Using a fifth-order aberration-corrected scanning transmission electron microscope, which provides a factor of 100 increase in signal over an uncorrected instrument, we demonstrated two-dimensional ...elemental and valence-sensitive imaging at atomic resolution by means of electron energy-loss spectroscopy, with acquisition times of well under a minute (for a 4096-pixel image). Applying this method to the study of a La₀.₇Sr₀.₃MnO₃/SrTiO₃ multilayer, we found an asymmetry between the chemical intermixing on the manganese-titanium and lanthanum-strontium sublattices. The measured changes in the titanium bonding as the local environment changed allowed us to distinguish chemical interdiffusion from imaging artifacts.

Introduction Les kinases ERK1/2 sont connues pour être impliquées dans la survie des cellules β et pourraient réguler la sécrétion d’insuline. Néanmoins le rôle spécifique de chaque kinase n’a jamais ...été étudié dans les cellules β. Le but de cette étude est de déterminer le rôle de ERK1 en utilisant des souris ERK1-/-. Matériels et méthodes La Ca2 + c et la secrétion d’insuline ont été mesurées sur des îlots périfusés. La masse des îlots a été quantifiée sur coupes histologiques. Les niveaux d’expression et l’activation des kinases ont été mesurés par western blot et immunocytochimie. Résultats In vivo , les souris ERK1–/– présentent des glycémies comparables aux souris ERK1 +/+. In vitro , les changements de la Ca2 + c et de la sécrétion d’insuline en réponse au glucose sont similaires dans les îlots ERK1 +/+ et ERK1–/–. En revanche l’analyse morphométrique des coupes de pancréas de souris ERK1–/– âgées de 5 mois, a montré un nombre comparable mais un appauvrissement en petits îlots et un enrichissement en gros îlots en comparaison des souris ERK1 +/+. Chez les animaux plus âgés (10 mois) ce décalage de taille ne s’observe plus. Néanmoins, les souris ERK1–/– semblent protégées de l’inflammation pancréatique liée à l’âge. L’absence de ERK1 n’a pas été com-pensée par ERK2 (messager, protéine et activation). Par contre les activations de MSK1 ( p < 0,01) et de p90RSK ( p < 0,05), deux cibles des ERK1/2, ont été supprimées ou diminuées, respectivement, en réponse au glucose. En conséquence, l’activation de CREB ( p < 0,05), la cible directe de p90RSK et MSK1, est également réduite ( p < 0,05). Conclusion La kinase ERK1 semble jouer un rôle dans la régulation de la masse des cellules ß mais pas dans la régulation de la sécrétion d’insuline. Abstract
Long-term memory formation (LTM) is a process accompanied by energy-demanding structural changes at synapses and increased spine density. Concomitant increases in both spine volume and ...postsynaptic density (PSD) surface area have been suggested but never quantified in vivo by clear-cut experimental evidence. Using novel object recognition in mice as a learning task followed by 3D electron microscopy analysis, we demonstrate that LTM induced all aforementioned synaptic changes, together with an increase in the size of astrocytic glycogen granules, which are a source of lactate for neurons. The selective inhibition of glycogen metabolism in astrocytes impaired learning, affecting all the related synaptic changes. Intrahippocampal administration of l-lactate rescued the behavioral phenotype, along with spine density within 24 hours. Spine dynamics in hippocampal organotypic slices undergoing theta burst-induced long-term potentiation was similarly affected by inhibition of glycogen metabolism and rescued by l-lactate. These results suggest that learning primes astrocytic energy stores and signaling to sustain synaptic plasticity via l-lactate.
As silicon-based transistors in integrated circuits grow smaller, the concentration of charge carriers generated by the introduction of impurity dopant atoms must steadily increase. Current ...technology, however, is rapidly approaching the limit at which introducing additional dopant atoms ceases to generate additional charge carriers because the dopants form electrically inactive clusters. Using annular dark-field scanning transmission electron microscopy, we report the direct, atomic-resolution observation of individual antimony (Sb) dopant atoms in crystalline Si, and identify the Sb clusters responsible for the saturation of charge carriers. The size, structure, and distribution of these clusters are determined with a Sb-atom detection efficiency of almost 100%. Although single heavy atoms on surfaces or supporting films have been visualized previously, our technique permits the imaging of individual dopants and clusters as they exist within actual devices.
