Regulatory T cells (Tregs) and interleukin-17-producing T helper (Th17) cells were mutually antagonistic in the pathogenesis of chronic hepatitis C virus (HCV) infection. However, the regulation of ...imbalance between Tregs and Th17 cells was poorly understood in HCV infection. A recent report revealed the immunomodulatory role of Toll-like receptor (TLR) 2 in regulating the balance of Tregs/Th17 functions in multiple sclerosis. Thus, the aim of the current study was to assess the effect of TLR2 stimulation on the suppressive function of Tregs and Th17 differentiation in chronic hepatitis C. A total of 65 patients with chronic hepatitis C receiving pegylated interferon-a2a and ribavirin therapy for 48 weeks, as well as 20 of normal controls (NCs) were enrolled. Cellular proliferation and cytokine production was tested in purified CD4(+)CD25(+)CD127(dim/-) Tregs in response to the stimulation of Pam3Csk4, an agonist of TLR2. In treatment-naive patients, Tregs, but not Th17 cells, from chronic hepatitis C patients expressed higher levels of TLR2 compared with NCs. Stimulation with Pam3Csk4 enhanced the suppressive function of Tregs and production of IL-10 in chronic hepatitis C more than in NCs. However, TLR2 stimulation did not promote Th17 differentiation of Tregs in chronic hepatitis C patients. Moreover, effective anti-HCV therapy resulted in the induction of IL-17-secreting phenotypic shift of Tregs without loss of inhibitive function upon TLR2 stimulation. These data provided a novel mechanism underlying modulating the balance of Tregs/Th17 cells in chronic hepatitis C. HCV infection shifted Tregs/Th17 cells through TLR2 stimulation by inducing Tregs to produce IL-10 and enhancing inhibitive function of effector T cells, resulting in viral persistence.

O systems. This probe was used directly on test papers to distinguish normal organic solvents using their emission colours under UV light based on its AIE and ICT nature. Thanks to the vinyl group in NI-2TPE, we synthesized a copolymer of NIPAM and NI-2TPE, termed P(NIPAM-co-NI-2TPE). The resulting polymer is highly soluble and fluorescent in water (Φ
= 15.4%). Due to the well-known thermo-responsive character of NIPAM, P(NIPAM-co-NI-2TPE) exhibits an interesting fluorescence change in response to various temperatures. Due to the thermo-induced shrinking of the PNIPAM chain, the fluorescence intensity gradually increased from 20 to 34 °C. As the temperature further increased from 34 to 90 °C, the fluorescence intensity decreased sharply, which was caused by the well-known thermal effects. Furthermore, we synthesized a P(HEA-co-NI-2TPE-TPP acrylate) copolymer, in which HEA is a hydrophilic unit, TPP is a mitochondria label and NI-2TPE a fluorescent probe. The corresponding polymer probe is highly soluble in water with FLQY = 7% and we have further applied this probe as a mitochondria targeted imaging tracker in HeLa cells successfully.
Objective
To evaluate the effect and safety of foot baths with Tangbi Waixi Decoction (TW) in treating patients with diabetic peripheral neuropathy (DPN).
Methods
It is a multicenter double-blinded ...randomized controlled trial. Participants with DPN were recruited between November 18, 2016 and May 30, 2018 from 8 hospitals in China. All patients received basic treatments for glycemic management. Patients received foot baths with TW herbal granules either 66.9 g (intervention group) or 6.69 g (control group) for 30 min once a day for 2 weeks and followed by a 2-week rest, as a therapeutic course. If the Toronto Clinical Scoring System total score (TCSS-TS) ⩾6 points, the patients received a total of 3 therapeutic courses (for 12 weeks) and were followed up for 12 weeks. The primary outcome was change in TCSS-TS score at 12 and 24 weeks. Secondary outcomes included changes in bilateral motor nerve conduction velocity (MNCV) and sensory nerve conduction velocity (SNCV) of the median and common peroneal nerve. Safety was also assessed.
