To investigate the prevalence of congenital heart disease (CHD) in China based on a large prospective multicenter screening study.
A total of 122 765 consecutive infants born at 18 hospitals ...throughout China between August 1, 2011, and November 30, 2012, were included. Cases of CHD were identified by echocardiography, clinical assessment, and telephone follow-up.
The overall prevalence of CHD was 8.98 per 1000 live births (critical, 1.46; serious, 1.47; significant, 5.00; nonsignificant, 1.07), including 7.15 in male infants and 11.11 in female infants. The most common CHD was ventricular septal defect (3.3), followed by atrial septal defect (1.7), patent ductus arteriosus (0.78), pulmonary stenosis (0.73), tetralogy of Fallot (0.47), and transposition of the great arteries (0.35). Female predominance was observed for all CHD and mild CHD (significant and nonsignificant), and male predominance was observed for the critical CHDs. The proportion of preterm newborns was substantially higher among the major CHD cases (critical and serious) compared with normal newborns. There were appreciably more low birth weight infants among the critical CHD cases compared with normal newborns. Significantly higher rates of ventricular septal defect and atrioventricular septal defect were found in infants born to mothers aged ≥35 years. Extracardiac anomalies were found in 9.3% of CHD cases. The risk of CHD was increased by approximately 3-fold when a first-degree relative had CHD.
Our estimates are concordant with data from Western studies. This screening study may provide more accurate and complete information on the overall prevalence of CHD in China.
As an important theme in global climate change and urban sustainable development, the changes of land surface temperature (LST) and surface urban heat island (SUHI) have been more and more focused by ...urban ecologists. This study used land-use data to identify the urban-rural areas in 285 cities in China and comparatively analyzed LST in urban-rural areas with the perspective of spatial-temporal dynamics heterogeneity. The results showed that, 98.9% of the cities exhibited SUHI effect in summer nighttime and the effect was stronger in northern cities than that in southern cities. In 2010, the mean SUHI intensity was the largest in summer daytime, with 4.6% of the cities having extreme SUHI of over 4°C. From 2001 to 2010, the nighttime LST of most cities increased more quickly in urban areas compared with rural areas, with an increasing tendency of the urban-rural LST difference. The difference in the urban- rural LST change rate was concentrated in the range of 0–0.1°C/year for 68.0% of cities in winter and 70.8% of cities in summer. For the higher LST increasing in urban areas compared with rural areas, there were more cities in summer than winter, indicating that the summer nighttime was the key temporal period for SUHI management. Based on the change slope of urban-rural LST, cities were clustered into four types and the vital and major zones for urban thermal environment management were identified in China. The vital zone included cities in Hunan, Hubei and other central rising provinces as well as the Beibu Gulf of Guangxi Province. The major zone included most of the cities in Central Plain Urban Agglomeration, Yangtze River Delta and Pearl River Delta. These results can provide scientific basis for SUHI adaptation in China.

Seasonal flowering time is an ecologically and economically important trait in temperate trees. Previous studies have shown that temperature in many tree species plays a pivotal role in regulating ...flowering time. However, genetic control of flowering time is not synchronised in different individual trees under comparable temperature conditions, the underlying molecular mechanism is mainly to be investigated.
In the present study, we analysed the transcript abundance in male cones and needles from six early pollen-shedding trees (EPs) and six neighbouring late pollen-shedding trees (LPs) in Pinus tabuliformis at three consecutive time points in early spring. We found that the EPs and LPs had distinct preferred transcriptional modules in their male cones and, interestingly, the expression pattern was also consistently maintained in needles even during the winter dormancy period. Additionally, the preferred pattern in EPs was also adopted by other fast-growing tissues, such as elongating new shoots. Enhancement of nucleic acid synthesis and stress resistance pathways under cold conditions can facilitate rapid growth and maintain higher transcriptional activity.
During the cold winter and early spring seasons, the EPs were more sensitive to relatively warmer temperatures and showed higher transcriptomic activity than the LPs, indicating that EPs required less heat accumulation for pollen shedding than LPs. These results provided a transcriptomic-wide understanding of the temporal regulation of pollen shedding in pines.
Objectives
The tyrosine kinase inhibitor (TKI)-sensitive mutations of the epidermal growth factor receptor (EGFR) gene is essential in the treatment of lung adenocarcinoma. To overcome the difficulty ...of EGFR gene test in situations where surgery and biopsy samples are too risky to obtain, we tried a noninvasive imaging method using radiomics features and random forest models.
Methods
Five hundred three lung adenocarcinoma patients who received surgery-based treatment were included in this study. The diagnosis and EGFR gene test were based on resections. TKI-sensitive mutations were found in 60.8% of the patients. CT scans before any invasive operation were gathered and analyzed to extract quantitative radiomics features and build random forest classifiers to identify EGFR mutants from wild types. Clinical features (sex and smoking history) were added to the image-based model. The model was trained on a set of 345 patients and validated on an independent test group (
n
= 158) using the area under the receiver operating characteristic curve (AUC), sensitivity, and specificity.
