Fishes are ecologically important organisms that have long lifespans, high mobilities, and diverse trophic levels. Due to their importance, fishes are used as bioindicators for monitoring aquatic ...environments. One method for monitoring fishes is based on environmental DNA (eDNA), which are the deoxynucleic acids released by organisms into the environment. However, there has been a problem with false positives because eDNA is relatively stable in the environment and could even likely represent dead or non-inhabiting organisms. To address this weakness, environmental RNA (eRNA), which degrades more rapidly than eDNA in the environment, can be utilized to complement eDNA. But, to date, few studies have used eRNA for freshwater fish monitoring. In this study, to determine the relative usefulness of eDNA and eRNA metabarcoding in freshwater fishes, we performed eDNA and eRNA metabarcoding on 12S rRNA targeting fish using water samples that were collected from three locations in the Han River. We then calculated the sensitivity and positive predictivity of this approach by comparing our data to the previous specimen capture survey (PSCS) data from the last six years. The results showed that 42 species were detected by eDNA and 19 by eRNA at the three locations. At all locations, compared to the PSCS data, the average sensitivity was higher for eDNA (46.1%) than for eRNA (34.6%), and the average positive predictivity was higher for eRNA (31.7%) than for eDNA (20.7%). This confirmed that eDNA metabarcoding has the advantage of broadly determining species presence or absence (including those that are no longer present or dead), but it also generates false positives; meanwhile, eRNA metabarcoding reports living fish species, but detects fewer species than eDNA. Combining eDNA and eRNA therefore emphasizes their advantages and compensates for their disadvantages, and conducting this may therefore be useful for identifying false positives and monitoring the fish species that are actually present in the environment. This metabarcoding technique can be used in the future to provide insights into the aquatic environment and the monitoring of fisheries.

The Chinese mitten crab (CMC) also known as Eriocheir sinensis has great significance in the aquaculture industry. The bacterial communities inhabiting the CMC’s gut may differ depending on the host ...habitat and can aid in their normal biological functioning. These microbes are also known to have certain effects on their flavor. In this study, we utilized MiSeq high-throughput sequencing technology to explore the diversity of bacterial communities in the gut of CMCs from three different geographical locations in Korea: the Geum (GD), Han (HD), and Tamjin (TD) rivers. Although most of the environmental parameters were similar at the three sites, significant differences in conductivity (CDS), dissolved oxygen (DO), and salinity were observed. The results show that CMCs sampled from these locations exhibited distinct microbial composition and abundance. For example, the genus Candidatus Hepatoplasma displayed significantly higher abundance in CMCs from HD than those in the other locations, suggesting nutritional stress. Similarly, the crabs collected from TD showed a higher abundance of pathogenic Helicobacter than those from HD and GD sites. We also observed differences in the amino acid, nucleotide, and lactic acid concentrations between different tissues such as the muscle, hepatopancreas, and testis of CMCs. However, only small differences were observed when these characteristics were compared in CMCs from different locations. Our results offer important insights into the intestinal bacterial composition in CMCs which in turn may help in designing better culturing strategies for these important species of crabs.
We investigated microcystin (MC) production by four cyanobacterial strains (three Microcystis aeruginosa and one Planktothrix agardhii) in response to different grazer densities (direct exposure: ...zero, two, four, or eight individuals per 300 mL) and infochemical concentrations (indirect exposure: 0%, 10%, 25%, and 50%) of Daphnia magna and Moina macrocopa. MC production increased after direct exposure to both zooplankton species and was higher with increasing concentration of infochemicals. This MC production was significantly different among the control and three zooplankton treatment levels. Upon direct and indirect exposure of cyanobacteria to zooplankton, intracellular MC peaked on days 3 and 4. In most cyanobacterial strains, the peak MC contents were significantly higher in direct treatment with the highest zooplankton density and in indirect treatment with the highest concentration of zooplankton culture media filtrate than with treatments with the lowest density and concentration, respectively. Extracellular MC concentrations were much lower than intracellular ones, but both showed similar temporal patterns over the course of the experiment. Cyanobacteria directly exposed to Daphnia released greater amounts of extracellular MC than did those exposed to Moina. This is the first study to provide evidence of an induced defense of increased MC production by cyanobacteria in response to increasing zooplankton grazer density and increased concentrations of infochemicals released by zooplankton. In addition to the induction of tolerance in Daphnia to toxic Microcystis, we discuss how these reciprocal defenses may explain the coexistence of zooplankton and toxic cyanobacteria in eutrophic freshwaters.
