder that due to the cessation of smallpox vaccination over 40 years ago, most of the human population remains susceptible to poxvirus infection. With only two antivirals approved for the treatment of smallpox infection in humans, the need for additional anti-poxvirus compounds is evident. Having shown that the bisbenzimide H33342 is a potent inhibitor of poxvirus gene expression and DNA replication, here we extend these findings to include a set of novel bisbenzimide compounds that show anti-viral activity against mpox and a drug-resistant prototype poxvirus mutant. These results suggest that further development of bisbenzimides for the treatment of pandemic potential poxviruses is warranted.
This study employs thermal and epithermal neutron activation analysis (NAA) to quantitatively and specifically determine absorption dose rates to various body parts from uranium, thorium and ...potassium. Specifically, a case study of bauxite residue (red mud) from an industrial facility was used to demonstrate the feasibility of the NAA approach for radiological safety assessment, using small sample sizes to ascertain the activities of
U,
U,
Th and
K. This proof-of-concept was shown to produce reliable results and a similar approach could be used for quantitative assessment of other samples with possible radiological significance.
U and
Th were determined by epithermal and thermal neutron activation analysis, respectively.
U was determined based on the known isotopic ratio of
U/
U.
K was also determined using epithermal neutron activation analysis to measure total potassium content and then subtracting its isotopic contribution. Furthermore, the work demonstrates the application of Monte Carlo Neutral-Particle (MCNP) simulations to estimate the radiation dose from large quantities of red mud, to assure the safety of humans and the surrounding environment. Phantoms were employed to observe the dose distribution throughout the human body demonstrating radiation effects on each individual organ.
Structured
Objectives
A prospective parallel cohort trial was conducted to compare outcomes of patients treated with maxillary protraction vs LeFort 1 maxillary advancement surgery.
Setting and ...Sample Population
The primary site for the clinical trial is Children's Hospital Los Angeles; the satellite test site is Seattle Children's Hospital. All patients have isolated cleft lip and palate and a skeletal Class III malocclusion.
Material and Methods
A total of 50 patients, ages 11‐14, will be recruited for the maxillary protraction cohort. The maxillary surgery cohort consists of 50 patients, ages 16‐21, who will undergo LeFort 1 maxillary advancement surgery. Patients with additional medical or cognitive handicaps were excluded from the study.
Results
Current recruitment of patients is on track to complete the study within the proposed recruitment period.
Conclusion
This observational trial is collecting information that will examine dental, skeletal, financial and quality‐of‐life issues from both research cohorts.
A fast pneumatic transfer facility was installed in Nuclear Engineering Teaching Laboratory (NETL) of the University of Texas at Austin for the purpose of cyclic thermal and epithermal neutron ...activation analysis. In this study efforts were focused on the evaluation of cyclic epithermal neutron activation analysis (CENAA). Various NIST and CANMET certified materials were analyzed by the system. Experiment results showed
110
Ag with its 25 s half-life as one of the isotopes favored by the system. Thus, the system was put into practical application in identifying silver in metallic ores. Comparison of sliver concentrations as determined by CENAA in CANMET certified reference materials gave very good results.
