Inflammatory factors and type I interferons (IFNs) are key components of host antiviral innate immune responses, which can be released from the pathogen-infected macrophages. African swine fever ...virus (ASFV) has developed various strategies to evade host antiviral innate immune responses, including alteration of inflammatory responses and IFNs production. However, the molecular mechanism underlying inhibition of inflammatory responses and IFNs production by ASFV-encoded proteins has not been fully understood. Here we report that ASFV infection only induced low levels of IL-1β and type I IFNs in porcine alveolar macrophages (PAMs), even in the presence of strong inducers such as LPS and poly(dA:dT). Through further exploration, we found that several members of the multigene family 360 (MGF360) and MGF505 strongly inhibited IL-1β maturation and IFN-β promoter activation. Among them, pMGF505-7R had the strongest inhibitory effect. To verify the function of pMGF505-7R
in vivo
, a recombinant ASFV with deletion of the MGF505-7R gene (ASFV-Δ7R) was constructed and assessed. As we expected, ASFV-Δ7R infection induced higher levels of IL-1β and IFN-β compared with its parental ASFV HLJ/18 strain. ASFV infection-induced IL-1β production was then found to be dependent on TLRs/NF-κB signaling pathway and NLRP3 inflammasome. Furthermore, we demonstrated that pMGF505-7R interacted with IKKα in the IKK complex to inhibit NF-κB activation and bound to NLRP3 to inhibit inflammasome formation, leading to decreased IL-1β production. Moreover, we found that pMGF505-7R interacted with and inhibited the nuclear translocation of IRF3 to block type I IFN production. Importantly, the virulence of ASFV-Δ7R is reduced in piglets compared with its parental ASFV HLJ/18 strain, which may due to induction of higher IL-1β and type I IFN production
in vivo
. Our findings provide a new clue to understand the functions of ASFV-encoded pMGF505-7R and its role in viral infection-induced pathogenesis, which might help design antiviral agents or live attenuated vaccines to control ASF.
In order to quantify air pollution effects on climate change, we investigated the climate response associated with anthropogenic particulate matters (PMs) by dividing fine PM (PM2.5, particle size ...≤2.5 μm) and coarse particulate matter (CPM, particle size >2.5 μm) in great detail in this work, with an aerosol‐climate coupled model. We find that the changes in PM2.5 and CPM are very different and thus result in different, even opposite effects on climate, especially on a regional scale. The column burden of PM2.5 increases globally from 1850 to the present, especially over Asia's southern and eastern parts, whereas the column concentration of CPM increases over high‐latitude regions and decreases over South Asia. The resulted global annual mean effective radiative forcing (ERF) values due to PM2.5 and CPM changes are −1.21 W·m−2 and −0.24 W·m−2, respectively. Increases in PM2.5 result in significant cooling effects on the climate, whereas changes in CPM produce small and even opposite effects. The global annual mean surface air temperature (SAT) decreases by 0.94 K due to PM2.5 increase. Coolings caused by increased PM2.5 are more apparent over Northern Hemisphere (NH) terrain and ocean at mid‐ and high latitudes. Increases in SATs caused by increased CPM are identified over high latitudes in the NH, whereas decreases are identified over mid‐latitude regions. Strong cooling due to increased PM2.5 causes a southward shift of the Intertropical Convergence Zone (ITCZ), whereas the Hadley circulation associated with CPM is enhanced slightly over both hemispheres, along with the weak movement of corresponding ITCZ. The global annual mean precipitation decreases by approximately 0.11 mm day−1 due to the increased PM2.5. Generally, PM2.5 concentration changes contribute more than 80% of the variation caused by all anthropogenic aerosols in ERF, SAT, cloud fraction, and precipitation.
Changes in PM2.5 account for about 95% of the total changes in total anthropogenic particulate matter from 1850 to the present.
The global annual mean effective radiative forcing values were −1.21 and −0.24 W·m−2 for the changes in PM2.5 and coarse particulate matter (CPM), respectively, from the year 1850 to the present.
Reducing PM2.5 would have a much higher impact on climate than reducing CPM.
Abstract
Though the single-layer solutions have been found for the δ-four-stream spherical harmonic expansion method (SHM) in radiative transfer, there is lack of a corresponding doubling–adding ...method (4SDA), which enables the calculation of radiative transfer through a vertically inhomogeneous atmosphere with multilayers. The doubling–adding method is based on Chandrasekhar's invariance principle, which was originally developed for discrete ordinates approximation. It is shown that the invariance principle can also be applied to SHM and δ-four-stream spherical harmonic expansion doubling–adding method (δ-4SDA) is proposed in this paper. The δ-4SDA method has been systematically compared to the δ-Eddington doubling–adding method (δ-2SDA), the δ-two-stream discrete ordinates doubling–adding method (δ-2DDA), and δ-four-stream discrete ordinates doubling–adding method (δ-4DDA). By applying δ-4SDA to a realistic atmospheric profile with gaseous transmission considered, it is found that the accuracy of δ-4SDA is superior to δ-2SDA or δ-2DDA, especially for the cloudy/aerosol conditions. It is shown that the relative errors of δ-4SDA are generally less than 1% in both heating rate and flux, while the relative errors of both δ-2SDA and δ-2DDA can be over 6%. Though δ-4DDA is slightly more accurate than δ-4SDA in heating rates, both of them are accurate enough to obtain the cloud-top solar heating. Here δ-4SDA is superior to δ-4DDA in computational efficiency. It is found that the error of aerosol radiative forcing can be up to 3 W m−2 by using δ-2SDA at the top of the atmosphere (TOA); such error is substantially reduced by applying δ-4SDA. In view of the overall accuracy and computational efficiency, δ-4SDA is suitable for application in climate models.
The NLRP3 inflammasome plays a major role in innate immune responses by activating caspase-1, resulting in secretion of IL-1β and inflammatory pathologic responses. Viral RNA can induce NLRP3 ...inflammasome activation. However, none of the components of NLRP3 inflammasome has the ability to bind viral RNA. Therefore, it had been proposed that there might have been some unidentified cytosolic RNA sensors that could bind viral RNA and NLRP3 to initiate NLRP3 inflammasome activation. In this study, DDX19A, a member of the DEAD/H-box protein family, was identified as a novel component of NLRP3 inflammasome using arterivirus infection as a model. We found that DDX19A interacted with viral RNA and NLRP3. Knockdown of DDX19A expression efficiently inhibited procaspase-1 cleavage and IL-1β secretion in porcine reproductive and respiration syndrome virus (PRRSV)-infected or PRRSV RNA-stimulated primary porcine alveolar macrophages. Overall, DDX19A was identified as a novel cytosolic RNA sensor that bridged PRRSV RNA and NLRP3 to activate NLRP3 inflammasome.