Results
The performance of the random forest model with 94 radiomics features reached an AUC of 0.802. Its AUC was further improved to 0.828 by adding sex and smoking history. The sensitivity and specificity are 60.6% and 85.1% at the best diagnostic decision point.
Conclusion
Our results showed that radiomics could not only reflect the genetic differences among tumors but also have diagnostic value and the potential to be a diagnostic tool.
Key Points
•
Radiomics provides a potential noninvasive method for the prediction of EGFR mutation status.
•
In situations where surgeries and biopsy are not available, CT image-based radiomics models could help to make treatment decisions.
•
The accuracy, sensitivity, and specificity still need to be improved before the image-based EGFR identifier could be used in clinics.
Introduction
Human umbilical cord blood-derived MSCs (hUC-MSCs) have the potential to differentiate into osteoblasts. This study investigated the function and potential mechanisms of a novel lncRNA ...LINC02381 in hUC-MSC osteogenic differentiation.
Materials and methods
hUC-MSCs were maintained in osteogenic differentiation medium. RT-qPCR assay was performed to assess LINC02381 expression. Alizarin Red S (ARS) and alkaline phosphatase (ALP) staining were performed to evaluate osteogenic differentiation. The interaction between miR-21 and LINC0238/KLF12 was determined by luciferase reporter and RNA immunoprecipitation (RIP) assays. Chromatin immunoprecipitation (ChIP) assay was used to confirm the transcriptional regulation of KLF12 on Wnt4 promoter. The nuclear translocation of β-catenin was evaluated using immunofluorescence. hUC-MSCs seeded on Bio-Oss Collagen scaffolds were transplanted into nude mice to assess in vivo osteogenesis. Bone formation was observed by H&E and Masson's trichrome staining. OSX and OPN levels were assessed by immunohistochemistry.
Results
LINC02381 was up-regulated in the clinical samples of osteoporotic patients. However, LINC02381 expression was reduced during osteogenic differentiation of hUC-MSCs. Enforced expression of LINC02381 suppressed the osteogenic differentiation of hUC-MSCs. Mechanistically, LINC02381 sponged miR-21 to enhance KLF12 expression, which led to the inactivation of Wnt/β-catenin signaling pathway. Furthermore, miR-21 mimics or KLF12 silencing counteracted LINC02381-induced inhibition of osteogenic differentiation, whereas IWP-4 (an inhibitor of Wnt pathway) abolished this effect.
Conclusion
In summary, LINC02381 repressed osteogenic differentiation of hUS-MSCs through sponging miR-21 to enhance KLF12-mediated inactivation of Wnt/β-catenin pathway, indicating that LINC02381 might be a therapeutic target for osteoporosis.
Conspectus The generalized energy-based fragmentation (GEBF) approach provides a very simple way of approximately evaluating the ground-state energy or properties of a large system in terms of ...ground-state energies of various small “electrostatically embedded” subsystems, which can be calculated with any traditional ab initio quantum chemistry (X) method (X = Hartree–Fock, density functional theory, and so on). Due to its excellent parallel efficiency, the GEBF approach at the X theory level (GEBF-X) allows full quantum mechanical (QM) calculations to be accessible for systems with hundreds and even thousands of atoms on ordinary workstations. The implementation of the GEBF approach at various theoretical levels can be easily done with existing quantum chemistry programs. This Account reviews the methodology, implementation, and applications of the GEBF-X approach. This method has been successfully applied to optimize the structures of various large systems including molecular clusters, polypeptides, proteins, and foldamers. Such investigations could allow us to elucidate the origin and nature of the cooperative interaction in secondary structures of long peptides or the driving force of the self-assembly processes of aromatic oligoamides. These GEBF-based QM calculations reveal that the structures and stability of various complex systems result from a subtle balance of many types of noncovalent interactions such as hydrogen bonding and van der Waals interactions. The GEBF-based ab initio molecular dynamics (AIMD) method also allows the investigation of dynamic behaviors of large systems on the order of tens of picoseconds. It was demonstrated that the conformational dynamics of two model peptides predicted by GEBF-based AIMD are noticeably different from those predicted by the classical force field MD method. With the target of extending QM calculations to molecular aggregates in the condensed phase, we have implemented the GEBF-based multilayer hybrid models, which could provide satisfactory descriptions of the binding energies between a solute molecule and its surrounding waters and the chain-length dependence of the conformational changes of oligomers in aqueous solutions. A coarse-grained polarizable molecular mechanics model, furnished with GEBF-X dipole moments of subsystems, exhibits some advantages of treating the electrostatic polarization with reduced computational costs. We anticipate that the GEBF approach will continue to develop with the ultimate goal of studying complicated phenomena at mesoscopic scales and serve as a practical tool to elucidate the structure and dynamics of chemical and biological systems.