Background
Soluble proteins and extracellular vesicles (EVs) are crucial wound repair mediators in cell-based therapy. Previous studies reported that EVs of perivascular cells stimulated migration ...and proliferation of cell types involved in the dermatological wound healing process. However, these studies only show effects of EVs from perivascular cells (PVCs) for in vitro models.
Methods
EVs were collected from 3D-cultured PVC (PVC-3D-EV) and compared with EVs from 2D-culture PVC (PVC-2D-EV) to investigate effects on wound contraction, angiogenesis, activation of myofibroblast, and collagen deposition.
Results
PVC-3D-EV was significantly improved in terms of wound contraction compared with PVC-2D-EV and the control. Activation of myofibroblast and collagen deposition in a rat skin wound model was significantly stimulated by PVC-3D-EV. In addition, angiogenesis and vascular endothelial growth factor expression were also highly stimulated by PVC-3D-EV. These results suggest that PVC-3D-EV was regulated in granulation tissue formation, angiogenesis, and wound contraction in healing of a rat skin wound. These results indicate a pivotal role of PVC-3D-EV in wound healing through multiple mechanisms.
Conclusions
3D-culture using a polystyrene scaffold is demonstrated to be a better system for providing better physiological conditions than the 2D-culture system, and EVs from 3D-cultured PVC could be a promising option for healing skin wound.

1. Conservation of freshwater animal populations requires their access to, as well as sufficient availability of, critical habitats, such as those for reproduction. Abundant small-scale barriers may ...cause extensive fragmentation of freshwater habitat but, by comparison to larger structures their effects are rarely considered by catchment managers. The relationship between the distribution of, and access to, spawning habitat in a regulated river, characterized by abundant small barriers, was examined for river lamprey Lampetra fluviatilis, a threatened migratory fish. 2. Telemetry of adult lamprey in the River Derwent, North East England was used to quantify upriver migration and access to spawning habitat, together with surveys of spawning habitat availability and spawning activity between 2002 and 2007. 3. Access in to the Derwent appeared severely restricted by a tidal barrage, beyond which lamprey migrated rapidly in unobstructed reaches. Of all lamprey tagged in the lower 4 km of river, or ascending the barrage, 64% and 17% passed the first and second weirs respectively, with high flows crucial for this. Although over 98% of lamprey spawning habitat occurred more than 51 km upstream, on average just 1.8% of river lamprey spawners were recorded there. 4. In order to protect or rehabilitate species or species assemblages, greater attention needs to be paid to the relative spatial distribution of low-head barriers and the resultant availability of key habitats within individual catchments. This is particularly important given the renewed emphasis internationally on low-head hydropower solutions as a source of renewable energy, and the rapid growth in numbers of low-head barriers in many catchments.
Micro-/nano-electromechanical (M/NEM) switches have received significant attention as promising switching devices for a wide range of applications such as computing, radio frequency communication, ...and power gating devices. However, M/NEM switches still suffer from unacceptably low reliability because of irreversible degradation at the contacting interfaces, hindering adoption in practical applications and further development. Here, we evaluate and verify graphene as a contact material for reliability-enhanced M/NEM switching devices. Atomic force microscopy experiments and quantum mechanics calculations reveal that energy-efficient mechanical contact–separation characteristics are achieved when a few layers of graphene are used as a contact material on a nickel surface, reducing the energy dissipation by 96.6% relative to that of a bare nickel surface. Importantly, graphene displays almost elastic contact–separation, indicating that little atomic-scale wear, including plastic deformation, fracture, and atomic attrition, is generated. We also develop a feasible fabrication method to demonstrate a MEM switch, which has high-quality graphene as the contact material, and verify that the devices with graphene show mechanically stable and elastic-like contact properties, consistent with our nanoscale contact experiment. The graphene coating extends the switch lifetime >103 times under hot switching conditions.