Recent studies of the past eastern Australian landscape from present‐day longitudinal river profiles and from mantle flow models suggest that the interaction of plate motion with mantle convection ...accounts for the two phases of large‐scale uplift of the region since 120 Ma. We coupled the dynamic topography predicted from one of these mantle flow models to a surface process model to study the evolution of the eastern Australian landscape since the Jurassic Period. We varied the rainfall regime, erodibility, sea level variations, dynamic topography magnitude, and elastic thickness across a series of experiments. The approach accounts for erosion and sedimentation and simulates catchment dynamics. Despite the relative simplicity of our model, the results provide insights on the fundamental links between dynamic topography and continental‐scale drainage evolution. Based on temporal and spatial changes in longitudinal river profiles as well as erosion and deposition maps, we show that the motion of the Australian plate over the convecting mantle has resulted in significant reorganization of the eastern Australian drainage. The model predicts that the Murray river drained eastward between 150 and ∼120 Ma, and switched to westward draining due to the tilting of the Australian plate from ∼120 Ma. First order comparisons of eight modeled river profiles and of the catchment shape of modeled Murray‐Darling Basin are in agreement with present‐day observations. The predicted denudation of the eastern highlands is compatible with thermochronology data and sedimentation rates along the southern Australian margin are consistent with cumulative sediment thickness.

A marked bend in the Hawaiian-Emperor seamount chain supposedly resulted from a recent major reorganization of the plate-mantle system there 50 million years ago. Although alternative mantle-driven ...and plate-shifting hypotheses have been proposed, no contemporaneous circum-Pacific plate events have been identified. We report reconstructions for Australia and Antarctica that reveal a major plate reorganization between 50 and 53 million years ago. Revised Pacific Ocean sea-floor reconstructions suggest that subduction of the Pacific-Izanagi spreading ridge and subsequent Marianas/Tonga-Kermadec subduction initiation may have been the ultimate causes of these events. Thus, these plate reconstructions solve long-standing continental fit problems and improve constraints on the motion between East and West Antarctica and global plate circuit closure.
The delafossite metalsPdCoO2,PtCoO2, andPdCrO2are among the highest conductivity materials known, with low-temperature mean free paths of tens of microns in the best as-grown single crystals. A key ...question is whether these very low resistive scattering rates result from strongly suppressed backscattering due to special features of the electronic structure or are a consequence of highly unusual levels of crystalline perfection. We report the results of experiments in which high-energy electron irradiation was used to introduce point disorder to the Pd and Pt layers in which the conduction occurs. We obtain the cross section for formation of Frenkel pairs in absolute units, and cross-check our analysis with first-principles calculations of the relevant atomic displacement energies. We observe an increase of resistivity that is linear in defect density with a slope consistent with scattering in the unitary limit. Our results enable us to deduce that the as-grown crystals contain extremely low levels of in-plane defects of approximately 0.001%. This confirms that crystalline perfection is the most important factor in realizing the long mean free paths and highlights how unusual these delafossite metals are in comparison with the vast majority of other multicomponent oxides and alloys. We discuss the implications of our findings for future materials research.
Introduction Des études relativement récentes suggèrent que nupr1 jouerait un rôle prépondérant dans les mécanismes de régulation de la sécrétion et de la réplication des cellules béta pancréatiques. ...L’objectif de cette étude est de caractériser in vivo le rôle de ce gène de stress quant à l’homéostasie du glucose. Matériels et méthodes Au cours de cette étude, nous avons utilisé le modèle de souris invalidées ou non pour le gène nupr1. La masse des cellules béta a été évaluée par immunohistochimie et leur fonction par quantification de l’insuline sécrétée en réponse au glucose. Les tests de tolérance au glucose et à l’insuline ont été effectués par mesure de la glycémie après administration i.p. de 2 g de glucose ou 0,75 unités d’insuline par kilogramme, respectivement. Les mécanismes moléculaires ont été étudiés par le biais de gènes rapporteurs, de sur-expression de gènes (nupr1, ccna2, tcf19) et par comparaison des profils d’expression génique. Résultats Les souris invalidées pour le gène nupr1 sont moins sensibles à l’insuline mais ne développent pas d’intolérance au glucose du fait d’une augmentation du nombre de cellules béta et du nombre total d’ilôts de Langerhans. Cette augmentation de masse est néanmoins associée à une réduction de la capacité sécrétrice des cellules béta. L’étude comparative des transcriptomes d’îlots de type sauvage et invalidés pour le gène nupr1 ainsi que les études de gènes rapporteurs et de prolifération ont ensuite permis de démontrer que nupr1 contrôle la réplication des cellules béta pancréatiques via la régulation de ccna2 et tcf19. Finalement, les souris invalidées pour le gène nupr1 semblent être protégées contre l’obésité et ne développent pas de résistance à l’insuline une fois soumises à un régime alimentaire riche en graisse. Conclusion Ces résultats suggèrent que la réduction de l’expression et/ou activité de nupr1 aurait le potentiel de protéger contre le diabète de type 2.
