Background: There are no published data on the training-load magnitude or distribution in elite international-level boxers preparing for a major competition nor on the training load’s relationship ...with objective and subjective training markers. Methods: Twelve elite boxers (eight males and four females) preparing for the 2018 Commonwealth Games were monitored during training for 12 weeks. The training load (TL), change in creatine kinase (ΔCK), and wellness variables were measured daily but were amalgamated into average weekly values over the 12-week period for weekly comparisons. The relationships between the TL, ΔCK, and wellness variables were also assessed. Results: The significant (p < 0.001) main effects of the week with large and moderate effect sizes were noted for the TL and ΔCK, respectively, with weeks 9 and 12 in the competition-specific and taper phases showing the greatest differences, respectively. For wellness, only the muscle condition showed a significant change over time (p < 0.001). There were significant (p < 0.05) small–moderate correlations between the TL, ΔCK, and wellness variables. Conclusions: This is the first study to describe the weekly training loads and responses to training of elite international boxers across a 12-week pre-competition training period in preparation for a major competition. The findings within this study report that elite international boxers have high chronic training loads that change between training blocks to put emphasis on different qualities. Monitoring the indirect muscle damage through CK may provide further information on the internal training responses in boxers.
Fruit quality is dependent on various factors including flavour, texture and colour. These factors are determined by the ripening process, either climacteric or non-climacteric. In grape berry, which ...is non-climacteric, the process is signalled by a complex set of hormone changes. Abscisic acid (ABA) is one of the key hormones involved in ripening, while sugar availability also plays a significant role in certain ripening aspects such as anthocyanin production. To understand the relative influence of hormone and sugar signalling
can prove problematic due to the physiological and environmental (abiotic and biotic) factors at play in vineyards. Here we report on the use of
detached berry culture to investigate the comparative significance of ABA and sugar in the regulation of Pinot noir berry anthocyanin production under controlled conditions. Using a factorial experimental design, pre-véraison berries were cultured on media with various concentrations of sucrose and ABA. After 15 days of
culture, the berries were analysed for changes in metabolites, hormones and gene expression. Results illustrated a stimulatory effect of sucrose and ABA on enhancing berry colour and a corresponding increase in anthocyanins. Increased ABA concentration was able to boost anthocyanin production in berries when sucrose supply was low. The sucrose and ABA effects on berry anthocyanins were primarily manifested through the up-regulation of transcription factors and other genes in the phenylpropanoid pathway, while in other parts of the pathway a down-regulation of key proanthocyanindin transcription factors and genes corresponded to sharp reduction in berry proanthocyanidins, irrespective of sucrose supply. Similarly, increased ABA was correlated with a significant reduction in berry malic acid and associated regulatory genes. These findings suggest a predominance of berry ABA over berry sugar in coordinating the physiological and genetic regulation of anthocyanins and proanthocyanins in Pinot noir grape berries.
Evidence from the literature suggests that different inoculation strategies using either active dry yeast (ADY) or freshly prepared yeast cultures affect wine yeast performance, thus altering biomass ...and many primary and secondary metabolites produced during fermentation. Here, we investigated how different inoculation methods changed the fermentation behaviour and metabolism of a commercial wine yeast. Using a commercial Sauvignon blanc (SB) grape juice, fermentation was carried out with two different inoculum preparation protocols using Saccharomyces cerevisiae X5: rehydration of commercial ADY and preparation of pre-inoculum in a rich laboratory medium. We also determined the effect of different numbers of yeast cells inoculation (varying from 1 × 106 to 1 × 1012) and successive inoculation on fermentation and end-product formation. The yeast inoculation method and number of cells significantly affected the fermentation time. Principal component analysis (PCA) using 60 wine metabolites showed a separation pattern between wines produced from the two inoculation methods. Inoculation methods influenced the production of amino acids and different aroma compounds, including ethyl and acetate esters. Varietal thiols, 3-mercaptohexanol (3MH), and 4-methyl-4-mercaptopentan-2-one (4MMP) in the wines were affected by the inoculation methods and numbers of inoculated cells, while little impact was observed on 3-mercaptohexyl acetate (3MHA) production. Pathway analysis using these quantified metabolites allowed us to identify the most significant pathways, most of which were related to central carbon metabolism, particularly metabolic pathways involving nitrogen and sulphur metabolism. Altogether, these results suggest that inoculation method and number of inoculated cells should be considered in the production of different wine styles.

Modern, nonlinear ballistic neutron guides are an attractive concept in neutron beam delivery and instrumentation because they offer increased performance over straight or linearly tapered guides. ...However, like other ballistic geometries they have the potential to create significantly non-trivial instrumental resolution functions. We address the source of the most prominent optical aberration, namely coma, and we show that for extended sources the off-axis rays have a different focal length from on-axis rays, leading to multiple reflections in the guide system. We illustrate how the interplay between coma, sources of finite size, and mirrors with non-perfect reflectivity can therefore conspire to produce uneven distributions in the neutron beam divergence, a source of complicated resolution functions. To solve these problems, we propose a hybrid elliptic–parabolic guide geometry. Using this new kind of neutron guide shape, it is possible to condition the neutron beam and remove almost all of the aberrations, whilst providing the same performance in beam current as a standard elliptic neutron guide. We highlight the positive implications for neutron scattering instruments that this new shape can bring.