BACKGROUND Dietary potassium has negative outcomes in patients with mildly impaired kidney function, while having positive outcomes in patients with hypertension. The association of dietary potassium ...intake with chronic kidney disease (CKD) development, with presence of hypertension, was studied in the Korean rural population with mildly impaired kidney function. MATERIAL AND METHODS From 3 rural areas of Korea, 5064 participants age ≥40 with CKD stage 2 at baseline were recruited. Patients were classified according to the quartile of dietary potassium intake. Newly developed CKD, defined as estimated glomerular filtration rate (eGFR) of <60 mL/min/1.73 m² at the time of follow-up, and eGFR decline, defined as eGFR decrease >15% at follow-up, were studied. The effect of dietary potassium on CKD development and eGFR decline were studied by Cox proportional hazard models. The association of potassium with blood pressures and C-reactive protein was also studied to examine the underlying mechanisms. RESULTS Compared to 8.6% in normotensives, 15.7% of hypertensives developed CKD. The hazard ratio (HR) (95% confidence interval) of CKD was lower in high potassium diet only in hypertensives, with 0.60 (0.37-0.99) in the highest quartile. The eGFR decline was also lower in patients with higher potassium diet, with 0.70 (0.50-0.98) in Q3 and 0.54 (0.34-0.85) in Q4. Potassium intake has also been shown to decrease high diastolic blood pressure development (>90 mmHg) in hypertensives at 0.45 (0.25-0.83). CONCLUSIONS Dietary potassium was associated with lower risk of CKD development and eGFR decline, and this association was observed only in hypertensives.
Due to the close relationship between pets and humans, pet owners are highly invested in proper diets for their pets. Even though pet food mislabeling is concerning, there are few studies on this ...topic. This study investigated pet food mislabeling in South Korea's market based on DNA barcoding. In total, 10 pet food products were purchased, and 200 sequences of the partial Cytochrome c oxidase subunit 1 (COI) gene were generated from clones of the samples. The obtained sequences were compared to available public databases to identify species present in the ingredients. The data analyses showed that the labeled species were consistent with species detected by COI sequences in 6 of the products. However, the expected species were not detected in 4 products, revealing possible mislabeling in these samples. Our findings indicated that DNA barcoding might represent a promising tool to detect pet food mislabeling.
The gut microbiota plays a crucial role in host physiology and the disruption of host–microbiota relationships caused by environmental stressors can impact host growth and survival. In this study, we ...used Daphnia galeata as a model organism to investigate the interactive effects of fish kairomones on the life-history traits and gut microbiota alterations of D. galeata , as well as the relationship between life-history traits and gut microbiota composition. The presence of fish kairomones enhanced fecundity, decreased growth, and altered gut microbiota, with significant changes in alpha diversity but not in beta diversity in the genotype KB5 of D. galeata . Statistical analysis revealed that the relative abundance of the Pseudomonadaceae family significantly increased upon exposure to fish kairomone, while the relative abundance of the Comamonadaceae family significantly decreased. The decreased growth in genotype KB5 may be associated with a significant increase in Pseudomonas , a member of the family Pseudomonadaceae, which is generally deficient in essential fatty acids, potentially negatively impacting growth. Meanwhile, it is speculated that the significant decrease in Limnohabitans belonging to the Comamonadaceae family is associated with the reduction of body size and increased fecundity of KB5 when exposed to fish kairomones. Furthermore, the genus Candidatus Protochlamydia was observed only under the fish kairomones-treated condition. These data suggest that variations in host life-history traits related to reproduction and growth are potentially associated with the relative abundance or presence of these microbial genera. Our research findings provide valuable insights into understanding the impact of biotic stress on the interaction between hosts and microbiota.