Our study aimed to identify the optimal scoring system for predicting the prognosis of patients with sepsis-associated acute respiratory failure (SA-ARF). All data were taken from the fourth version ...of the Markets in Intensive Care Medicine (MIMIC-IV) database. Independent risk factors for death in hospitals were confirmed by regression analysis. The predictive value of the five scoring systems was evaluated by receiving operating characteristic (ROC) curves. Kaplan‒Meier curves showed the impact of acute physiology score III (APSIII) on survival and prognosis in patients with SA-ARF. Decision curve analysis (DCA) identified a scoring system with the highest net clinical benefit. ROC curve analysis showed that APS III (AUC: 0.755, 95% Cl 0.714-0.768) and Logical Organ Dysfunction System (LODS) (AUC: 0.731, 95% Cl 0.717-0.7745) were better than Simplified Acute Physiology Score II (SAPS II) (AUC: 0.727, 95% CI 0.713-0.741), Oxford Acute Severity of Illness Score (OASIS) (AUC: 0.706, 95% CI 0.691-0.720) and Sequential Organ Failure Assessment (SOFA) (AUC: 0.606, 95% CI 0.590-0.621) in assessing in-hospital mortality. Kaplan‒Meier survival analysis patients in the high-APS III score group had a considerably poorer median survival time. The DCA curve showed that APS III may provide better clinical benefits for patients. We demonstrated that the APS III score is an excellent predictor of in-hospital mortality.
Pathway enrichment extensively used in the analysis of Omics data for gaining biological insights into the functional roles of pre-defined subsets of genes, proteins and metabolites. A large number ...of methods have been proposed in the literature for this task. The vast majority of these methods use as input expression levels of the biomolecules under study together with their membership in pathways of interest. The latest generation of pathway enrichment methods also leverages information on the topology of the underlying pathways, which as evidence from their evaluation reveals, lead to improved sensitivity and specificity. Nevertheless, a systematic empirical comparison of such methods is still lacking, making selection of the most suitable method for a specific experimental setting challenging. This comparative study of nine network-based methods for pathway enrichment analysis aims to provide a systematic evaluation of their performance based on three real data sets with different number of features (genes/metabolites) and number of samples.
The findings highlight both methodological and empirical differences across the nine methods. In particular, certain methods assess pathway enrichment due to differences both across expression levels and in the strength of the interconnectedness of the members of the pathway, while others only leverage differential expression levels. In the more challenging setting involving a metabolomics data set, the results show that methods that utilize both pieces of information (with NetGSA being a prototypical one) exhibit superior statistical power in detecting pathway enrichment.
The analysis reveals that a number of methods perform equally well when testing large size pathways, which is the case with genomic data. On the other hand, NetGSA that takes into consideration both differential expression of the biomolecules in the pathway, as well as changes in the topology exhibits a superior performance when testing small size pathways, which is usually the case for metabolomics data.
ABSTRACT
Relativistic magnetohydrodynamic shocks are efficient particle accelerators, often invoked in the models of gamma-ray bursts (GRBs) and shock-powered fast radio bursts (FRBs). Most ...theoretical studies assume a perpendicular shock with an ordered magnetic field perpendicular to the shock normal. However, the degree of magnetization σ and the magnetic field geometry in shock-powered GRB/FRB scenarios are still poorly constrained by observations. Analogous to the magnetization σ associated with the total field strength, we define a tangential magnetization σ⊥ associated with the tangential field component. We explore the jump conditions of magnetized relativistic shocks, either with an ordered field of arbitrary inclination angle or with a random field of arbitrary anisotropy. In either case, we find that the jump conditions of relativistic shocks are governed by the tangential magnetization σ⊥ instead of the total magnetization σ, insensitive to the inclination angles or the anisotropy of the pre-shock magnetic field. The approximated analytical solution developed in this work could serve as a quick check for numerical simulations and apply to theoretical studies of GRBs/FRBs with a more general field geometry.
Biomass-derived photothermal conversion materials are considered to be promising evaporator choices for cost-effective, sustainable, and environmentally friendly solar vapor generation. Herein we ...demonstrate a double-layer flamed straw, which is a typical solar-driven interfacial evaporator that can directly convert solar energy into heat and then localize heat at interface for vapor generation. Benefiting from the unique structure of a natural corn straw, the flamed straw exhibits a high solar absorbance of 91%, ultralow thermal conductivity (0.042 W m–1 K–1), and a sufficient water supply. Notably, the flamed-straw evaporator achieves a fast evaporation rate of 1.497 kg m–2 h–1 and a high photothermal efficiency of 86% under 1 sun illumination, showing comparable efficiency with the reported studies. Our work highlights the promise of using the low-cost biomass-derived materials as highly effective solar vapor generators in the realm of seawater desalination and wastewater treatment.