Incorporation of mesopores and active sites into metal-organic framework (MOF) materials to uncover new efficient catalysts is a highly desirable but challenging task. We report the first example of ...a mesoporous MOF obtained by templated electrosynthesis using an ionic liquid as both electrolyte and template. The mesoporous Cu(II)-MOF MFM-100 has been synthesised in 100 seconds at room temperature, and this material incorporates crystal defects with uncoupled Cu(II) centres as evidenced by confocal fluorescence microscopy and electron paramagnetic resonance spectroscopy. MFM-100 prepared in this way shows exceptional catalytic activity for the aerobic oxidation of alcohols to produce aldehydes in near quantitative yield and selectivity under mild conditions, as well as having excellent stability and reusability over repeated cycles. The catalyst-substrate binding interactions have been probed by inelastic neutron scattering. This study offers a simple strategy to create mesopores and active sites simultaneously via electrochemical formation of crystal defects to promote efficient catalysis using MOFs.
African swine fever virus (ASFV) is the etiological agent of African swine fever (ASF), and it is an enveloped, icosahedral, double-stranded DNA virus with a genome length ranging from 170 to 193 kb. ...The ASFV genome contains at least five multigene families (MGFs): MGF100, MGF110, MGF300, MGF360, and MGF505 at the right and left terminal variable regions. The members of the same MGF family are most similar and have conserved sequence motifs, whereas the genetic diversity of different MGF families varies widely. MGF genes play a crucial role in determining ASFV host range, virulence and reducing early cell death post-infection. pMGF505-7R is a multifunctional protein. Recent research advances of pMGF505-7R provide a few new clues to understand the functions of pMGF505-7R either in antagonizing the induction of type I IFN production and IFN downstream signaling or in suppressing inflammatory responses by inhibition of NF-κB signaling and NLRP3 inflammasome, which may be related to ASFV infection-induced pathogenesis.

After the DeepSORT model is constructed, YOLOv5 is introduced into the model, and finally, the YOLOv5-DeepSORT model for sports element detection and tracking is constructed and completed. The ATT-BERT sports culture element entity recognition model is constructed from four levels: the BERT layer, the BiLSTM layer, the attention layer, and the CRF layer, which results in an application framework for the integration of traditional sports culture into teaching. The influence factors of sports culture perception and sports teaching, as well as the integration of traditional ethnic sports culture into sports teaching in Z, were empirically examined. The study shows that the correlation coefficient between the two is around 0.465, and there is a positive and strong correlation between the two. The correlation coefficients of the dimensions for physical education students and non-physical education students were 0.513, 0.483, 0.485, and 0.462, respectively. The most common teaching methods used by teachers in physical education classrooms integrating traditional sports culture were explanation, demonstration, and complete decomposition, with ratios of 0.78 and 0.7, respectively. The integration of sports culture into physical education has resulted in good results. The outcome was satisfactory.

The autocorrelation function (ACF) and its relationship to fluctuation analysis (FA) are discussed, based on the reanalysis monthly mean geopotential height at 500 hPa from ECMWF (ERA-20C). ACF ...provides a measure of the influence of correlation at different time points. The principal pattern ACF shows relatively large positive correlations between every consecutive years in the central Asian continent and the North African continent. However, the confidence intervals of the ACF distributions are generally too wide, and so of low confidence level. It is found that the scaling rule of the ACF breaks down typically, while the distributions of the critical exponent of FA always show patterns. It is also found that the larger values of critical exponent appear in the tropical region, which reflects a larger climate persistence. The critical exponent becomes smaller in the jet stream regions, indicating less year to year reappearance. The characteristic time accounts for the weighted summation of all possible ACFs, which represents a memory length of a certain climate events. In the boreal winter, some robust patterns can be identified by the corresponding distribution of the characteristic time, which provides a unified way of describing the persistence of climate. An approximate relation between the critical exponent of FA and the characteristic time has been found, and such relation is verified by the distributions of these two physical variables.