Results
Totally 632 patients were enrolled, and 317 and 315 were randomized to the intervention and control groups, respectively. After the 12-week intervention, patients in both groups showed significant declines in TCSSTS scores, and significant increases in MNCV and SNCV of the median and common peroneal nerves compared with pre-treatment (
P
<0.05). The reduction of TCSS-TS score at 12 weeks and the increase of SNCV of median nerve at 24 weeks in the control group were greater than those in the intervention group (
P
<0.05). The number of adverse events did not differ significantly between groups (
P
>0.05), and no serious adverse event was related with treatment.
Conclusion
Treatment of TW foot baths was safe and significantly benefitted patients with DPN. A low dose of TW appeared to be more effective than a high dose. (Registry No. ChiCTR-IOR-16009331).
Parallel computing techniques have been introduced into digital image correlation (DIC) in recent years and leads to a surge in computation speed. The graphics processing unit (GPU)-based parallel ...computing demonstrated a surprising effect on accel- erating the iterative subpixel DIC, compared with CPU-based parallel computing. In this paper, the performances of the two kinds of parallel computing techniques are compared for the previously proposed path-independent DIC method, in which the initial guess for the inverse compositional Gauss-Newton (IC-GN) algorithm at each point of interest (POI) is estimated through the fast Fourier transform-based cross-correlation (FFT-CC) algorithm. Based on the performance evaluation, a heterogeneous parallel computing (HPC) model is proposed with hybrid mode of parallelisms in order to combine the computing power of GPU and multicore CPU. A scheme of trial computation test is developed to optimize the configuration of the HPC model on a specific computer. The proposed HPC model shows excellent performance on a middle-end desktop computer for real-time subpixel DIC with high resolution of more than 10000 POIs per frame.
Anomalous phase separation in metallic glasses is a longstanding issue that has been controversial since it was first implied in 1969 due to the influence of microstructure artifacts and the lack of ...direct large-scale (over nanoscale) evidence. This study confirms an ultralarge scale anomalous glass phase separation phenomenon in a Cr-Fe-Co-Ni-Zr high-entropy metallic glass in which any pair of atomic components has a nonpositive heat of mixing using interferencefree visible-light, atomic force, scanning electron, and double spherical aberration-corrected atomic resolution analytical electron microscopes. The second glassy phase separated from the glassy matrix has an ultralarge size of tens of micrometers, which can be controlled by the cooling rate during sample preparation. A mechanism of before-monotectic-like liquid immiscibility is introduced to interpret the phenomenon, which helps understand the anomalous phase separation in metallic glasses and in other fields, such as liquid-liquid phase separation in cells.

Background
As a therapy to prevent and treat hypertension, exercise is widely used in clinical practice. But due to the lack of documentary evidence, Baduanjin as a relaxed and convenient mode of ...exercise is not currently recommended by professional health organizations to treat hypertension. The purpose of this article is to examine the efficacy of Baduanjin as an antihypertensive exercise therapy.
Methods
Our systematic retrieved of the entire relevant literatures in 12 databases. Finally, 28 eligible trials involving Baduanjin intervention in hypertension were included. After the quality assessment and bias risk assessment of the included trials, we analyzed the blood pressure values before and after the intervention, and performed meta-analysis on the random effect results. In order to explore the factors influencing the decrease of blood pressure, we also performed a subgroup analysis of the results.
Results
Participants (
n
= 2121) were adults (61.74 ± 5.85years of age, mean ± SD), with baseline blood pressure (systolic blood pressure (SBP) = 150.7 ± 9.2 mmHg, diastolic blood pressure (DBP) = 93.2 ± 8.8 mmHg). Baduanjin was practiced 7.5 ± 3.8 sessions / week for 28.2 ± 12.8 min /session for 16.7 ± 9.2 weeks. Overall, Baduanjin resulted in SBP (−9.3 mmHg, d = −1.49, 95%CI: −1.73 to −1.13) and DBP (−6.3 mmHg, d = −1.20, 95%CI: −1.51 to −0.88) vs. the control group (
p
< 0.001). After a subgroup analysis of age, we found that SBP heterogeneity was significantly reduced in the elderly group.
Conclusion
Our results indicate that Baduanjin can effectively reduce blood pressure (i.e., 9.3 mmHg and 6.3 mmHg of SBP and DBP reductions, respectively), and reduce the incidence rate of cardiovascular disease in hypertensive patients. In addition, we will be more likely to recommend that the elderly exercise Baduanjin.
